Summary Direct, simple and reliable means for characterization of the common amino acids, using mixtures of structural analogues of amino acids and combinations of low-voltage electrophoresis (1.04 mol dm–3 formic acid) and chromatography (tert-butanol/methanol/pyridine/formic acid/water, 33:43:9.6:0.4:20, v/v, methanol/pyridine/formic acid/water, 70:9.6:0.4:20, v/v, or tertpentanol/methylethylketone/pyridine/water, 37.5:37.5:5:20, v/v) on amorphous cellulose thin-layer are given. Summary High-performance liquid chromatographic and gas chromatographic methods were developed for the separation of unusual secondary aromatic amino acids. Amino acids containing 1,2,3,4-tetrahydroisoquinoline, 1,2,3,4-tetrahydronorharmane-1-carboxylic acid and 1,2,3,4-tetrahydro-3-carboxy-2-carboline moieties were synthetized in racemic or chiral forms. The high-performance liquid chromatography was carried out either on a teicoplanin-containing chiral stationary phase or on an achiral C18 column. In the latter case the diastereomers of the amino acids formed by precolumn derivatization with the chiral reagents 2,3,4,6-tetra-O-acetyl-β-D-glucopyranosyl isothiocyanate or 1-fluoro-2,4-dinitrophenyl-5-L-alanine amide were separated. The gas chromatographic analyses were based on separation on a Chirasil-L-Val column. A variety of amino acids were enantioresolved on a vancomycin bonded chiral phase using the polar-organic mobile phases after their pre-column derivatization with electrophilic reagents in alkaline medium. The resolution was highly dependent on the analyte's structure and was enhanced as the aromatic side-chain group on the skeleton of analyte for pi-pi interaction with the chiral selector became available. The steric hindrance resulting from the bulky side-chain group on the analyte also affected the resolution. Elution reversal, not found on the teicoplanin phase under the same chromatographic conditions, was possible through altering the type of reagent used in the derivatizing reaction (e.g. 2,4-difluorophenylisothiocyanate to 2,4-difluorophenylisocyanate). It is believed that the steric hindrance, as a result of the bulky sulfur atom in reagents such as methylisothiocyanate and others examined in this study, was responsible for the reversed elution order. The bulkiness of the substituent on the aromatic ring of derivatizing reagents (i.e. 2,3- and 3,5-dichlorophenylisothiocyanate) was observed to affect the resolution and alter the elution order as well.  相似文献

alpha,alpha-Disubstituted amino acids are alpha-amino acids in which the hydrogen atom at the alpha-position of the L-alpha-amino acid is replaced with an alkyl substituent. The introduction of an alpha-alkyl substituent changes the properties of amino acids, with the conformational freedom of the side chain in the amino acids and the secondary structure of their peptides being especially restricted. The author developed a synthetic route of optically active alpha-ethylated alpha,alpha-disubstituted amino acids using chiral cyclic 1,2-diol as a chiral auxiliary. It was found that the preferred secondary structure of peptides composed of chiral alpha-ethylated alpha,alpha-disubstituted amino acids is a fully extended C5-conformation, whereas that of peptides composed of chiral alpha-methylated alpha,alpha-disubstituted amino acids is a 3(10)-helical structure. Also, a new chiral cyclic amino acid; (3S,4S)-1-amino-3,4-di(methoxy)cyclopentanecarboxylic acid {(S,S)-Ac5c(dOM)}, and a bicyclic amino acid; (1R,6R)-8-aminobicyclo[4.3.0]non-3-ene-8-carboxylic acid {(R,R)-Ab5,6= c}, in which the alpha-carbon atom is not the chiral center but chiral centers exist at the side-chain cycloalkane skeleton, were designed and synthesized. The (S,S)-Ac5c(dOM) hexa- and octapeptides preferentially formed left-handed (M) helices, in which the helical-screw direction is exclusively controlled by the side-chain chiral centers. Contrary to the left-handed helices of (S,S)-Ac(5)c(dOM) peptides, the (R,R)-Ab5,6= c hexapeptide formed both diastereomeric right-handed (P) and left-handed (M) helices, and the twelve chiral centers at the side chain showed no preference for helical-screw direction. Thus, the chiral environment at the side chain is important for the control of helical-screw direction. Furthermore, the author designed a new class of chiral cyclic alpha,alpha-disubstituted amino acids that have pendant chiral centers at the substituent of the delta-nitrogen atom. The synthetic route would provide various optically-active cyclic alpha,alpha-disubstituted amino acids bearing a pendant chiral moiety.  相似文献

Five new chiral derivatizing reagents 5-hydrazino-2,4-dinitrophenyl-l-alaninamide (HDNP-l-Ala-NH2), 5-hydrazino-2,4-dinitrophenyl-l-phenylalaninamide (HDNP-l-Phe-NH2), 5-hydrazino-2,4-dinitrophenyl-l-valinamide (HDNP-l-Val-NH2), 5-hydrazino-2,4-dinitrophenyl-l-leucinamide (HDNP-l-Leu-NH2) and 5-hydrazino-2,4-dinitrophenyl-l-phenylglycinamide (HDNP-d-Phg-NH2) were synthesized by straightforward two-step synthesis starting from 1,5-difluoro-2,4-dinitrobenzene. Nucleophilic substitution of one fluorine atom in DFDNB with different amino acid amides yielded Marfey's reagent (5-fluoro-2,4-dinitrophenyl-l-alaninamide) and its structural variants (5-fluoro-2,4-dinitrophenyl-l-phenylalaninamide, 5-fluoro-2,4-dinitrophenyl-l-valinamide, 5-fluoro-2,4-dinitrophenyl-l-leucinamide and 5-fluoro-2,4-dinitrophenyl-d-phenylglycinamide). Chiral hydrazine reagents were prepared by nucleophilic substitution of remaining fluorine atom in Marfey's reagent and its variants with hydrazine under basic conditions. These reagents react quantitatively with chiral carbonyl compounds under mild conditions (30 degrees C, 30 min) to form hydrazone diastereomers. The labeling reaction occurs only in the presence of acid which has a catalytic action and diastereomers have strong absorbance around 348 nm. The separation of diastereomers was tried on a reversed-phase C18 HPLC column using different binary solvent combinations. Excellent separation was achieved in case of cyclic ketones having substitution at alpha-position. Optimization for derivatization yield, limit of detection, limit of quantification, linearity, accuracy and precision was carried out with respect to HDNP-l-Val-NH2. Studies related to effects of structural modification in reagents and analytes on chromatographic behavior of diastereomers were also analyzed.  相似文献

发展了一种饲料中含硫氨基酸的检测方法。样品经过甲酸氧化,将其中的胱氨酸和蛋氨酸分别氧化为磺基丙氨酸和蛋氨酸砜;再经酸水解后,采用2,4-二硝基氟苯进行柱前衍生化,衍生物经高效液相色谱分析。使用Elite AAK C18色谱柱(250 mm×4.6 mm, 5 μm)分离;以0.05 mol/L乙酸钠和乙腈-水(50:50, v/v)为流动相,梯度洗脱,流速为1.2 mL/min;检测波长为360 nm;柱温为31 ℃。结果表明,胱氨酸在0.4~16.0 mg/L、蛋氨酸在0.7~29.6 mg/L范围内的线性相关系数分别为0.9999、0.9998;胱氨酸和蛋氨酸的定量限(信噪比(S/N)=10)分别为2.6 μg/kg和3.1 μg/kg;3次样品平行测定的胱氨酸和蛋氨酸含量的相对标准偏差(RSD)分别为0.79%和2.56%,加标回收率分别为100.28%~102.00%和105.72%~107.89%。该方法分析成本低,测定结果准确,灵敏度高,符合饲料中含硫氨基酸的检测要求。  相似文献

Two new solvent systems, n-hexane + propionic acid (26:5, v/v) and chloroform + acetone (29:3, v/v), for the rapid resolution and identification of an 18-component mixture of phenylthiohydantoin amino acids are reported. Using these systems certain difficult combinations of phenylthiohydantoin amino acids are resolved. Two more solvent systems, viz chloroform + acetic acid (27:3, v/v) and chloroform + methanol (30:4, v/v), are developed to resolve phenylthiodantoin derivatives of aspartic and glutamic acids.  相似文献

A general route to alpha-pyrones and 3-substituted isocoumarins from (Z)-iodovinylic acids 1a-f or 2-iodobenzoic acids 4a-c is described, including compounds bearing a substituent on the aromatic ring. Treatment of (Z)-beta-iodovinylic acids 1a-f or 2-iodobenzoic acids 4a-c with various allenyltributyltin reagents in the presence of palladium acetate, triphenylphosphine, and tetrabutylammonium bromide in dimethylformamide provided good yields of the corresponding alpha-pyrones 3a-k or 3-substituted isocoumarins 5a-g via tandem Stille reaction and 6-endo-dig oxacyclization.  相似文献

Summary Thin-layer chromatography (TLC) is a rapid, reliable and inexpensive screening technique for diagnosis of inherited metabolic diseases (IMD). Our screening program encompasses five main situations where the use of TLC is considered to be vindicated: (i) analysis of amino acids; (ii) screening for sugar defects; (iii) detection of pathological oligosaccharidurias; (iv) screening for organic acid disorders; and (v) detection of abnormalities in tryptophan metabolism. 利用电喷雾质谱、荧光、核磁和理论计算研究了ATP与19种氨基酸的弱相互作用.在质谱中发现除甘氨酸(Gly)、丙氨酸(Ala)、缬氨酸(Val)外,其它氨基酸均可观测到与ATP因弱相互作用形成的复合物离子.利用不同质谱锥孔电压下复合物稳定性的不同,分析了侧链基团对ATP与19种氨基酸弱相互作用的影响.并利用荧光光谱和核磁共振波谱法研究了芳香性氨基酸与ATP的弱相互作用.结果表明,氨基酸与ATP的弱相互作用强弱顺序为:色氨酸(Trp)>苯丙氨酸(Phe)>具有R‖C-NH2的氨基酸>具有-RCOOH、-R-NH2的氨基酸>具有-RSH、-ROH的氨基酸>R为长链的氨基酸>R为短链的氨基酸.不同官能团的氨基酸与ATP的弱相互作用的模拟计算也证实了此结论,并发现氨基酸的主侧链基团与ATP分子基团间的多个分子间因氢键作用使复合物能稳定存在.这一结果将为预测蛋白与ATP结合位点及研究ATP的识别机理提供依据.  相似文献

Abstract

Proteins are among the most important components of all living systems. Their function range from catalysts 9enzymes) to regulators to structural components. The building blocks and language of proteins are about 20 amino acids (H2N CHR COOH), linked together By peptide bonds ([sbnd]CO[sbnd]NH[sbnd]) in chains that may consist of a few dozen to more than 1000 amino acides. The determination of primary structure of proteins, namely the sequence (arrangement) of the various amino acids along the chain is still a challenging tast. Edman reaction lies virtually at the core of all modern sequencing strategies [1,2]. The N-terminal polypeptide is first coupled to phenyl isothiocyanate to from the phenylthio carbamyl peptide; this derivative is then cleaved with anhydrous acid to expose a new N-terminus and to release the original N-terminal amino acid as a 5′thiazolinone (Scheme-1). The excess reagents and by products are extracted by an organic solvent wash. The extract of thiazolinone amino acid (obtained either from liquid-phase or solid-phase degration) is evaporated and converted to the phenylhtiohydantion derivative by 5 n HCL/CH3COOH (1:2 v/v) at 52°C for 50 min. The sample is extracted with ethyl acetate, dried and redissolved in a suitable volume of ethanol for TLC identification. Repetition of this process with identification of the released PTH-amino acidsfro the N-terminal end. For smaller peptides PITC may be used to remove the amino terminal amino acid while a chromphore or fluorophore such as dansyl chloride or DABITC, which react with the newly exposed amino terminus, is used to identify the new amino terminus. Both manual and automated methodologies are currently used for small and large polypeptides which rely upon identification of amino terminal amino acid as PTH derivative. A large number of papers have been and continue to be publihsed on the analysis of the PTH derivatives of amino acides.  相似文献

Summary The introduction of a cyano-modified, pre-coated layer substantially widens the selectivity of stationary phases in thin-layer chromatography. This is a moderately polar sorbent based on silica gel 60, which can be used both in adsorption chromatography and in reversed-phase chromatography. This new pre-coated layer is particularly suitable for separation of steroids, alkaloids and derivatized amino acids. The possibility of separating habitforming drugs and preservatives in the presence of ionpair reagents is also discussed.  相似文献

In the advanced Marfey's method, the resolution between the diastereomers derivatized with 1-fluoro-2,4-dinitrophenyl-5-l-leucinamide (l-FDLA) and 1-fluoro-2,4-dinitrophenyl-5-d-leucinamide (d-FDLA) is reflected by the difference of hydrophobicity of the two functional groups at the asymmetric carbon. However, no effective method has been developed for the estimation of hydrophobicity so far. For this purpose, we introduced log D from the ACD Labs LogD and applied it to relatively simple primary amines, amino acids and secondary alcohols in the present study. It was found that the difference of the retention times (ΔtR) correlated with that of log D (Δlog D) for both diastereomers based on the obtained experimental results. Based on these results, the following procedure was proposed for the non-empirical determination of the absolute configuration of primary amines including amino acids and secondary alcohols: (1) estimate the hydrophobicity by the calculation of log D for the two substituent groups at the asymmetric carbon, (2) locate the trans-type arrangement of the two more hydrophobic substituents in the l-DLA derivative and judge the asymmetric carbon to be R or S in the trans-type that is eluted first, (3) derivatize the desired compound with l- or d-FDLA and analyze by LC/MS, and (4) compare the elution order with the prospective one and determine the absolute configuration at the asymmetric carbon. Furthermore, log D could also be used to predict the retention times of unavailable amino acids and small peptides, indicating that the combination of the advanced Marfey's method with log D would provide more reliable structural information on a mixture composed of amino acids and small peptides. The developed method is being applied to more complicated compounds.  相似文献

A new high-performance liquid chromatography method is described for the simultaneous quantitation of amino acids and amines for 37 compounds (20 amino acids + 17 amines), as their o-phthaldialdehyde (OPA)-3-mercaptopropionic acid derivatives, within 53 min. Based on previously documented stoichiometric and reaction mechanism studies, derivatizations have been carried out with the OPA-SH-group = 1:50 containing reagents. Reliability and reproducibility of analyses have been considerably improved. Average reproducibility data in a wide concentration range of derivatives had RSD < or = 3.4%.  相似文献

The oxazoline ring system, first prepared in 1884, has only in the last five years been shown to possess considerable utility in the synthesis of a variety of functionalized organic compounds. By appropriate metalation of the 2-alkyl substituent it is possible to prepare a variety of homologated carboxylic acid derivatives, both chiral and achiral. Various other manipulations of the oxazoline ring lead to aldehydes, ketones, lactones, amino acids, thiiranes, and olefins. Due to the inertness of the oxazoline ring system to various reagents (RMgX, LiAlH4, CrO3, mild acid or alkali) it may be utilized as a protecting group for carboxylic acids during the transformations involving the above reagents. Two major new developments in oxazoline chemistry involve (a) the use of a chiral oxazoline as a reagent for asymmetric synthesis of chiral carboxylic acids, lactones, alcohol, thiiranes, and olefins and (b) a new mode of aromatic substitution in which the aryloxazolines act as nucleophilic or electrophilic reagents. This behavior has led to a variety of polysubstituted benzenes and biphenyls which would be inaccessible by classical routes. A variety of examples illustrating the utility of oxazolines indicated above are presented so that the reader may appreciate the vast potential of this simple ring system in synthesis.  相似文献

Chromatographic systems for the separation of amino acid mixtures on RP-18 as a stationary phase have been elaborated. The best results were obtained using methanol-water (1:1, v/v; 1:3, v/v; 1:5, v/v) as a mobile phase. The following amino acids have been examined: asparagine, arginine monohydrochloride, cystine, cysteine chloride, glycine, histidine monohydrochloride, hydroxyproline, isoleucine, leucine, lysine monochloride, methionine, ornithine monohydrochloride, phenylalanine, proline, threonine, tryptophan, tyrosine, serine, valine. Histidine (as monohydrochloride) and methionine were determined by first-, second- and third-derivative spectrophotometry in a mixture of several amino acids.  相似文献

Diphenylparabanic acid was found to react with >2 equiv of organolithiums at -78 °C to effectively give the corresponding symmetrical α-diketones. However, upon treatment with 1 equiv of organolithium, the parabanic acid gave mainly 5-substituted 5-hydroxyimidazolidine-2,4-diones. On the other hand, Grignard reagents were less reactive toward the parabanic acid at low temperature, and selectively gave the corresponding 5-hydroxyimidazolidine-2,4-diones even if more than 1 equiv of the reagents was used. A tandem process in which the parabanic acid was first reacted with a Grignard reagent and then reacted in one-pot with an organolithium effectively gave the unsymmetrical α-diketone. 5-Substituted 5-hydroxyimidazolidine-2,4-diones were useful as versatile precursors for preparing α-ketocarboxylic acids as well as unsymmetrical α-diketones.  相似文献

Summary This paper reports on the use of uni-dimensional high performance thin layer chromatography on silica gel for the separation and detection of ether phospholipids involving in situ reflectance densitometry. Dipping reagents for charring (cupric sulfate-phosphoric acid and manganese chloride-sulfuric acid) or fluorescence enhancement (1-anilino-8-naphthalene sulfonate) were evaluated. Two lipophilic fluorescence reagents (2,5-bis-5′-tert-butylbenzoxazolyl-(2′))thiophene and 4-(N,N-dihexadecyl)amino-7-nitrobenz-2-oxa-1,3-diazole) which move in the mobile phase were tested. The latter reagent gave the more sensitive and stable measurements: nanogram amounts of ether phospholipids could be detected in a reproducible way. Since alkyl lysophospholipids are highly polar molecules, high performance thin layer chromatography is a challenge. Characteristics of sorbents and influence of mobile phases on detection and separation are discussed for the NBD-dihexadecylamine fluorescence detection method.  相似文献
