Considering the frightening high level of mortality from cancer, studies of anticancer agents are vital nowadays. The 24 thioderivatives of 2‐alkyl(aryl)‐quinazolin‐4(3H)‐thiones and 20 thioderivatives of [1,2,4]triazolo[1,5‐c]quinazoline‐2‐thiones were synthesized and evaluated for preliminary in vitro anticancer activity with subsequent in silico QSAR analysis. The substance 18 had the best results inhibiting growth of eight cancer cell lines: CCRF‐CEM of leukemia; SF‐539, SNB‐75, and U251 of CNS cancer; 786, RXF393, and UO‐31 of renal cancer; and MDA‐MB‐231/ATCC of breast cancer (?31.50 – 47.41% of cell growth) with low procancer effect. Calculated QSAR‐models for CCRF‐CEM of leukemia, T‐47D and HS 578T of breast cancer, and mean cell growth demonstrated good rate of anticancer activity prediction (r2 = 0.7 – 0.8,  = 0.5 – 0.7).  相似文献

A novel, functional‐group‐tolerant, and highly regioselective one‐pot synthesis of six 4‐chloro‐1‐aryl‐3‐oxypyrazoles, 8a – 8f , containing an oximino ester moiety has been developed. Their structures were characterized by 1H‐ and 13C‐NMR, IR, MS, and elemental analyses. The regioselectivity of the reaction was also determined by single‐crystal X‐ray diffraction analysis of product 8d . The reaction pathway, proposed with the aid of DFT calculations, likely proceeds via a DMF‐catalyzed mechanism, which involves an electrophilic attack by SOCl2 and two nucleophilic substitutions by benzyl bromide (BnBr) and Cl?, respectively, as the key steps. A preliminary in vitro bioassay indicated that most compounds exhibited good fungicidal activities against Sclerotinia sclerotiorum and Gibberella zeae. Especially, 8d and 8e displayed higher or similar fungicidal activities compared with pyraclostrobin at the concentration of 10 μg/ml.  相似文献

A grid potential analysis employing a novel approach of 3D quantitative structure–activity relationships (QSAR) as AutoGPA module in MOE2009.10 was performed on a dataset of 42 compounds of N‐arylsulfonylindoles as anti‐HIV‐1 agents. The uniqueness of AutoGPA module is that it automatically builds the 3D‐QSAR model on the pharmacophore‐based molecular alignment. The AutoGPA‐based 3D‐QSAR model obtained in the present study gave the cross‐validated Q2 value of 0.588, r2pred value of 0.701, r2m statistics of 0.732 and Fisher value of 94.264. The results of 3D‐QSAR analysis indicated that hydrophobic groups at R1 and R2 positions and electron releasing groups at R3 position are favourable for good activity. To find similar analogues, virtual screening on ZINC database was carried out using generated AutoGPA‐based 3D‐QSAR model and showed good prediction. In addition to those mentioned earlier, in‐silico ADME absorption, distribution, metabolism and excretion profiling and toxicity risk assessment test was performed, and results showed that majority of compounds from current dataset and newly virtually screened hits generated were within their standard limit. Metal‐catalyzed living radical polymerization of methyl methacrylate initiated with N‐chloro amides (N‐chloro N‐ethyl propionamide, N‐chloro benzanilide, N‐chloro methylbenzamide, and N‐chloro acetanilide), lactams (N‐chloro caprolactam and N‐chloro 2‐pyrrolidinone), carbamates or urethanes (N‐chloro ethylcarbamate or N‐chlorourethane), imides (N‐chloro phtalimide, N‐chloro succinimide, trichloroisocyanuric acid, and N‐chloro saccharin) and catalyzed with the self‐regulated catalytic system Cu2S/2,2′‐bipyridine is reported. The initiation efficiency of these initiators is determined by their structure. Regardless of the initiator efficiency, in all cases, poly (methyl methacrylate) with narrow molecular weight distribution and functionalized chain‐ends was obtained. These new classes of initiators open new strategies for the functionalization of polymer chain‐ends and for the synthesis of complex architectures by graft copolymerization initiated from N‐chloro proteins, aliphatic, aromatic and semiaromatic polyamides, and polyurethanes. © 2005 Wiley Periodicals, Inc. New functional monomer methacryloyl isocyanate containing 4‐chloro‐1‐phenol (CPHMAI) was prepared on reaction of methacryloyl isocyanate (MAI) with 4‐chloro‐1‐phenol (CPH) at low temperature and was characterized with IR, 1H, and 13C‐NMR spectra. Radical polymerization of CPHMAI was studied in terms of the rate of polymerization, solvent effect, copolymerization, and thermal properties. The rate of polymerization of CPHMAI has been found to be smaller than that of styrene under the same conditions. Polar solvents such as dimethylsulfoxide (DMSO) and N,N‐dimethyl formamide (DMF) were found to slow the polymerization. Copolymerization of CPHMAI (M1) with styrene (M2) in tetrahydrofuran (THF) was studied at 60°C. 本文介绍了用结合化学、生物实验以及计算机辅助分子设计方法优化对瓜类白粉病毒有抑制作用的先导化合物的研究。用3-取代氨基-1-芳基丙酮-1-肟以及卤代烃合成了44个取代苯丙酮肟衍生物。生物测试的结果表示这些化合物大部分对瓜类白粉病毒有抑制作用。基于这些生物测试,对这44个化合物做了QSAR研究。根据所得CoMFA (rcv2, S 以及 r2 分别为0.577, 0.258, 0.962) 和 CoMSIA (rcv2, S 以及 r2 分别为0.583, 0.343, 0.932) 模型,设计了3个新化合物,而且预测结果显示,它们无致癌和致突变毒性。测试结果显示预测活性与实验活性相对应,说明这两个模型具有较高的预测准确率。  相似文献

Gambogic acid (GA, 1 ), the most prominent member of Garcinia natural products, has been reported to be a promising anti‐tumor agent. Previous studies have suggested that the planar B ring and the unique 4‐oxa‐tricyclo[4.3.1.03,7]dec‐2‐one caged motif were essential for anti‐tumor activity. To further explore the structure‐activity relationship (SAR) of caged Garcinia xanthones, two new series of B‐ring modified caged GA analogues 13a – 13e and 15a – 15e were synthesized utilizing a Claisen/Diel‐Alder cascade reaction. Subsequently, these compounds were evaluated for their in vitro anti‐tumor activities against A549, MCF‐7, SMMC‐7721 and BGC‐823 cancer cell lines by MTT assay. Among them, 13b – 13e exhibited micromolar inhibition against several cancer cell lines, being approximately 2–4 fold less potent in comparison to GA. SAR analysis revealed that the peripheral gem‐dimethyl groups are essential for maintaining anti‐tumor activity and substituent group on C1 position of B‐ring has a significant effect on potency, while modifications at C‐2, C‐3 and C‐4 positions are relatively tolerated. These findings will enhance our understanding of the SAR of Garcinia xanthones and lead to the development of simplified analogues as potential anti‐tumor agents.  相似文献

The three-dimensional quantitative structure-activity relationships of a series of 5-[ 1-aryl-1,4-dihydro-6-methylpyridazin-4-one-3-yl]-2-arylamino-1,3,4-thiadiazoles, related to the fungicidal activity, were studied using the comparative molecular field analysis (CoMFA). The results show that the contributions of steric and electrostatic fields to the activity are 0.505 and 0.495, respectively. The cross-validated q^2 and the correlation coefficient r^2 for the model established by the study are 0.769 and 0.938, respectively, with the F value of 60.996, and the standard deviation s of 0.074. These values indicate that the model is significant and has good predictability. The analysis results are in good agreement well with the study of 2D-QSAR, and offered important structural insights into designing highly active compounds prior to synthesis.  相似文献

Based on the structure of natural product 2‐aryl‐4,5‐dihydrothiazole‐4‐carboxylic acid, a series of novel (R)‐2‐aryl‐4,5‐dihydrothiazole‐4‐carboxylic acid derivatives were designed and synthesized. Their structures were characterized by 1H NMR, 13C NMR and HRMS. The single crystal structure of compound 9b was determined by X‐ray diffraction analysis. The antifungal activities were evaluated for the first time. The bioassay results indicated that most compounds exhibited moderate to good antifungal activities. The antifungal activities of compound 13a (against Cercospora arachidicola Hori), 13d (against Alternaria solani), and 16e (against Cercospora arachidicola Hori) were 61.9%, 67.3% and 61.9%, respectively, which are higher than those of the commercial fungicides chlorothalonil and carbendazim. Moreover, compound 13d exhibited excellent antifungal activities against 7 kinds of the fungi tested (66.7%, 77.3%, 63.0%, 87.9%, 70.0%, 70.0% and 80.0% at 50 µg?mL). Therefore, 13d can be used as a new lead structure for the development of antifungal agents.  相似文献

Kinetics of the reaction of 2‐chloro‐3,5‐dinitrobenzotriflouride with aniline were studied in toluene, methanol‐toluene binary solvents, benzene and chloroform. The reaction in toluene exhibits third‐order kinetics consistent with aggregates of aniline. Thermodynamic parameters (H#, (S# and (G#are calculated and discussed for the reaction of 2‐chloro‐3,5‐dinitrobenzotriflouride with aniline in methanol‐toluene. Molecular complexes between aniline and the substrate are rejected spectrophotometricaly. The mechanism is studied and compared with the reaction in presence of pyridine. It shows an amine dependence and formation of homo and/or hetero mixed aggregates between aniline and pyridine i.e. dimer mechanism.  相似文献

Molecular modelling studies were performed to identify the essential structural requirements of quinoline-based derivatives for improving their antimalarial activity. The developed CoMFA, CoMSIA and HQSAR models for a training set comprising 37 derivatives showed good statistical significance in terms of internal cross validation (q2) 0.70, 0.69 and 0.80 and non-cross validation (r2) 0.80, 0.79 and 0.80. Also, the predicted r2 values (r2pred) of 0.63, 0.61 and 0.72 for a test set consisting of 12 compounds suggested significant predicting ability of the models. Structural features were correlated in terms of steric, electrostatic, hydrophobic, hydrogen bond donor and hydrogen bond acceptor interactions. Furthermore, the bioactive conformation was explored and explained by docking compounds #28, 32 and 40 into the active binding site of lactate dehydrogenase of Plasmodium falciparum. The QSAR models, contour map and docking binding affinity obtained could be successfully utilized as a guiding tool for the design and discovery of novel quinoline-based derivatives with potent antimalarial activity.  相似文献

A new nickel(II) σ‐aryl complex, trans‐chloro(9‐phenanthrenyl)bis(triphenylphosphine)nickel(II), was used as a precatalyst for the Suzuki–Miyaura coupling reactions of aryl chlorides. The catalytic conditions were optimized by investigating the cross‐coupling of p‐chloroanisole with phenylboronic acid. The results show that this complex is efficient for both electron‐rich and electron‐deficient aryl chlorides, though it gives better yields for activated arylboronic acids than deactivated ones. All isolated cross‐coupled biaryl products have been characterized by 1H and 13C NMR, and their spectral data are consistent with those reported. Side products from the coupling of arylboronic acid with the precatalyst complex have also been isolated and characterized, which is helpful for understanding the coupling mechanism. 脂质体电动色谱 (Liposome electrokinetic chromatography,LEKC)是一种简单快速的评价药物与生物膜相互作用的方法。本文建立了脂质体电动色谱作为高通量筛选皮肤渗透性的体外分析方法。将脂质体电动色谱中保留因子的对数值(log k)作为自变量建立了定量保留活性关系式。采用SPSS分析软件对于16种结构不同的化合物进行分析,结果表明log k与皮肤渗透性常数线性相关性良好( R2=0.886)。采用交互验证评价了该模型的预测能力。在定量保留活性关系中的一个变量和传统定量构效关系中的三个变量可解释的能力( R2 =0.704)相似。文中建立的定量保留活性关系模型对于新化合物早期的筛选可提供一种有效快捷的方法。  相似文献
