5(4)-Chloro-1-(2,4-dinitrophenyl)pyrazoles from 2,4-Dinitrophenylhydrazones of Chlorovinyl Ketones and β,β-Dichloroacrolein

A method has been developed for obtaining 3-alkyl(phenyl)-4(5)-chloro-1-(2,4-dinitrophenyl)pyrazoles from appropriate dinitrophenylhydrazones of 1-chloro-, 1,2-, and 2,2-dichlorovinyl ketones by heating the latter in polyphosphoric acid. 3-(3-吡啶基)-4-氨基-5-芳胺基-1,2,4-三唑的合成与表征

以烟酸与芳胺为基本原料,经多步反应,制得1－烟酰基－4－芳基氨基硫脲;后者在水合肼存在下关环,合成了一系列新化合物3－（3－吡啶基）－4－氨基－5－芳胺基－1,2,4－三唑。化合物结构经元素分析、IR,^1H NMR,^13C NMR确证。     

2-Polyfluoroalkylchromones. 14. Synthesis of 4-chloro-3(5)-(2-hydroxyaryl)-5(3)-polyfluoroalkylpyrazoles

3-Chloro-2-polyfluoroalkyl- and 3-chloro-6-nitro-2-polyfluoroalkylchromones were synthesized. These compounds react with N₂H₄·2HCl on boiling in ethanol to form 4-chloro-3(5)-(2-hydroxyaryl)-5(3)-polyfluoroalkylpyrazoles.     

(3S,4S)-4-氨基-3-羟基-6-甲基庚酸的合成

价廉易得的L-亮氨酸先以苄基同时保护氨基及羧基得(S)-2-(二苄胺基)-4-甲基戊酸苄酯,进而在碱性条件下与乙腈发生亲核取代反应得(S)-4-(二苄胺基)-6-甲基-3-氧代庚腈,再经硼氢化钠选择性还原羰基得(3S,4S)-4-(二苄胺基)-3-羟基-6-甲基庚腈,用双氧水氧化得(3S,4S)-4-(二苄胺基)-3-羟基-6-甲基庚酸,最后在Pd(OH)2/C-H2作用下脱掉苄基得到(3S,4S)-4-氨基-3-羟基-6-甲基庚酸,即(3S,4S)-statine。整个合成路线总产率为33.6%。     

Carbon-13 NMR Studies of 3-Aryl-4-(5-aryl-Δ 2-1,2,4-oxadiazolin-3-yl)sydnones and 3-Aryl-4-(5-aryl-1,2,4-oxadiazol-3-yl)sydnones

The ¹³C NMR spectra of twelve 3-aryl-4-(5-aryl-Δ²-l,2,4-oxadiazolin-3-yl)sydnones and twelve 3-aryl-4-(5-aryl-l,2,4-oxadiazol-3-yl)sydnones have been measured and assigned by means of proton-noise decoupling and DEPT-experiments. The coupling constants were determined by means of gated decoupling, and NOE effects were observed by comparison of proton-decoupled and inverse-gated decoupled spectra. Differences shown by the oxadiazoline and the oxadiazole rings and the substitution effects are discussed.     

Synthesis of 6-amino-4-aryl-5-cyano-3-(3-cyanopyridin-2-ylthiomethyl)-2,4-dihydropyrano[2,3-c]pyrazoles and their hydrogenated analogs. Molecular structure of 6-amino-5-cyano-3-(3-cyano-4,6-dimethylpyridin-2-ylthiomethyl)-4-(2-nitrophenyl)-2,4-dihydropyrano[2,3-c]pyrazole

Substituted 3-hydroxypyrazoles, which were prepared based on ethyl esters of substituted 4-(pyridin-2-ylthio)- or 4-(1,4-dihydropyridin-2-ylthio)acetoacetic acids and hydrazine hydrate, were used in the synthesis of 6-amino-4-aryl-5-cyano-3-(pyridin-2-ylthiomethyl)-2,4-dihydropyrano[2,3-c]pyrazoles. The molecular and crystal structure of 6-amino-5-cyano-3-(3-cyano-4,6-dimethylpyridin-2-ylthiomethyl)-4-(2-nitrophenyl)-2,4-dihydropyrano[2,3-c]pyrazole was established by X-ray diffraction analysis.     

D-脯氨酸催化的(3S,4S)-4-氨基-3-羟基-5-苯基戊酸衍生物的合成

报道了一条合成(3S,4S)-4-氨基-3-羟基-5-苯基戊酸(Ahppa)衍生物的新路线。以氨基保护的L-苯丙氨酸为起始原料,依次经Weinreb胺缩合、还原、aldol缩合及溴仿4步反应合成了3个Ahppa衍生物,总收率5.8%~6.7%,其结构经¹H NMR, ¹³C NMR和ESI-MS确证。对反应条件进行了探讨,结果表明：催化剂D-脯氨酸用量对反应收率影响不大,对立体选择性影响较大;氨基上保护基体积较大有利于提高反应立体选择性。     

6-氨基-5-氰基-3-甲基-4-(3-硝基苯)-1-苯基吡唑[3,4-b]并吡啶与人血清白蛋白结合作用的光谱特征

采用紫外光谱法和荧光光谱法研究了6-氨基-5-氰基-3-甲基-4-(3-硝基苯)-1-苯基吡唑[3,4-b]并吡啶(6A)与人血清白蛋白(HSA)的结合作用,利用同步荧光法和三维荧光法研究了6A与HSA作用前后人血清白蛋白的构象变化。观测到生理pH7.4条件下6A使HSA的紫外吸收峰增强,特征荧光峰猝灭。Stern-Volmer曲线显示,6A对HSA的荧光猝灭可能是一个单一的静态猝灭过程,并且得出18℃和37℃时的结合位点数和结合常数。根据F rster非辐射转移理论可求出6A与HSA作用距离r=3.73 nm;根据基本热力学参数ΔH、ΔS和ΔG判断6A和HSA主要通过氢键和范德华力发生相互作用。     

Novel synthesis of 2-amino-3-hetaryl-4(5H)-oxothiophenes

3-Cyano-3-hetaryl-2-oxopropyl thioacetates were obtained by the acylation of hetarylacetonitriles with acetylmercaptoacetyl chloride. A highly enantioselective synthesis of 5-(l-menthyloxy )-4-substituted-3-chloro-2(5H )-furanones

In this paper, stereocontrolled tandem Michael addition-elimination reaction of the novel chiral source, 5-(l-menthyloxy)-3,4-dichloro-2(5H)-furanone, with various thiols and amines has been investigated. A series of new enantiomerically pure compounds, 5–(l-menthyloxy)-4-substituted-3-chloro-2(5H)-furanones, were obtained in good yields with d. e. ? 98% under mild conditions.     

Synthesis of 3-amino-5-nitrobenzaldehyde oxime and its conversion into 3,4-bis(3-amino-5-nitrophenyl)furoxan and isomeric 3(4)-(3-amino-5-nitrophenyl)-4(3)-nitrofuroxans

The oxime of the hitherto unknown 3-amino-5-nitrobenzaldehyde was synthesized by the reaction of 3-amino-5-nitrobenzaldehyde phenylhydrazone with excess of (NH₂OH)₂·H₂SO₄. The oxime obtained was used as the starting compound for the synthesis of novel diaryl- and arylnitrofuroxans. Translated fromIzvestiya Akademii Nauk. Synthesis of 4-amino-5-(4,6-diphenyl-2-pyrimidinyl)-3,4-dihydro-2H-1,2,4-triazole-3-thione and its reactions with C-electrophiles

4-Amino-5-(4,6-diphenyl-2-pyrimidinyl)-3,4-dihydro-2H-1,2,4-trazole-3-thione is formed from the reaction of 4,6-diphenylpyrimidinecarboxylic acid or its ethyl ester with thiocarbonyl hydrazide. Alkylation of the product leads to S-alkyl derivaties or 6-substituted 3-(4,6-diphenyl-2-pyriimidinyl)-7H-1,2,4-triazolo[3,4-b]-1,3,4-thiadiazine. Acetylation of 4-amino-5-(4,6-diphenyl-2-pyrimidinyl)-3,4-dihydro-2H-1,2,4-triazole-3-thione gave under different conditions monoacetyl-, diacetyl, and triacetyl derivatives at the amino group and the N₍₂₎ atom, whereas benzoylation gave a benzoyl group at the amino group and 3-(4,6-diphenyl-2-pyrimidinyl)-6-phenyl-1,2,4-triazolo[3,4-b]-1,3,4-thiadiazole. __________ Translated from Khimiya Geterotsiklicheskikh Soedinenii, No. 7, 1088–1094, July, 2007.     

4-(4-羟基丁基－2－炔氧基)－3-（苯磺酰基）－1，2，5－噁二唑－2－氧化物与牛血清白蛋白结合作用的光谱法研究

The interaction between 4-(4-hydroxybut-2-ynyloxy)-3-(phenylsulfonyl)-1,2,5-oxadiazole-2-oxide(FB)andbovine serum albumin(BSA)was studied by spectroscopic methods including fluorescence and UV-Vis absorptionspectroscopy.The quenching mechanism of fluorescence of BSA by FB was considered to be a dynamic quenchingprocedure.The number of binding sites n and apparent binding constant K were measured by fluorescence quench-ing method.The results indicate that there is FB molecular binding with BSA,and forming 1∶1 complex.Thethermodynamic parameters such as ΔH,ΔG and ΔS,etc.,were calculated.The results indicate that the binding reac-tion is mainly entropy-driven and hydrophobic forces play major role in the binding reaction.The distance r be-tween donor(BSA)and acceptor(FB)was obtained according to Frster theory of non-radioactive energy transfer.

3-氯-4-二苄胺基-5-甲氧基-2(5H)-呋喃酮的合成与晶体结构

在THF溶液中合成了标题化合物3-氯-4-二苄胺基-5-甲氧基-2(5H)-呋喃酮,并用FT-IR、UV-Vis、1HNMR、13C NMR、MS、元素分析和X-射线衍射等进行了表征。结果表明此化合物属正交晶系,空间群为Pbca,晶胞参数为:a=15.891(16),b=11.126(11),c=19.778(19),α=β=γ=90°,V=3497(6)3,Z=8,Dc=1.306Mg/m3,μ=0.234 mm-1,F(000)=1440。在化合物的分子结构中,两个苯环几乎垂直于呋喃酮平面,且它们与呋喃酮平面的两面角分别为89.38°和88.19°。     

Laser photolysis of 1-(4-nitrophenyl)-3-methylpyrazole and 1-ethyl-3-(4-nitrophenyl)-5-chloropyrazole

The spectral kinetic characteristics of the triplet states of 1-(4-nitrophenyl)-3-methylpyrazole (1) and 1-ethyl-3-(4-nitrophenyl)-5-chloropyrazole (2) were studied by the laser nanosecond photolysis technique in different solvents. The triplet lifetimes (τ_T) of molecules 1 and 2 were found to depend strongly on the solvent nature. An increase in τ_T by approximately two orders of magnitude on going from nonpolar and polar aprotic solvents (τ_T ≤ 15 ns) to aqueous-acetonitrile solutions (τ_T = 1100 ns for a volume acetonitrile to water ratio of 1 : 3) was analyzed, taking into account the influence of the medium on the relative contribution of the n,π*- and π,π*-configurations to the lowest triplet state. __________ Published in Russian in Izvestiya Akademii Nauk. Synthesis, Crystal Structure and Antitumor Activity of （E）-4-tert-Butyl-N-（2,4-dichlorobenzylidene）-5-（1,2,4-triazol-1-yl）-thiazol-2-amine

The title compound (E)-4-tert-butyl-N-(2,4-dichlorobenzylidene)-5-(1,2,4-triazol-1-yl)-thiazol-2-amine was synthesized by the reaction of 4-tert-butyl-5-(1,2,4-triazol-1-yl)-thiazol-2-amine with 2,4-dichlorobenzaldehyde, and its crystal structure was determined by singlecrystal X-ray diffraction. The crystal belongs to the triclinic system, space group P1 with a = 7.9748(4), b = 10.1803(5), c = 11.4603(6), α = 102.882(1), β = 100.253(1), γ = 104.457(1)°, V = 850.95(7)3, Z = 2, F(000) = 392, C16H15Cl2N5S, Mr = 380.29, Dc = 1.484 g/cm3, S = 1.095, μ = 0.512 mm-1, the final R = 0.0301 and wR = 0.0965 for 3334 observed reflections (I 2σ(I)). The preliminary antitumor activity shows that for the title compound the IC50 of Hela is 0.175 μmol/mL and that of Bel7402 is 0.156 μmol/mL.     

Synthesis and Crystal Structure of (Z)-Ethyl 5-(phenylamino)-3-(phenylimino)-3H-1,2-dithiole-4-carboxylate

The compound (Z)-ethyl 5-(phenylamino)-3-(phenylimino)-3H-1,2-dithiole-4-carboxylate 3 has been synthesized by the reaction of ethylacetoacetate 1 and phenylisothiocyanate 2. Its structure has been established by ¹H NMR, ¹³C NMR, infrared, mass spectra, and x-ray crystallography.      

Preparation of 3-Aryl-4-(5-ARYL-Δ2-1,2,4-Oxadiazolin-3-YL)Sydnones and their Conversion into 3-ARYL-4-(5-ARYL-1,2,4-Oxadiazol-3-YL)Sydnones

3-Aryl-4-(5-aryl-Δ²-1,2,4-oxadiazolin-3-yl)sydnones (5) were synthesized in high yields by the reaction of 3-arylsydnone-4-carboxamide oximes (prepared from the corresponding 3-arylsydnone-4-carbonitriles) with aromatic aldehydes in the presence of acid catalysts. No reaction occurred when aliphatic aldehydes were used. The oxadiazolin-3-ylsydnones (5) were easily converted into the corresponding 3-aryl-4-(5-aryl-1,2,4-oxadiazol-3-yl)sydnones by N-bromosuccinimide oxidation. The 3-arylsydnone-4-carbonitrile oxides were synthesized in good yields by N-bromosuccinimide oxidation of the corresponding 3-arylsydnone-4-carboxaldehyde oximes.
