A doubly hydrophilic triblock copolymer of poly(N,N‐dimethylamino‐2‐ethyl methacrylate)‐b‐Poly(ethylene glycol)‐b‐poly(N,N‐dimethylamino‐2‐ethylmethacrylate) (PDMAEMA‐b‐PEG‐b‐PDMAEMA) with well‐defined structure and narrow molecular weight distribution (Mw/Mn = 1.21) was synthesized in aqueous medium via atom transfer radical polymerization (ATRP) of N,N‐dimethylamino‐2‐ethylmethacrylate (DMAEMA) initiated by the PEG macroinitiator. The macroinitiator and triblock copolymer were characterized with 1H NMR and gel permeation chromatography (GPC). Fluorescence spectroscopy, dynamic light scattering (DSL), transmittance measurement, and rheological characterization were applied to investigate pH‐ and temperature‐induced micellization in the dilute solution of 1 mg/mL when pH > 13 and gelation in the concentrated solution of 25 wt % at pH = 14 and temperatures beyond 80 °C. The unimer of Rh = 3.7 ± 0.8 nm coexisted with micelle of Rh = 45.6 ± 6.5 nm at pH 14. Phase separation occurred in dilute aqueous solution of the triblock copolymer of 1 mg/mL at about 50 °C. Large aggregates with Rh = 300–450 nm were formed after phase separation, which became even larger as Rh = 750–1000 nm with increasing temperature. The gelation temperature determined by rheology measurement was about 80 °C at pH 14 for the 25 wt % aqueous solution of the triblock copolymer. © 2008 Wiley Periodicals, Inc. Triblock copolymers of poly(styrenesulfonate)‐b‐poly(ethylene glycol)‐b‐poly(styrenesulfonate) with narrow molecular weight distribution (Mw/Mn = 1.28–1.40) and well‐defined structure have been synthesized in aqueous solution at 70 °C via reversible addition‐fragmentation chain transfer polymerization. Poly(ethylene glycol) (PEG) capped with 4‐cyanopentanoic acid dithiobenzoate end groups was used as the macro chain transfer agent (PEG macro‐CTA) for sole monomer sodium 4‐styrenesulfonate. The reaction was controllable and displayed living polymerization characteristics and the triblock copolymer had designed molecular weight. The reaction rate depended strongly on the CTA and initiator concentration ratio [CTA]0/[ACPA]0: an increase in [CTA]0/[ACPA]0 from 1.0 to 5.0 slowed down the polymerization rate and improved the molecular weight distribution with a prolonged induction time. 1‐Phosphabicyclo[3.2.1]octanes 1‐Phosphabicyclo[3.2.1]octane has been obtained by free‐radical cyclization of (2‐vinyl‐4‐pentenyl)‐phosphane in the presence of AIBN. Another approach to 1‐phosphabicyclo[3.2.1]octanes involves free‐radical cyclization of 2‐methyl‐4‐(2‐propenyl)‐phospholane synthesized by the reaction of [2‐(2‐propenyl)‐4‐pentenyl]‐phosphane with KPH2/[18]crown‐6 in THF. The bicyclic phosphanes are characterized by reactions with CS2, selenium, sulfur, NO, CH3I, and HSO3F, respectively, structural and analytical data as well as 1H, 13C, 31P, 77Se NMR spectral measurements. The steric crowding of the phosphanes as complex ligands has been estimated from 31P–1H coupling constants according to the Tolman model. The configuration of the methyl substituents as well as the conformation of the six‐membered ring were determined by NMR parameters (coupling constants, noe's) and proved by X‐ray crystal structure analysis.  相似文献

A series of well‐defined amphiphilic triblock copolymers, poly(ethylene glycol)‐b‐poly(tert‐butyl acrylate)‐b‐poly(2‐hydroxyethyl methacrylate) (PEG‐b‐PtBA‐b‐PHEMA), were synthesized via successive atom transfer radical polymerization (ATRP). ATRP of tBA was first initiated by PEG‐Br macroinitiator using CuBr/N,N,N′,N″,N′″‐pentamethyldiethylenetriamine as catalytic system to give PEG‐b‐PtBA diblock copolymer. This copolymer was then used as macroinitiator to initiate ATRP of HEMA, which afforded the target triblock copolymer, PEG‐b‐PtBA‐b‐PHEMA. The critical micelle concentrations of obtained amphiphilic triblock copolymers were determined by fluorescence spectroscopy using N‐phenyl‐1‐naphthylamine as probe. The morphology and size of formed aggregates were investigated by transmission electron microscopy and dynamic light scattering, respectively. Finally, an acid‐sensitive PEG‐b‐PtBA‐b‐P(HEMA‐CAD) prodrug via cis‐aconityl linkage between doxorubicin and hydroxyls of triblock copolymers with a high drug loading content up to 38%, was prepared to preliminarily explore the application of triblock copolymer in drug delivery. © 2012 Wiley Periodicals, Inc. A novel, near‐monodisperse, well‐defined ABA triblock copolymer, poly[2‐(dimethylamino)ethyl methacrylate]‐b‐poly(propylene oxide)‐b‐poly[2‐(dimethylamino)ethyl methacrylate], was synthesized via oxyanion‐initiated polymerization. The initiator was a telechelic‐type potassium alcoholate prepared from poly(propylene glycol) and KH in dry tetrahydrofuran. The copolymers produced were characterized by Fourier transform infrared, 1H NMR, and gel permeation chromatography (GPC). GPC and 1H NMR analyses showed that the products obtained were the desired copolymers, with narrow molecular weight distributions (ca. 1.09–1.11) very close to that of the original poly(propylene glycol). 1H NMR, surface tension measurements, and dynamic light scattering all indicated that the triblock copolymer led to interesting aqueous solution behaviors, including temperature‐induced micellization and very high surface activity. © 2002 John Wiley & Sons, Inc. Biodegradable, amphiphilic, diblock poly(ε‐caprolactone)‐block‐poly(ethylene glycol) (PCL‐b‐PEG), triblock poly(ε‐caprolactone)‐block‐poly(ethylene glycol)‐block‐poly(ε‐caprolactone) (PCL‐b‐PEG‐b‐PCL), and star shaped copolymers were synthesized by ring opening polymerization of ε‐caprolactone in the presence of poly(ethylene glycol) methyl ether or poly(ethylene glycol) or star poly(ethylene glycol) and potassium hexamethyldisilazide as a catalyst. Polymerizations were carried out in toluene at room temperature to yield monomodal polymers of controlled molecular weight. The chemical structure of the copolymers was investigated by 1H and 13C NMR. The formation of block copolymers was confirmed by 13C NMR and DSC investigations. The effects of copolymer composition and molecular structure on the physical properties were investigated by GPC and DSC. For the same PCL chain length, the materials obtained in the case of linear copolymers are viscous whereas in the case of star copolymer solid materials are obtained with low Tg and Tm temperatures. © 2007 Wiley Periodicals, Inc. A series of well‐defined amphiphilic triblock copolymers [polyethylene glycol monomethyl ether]‐block‐poly(ε‐caprolactone)‐block‐poly[2‐(dimethylamino)ethyl methacrylate] (mPEG‐b‐PCL‐b‐PDMAEMA or abbreviated as mPEG‐b‐PCL‐b‐PDMA) were prepared by a combination of ring‐opening polymerization and atom transfer radical polymerization. The chemical structures and compositions of these copolymers have been characterized by Fourier transform infrared spectroscopy, 1H NMR, and thermogravimetric analysis. The molecular weights of the triblock copolymers were obtained by calculating from 1H NMR spectra and gel permeation chromatography measurements. Subsequently, the self‐assembly behavior of these copolymers was investigated by fluorescence probe method and transmission electron microscopy, which indicated that these amphiphilic triblock copolymers possess distinct pH‐dependent critical aggregation concentrations and can self‐assemble into micelles or vesicles in PBS buffer solution, depending on the length of PDMA in the copolymer. Agarose gel retardation assays demonstrated that these cationic nanoparticles can effectively condense plasmid DNA. Cell toxicity tests indicated that these triblock copolymers displayed lower cytotoxicity than that of branched polyethylenimine with molecular weight of 25 kDa. In addition, in vitro release of Naproxen from these nanoparticles in pH buffer solutions was conducted, demonstrating that higher PCL content would result in the higher drug loading content and lower release rate. Macroinitiator‐amino terminated poly(ethylene glycol) (PEG) (NH2‐PEO‐NH2) was prepared by converting both terminal hydroxyl groups of PEG to more reactive primary amino groups. The synthetic route involved reactions of chloridize, phthalimide and finally hydrazinolysis. Furthermore, poly(γ‐benzyl‐L ‐glutamate)‐poly(ethylene oxide)‐poly(γ‐benzyl‐L ‐glutamate) (PBLG‐PEO‐PBLG) triblock copolymer was synthesized by polymerization of γ‐benzyl‐L ‐glutamate N‐carboxyanhydride (Bz‐L‐GluNCA) using NH2‐PEO‐NH2 as macroinitiator. The resultant NH2‐PEO‐NH2 and triblock copolymer were characterized by FT‐IR, 1H‐NMR and gel permeation chromatography (GPC) techniques. The results demonstrated that the degree of amination of the NH2‐PEO‐NH2 could be up to 1.95. The molecular weight of the PBLG‐PEO‐PBLG triblock copolymer could be adjusted easily by controlling the molar ratio of Bz‐L ‐Glu NCA to the macroinitiator NH2‐PEO‐NH2. Most of the strategies for the synthesis of 6,8‐dioxabicyclo[3.2.1]octanes (BCO) and cyclopentenols (CP), which show high biological and pharmaceutical activity, are multistage and/or require hardly accessible starting materials and catalysts. The general method for BCO and CP preparation in a single preparative stage from two simple reagents, ketones and acetylene, under the action of an available super‐basic catalytic system KOH/DMSO becomes promising for the development of general approaches to the design of these classes of biologically active compounds. The mechanism of competing reactions yielding both 7‐methylene‐6.8‐dioxabicyclo[3.2.1]‐octanes and functionalized cyclopentenoles from their common intermediate 1,5‐diketones and acetylene in the KOH/DMSO superbasic media is investigated using the B2PLYP/6‐311++G**//B3LYP/6‐31+G* calculations with particular attention to the diastereoselectivity aspects of individual stages.  相似文献

Nontoxic and biodegradable poly(?‐caprolactone)‐b‐poly(ethylene glycol)‐b‐poly(?‐caprolactone) triblock copolymers were synthesized by the solution polymerization of ?‐caprolactone in the presence of poly(ethylene glycol). The chemical structure of the resulting triblock copolymer was characterized with 1H NMR and gel permeation chromatography. In aqueous solutions of the triblock copolymers, the micellization and sol–gel‐transition behaviors were investigated. The experimental results showed that the unimer‐to‐micelle transition did occur. In a sol–gel‐transition phase diagram obtained by the vial‐tilting method, the boundary curve shifted to the left, and the gel regions expanded with the increasing molecular weight of the poly(?‐caprolactone) block. In addition, the hydrodynamic diameters of the micelles were almost independent of the investigated temperature (25–55 °C). The atomic force microscopy results showed that spherical micelles formed at the copolymer concentration of 2.5 × 10?4 g/mL, whereas necklace‐like and worm‐like shapes were adopted when the concentration was 0.25 g/mL, which was high enough to form a gel. © 2006 Wiley Periodicals, Inc. Biodegradable star‐shaped poly(ethylene glycol)‐block‐poly(lactide) copolymers were synthesized by ring‐opening polymerization of lactide, using star poly(ethylene glycol) as an initiator and potassium hexamethyldisilazide as a catalyst. Polymerizations were carried out in toluene at room temperature. Two series of three‐ and four‐armed PEG‐PLA copolymers were synthesized and characterized by gel permeation chromatography (GPC) as well as 1H and 13C NMR spectroscopy. The polymerization under the used conditions is very fast, yielding copolymers of controlled molecular weight and tailored molecular architecture. The chemical structure of the copolymers investigated by 1H and 13C NMR indicates the formation of block copolymers. The monomodal profile of molecular weight distribution by GPC provided further evidence of controlled and defined star‐shaped copolymers as well as the absence of cyclic oligomeric species. The effects of copolymer composition and lactide stereochemistry on the physical properties were investigated by GPC and differential scanning calorimetry. For the same PLA chain length, the materials obtained in the case of linear copolymers are more viscous, whereas in the case of star copolymer, solid materials are obtained with reduction in their Tg and Tm temperatures. © 2007 Wiley Periodicals, Inc. A series of well‐defined double hydrophilic graft copolymers containing poly[poly(ethylene glycol) methyl ether acrylate] (PPEGMEA) backbone and poly[poly(ethylene glycol) ethyl ether methacrylate] (PPEGEEMA) side chains were synthesized by the combination of single electron transfer‐living radical polymerization (SET‐LRP) and atom transfer radical polymerization (ATRP). The backbone was first prepared by SET‐LRP of poly(ethylene glycol) methyl ether acrylate macromonomer using CuBr/tris(2‐(dimethylamino)ethyl)amine as catalytic system. The obtained comb copolymer was treated with lithium diisopropylamide and 2‐bromoisobutyryl bromide to give PPEGMEA‐Br macroinitiator. Finally, PPEGMEA‐g‐PPEGEEMA graft copolymers were synthesized by ATRP of poly(ethylene glycol) ethyl ether methacrylate macromonomer using PPEGMEA‐Br macroinitiator via the grafting‐from route. The molecular weights of both the backbone and the side chains were controllable and the molecular weight distributions kept narrow (Mw/Mn ≤ 1.20). Dihydroxyl capped biodegradable poly(DTC‐b‐PEG‐b‐DTC) (BCB) triblock copolymer and poly(TMC‐b‐DTC‐b‐PEG‐b‐DTC‐b‐TMC) (ABCBA) pentablock copolymer have been synthesized by PEG and BCB copolymer as macroinitiator in the presence of yttrium tris(2,6‐di‐tert‐butyl‐4‐methylphenolate). The copolymers without random segments have been thoroughly characterized by 1H, 13C‐NMR, SEC, and DSC. Molecular weights of the obtained copolymers are dependent on the amount of PEGs and coincide with the theoretical values. The exchange reaction of yttrium alkoxide and hydroxyl end group is essential for controlling the products' molecular weight. Their thermal behaviors are relevant to the chain lengths of PEG and PDTC segments. The Monte Carlo method has been developed to estimate the chain propagation constant and exchange reaction constant. Two samples of ABCD 4‐miktoarm star quarterpolymer with A = polystyrene (PS), B = poly(ε‐caprolactone) (PCL), C = poly(methyl methacrylate) (PMMA) or poly(tert‐butyl acrylate) (PtBA), and D = poly(ethylene glycol) (PEG) were prepared using click reaction strategy (Cu(I)‐catalyzed Huisgen [3 + 2] reaction). Thus, first, predefined block copolymers of different polymerization routes, PS‐b‐PCL with azide and PMMA‐b‐PEG and PtBA‐b‐PEG copolymers with alkyne functionality, were synthesized and then these blocks were combined together in the presence of Cu(I)/N,N,N′,N″,N″‐pentamethyldiethylenetriamine as a catalyst in DMF at room temperature to give the target 4‐miktoarm star quarterpolymers. AB block copolymers composed of hydrophilic poly(ethylene glycol) (PEG) and hydrophobic poly(amino acid) with a carboxyl group at the end of PEG were synthesized with α‐carboxylic sodium‐ω‐amino‐PEG as a macroinitiator for the ring‐opening polymerization of N‐carboxy anhydride. Characterizations by 1H NMR, IR, and gel permeation chromatography were carried out to confirm that the diblock copolymers were formed. In aqueous media this copolymer formed self‐associated polymer micelles that have a carboxyl group on the surface. The carboxyl groups located at the outer shell of the polymeric micelle were expected to combine with ligands to target specific cell populations. A series of well‐defined poly[methyl(3,3,3‐trifluoropropyl)siloxane]‐b‐polystyrene‐b‐poly(tert‐butyl acrylate) (PMTFPS‐b‐PS‐b‐PtBA) triblock copolymers were prepared by a combination of anionic ring‐opening polymerization of 1,3,5‐trimethyl‐1,3,5‐tris(3′,3′,3′‐trifluoropropyl)cyclotrisiloxane (F3), and atom transfer radical polymerization (ATRP) of styrene (St) and tert‐butyl acrylate (tBA), using the obtained α‐bromoisobutyryl‐terminal PMTFPS (PMTFPS‐Br) as the macroinitiators. The ATRP of St from PMTFPS‐Br, as well as the ATRP of tBA from the obtained PMTFPS‐b‐PS‐Br macroinitiators, has typical characteristic of controlled/living polymerization. The results of contact angle measurements for the films of PMTFPS‐b‐PS‐b‐PtBA triblock copolymers demonstrate that the compositions have an effect on the wetting behavior of the copolymer films. For the copolymer films with different compositions, there may be different macroscale or nanoscale structures on the outmost layer of the copolymer surfaces. The films with high content of PtBA blocks exhibit almost no ordered microstructures on the outmost layer of the copolymer surfaces, even though they have microphase‐separated structures in bulk. © 2012 Wiley Periodicals, Inc. A novel triblock copolymer PS–PHB–PS based on the microbial polyester Poly[(R)‐3‐hydroxybutyrate)] (PHB) and poly(styrene) (PS) was prepared to be used as compatibilizer for the corresponding PHB/PS blends. It was prepared in a three‐step procedure consisting of (i) transesterification reaction between ethylene glycol and a high‐molecular‐weight PHB, (ii) synthesis of bromo‐terminated PHB macroinitiator, and (iii) atom transfer radical polymerization polymerization of styrene initiated by the PHB‐based macroinitiator. Fourier transform infrared, gel permeation chromatography, 1H‐, and 13C‐NMR spectroscopies were used to determine the molecular structure and/or end‐group functionalities at each step of the procedure. Although thermogravimetric analysis showed that the block copolymer underwent a stepwise thermal degradation and had better thermal stability than their respective homopolymers, differential scanning calorimetry displayed that the PHB block in the copolymer could not crystallize, and thus generating a total amorphous structure. Atomic force microscopy images indicated that the block copolymer was phase segregated in a well‐defined morphological structure with nanodomain size of ~40 nm. Contact angle measurements proved that the wettability properties of the block copolymer were in between those of the PHB and PS homopolymers. Blends analyzed for their morphology and thermal properties showed good miscibility and had well‐defined morphological features. Polymer blends exhibited lower crystallinity and decreased stiffness which was proportional to the amount of compatibilizer content in the blends. © 2012 Wiley Periodicals, Inc. H‐shaped quintopolymer containing different five blocks: poly(ε‐caprolactone) (PCL), polystyrene (PS), poly(ethylene glycol) (PEG), and poly(methyl methacrylate) (PMMA) as side chains and poly(tert‐butyl acrylate) (PtBA) as a main chain was simply prepared from a click reaction between azide end‐functionalized PCL‐PS‐PtBA 3‐miktoarm star terpolymer and PEG–PMMA‐block copolymer with alkyne at the junction point, using Cu(I)/N,N,N′,N″,N″‐pentamethyldiethylenetriamine (PMDETA) as a catalyst in DMF at room temperature for 20 h. The H‐shaped quintopolymer was obtained with a number–average molecular weight (Mn) around 32,000 and low polydispersity index (Mw/Mn) 1.20 as determined by GPC analysis in THF using PS standards. Novel calixarene‐centered amphiphilic A2B2 miktoarm star copolymers composed of two PCL arms and two PEG arms with calix[4]arene as core moiety were synthesized by the combination of CROP and “click” chemistry. First, a heterotetrafunctional calix[4]arene derivative with two hydroxyl groups and two alkyne groups was designed as a macroinitiator to prepare calixarene‐centered PCL homopolymers (C4‐PCL) by CROP in the presence of Sn(Oct)2 as catalyst at 110 °C. Next, azide‐terminated PEG (A‐PEG) was synthesized by tandem treating methoxy poly(ethylene glycol)s (mPEG) with 4‐chlorobutyryl chloride and NaN3. Finally, copper(I)‐catalyzed cycloaddition reaction between C4‐PCL and A‐PEG led to A2B2 miktoarm star copolymer [C4S(PCL)2‐(PEG)2]. 1H NMR, FT‐IR, and SEC analyses confirmed the well‐defined miktoarm star architecture. These amphiphilic miktoarm star copolymers could self‐assemble into multimorphological aggregates in water. The calix[4]arene moieties with a cavity <1 nm on the hydrophilic/hydrophobic interface of these aggregates may provide potential opportunities to entrap guest molecules for special applications in supermolecular science. © 2010 Wiley Periodicals, Inc. Poly[N‐isopropylacrylamide‐g‐poly(ethylene glycol)]s with a reactive group at the poly(ethylene glycol) (PEG) end were synthesized by the radical copolymerization of N‐isopropylacrylamide with a PEG macromonomer having an acetal group at one end and a methacryloyl group at the other chain end. The temperature dependence of the aqueous solutions of the obtained graft copolymers was estimated by light scattering measurements. The intensity of the light scattering from aqueous polymer solutions increased with increasing temperature. In particular, at temperatures above 40°C, the intensity abruptly increased, indicating a phase separation of the graft copolymer due to the lower critical solution temperature (LCST) of the poly(N‐isopropylacrylamide) segment. No turbidity was observed even above the LCST, and this suggested a nanoscale self‐assembling structure of the graft copolymer. The dynamic light scattering measurements confirmed that the size of the aggregate was in the range of several tens of nanometers. The acetal group at the end of the PEG graft chain was easily converted to the aldehyde group by an acid treatment, which was analyzed by 1H NMR. Such a temperature‐induced nanosphere possessing reactive PEG tethered chains on the surface is promising for new nanobased biomedical materials. © 2006 Wiley Periodicals, Inc.
