Summary Seasonal dependence of 20-hydroxyecdysone content ofSerratula tinctoria andSerratula wolffii (Asteraceae) was investigated by micellar electrokinetic chromatography (MEKC). Samples were collected each month through the vegetation period. The leaves were dried, milled and extracted with methanol. Clean-up of the extracts was by solid-phase extraction using a polyamide micro-column to remove flavonoids and other plant phenolics which can interfere with the analysis. This work deals with the separation of 20-hydroxyecdysone from polypodine B and the seasonal variation of 20-hydroxyecdysone concentration. Determinations have been performed by both thin-layer chromatography and capillary electrophoresis using micellar electrokinetic chromatography. The limited peak capacity of neutral compounds in micellar electrokinetic chromatography (MEKC) causes peak overlap in a simple 38-compound sample that is predicted by statistical-overlap theory (SOT). The low-concentration sample was prepared in-house from several compound classes to span the entire migration-time range and was resolved partially in a pH=7 phosphate buffer containing 50 mM sodium dodecyl sulfate. Peaks, singlets, doublets, and other multiplets were identified on the basis of known migration times and were counted at 13 voltages spanning 4 – 26 kV. These numbers agreed well with predictions of a simple SOT based on the assumption of an inhomogeneous Poisson distribution of migration times. Because the dispersion theory of MEKC is simple, the standard deviations of single-component peaks were modeled theoretically. As part of a new way to implement SOT, probability distributions of the numbers of peaks, singlets, and so on, were computed by Monte Carlo simulation. These distributions contain all theoretical information on peak multiplicity predictable by SOT and were used to evaluate the agreement between experiment and theory. The peak capacity of MEKC was calculated numerically and substituted into the simplest equations in SOT, affirming that peak overlap arises from limited peak capacity.  相似文献

A method based on magnetic separation was developed for the extraction of several estrogens (including diethylstilbestrol, estrone and estriol) in water followed by sweeping micellar electrokinetic chromatography (MEKC) analysis with UV detection. Novel magnetic octadecylsilane (ODS) particles were prepared using a silanization method with octadecyl trimethoxysilane as the surface modification reagent of magnetic Fe3O4 particles. Octadecyl trimethoxysilane was covalently immobilized on the magnetic iron oxide particles. The particles were used as the sorbents in the magnetic separation for the extraction of trace amounts of estrogens from water. The extraction condition and efficiency of the particles for the estrogens were investigated. Combining the magnetic ODS particles extraction and sweeping MEKC with UV detection, the estrogens at concentrations as low as ng/mL in water can be detected without interference from other substances in the sample matrix.  相似文献

In-hospital deaths caused by the infection of methicillin-resistant Staphylococcus aureus (MRSA) are on the increase worldwide. Teicoplanin is a potent glycopeptide antibiotic against MRSA. A rapid and cost-saving micellar electrokinetic chromatography (MEKC) method combined with solid phase extraction (SPE) was developed and then validated to quantify teicoplanin in patient serum in this work. The method includes the following steps: (1) pretreatment of the serum samples with 10 M urea to denature proteins, (2) application of SPE by using an OASIS HLB cartridge to clean up and concentrate the serum samples, and (3) use of MEKC for sample analysis. Under the optimized conditions, the SPE recovery of teicoplanin is higher than 90%. The six major components of teicoplanin could be baseline-separated from one another and endogenous materials in 12 min with a background electrolyte composed of 20 mM sodium tetraborate buffer pH 8.8, 40 mM sodium dodecyl sulfate, and 11% (v/v) ACN. The relative standard deviation (R.S.D.) of the peak area ratios for method repeatability (n = 6) and intermediate precision (inter-day, n = 3) were found to be lower than 4.18% and 5.30%, respectively. The calibration curves were linear between the chromatographic response and total teicoplanin concentration over the range of 5 μg/mL to 55 μg/mL. Limit of detection (LOD) for each of the six components was found to be lower than 0.06 μg/mL. Pearson’s correlation revealed that a good correlation (r = 0.98) was obtained between the SPE-MEKC method and the fluorescence polarization immunoassay (FPIA) method. The developed method can be used to quantitatively determine serum teicoplanin concentration in patients for dose monitoring and clinical research.  相似文献

Micellar electrokinetic chromatography (MEKC), a mode of capillary electrophoresis (CE), is considered an efficient analytical technique allowing for the reduction of organic solvent consumption during the experimental procedure. However, during sample preparation of natural products, the usage of large amount of organic solvent is generally unavoidable. In this article, therefore, a fast, simple, efficient, highly automatic and organic solvent-free sample preparation method, namely surfactant-assisted pressurized liquid extraction (PLE), was developed for the extraction of flavonoids in Costus speciosus flowers before MEKC analysis. The various experimental parameters such as the type and concentration of surfactant, and extraction time were evaluated systematically. Under the optimized conditions, the extraction efficiencies of surfactant-assisted PLE methods were comparable with Soxhlet extraction using organic solvent. The combination of surfactant-assisted PLE and MEKC was shown to be a green, rapid and effective approach for extraction and analysis of flavonoids in C. speciosus flowers.  相似文献

A capillary electrophoretic method with UV detection at 278 nm has been developed for analysis of the immunosuppressant rapamycin (sirolimus) in human blood at low microgram per liter levels. Separation has been achieved in an acidic carrier electrolyte containing sodium dodecylsulfate and 30% (v/v) acetonitrile. For sample clean-up and preconcentration, an off-line solid-phase extraction step using a silica-based reversed-phase material and an on-capillary focussing technique were employed. The latter allows the injection of increased sample volumes without excessive band broadening. Although this new method is less sensitive than existing liquid chromatographic procedures combined with mass spectrometry, it is fully suited to routine analysis of rapamycin in blood from patients treated with this drug. Last but not least the low costs make it an attractive alternative to established methods.  相似文献

A sensitive method for simultaneous determination of six phenolic whitening agents,including arbutin, phenol,resorcinol,hydroquinone,kojic acid,and salicylic acid in cosmetics has been developed using micellar electrokinetic capillary chromatography with amperometric detection(MECC-AD).Effects of several factors,such as the pH value and concentration of running buffer,potential applied to the working electrode,separation voltage,and injection time were investigated to obtain optimum conditions for separation and detection.With a 75 cm long fused-silica capillary tube,well-defined separation of six phenolic compounds was achieved in 10mmol/L SDS/40 mmol/L H3BO3-Na2B4O7 running buffer(pH 9.0).Good linear relationship was obtained for each analyte over three orders of magnitude with correlation coefficients(r2) between 0.9985 and 0.9994,and the detection limit(S/N=3) ranged from 0.04μg/mL to 0.45μg/mL The proposed method has been successfully applied for the determination of phenolic whitening agents in real cosmetic samples with satisfactory results,providing an alternative monitoring method for cosmetics safety regulation.  相似文献

Arnica montana preparations have been used in Europe for centuries to treat skin disorders. Among the biologically active ingredients in the flower heads of the plant are sequiterpenes, flavonoids and phenolic acids. For the simultaneous determination of compounds belonging to the latter two groups a micellar electrokinetic capillary chromatography (MEKC) method was developed and validated. By using an electrolyte solution containing 50 mM borax, 25 mM sodium dodecyl sulfate and 30% of acetonitrile the separation of seven flavonoids and four caffeic acid derivatives was feasible in less than 20 min. The optimized system was validated for repeatability (σrel ≤ 4.4%), precision (inter-day σrel ≤ 8.13%, intra-day σrel ≤ 4.32%), accuracy (recovery rates from 96.8 to 102.4%), sensitivity (limit of detection (LOD) ≤ 4.5 μg mL−1) and linearity (R2 ≥ 0.9996), and then successfully applied to assay several plant samples. In all of them the most dominant flavonoid was found to be quercetin 3-O-glucuronic acid, whereas 3,5-dicaffeoylquinic acid was the major phenolic acid; the total content of flavonoids and phenolic acids varied in the samples from 0.60 to 1.70%, and 1.03 to 2.24%, respectively.  相似文献

Summary The separation of seven analogous anabolic steroids was studied by micellar electrokinetic capillary chromatography (MECC). The retention order was found to be dependent on polarity. All of these steroids were well separated by the addition of organic modifiers to the separation buffer. Of the organic modifiers tested, 1-propanol gave the best separation, better than methanol or acetonitrile.  相似文献

A fast and simple micellar electrokinetic capillary chromatographic method was developed for the analysis of piribedil in pharmaceutical formulations. The effects of buffer concentration, buffer pH, sodium dodecyl sulphate (SDS) concentration, organic modifier, applied voltage and injection time were investigated. Optimum results were obtained with a 50 mM borate buffer at pH 8.0 containing 50 mM SDS by using a fused silica capillary (50 m internal diameter, 72 cm effective length). The sample was injected hydrodynamically for 4 s at 50 mbar pressure and the applied voltage was +30 kV. The detection wavelength was set at 205 nm. Diflunisal was used as an internal standard. The analysis was performed at 25 °C and the total run time was 14 min. The method was suitably validated with respect to linearity range, limit of detection and quantification, precision, accuracy, specificity and robustness. The linear calibration range was 5–100 g mL–1 and the limit of detection was determined as 1 g mL–1. The method developed was successfully applied to the determination of piribedil in pharmaceutical formulations. The results were compared with a spectrophotometric method reported in the literature and no significant difference was found statistically.  相似文献

A comparison between chiral cyclodextrin‐modified microemulsion electrokinetic chromatography (CD‐MEEKC) and cyclodextrin‐modified micellar electrokinetic chromatography (CD‐MEKC) for the enantiomeric separation of esbiothrin was carried out. For both methods, the separation conditions were optimized by varying CD types and concentration, running buffer pH and compositions, organic modifiers, and temperature. The optimal CD‐MEEKC conditions were 0.8% n‐heptane, 2.3% SDS, 6.6% n‐butanol, 90.3% 10 mM sodium tetraborate containing 3% (w/v, the ratio of CD mass to microemulsion volume) methyl‐β‐cyclodextrin, pH 10, 25°C. The optimized CD‐MEKC conditions were 3.3% SDS, 96.7% 10 mM sodium tetraborate containing 5% (w/v) β‐CD, pH 10, 25°C. The difference in physicochemical properties of the buffer and CDs resulted in different optimal CD type. The competitive distribution between the microemulsion (or micelle) and chiral CD contributed to the chiral separation. Both methods provided excellent separation (Rs ~? 3) with similar migration time (ca. 15 min). CD‐MEEKC provided higher separation efficiencies (>300000) than CD‐MEKC (>200000). The LODs for CD‐MEEKC and CD‐MEKC were 4.7 μg/mL and 3.2 μg/mL, respectively. The RSDs of migration time and peak area for CD‐MEEKC were slightly higher than for CD‐MEKC. Both the demonstrated CD‐MEEKC and CD‐MEKC methods provided high efficiencies, low LODs, and reproducible enantioseparations of esbiothrin.  相似文献

An analytical strategy micelle to trapping solution stacking (MSS) was developed in acidic buffer in micellar electrokinetic chromatography (MEKC). The stacking mechanism is based on the transport, release, capturing of molecules bound to micelle carriers that are made to collapse into trapping solution (TS) to serve as the medium to contain and stacking the analytes. Tetrandrine and fangchinoline were selected as model mixture using sodium dodecyl sulfate (SDS) micelles as carrier to demonstrate this stacking method. The experiments by MSS-MEKC were carried out and further compared with those by normal MEKC. The results reveal that 113–123-fold improvements in the detection sensitivity was obtained for the analytes, and separation and determination of tetrandrine and fangchinoline in Stephaniae tetrandrae S. Moore and Fengtongan capsules were finished under optimum conditions using the sample matrix containing 8.0 mM SDS and TS containing 50 mM H3PO4–55% (v/v) ethanol.  相似文献

The separation and determination of aminophenols and phenylenediamines were investigated by liquid chromatography (LC) and micellar electrokinetic chromatography (MEKC) in this study. Aminophenols and phenylenediamines are commonly used components in commercial hair colorants. The problem of tailing peaks in LC was improved by the technique of using mobile phase containing 15 mM triethylamine at pH 8.0. The analysis of o-aminophenol was not succeeded with LC even though the modifier of triethylamine was added. But it could be quantitative successfully by MEKC. The optimum separation condition of MEKC was achieved by employing 55 mM cetyltrimethyl ammonium chloride in 50 mM borate buffer (pH 9.2) with electric field strength of −145 V cm−1. Finally, the commercial hair dyes were analyzed by developing methods of LC and MEKC. From both the results, there is no significant difference presence at 99.5% confidence level. Summary Coelectroosmotic micellar electrokinetic chromatography (coelectroosmotic MEKC) has been investigated for the separation of twelve aromatic sulfonate compounds. The advantage of this method is that it combines the efficient separation characteristic of MEKC and the short analysis time of the coelectroosmotic mode. MEKC was performed with either cetyltrimethylammonium bromide (CTAB) or polyethylene glycol dodecyl ether (Brij 35) surfactants as pseudostationary phases and 2-propanol as organic modifier. The electroosmotic Flow (EOF) was reversed by adding two types of EOF modifier, an alkylammonium salt (cetyltrimethylammonium bromide, CTAB) or a cationic polyelectrolyte (hexadimetrine bromide, HDB). The surfactant concentration, applied voltage, and temperature were optimized, the influence of 2-propanol on the MEKC resolution of the compounds was studied. The effect of the osmotic modifier on the separation was also investigated.  相似文献
