Morphology is presented as a powerful tool to control the in vitro degradation and drug release characteristics of novel drug delivery microspheres prepared from homopolymer blends of 1,5‐dioxepan‐2‐one, DXO, and L ‐lactide, L‐LA. Their performance in this respect was compared to analogous P(L‐LA‐co‐DXO) microspheres. Blends formed denser and less porous microspheres with a higher degree of matrix crystallinity than copolymers of corresponding L‐LA:DXO composition. The morphology differences of blends and copolymers, further adjustable by means of component ratio, are shown to have a vital impact on the in vitro performance. Sustained drug delivery was obtained from both copolymers and blends. Molecular weight loss was retarded and diffusion‐mediated release was inhibited in the latter case, further delaying the release process. The effects of storage on the physicochemical properties of these systems were evaluated under desiccated and moist conditions for 5 months. Storage‐induced physicochemical changes, such as matrix crystallization and molecular weight decrease, were accelerated at higher relative humidities. P(L‐LA‐co‐DXO) demonstrated higher moisture sensitivity than a PLLA‐PDXO blend of corresponding composition. The more crystalline and dense morphology of blend microspheres may thus be considered an improvement of the storage stability. © 2000 John Wiley & Sons, Inc. Novel microspheres from copolymers of 1,5-dioxepan-2-one (DXO) and L -lactide (L -LA) were prepared by oil-in-water solvent evaporation and oil-in-oil solvent removal. The two preparation techniques were evaluated for sphere formulation and incorporation of two different drugs. Sustained release of these therapeutic substances was obtained. The consequences of altering the DXO : LA ratio, preparation method, and drug hydrophilicity were explored and identified as factors governing sphere quality, in vitro degradation, and drug release characteristics. Star‐shaped homo‐ and copolymers were synthesized in a controlled fashion using two different initiating systems. Homopolymers of ε‐caprolactone, L ‐lactide, and 1,5‐dioxepan‐2‐one were firstly polymerized using (I) a spirocyclic tin initiator and (II) stannous octoate (cocatalyst) together with pentaerythritol ethoxylate 15/4 EO/OH (coinitiator), to give polymers with identical core moieties. Our gained understanding of the versatile and controllable initiator systems kinetics, the transesterification reactions occurring, and the role which the reaction conditions play on the material outcome, made it possible to tailor the copolymer microstructure. Two strategies were used to successfully synthesize copolymers of different microstructures with the two initiator systems, i.e., a more multiblock‐ or a block‐structure. The correct choice of the monomer addition order enabled two distinct blocks to be created for the copolymers of poly(DXO‐co‐LLA) and poly(CL‐co‐LLA). In the case of poly(CL‐co‐DXO), multiblock copolymers were created using both systems whereas longer blocks were created with the spirocyclic tin initiator. © 2008 Wiley Periodicals, Inc. Fingerprinting of the degradation product patterns by electrospray ionization mass spectrometry (ESI‐MS) was evaluated as a tool to monitor the degree of degradation in polyester‐ether networks. Four different crosslinked caprolactone (CL) and/or 1,5‐dioxepan‐2‐one (DXO) networks were subjected to hydrolytic degradation in aqueous solution at 37 °C for up to 147 days. After predetermined time periods, the water‐soluble degradation products were analyzed by ESI‐MS and tandem ESI‐MS. In addition, changes in pH, mass loss, and copolymer composition were determined. In the case of more slowly hydrolyzed CL‐rich (co)polymers, CL and/or DXO oligomers terminated by hydroxyl and carboxyl end groups were predominantly formed as degradation products. However, on prolonged hydrolysis oligomers with attached crosslinking agent dominated the degradation product patterns of more easily hydrolyzed DXO‐rich (co)polymers. It was shown that in the recorded mass spectra the variation of intensities in the series of ions corresponding to DXO and CL/DXO oligomers with or without attached crosslinking agent could be utilized to monitor the extent of hydrolytic degradation in the polyester matrix and the disruption of the network structure. © 2008 Wiley Periodicals, Inc. Summary: Biodegradable poly(1,5‐dioxepan‐2‐one) (PDXO) was grown directly from Si OH groups of a silica nanoparticle by surface‐initiated, ring‐opening polymerization (SI‐ROP) of 1,5‐dioxepan‐2‐one (DXO). The direct SI‐ROP of DXO was achieved by heating a mixture of Sn(Oct)2, DXO, and the silica nanoparticles (316 nm in diameter) in anhydrous toluene. The resulting silica/PDXO hybrid nanoparticles were characterized by means of 1H NMR spectroscopy, IR spectroscopy, thermogravimetric analysis, and field‐emission scanning electron microscopy.

Environmental switches may be fabricated for the controlled release of pharmaceutical drug using a thermally responsive polymer with the intrinsic chemical and physical nature of stimuli‐sensitive smart materials. Particularly, much attention has been paid to the biomedical applications of poly(N‐isopropyl acrylamide) (PNIPAAm) because of its unique reversible transition at a specific lower critical solution temperature (LCST).Thermally sensitive block copolymers, poly(N‐isopropyl acrylamide‐b‐poly(L ‐lactide‐co‐glycolide) (PNIPAAm‐b‐PLGA), and polyethylene glycol‐poly (lactide‐co‐glycolide) (PEG‐PLGA) triblock copolymers with different compositions and length of PLGA block were synthesized via ring‐opening polymerization of lactide and glycolide in the presence of OH‐terminated PNIPAAm or PEG. The composition and structure of the polymer were determined by NMR and FTIR. The effect of important factors, such as ionic strength, pH, and polymer concentration on the phase transition behavior of temperature‐sensitive polymers, were investigated by cloud point measurements. The resulting thermosensitive polymers were used for the entrapment of a narcotic antagonist drug, naltrexone, as the model drug. The loading efficiency and drug release behavior of naltrexone‐loaded hydrogels were investigated. The naltrexone loaded thermosensitive polymers were able to sustain the release of naltrexone for different periods of time, depending on the polymer composition, and concentration. In vitro release studies showed that these thermosensitive polymers are able to deliver naltrexone in biologically active forms at a controlled rate for 3–8 weeks. To avoid organometallic catalysts in the synthesis of poly(1,5‐dioxepan‐2‐one), the enzymatic ring‐opening polymerization of 1,5‐dioxepan‐2‐one (DXO) was performed with lipase CA (derived from Candida antarctica) as a biocatalyst. A linear relationship between the number‐average molecular weight and monomer conversion was observed, and this suggested that the product molecular weight could be controlled by the stoichiometry of the reactants. The monomer consumption followed a first‐order rate law with respect to the monomer, and no chain termination occurred. Water acted as a chain initiator, but it could cause polymer hydrolysis when it exceeded an optimum level. An initial activation via the heating of the enzyme was sufficient to start the polymerization, as the monomer conversion occurred when samples were left at room temperature after an initial heating at 60 °C. A high lipase content led to a high monomer conversion as well as a high molecular weight. An increase in the monomer conversion and molecular weight was observed when the polymerization temperature was increased from 40 to 80 °C. A further increase in the polymerization temperature led to a decrease in the monomer conversion and molecular weight because of the denaturation of the enzyme at elevated temperatures. The polymerization behavior of DXO under lipase CA catalysis was compared with that of ε‐caprolactone (CL). The rate of monomer conversion of DXO was much faster than that of CL, and this may have been due to differences in their specificity toward lipase CA. © 2005 Wiley Periodicals, Inc. A series of copolymers composed of methoxy poly(ethylene glycol) and a hydrophobic block of poly(ɛ-caprolactone-co-propargyl carbonate) grafted with poly(2-[dimethylamino]ethyl methacrylate) was synthesized by combining ring opening polymerization, azide-alkyne click reaction, and atom transfer radical polymerization (ATRP). Well-defined copolymers with a target composition and a tailored structure were achieved via the grafting from approach by using a single catalytic system for both click reaction and ATRP. Kinetic studies demonstrated the controlled/living character of the employed polymerization methods. The thermal properties and self-assembly in aqueous medium of the graft copolymers were dependent on their composition. The resulting polymeric materials showed low cytotoxicity toward L929 cells, demonstrating their potential for biomedical applications. This type of materials containing cationic side chains tethered to biocompatible and biodegradable segments could be the basis for promising candidates as drug and gene delivery systems.  相似文献

Starburst triblock copolymers consisting of 8‐arm poly(ethylene glycol) (8‐arm PEG) and biodegradable poly(L ‐lactide) (PLLA) or its enantiomer poly(D ‐lactide) (PDLA), 8‐arm PEG‐b‐PLLA‐b‐PEG ( Stri‐L ), and 8‐arm PEG‐b‐PDLA‐b‐PEG ( Stri‐D ) were synthesized. An aqueous solution of a 1:1 mixture ( Stri‐Mix ) of Stri‐L and Stri‐D assumed a sol state at room temperature, but instantaneously formed a physically crosslinked hydrogel in response to increasing temperature. The resulting hydrogel exhibited a high‐storage modulus (9.8 kPa) at 37 °C. Interestingly, once formed at the transition temperature, the hydrogel was stable even after cooling below the transition temperature. The hydrogel formation process was irreversible because of the formation of stable stereocomplexes. In aqueous solution, gradual hydrolytic erosion was observed because of degradation of the hydrogel. The combination of rapid temperature‐triggered irreversible hydrogel formation, high‐mechanical strength, and degradation behavior render this polymer mixture system suitable for use in injectable biomedical materials, for example, as a drug delivery system for bioactive reagents or a biodegradable scaffold for tissue engineering. © 2008 Wiley Periodicals, Inc. ABSTRACT

Copolymers of 1,5-dioxepan-2-one (DXO) and e-caprolactone (?-CL), δ-valerolactone (δ-VL) or L-lactide (LLA) have been synthesized and characterized. High molecular weight copolymers were obtained using stannous-2-ethyl hexanoate as catalyst in bulk. Reactivity ratios for the copolymerization of DXO and δ-VL were determined at 110°C as rVL=0.5 and rDXO=2.3. At high conversion, depolymerization of δ-VL occurred, resulting in lower molecular weight and variations in the copolymer composition.

Physical properties, such as crystallinity and melting temperature of the DXO-copolymers proved to be strongly dependent on the choice of comonomer and on the molar composition of the copolymers. DXO appears to be incorporated into the poly-?-caprolactone (PCL) crystals and to some extent into the poly-δ-valerolactone (PVL) crystals, resulting in a more gradual decrease in crystallinity with increasing amount of DXO.  相似文献

Poly(lactones) may be crosslinked by ring-opening polymerization of the corresponding cyclic esters in the presence of tetrafunctional bis(ϵ-caprolactone). The homopolymer of 1.5-dioxepan-2-one (DXO) has poor mechanical properties but also some very good properties, such as biocompatibility and degradability. Crosslinking of degradable polymer based on the poly(ether-ester) DXO was performed with crosslinkers having the same reactivity as the monomer. 2,2-Bis(ϵ-caprolactone-4-yl)propane (BCP) and bis(ϵ-caprolactone-4-yl) (BCY) with tetrafunctionalities were synthesized from the corresponding diols and then used as comonomers during the polymerization of DXO. The comonomers showed the same reactivity to the initiator, stanneous 2-ethylhexanoic acid, as DXO and perfectly random crosslinked films were obtained. The crosslinked films showed a high degree of swelling already at 2–3 mol % BCP or BCY. The BCP crosslinker was somewhat less soluble in DXO at lower temperatures, but all BCP was soluble at 180°C. These polymeric films were elastic with no crystallinity and the Tg values increased from −39°C for pure DXO to −35°C for BCP crosslinked films and −21°C for BCY crosslinked ones. © 1997 John Wiley & Sons, Inc. In situ forming biodegradable polymeric systems loaded with betamethasone (BTM) and betamethasone acetate (BTMA) were prepared using poly(DL ‐lactide‐co‐glycolide) (PLGA), ethyl heptanoate (EH), and N‐methyl‐2‐pyrrolidone (NMP) as the biodegradable polymer, additive, and solvent, respectively. The drug release studies were carried out in buffer (pH = 7.4, 37°C) using high performance liquid chromatography (HPLC). 1H‐NMR was used to determine the polymer degradation behavior, release mechanism, and interactions between the polymer and drug. The 1H‐NMR spectra showed that all interactions between the polymer and drug were hydrogen bonding. Hydroxyl groups and fluorine in drugs were involved in hydrogen bonding with PLGA polymer. In 1H‐NMR studies, we found that the degradation rate in the systems loaded with BTMA was higher than the systems loaded with BTM because BTMA is only slightly soluble and accelerates the hydrolysis of PLGA chains. The formulations loaded with BTM had obviously lower burst release compared with BTMA loaded samples. Medium to high molecular weight random copolymers of 1,5-dioxepan-2-one (DXO) and L-lactide (L-LA) or ε-caprolactone (ε-CL) of different compositions have been investigated. Polymerization was conducted in bulk at 110°C using stannous 2-ethylhexanoate as catalyst. Poly(DXO-co-L-LA) is a hydrolysable material with a glass transition temperature (Tg) ranging from −36 up to 58°C depending on the molar composition. The material exhibited crystallinity as long as the amount of DXO did not exceed 50 weight%. Reactivity ratios were determined to rL-LA=10 and rDXO=0.1, giving a more blocky structure than expected in a random copolymer. The copolymer between ε-CL and DXO was shown to be a truly random copolymer by 13C NMR, as expected from the reactivity ratios, rDXO=1.6 and rε-CL=0.6. Tg of the material was ranging from −64 up to −39°C. The ability of the poly(DXO-co-ε-CL) to crystallize was retained up to a DXO content of 40 weight%. The melting temperature and crystallinity of both copolymers decrease with increasing amount of DXO. Incorporation of semicrystalline comonomers, L-LA or ε-CL, into the amorphous poly(DXO) creates materials with adjustable properties depending on the molar composition.  相似文献

The synthesis of a new cyclic carbonate monomer containing an allyl group was reported and its biodegradable amphiphilic block copolymer, poly(ethylene glycol)‐block‐poly(L ‐lactide‐co‐5‐methyl‐5‐allyloxycarbonyl‐propylene carbonate) [PEG‐b‐P(LA‐co‐MAC)] was synthesized by ring‐opening polymerization (ROP) of L ‐lactide (LA) and 5‐methyl‐5‐allyloxycarbonyl‐1,3‐dioxan‐2‐one (MAC) in the presence of poly (ethylene glycol) as a macroinitiator, with diethyl zinc as a catalyst. 13C NMR and 1H NMR were used for microstructure identification of the copolymers. The copolymer could form micelles in aqueous solution. The core of the micelles is built of the hydrophobic P(LA‐co‐MAC) chains, whereas the shell is set up by the hydrophilic PEG blocks. The micelles exhibited a homogeneous spherical morphology and unimodal size distribution. By using the cyclic carbonate monomer containing allyl side‐groups, crosslinking of the PEG‐b‐P(LA‐co‐MAC) inner core was possible. The adhesion and spreading of ECV‐304 cells on the copolymer were better than that on PLA films. Therefore, this biodegradable amphiphilic block copolymer is expected to be used as a biomaterial for drug delivery and tissue engineering. © 2007 Wiley Periodicals, Inc. Biocompatible and biodegradable ABC and ABCBA triblock and pentablock copolymers composed of poly(ε‐caprolactone) (PCL), poly(L ‐lactide) (PLA), and poly(ethylene glycol) (PEO) with controlled molecular weights and low polydispersities were synthesized by a click conjugation between alkyne‐terminated PCL‐b‐PLA and azide‐terminated PEO. Their molecular structures, physicochemical and self‐assembly properties were thoroughly characterized by means of FT‐IR, 1H‐NMR, gel permeation chromatography, differential scanning calorimetry, wide‐angle X‐ray diffraction, dynamic light scattering, and transmission electron microscopy. These copolymers formed microphase‐separated crystalline materials in solid state, where the crystallization of PCL block was greatly restricted by both PEO and PLA blocks. These copolymers self‐assembled into starlike and flowerlike micelles with a spherical morphology, and the micelles were stable over 27 days in aqueous solution at 37 °C. The doxorubicin (DOX) drug‐loaded nanoparticles showed a bigger size with a similar spherical morphology compared to blank nanoparticles, demonstrating a biphasic drug‐release profile in buffer solution and at 37 °C. Moreover, the DOX‐loaded nanoparticles fabricated from the pentablock copolymer sustained a longer drug‐release period (25 days) at pH 7.4 than those of the triblock copolymer. The blank nanoparticles showed good cell viability, whereas the DOX‐loaded nanoparticles killed fewer cells than free DOX, suggesting a controlled drug‐release effect. © 2010 Wiley Periodicals, Inc. In this study, a facile method to fabricate reduction‐responsive core‐crosslinked micelles via in situ thiol‐ene “click” reaction was reported. A series of biodegradable poly(ether‐ester)s with multiple pendent mercapto groups were first synthesized by melt polycondensation of diol poly(ethylene glycol), 1,4‐butanediol, and mercaptosuccinic acid using scandium trifluoromethanesulfonate [Sc(OTf)3] as the catalyst. Then paclitaxel (PTX)‐loaded core‐crosslinked (CCL) micelles were successfully prepared by in situ crosslinking hydrophobic polyester blocks in aqueous media via thiol‐ene “click” chemistry using 2,2′‐dithiodiethanol diacrylate as the crosslinker. These PTX‐loaded CCL micelles with disulfide bonds exhibited reduction‐responsive behaviors in the presence of dithiothreitol (DTT). The drug release profile of the PTX‐loaded CCL micelles revealed that only a small amount of loaded PTX was released slowly in phosphate buffer solution (PBS) without DTT, while quick release was observed in the presence of 10.0 mM DTT. Cell count kit (CCK‐8) assays revealed that the reduction‐sensitive PTX‐loaded CCL micelles showed high antitumor activity toward HeLa cells, which was significantly higher than that of reduction‐insensitive counterparts and free PTX. An intelligent drug delivery nanosystem has been developed based on biodegradable supramolecular polymer micelles (SMPMs). The drug release can be triggered from SMPMs responsively by a bioactive agent, L ‐phenylalanine in a controlled fashion. The SMPMs are constructed from ethylcellulose‐graft‐poly(ε‐caprolactone) (EC‐g‐PCL) and α‐cyclodextrin (α‐CD) derivate via host–guest and hydrophobic interactions. It has been found that these SMPMs have disassembled rapidly in response to an additional L ‐phenylalanine, due to great affinity discrepancy to α‐CD between L ‐phenylalanine and PCL. Experiments have been carried out on trigger‐controlled in vitro drug release of the SMPMs loaded with a model porphyrin based photosensitizer THPP. The result shows that the SMPMs released over 85% THPP in 6 h, which is two orders magnitudes faster than that of control. Also investigated is the photodynamic therapy (PDT) of THPP‐loaded SMPMs with and without L ‐phenylalanine on MCF‐7 carcinoma cell line. An effective trigger‐concentration dependent lethal effect has been found showing promise in clinical photodynamic therapy.

Novel, biodegradable poly(?‐caprolactone)‐block‐poly(trans‐4‐hydroxy‐N‐benzyloxycarbonyl‐L ‐proline)‐block‐poly(?‐caprolactone) triblock copolymers were synthesized by ring‐opening polymerization from dihydroxyl‐terminated macroinitiator poly(trans‐4‐hydroxy‐N‐benzyloxycarbonyl‐L ‐proline) (PHpr) and ?‐caprolactone (?‐CL) with stannous octoate as the catalyst. The molecular weights were characterized with gel permeation chromatography and matrix‐assisted laser desorption/ionization time‐of‐flight mass spectrometry. With an increase in the contents of ?‐CL incorporated into the copolymers, a decrease in the glass‐transition temperature (Tg) was observed. The Tg values of copoly(4‐phenyl‐?‐caprolactone) and copoly(4‐methyl‐?‐caprolactone) were higher than Tg of copoly(?‐caprolactone). Their micellar characteristics in an aqueous phase were investigated with fluorescence spectroscopy, dynamic light scattering, and transmission electron microscopy. The block copolymers formed micelles in the aqueous phase with critical micelle concentrations in the range of 1.00–1.36 mg L?1. With higher molecular weights and hydrophobic components in the copolymers, a higher critical micelle concentration was observed. As the feed weight ratio of antitriptyline hydrochloride (AM) to the polymer increased, the drug loading increased. The micelles exhibited a spherical shape, and the average size was less than 250 nm. We prepared well‐defined diblock copolymers of thermoresponsive poly(N‐isopropylacrylamide‐coN,N‐dimethylacrylamide) blocks and biodegradable poly(D ,L ‐lactide) blocks by combination of reversible addition‐fragmentation chain transfer radical (RAFT) polymerization and ring‐opening polymerization. α‐Hydroxyl, ω‐dithiobenzoate thermoresponsive polymers were synthesized by RAFT polymerization using hydroxyl RAFT agents. Biodegradable blocks were prepared by ring‐opening polymerization of D ,L ‐lactide initiated by α‐hydroxyl groups of thermoresponsive polymers, which inhibit the thermal decomposition of ω‐dithioester groups. Terminal dithiobenzoate (DTBz) groups of thermoresponsive blocks were easily reduced to thiol groups and reacted with maleimide (Mal). In aqueous media, diblock copolymer products formed surface‐functionalized thermoresponsive micelles. These polymeric micelles had a low critical micelle concentration of 22 μg/L. In thermoresponsive studies of the micelles, hydrophobic DTBz‐surface micelles demonstrated a significant shift in lower critical solution temperature (LCST) to a lower temperature of 30.7 °C than that for Mal‐surface micelles (40.0 °C). In addition, micellar LCST was controlled by changing bulk mixture ratios of respective heterogeneous end‐functional diblock copolymers. Micellar disruption at acidic condition (pH 5.0) was completed within 5 days due to hydrolytic degradation of PLA cores, regardless of showing a slow disruption rate at physiological condition. Furthermore, we successfully improved water‐solubility of hydrophobic drug, paclitaxel by incorporating into the micellar cores. © Wiley Periodicals, Inc. New biodegradable/biocompatible ABC block copolymers, poly(ethylene oxide)‐b‐poly(glycidol)‐b‐poly(L ,L ‐lactide) (PEO‐PGly‐PLLA), were synthesized. First, PEO‐b‐poly(1‐ethoxyethylglycidol)‐b‐PLLA was synthesized by a successive anionic ring‐opening copolymerization of ethylene oxide, 1‐ethoxyethylglycidyl ether, and L ,L ‐lactide initiated with potassium 2‐methoxyethanolate. In the second step, the 1‐ethoxyethyl blocking groups of 1‐ethoxyethylglycidyl ether were removed at weakly acidic conditions leaving other blocks intact. The resulting copolymers were composed of hydrophilic and hydrophobic segments joined by short polyglycidol blocks with one hydroxyl group in each monomeric unit. These hydroxyl groups may be used for further copolymer transformations. The PEO‐PGly‐PLLA copolymers with a molecular weight of PLLA blocks below 5000 were water‐soluble. Above the critical micellar concentration (ranging from 0.05 to1.0 g/L, depending on the composition of copolymer), copolymers formed macromolecular micelles with a hydrophobic PLLA core and hydrophilic PEO shell.
