Treatment of 3-(or-hydroxybenzyl)pyrazolo[1,5-a]pyridines with trifluoroacetic acid in dichloromethane resulted in the formation of pyrazolo[1,5-a]pyridines, bis[α-(pyrazolo[1,5-a]pyrid-3-yl)benzyl] ethers, and phenylbis(pyrazolo[1,5-a]pyrid-3-yl)methanes, depending upon the presence or absence of the substituents at the 2- and/or 4-positions and the reaction conditions employed.  相似文献

An experimental ((1)H NMR) and theoretical (DFT) study of the ring-chain-ring isomerization of 3-(2-pyridyl)-[1,2,3]triazolo[1,5-a]pyrid-7-yl derivatives (A) into 6-{[1,2,3]triazolo[1,5-a]pyrid-3-yl}-2-pyridyl derivatives (B) has been carried out. Based on the calculations, a mechanism of several steps will be proposed. The experimental results as well as the calculations lead to the conclusion that the A-B ratio depends on the electronic properties of the substituents.  相似文献

Pyridinium dichloromethylids, formed in the reaction of dichlorocarbene with 2-(benzylideneamino)pyridines, undergo intramolecular 1,5-cyclization to give 2-aryl-3-chloroimidazo[1,2-a]pyridines, which undergo partial conversion to 2-aryl-3-chloro-4H-pyrido[1,2-a]pyrimidin-4-ones under the reaction conditions. 2-Aryl-3-bromoimidazo[1,2-a]pyridines and 2-[N-(1-aryl-2,2,2-tribromoethyl)amino]pyridines, respectively, are obtained in the reaction of dibromocarbene and the tribromomethyl onion generated in the reaction of bromoform with potassium hydroxide.Translated from Khimiya Geterotsiklicheskikh Soedinenii, No. 6, pp. 810–816, June, 1991.  相似文献

We have studied the effect of structural factors and the nature of the solvent on the tautomeric composition of aromatic substituted dihydro-1,2,4-triazolo[1,5-a]pyrimidines. Reaction of 3-aryl-10-methyl-2H-1,3,4-thiadiazino[3,2-a]benzimidazolium salts with triethylamine led to ring contraction of the thiadiazine ring to a pyrazole ring with the formation of a mixture of derivatives of 3-mercaptopyrazolobenzimidazole and di(pyrazolo[1,5-a]benzimidazolyl-3) disulfide. The disulfides were formed exclusively when these salts react with ethanolic alkali. Pyrazolo[1,5-a]pyridines and 6-iodopyrazolo[1,5-a]pyridines were synthesized by gold-catalyzed and iodine-mediated cyclization of enynylpyrazoles in good to excellent yields, respectively. The iodinated adducts were further converted to 6-arylpyrazolo[1,5-a]pyridines via Suzuki-Miyaura coupling reaction and 6-cyanopyrazolo[1,5-a]pyridine by Ullmann condensation reaction. One of the cyclization adducts, 2-(4-fluorophenyl)pyrazolo[1,5-a]pyridine, was converted to a p38 kinase inhibitor, 2-(4-fluorophenyl)-3-(4-pyridinyl)pyrazolo[1,5-a]pyridine, in two steps.  相似文献

Condensation in ethanol of ethyl ethoxymethyleneacetoacetate with systems containing an amidine fragment (substituted 3-aminopyrazoles and 3-amino-1,2,4-triazole) gave 6-carbethoxy-7-methylpyrazolo[1,5-a]pyrimidines. Addition of base to solutions of the obtained bicyclic carbethoxy derivatives in the course of several minutes caused rearrangement to 6-acetyl-7-hydroxypyrazolo[1,5-a]pyrimidine and 6-acetyl-7-hydroxy-1,2,4-triazolo[1,5-a]pyrimidine respectively. 3-(-haloacyl)-2,4-dialkylpyrazolo[1,5-a]benzimidazoles can be obtained either by brominating 3-acetylpyrazolo[1,5-a]benzimidazoles with bromine in acetic acid, or by acylating the 3-unsubstituted pyrazolobenzimidazoles with haloacetic halides. Halogenation of 3-acetylpyrazolo[1,5-a]benzimidazoles with bromine in acetic acid in the presence of sodium acetate, and bromination with N-bromosuccinimide or 1-chlorobenzotriazole, result in deacylation to give 3,6(7)-dibromo- and 3-chloropyrazolo[1,5-a]benzimidazoles. In our effort to identify potent purinergic P2Y(1) receptor antagonists as potent platelet aggregation inhibitors with enhanced metabolic stability, we developed an efficient route for the large-scale preparation of 2'-deoxy-C-nucleosides of pyrazolo[1,5-a]-1,3,5-triazine. The key strategic elements of this novel synthetic approach involved the following: (i) the use of a novel activating group, the N-methyl-N-phenylamino group, which was easily generated in high yield by treatment of the pyrazolo[1,5-a]-1,3,5-triazin-4-one (5) with phosphorus oxychloride and dimethylaniline under high pressure, (ii) a regio- and stereospecific palladium-mediated coupling reaction of the readily available unprotected glycal 1,4-anhydro-2-deoxy-D-erythro-pent-1-enitol (4b) and the 8-iodo derivative (16), and (iii) the stereoselective reduction of the ketone group of the furanosyl ring followed by the subsequent displacement of the N-methyl-N-phenylamino group upon treatment with methylamine. The beta configuration at the anomeric C-1' position of the glycal moieties was perfectly retained throughout this conversion. This procedure afforded 8-(2'-deoxy-beta-D-ribofuranosyl)-2-methyl-4-(N-methylamino)pyrazolo[1,5-a]-1,3,5-triazine (21) and 8-(2'-deoxy-beta-D-xylofuranosyl)-2-methyl-4-(N-methylamino)pyrazolo[1,5-a]-1,3,5-triazine (24) with an overall yield of 50% and 39%, respectively. Finally, the conversion of nucleosides 21 and 24 to the pyrazolotriazine C-nucleotides 3',5'-bisphosphate 2 and 3',5'-cyclophosphate 26 is also described herein and represents the first reported nucleotide derivatives within the pyrazolo[1,5-a]-1,3,5-triazine series. Preliminary biological testing has shown that compound 2 strongly inhibits ADP-induced human platelet aggregation and shape change and possesses significant efficacies 30 min after injection in rat, highlighting a strong P2Y(1)-receptor antagonist activity in vitro combined with a prolonged duration of action in vivo.  相似文献

Reaction of 6-acetyl-7-(2-dimethylaminovinyl)pyrazolo[1,5-a]pyrimidine 1 with 1,3- and 1,4-bisnucleophiles has been investigated; obtainment of new polycyclic heterocyclic derivatives is reported. 2-巯基-5,7-二甲基-1,2,4-三唑并[1,5-a]嘧啶(6)与氯乙醛或氯丙酮反应生成5,7-二甲基-1,2,4-三唑并[1,5-a]嘧啶-2-硫丙酮(硫乙醛) (7). 中间体7与O-烷基(芳基)羟胺(5)反应得到目标化合物1-(5,7-二甲基-1,2,4三唑[1,5-a]嘧啶-2-硫)乙醛-O-烷基肟醚(9), 2-(5,7-二甲基-1,2,4三唑[1,5-a]嘧啶-2-硫)丙酮O-烷基肟醚(10). 初步生物活性测试结果表明部分化合物具有较好的除草活性.  相似文献

The crystal structure of the title compound 1-(4-fluorophenyl) -2-hexylthio-benzo [4,5]furo[3,2-d]-1,2,4-triazolo[1,5-a]pyrimidin-5(1H) -one(C23H21FN4O2S,Mr = 436.5) has been prepared and determined by single-crystal X-ray diffraction. The crystal is of monoclinic,space group P21/n with a = 13.9854(3) ,b = 17.2678(4) ,c = 18.1828(5) ,β = 99.364(2) °,V = 4332.58(18) 3,Z = 4,Dc = 1.338,F(000) =1824,μ = 0.185 mm-1,MoKa radiation(λ = 0.71073) ,R = 0.0538 and wR = 0.1162 for 4728 observed reflections with I > 2σ(I) . X-ray diffraction analysis reveals the fused rings of benzo[4,5]furo[3,2-d]-1,2,4-triazolo[1,5-a] pyrimidin-5(1H) -one system are nearly coplanar. The crystal packing is mainly stabilized by weak intermolecular C-H···O hydrogen bond and π-π interactions.  相似文献

Under regular heating and microwave irradiation, 3-alkyl substituted imidazo[1,5-a] pyridines were synthesized from 2,2'- pyridil, di-2-pyridyl ketone and aliphatic aldehydes in the presence of ammonium acetate and acetic acid. Compared to the traditional heating condition, the reaction time under microwave irradiation was shorter and 3-alkyl imidazo[1,5-a]pyddines were given in higher yield.  相似文献

1-Alkynyl- and 1-alkenyl-3-arylimidazo[1,5-a]pyridines were synthesized. The Sonogashira coupling of 3-aryl-1-iodoimidazo[1,5-a]pyridines and various terminal alkynes with Pd(PPh(3))(2)Cl(2) (10 mol %) and CuI (10 mol %) in triethylamine at 80 °C for 12 h afforded the corresponding 1-alkenyl-3-arylimidazo[1,5-a]pyridines in good to excellent yields. The Mizoroki-Heck reaction of 3-aryl-1-iodoimidazo[1,5-a]pyridines and various styrene derivatives proceeded smoothly with Pd(OAc)(2) (5 mol %), IMes·HCl (10 mol %), and Cs(2)CO(3) (2 equiv) in DMA at 130 °C for 20 h to give the alkenylated imidazo[1,5-a]pyridines in moderate to high yields. The fluorescence maxima and fluorescence quantum yields of the alkynylated products were 458-560 nm and Φ(F) = 0.08-0.26 in chloroform solution, and those of the alkenylated imidazopyridines were 479-537 nm and Φ(F) = 0.03-0.13. The absorption behaviors of the obtained alkynylated and alkenylated imidazo[1,5-a]pyridines showed a good fit to the values predicted by TDDFT calculations at the B3LYP/6-311++G(d,p) level. In addition, the alkynylated imidazo[1,5-a]pyridines obtained showed linear correlations between the Hammett substituent constants of the substituents on the arylalkynyl group and their fluorescence wavelengths.  相似文献
