N-Tolylglycosylamines were synthesized from o-, m-, and p-toluidines and aldoses (D-glucose, D-galactose, D-mannose, L-rhamnose, D-xylose, and L-arabinose). Derivatives of N-(2-hydroxyethyl)cytisine, N-(2-hydroxypropyl)-, N-(2-hydroxy-2-(1-adamantyl)ethyl)-, and N-(2-hydroxy-2-phenylethyl)cytisine, were synthesized by reduction of N-(2-oxopropyl)-, N-(2-oxo-2-(1-adamantyl)ethyl)-and N-(2-oxo-2-phenylethyl)cytisine with metal hydrides. New substituted 4-chloromethylcoumarins that were used as alkylating agents to modify cytisine were synthesized by Pechmann condensation. A series of 4-(12-cytisylmethyl)coumarins containing pharmacophores of the natural heterocycles coumarin and cytisine in a single molecule was prepared. Various approaches to the synthesis of per-O-acetylated thioglycosides of N-acetylneuraminic acid (Neu5Ac) containing an unprotected carboxy group starting from the corresponding methyl esters were comparatively studied. One-step demethylation of methyl thioglycoside (LiI, Py, reflux) proceeded inefficiently in contrast to the analogous smooth reaction of phenyl thioglycoside. An indirect route to derivatives with a free carboxy group involving saponification followed by acetylation of hydroxy groups proved to be more efficient for methyl -thioglycoside of Neu5Ac.  相似文献

A parallel solid-phase synthesis of combinatorial library of 436 amides of 4-(3,7-diorganyl-3,7-diazabicyclo[3.3.1]nonan-2-yl)butanoic acid has been accomplished starting from natural alcaloid (−)-cytisine. A five-step liquid-phase synthesis resulted in the conversion of cytisine to 7-benzyl-3-[(9H-fluoren-9-yl)methyl]-substituted acids, which were further diversified with the use of solid-phase technology on the acid-susceptible amine resins. Summary. The preparation of N-acylsulfonamides is described using primary amines, arylsulfonyl chlorides and acyl chlorides. Reaction of primary aryl amines with arylsulfonyl chlorides in the presence of NaHCO3 produced N-arylsulfonamides, which reacted in situ with benzoyl chloride furnishing the corresponding N-benzoyl-N-arylsulfonamides in 72–96% yields. Accordingly, 4-nitrobenzoyl chloride and 3,5-dinitrobenzoyl chloride were used as acylating agents. All the reactions were carried out under solvent-free conditions at room temperature and the products were isolated after simple work-up in high yields and purity.  相似文献

Three series of nonionic N-alkylaldonamides, N-alkyl-N-methylgluconamides (Cn-MGA, Cn: n-C(10)H(21), n-C(12)H(25), n-C(14)H(29), n-C(16)H(33), and n-C(18)H(37)), N-alkyl-N-methyllactobionamides (Cn-MLA, alkyl as above-mentioned), and N-oleyl-N-methylglucon/lactobionamide, were synthesized in the reaction of an appropriate N-alkyl-N-methylamine with delta-D-glucolactone and lactobionic acid, respectively. Krafft temperatures of aqueous solutions and surface properties of these surfactants at 20 degrees C, i.e., surface excess concentration, Gamma(cmc), surface area demand per molecule, A(min), efficiency in surface tension reduction, pC(20), effectiveness in surface tension reduction, Pi(cmc), critical micelle concentration, CMC, and CMC/C(20) parameter as well as standard free energies of adsorption, DeltaG degrees (ads), and of micellization, DeltaG degrees (mic), were determined. It was shown that introduction of the methyl group to the amide nitrogen increased the solubility of the surfactants, which was confirmed by their Krafft temperatures. Lactobionamides are more water soluble than gluconamides. On the other hand, the Cn-MGA surfactants are more surface active than the respective Cn-MLA ones. This observation is based on the determined adsorption and micellization parameters. The presence of one double bond in a hydrocarbon chain as in oleyl-amides increases their hydrophilic character compared with that of saturated C18 derivatives. No distinct differences were observed between the A(min) values obtained for both series studied, although they differ markedly in the size of the hydrophilic groups.
