Photophysical properties of 3-acetyl-4-oxo-6,7-dihydro-12H indolo-[2,3-a] quinolizine (AODIQ) have been studied in different aqueous micellar environments using steady-state and time-resolved emission spectroscopy. The charge transfer (CT) fluorescence exhibits appreciable hypsochromic shift, along with an enhancement in the fluorescence intensity in all the micellar media. This is associated with an increase in the fluorescence anisotropy (r), which suggests that the fluorophore molecule experiences motionally restricted environments upon binding with the micelles. Fluorescence spectral position and fluorescence quenching studies suggest that the fluorescing moiety does not penetrate into the core of the micellar units; rather it binds at the micelle-water interfacial region. The binding constant and free energy change during probe-micelle binding have been evaluated from relevant fluorescence data. Light has been thrown on the mode of action of urea on micelle bound probes. The results are interpreted in terms of the model that urea displaces water molecules from the micellar interface and the consequent destabilization leads to the expulsion of the probe molecules from the interfacial region. Polarity and viscosity of the microenvironments around the probe have been determined in the micellar systems.  相似文献

In continuation of our recent study on the steady state photophysics of a biologically active beta-carboline derivative, 3-acetyl-4-oxo-6,7-dihydro-12H indolo-[2,3-a] quinolizine (AODIQ), in the present article we have investigated the effect of nanocavity confinement on the excited state dynamics and rotational relaxation of the probe using picosecond time resolved fluorescence and fluorescence anisotropy techniques. The polarity dependent intramolecular charge transfer process is responsible for the remarkable sensitivity of this biological fluorophore in micellar environments. The fluorescence anisotropy decay of AODIQ incorporated inside the micelle is biexponential. The rotational motion of the probe was interpreted on the basis of a two step model consisting of a fast restricted rotation of the probe and a slow lateral diffusion of the probe in the micelle; both coupled to the overall rotation of the micelle. Experimental results reveal that micellar environment causes significant retardation of both the wobbling as well as the translational motion of the probe.  相似文献

Interaction of 3-acetyl-4-oxo-6,7-dihydro-12H indolo-[2,3-a] quinolizine (AODIQ), a biologically active molecule, with model transport proteins, bovine serum albumin (BSA) and human serum albumin (HSA) have been studied using steady state and picosecond time-resolved fluorescence and fluorescence anisotropy. The polarity dependent intramolecular charge transfer (ICT) process is responsible for the remarkable sensitivity of this biological fluorophore to the protein environments. The CT fluorescence exhibits appreciable hypsochromic shift along with an enhancement in the fluorescence yield, fluorescence anisotropy (r) and fluorescence lifetime upon binding with the proteins. The reduction in the rate of ICT within the hydrophobic interior of albumins leads to an increase in the fluorescence yield and lifetime. Marked increase in the fluorescence anisotropy indicates that the probe molecule is located in a motionally constrained environment within the proteins. Micropolarities in the two proteinous environments have been determined following the polarity sensitivity of the CT emission. Addition of urea to the protein-bound systems leads to a reduction in the fluorescence anisotropy indicating the denaturation of the proteins. Polarity measurements and fluorescence resonance energy transfer (FRET) studies throw light in assessing the location of the fluorophore within the two proteinous media.  相似文献

2-Hydroxy-4H-benzo[4,5]thiazolo[3,2-a]pyrimidin-4-one 2a and 7-hydroxy-5H-thiazolo[3,2-a]pyrimidin-5-one 2b, were obtained in high yields under mild conditions from the cyclization reactions of bis-(2,4,6-trichlorophenyl) malonate and 2-aminobenzothiazole or 2-aminothiazole, respectively. A new class of compounds, 2,3-dihydro-4H-benzo[4,5]thiazolo[3,2-a]furo[2,3-d]pyrimidin-4-ones and 6,7-dihydro-5H-furo[2,3-d]thiazolo[3,2-a]pyrimidin-5-ones, were synthesized via the microwave assisted radical addition of compounds 2a and 2b to various alkenes using manganese(III) acetate. A preliminary acetylcholine esterase (AchE) inhibition test of compound 4e showed excellent (92%) inhibitory potential, comparable with the standard drug Donapezil®.  相似文献

We have studied the reaction of 2,3-dihydro-4H-[1,3]thiazino[3,2-a]benzimidazol-4-one and 2-aryl-2,3-dihydro-4H-[1,3]thiazino[3,2-a]benzimidazol-4-ones with amines, alkylating reagents, and hydrogen peroxide. By the condensation of isatin, N-methyl-, N-propyl-, N-benzoylisatin with 4-methyl-1,2-benzeneldiamine 3-methyl-6H-indolo[2,3-b]quinoxalines were obtained in high yields. The reaction occurs stereoselectively, in the mixture only one of possible isomers is present. By the oxidation of methyl group of the quinoxaline fragment new carbonyl derivatives were obtained, and by the reduction of nitro group, the corresponding indoloquinoxalinamines.  相似文献

The reduction of 1-[3-(thienyl-2-carbonitrile)]pyrrole, formed by condensation of 3-aminothiophene-2-carbonitrile with 2,5-dimethoxytetrahydrofuran, gave 1-[3-(thienyl-2-aminomethyl)]pyrrole. This compound was found to be a convenient intermediate for the preparation of 4-aryl-5,6-dihydro-4H-pyrrolo[1,2-a]thieno-[2,3-f][1,4]diazepines which was accomplished by two different synthetic routes.  相似文献

The reactions of 3-acetyl-4-ethoxycarbonyl- or 3,4-diethoxycarbonylpyrrolo[1,2-a]pyrimidine derivatives 7a,b , which were prepared by condensation of the 2-aminopyrrole ( 4 ) with ethyl 3-ethoxymethylene-2,4-dioxovalerate ( 5a ) or ethyl ethoxymethyleneoxaloacetate ( 5b ), with diazomethane are described. Thus, reaction of 7a , with diazomethane gave ethyl 2a-acetyl-7-cyano-2a,3a-dihydro-5,6-dimethyl-3H -cyclopropa[e]pyrrolo[1,2-a]pyrimidine-3a-carboxylate ( 11 ) in 74% yield, which was readily transformed into the 1-pyrrol-2-yl-pyrrole ( 18 ) by treatment with potassium hydroxide. On the other hand, reaction of 7b with diazomethane afforded three products whose structures were assigned as diethyl 7-cyano-2a,3a-dihydro-5,6-dimethyl-3H-cyclopropa[e]pyrrolo[1,2-a]pyrimidine-2a,3a-carboxylate ( 20 ), 6-cyano-7,8-dimethyl-3a,3b,5,9a-tetrahydro-4H -aziridino[c]-1H or 3H-pyrazolo[3,4-e]pyrrolo[1,2-a]pyrimidine-3a,9a-dicarboxylates ( 21,22 ). Reaction of 2,3,3-trimethyl-, 2,3,3,5- and 2,3,3,7-tetramethyl-3H-indoles with 4-nitro-2-chloromethylphenol has given the 5a,6-dihydro-12H-indolo-[2,1-b][1,3]-benzoxazines, which have been examined for ring-chain interconversion by NMR spectroscopy. Anions of β,β-disubstituted acetyl acetates (2, CH2COCR1R2COOR3; R1= R2 = Me2 R1 = Et, R2 = COOMe, R1 = Et, R2 = SPh) add to 1-tosyl-3,4,4-trimethyl-Δ -imidazolinium iodide (3) to give the corresponding imidazolines (4), which are regarded as models of methylenetetrahydrofolate These models transfer C(2) of the imidazalodine -with its appended carbon fragment- to tryptamine and 2-(3,4-dimethoxybenzene)ethyl amine to yield enaminoketo ester intermediates which can be converted into benzo[a]quinolizine and indolo[2,3-a]quinolizine derivatives, respectively, in two cyclization steps. The intermediate derived from transfer of carbon fragment CH-CH2COC(SPh) (Et) COOMe to tryptamine exhibits reactions which involve nucleophilic displacement at a sulfur atom of a thio ether bond.  相似文献

We present a novel, convenient, and efficient method for synthesizing polysubstituted 3-oxo-2,3-dihydro-5H-[1,3]thiazolo[3,2-a]pyrimidines via a three-component reaction. The zwitterions generated from the reaction of isocyanides and dialkyl acetylenedicarboxylates react with 2-imino-1,3-thiazolidin-4-one to produce the title compounds in good yields.  相似文献

Synthesis of 1-(2- or 3-thienylmethyl)methyl)pyrrole and some derivatives are described from halogenomethylthiophenes. The intramolecular cyclisation of carboxylic acid 19 and 20 leads to pyrrolothienopyridines 22 and 23. On the other hand azide-aldehydes 29 and 30 in aqueous acid mixture gave the pyrrolo[1,2a]thieno[2,3-e][1,4]diazepine.  相似文献
