Benzo-1,2,3,4-tetrazine 1,3-dioxides annulated with tetraazapentalene systems 3. Annulation at the C(7)—C(8) bond

Thermolysis of 7-azido-8-(2H-1,2,3-triazol-2-y1)-1,2,3,4-benzotetrazine 1,3-dioxide and 7-azido-8-(l H-1,2,3-triazol-l-yl)-1,2,3,4-benzotetrazine 1,3-dioxide afford new heterocyclic systems, viz., 7,11-dehydro-7H, 11H [1,2,3]triazolo[l′,2′:2,3][1,2,3]triazolo[4,5-f]-[1,2,3,4]benzotetrazine 1,3-dioxide and 7,9-dehydro-7 H, 9H [1,2,3]triazolo[2′,1′:2,3]-[1,2,3]triazolo[4,5-f][1,2,3,4]benzotetrazine 1,3-dioxide, respectively, and their thermal stability has been studied.     

Benzo-1,2,3,4-tetrazine 1,3-dioxides annulated with tetraazapentalenes

New heterocyclic system, viz., 8,12-dehydro-8H,12H-[1,2,3]triazolo[2′,1′:1,2][1,2,3]triazolo-[4,5-f][1,2,3,4]benzotetrazine 2,4-dioxide, has been synthesized by thermolysis of 5-azido-6-(1,2,3-triazol-2-yl)benzotetrazine 1,3-dioxide, and its thermal stability and spectral properties have been studied. An unusual cascade of <Emphasis Type="Italic">S</Emphasis><Subscript>N</Subscript> reactions of benzotetrazine 1,3-dioxide derivatives

An unusual cascade of S _NAr reactions was discovered in the series of benzo-1,2,3,4-tetrazine 1,3-dioxides containing two adjacent nucleofuges X and Y in the benzene ring. First, the 1,2,3-triazole anion displaces the anion X^s- from the more reactive site. Then the nucleo-phile X^s- displaces the adjacent group Y. For instance, 1,2,3-triazole reacts with 6-azido-5-nitrobenzotetrazine 1,3-dioxide to give 5-azido-6-(1,2,3-triazol-2-yl)benzotetrazine 1,3-dioxide, with 8-azido-7-nitrobenzotetrazine 1,3-dioxide to give 7-azido-8-(1,2,3-triazol-2-yl)benzotetrazine 1,3-dioxide and 7-azido-8-(1,2,3-triazol-1-yl)benzotetrazine 1,3-dioxide, and with 7-bromo-6-(phenylthio)benzotetrazine 1,3-dioxide to give 7-phenylthio-6-(1,2,3-triazol-2-yl)benzotetrazine 1,3-dioxide.     

Behavior of [1,2,5]oxadiazolo[3,4-<Emphasis Type="Italic">e</Emphasis>][1,2,3,4]tetrazine 4,6-dioxide in various media

A behavior of [1,2,5]oxadiazolo[3,4-e][1,2,3,4]tetrazine 4,6-dioxide (1) in organic solvents and aqueous solutions of various acidity has been studied by UV spectrophotometry. Kinetic laws for the N,N′-dioxide 1 hydrolysis have been established and decomposition process constants at various temperatures in the media with pH 6.86 have been determined. A product of the reaction with water, viz., 5H-[1,2,3]triazolo[4,5-c][1,2,5]oxadiazole, has been isolated. 5-(tert-Butyl)-5H-[1,2,3]triazolo[4,5-c][1,2,5]oxadiazole has been synthesized as the model compound. Synthesis and selected transformations of 1-(5-methyl-1-aryl-1H-1,2,3-triazol-4-yl)ethanones and 1-[4-(4-R-5-methyl-1H-1,2,3-triazol-1-yl)phenyl]ethanones

By cycloaddition of arylazides to acetylacetone are obtained derivatives of 1,2,3-triazole. In the reaction of 1-[5-methyl-1-(R-phenyl)-1H-1,2,3-triazol-4-yl] ethanones (IIa–IIe) and 1-[4-(4-R-5-methyl-1H-1,2,3-triazol-1-yl)phenyl] ethanones (VIIa-VIIe) with isatin are obtained 2-[1-(R-phenyl)-5-methyl-1H-1,2,3-triazol-4-yl]-4-quinolinecarboxylic acids (IIIa–IIIe) and 2-[4-(4-R-5-methyl-1H-1,2,3-triazol-1-yl)phenyl] -4-quinolinecarboxylic acids (IXa, IXb), respectively. We found that 1-[5-methyl-1-(R-phenyl)-1H-1,2,3-triazol-4-yl] ethanones (IIa–IIe) readily transform into [5-methyl-1-(R-phenyl)-1H-1,2,3-triazol-4-yl] acetic acids (IVa–IVc) by the method of Wilgerodt-Kindler. The (5-methyl-1-phenyl-1H-1,2,3-triazol-4-yl)acetic acid reacts with 5-phenyl-4-amino-4H-1,2,4-triazol-3-thiol affording 6-[(5-methyl-1-phenyl-1H-1,2,3-triazol-4-yl) methyl]-3-phenyl[1,2,4] triazolo[3,4-b] [1,3,4] thiadiazole (VI). Alkylation of 1-hydroxy-1H-[1,2,3]triazolo[4,5-e][1,2,3,4]tetrazine 5,7-dioxide

Alkylation of 1-hydroxy-1H-[1,2,3]triazolo[4,5-e][1,2,3,4]tetrazine 5,7-dioxide 1 and its silver salt 10 with different alkylating agents (diazomethane, diazoacetone, bromoacetone, α-bromoacetophenone, methyl iodide, methyl vinyl ketone) was studied. Alkylation of compound 1 with diazo compounds and salt 10 with halocompounds results predominantly in O-alkylation products, 1-alkoxy-1H-[1,2,3]triazolo[4,5-e][1,2,3,4]tetrazine 5,7-dioxides. The Michael reaction of compound 1 with methyl vinyl ketone involves the triazole nitrogen atom to give 1-(3-oxobutyl)-1H-[1,2,3]triazolo[4,5-e][1,2,3,4]tetrazine 3,4,6-trioxide. The structures of the compounds synthesized were established by ¹H, ¹³C, ¹⁴N NMR spectroscopy and mass spectrometry.     

Synthesis and transformations of 4-amino-4′-(4-nitro-1<Emphasis Type="Italic">H</Emphasis>-1,2,3-tri-azol-1-yl)- and 4-amino-4′-(1,2,3-triazolo[4,5-<Emphasis Type="Italic">c</Emphasis>]-1,2,5-oxadiazol-5-yl)- 3,3′-(NNO)-azoxy-1,2,5-oxadiazoles

Methods for the synthesis of 4-amino-4′-R-3,3′-(NNO)-azoxy-1,2,5-oxadiazoles (amino-azoxyfurazans) with 4-nitro-1,2,3-triazol-1-yl and 1,2,3-triazolo[4,5-c]furazan-5-yl substituents (R) by cycloaddition of morpholinonitroethylene to 4′-amino-4-azido-3,3′-(ONN)-azoxyfurazan or oxidative condensation of 3-R-4-aminofurazans with 4-trifluoroacetylamino-3-nitrosofurazan were elaborated. Dense energetic compounds of C,H, N,and O atoms. III. 5-[4-nitro-(1,2,5)oxadiazolyl]-5H-[1,2,3]triazolo[4,5-c][1,2,5]oxadiazole

Diazidoazofurazan 8 was obtained from the bis-diazonium salt of diaminoazofurazan 7 by treatment with sodium azide and underwent thermolysis to 5-[4-azido-(1,2,5)oxadiazolyl]-5H-[1,2,3]triazolo[4,5-c][1,2,5] oxadiazole 5 . The corresponding amine 13 was obtained from the azide 5 by reduction with stannous chloride and was oxidized by ammonium persulfate to 5-[4-nitro-(1,2,5)oxadiazolyl]-5H-[1,2,3]triazolo[4,5-c][1,2,5]oxadiazole 1 . The azide 5 was converted to a phosphinimine 9 in a reaction with triphenylphosphine.     

Reactions of 1,2,4-triazole derivatives with a “model” thiirane

Reactions of 3-mono- and 3,5-disubstituted 1,2,4-triazoles with a “model” thiirane, 8-bromo-1,3-dimethyl-7-(thiiran-2-ylmethyl)-3,7-dihydro-1H-purine-2,6-diones proceed at the positions N¹ and N² of the triazole ring and yield 7-(5-R-3-R′-1,2,4-triazol-1-yl)methyl- and/or 7-(5-R′-3-R-1,2,4-triazol-1-yl)methyl-1,3-dimethyl-6,7-dihydro[1,3]thiazolo[2,3-f]-purine-2,4-(1H,3H)-diones. 3-Methylsulfonyl-1,2,4-triazole reacted regiospecifically at the position N¹ forming 1,3-dimethyl-7-[(3-methyl-sulfonyl-1,2,4-triazole-1-yl)-methyl]-6,7-dihydro[1,3]thiazolo-[2,3-f]purine-2,4(1H,3H)-dione.     

Novel Selected Tandem Transformations of the Amino and Carbonyl/Nitrile Groups in the Gewald Thiophenes

Novel transformations of the amino and carbonyl/nitrile groups in the Gewald thiophenes were studied for thienopyrimidine synthesis. It was found that 2-amino-thiophene-3-carboxamides and ethyl 2-(acetylamino)-4,5,6,7-tetrahydro-1-benzothiophene-3-carbo- xylate did not yield tetrazole derivatives, neither in the reaction with triethyl orthoformate and sodium azide, nor in the reaction with phosphorus oxychloride and sodium azide, correspondingly. On the contrary, derivatives of thieno[2,3-d]pyrimidin-4(3H)-one and thieno[2,3-d][1,3]oxazin-4-one were isolated. New 2-azidothiophenes [2-azido-4,5,6,7-tetra hydro-1-benzothiophene-3-carbonitrile and 2-azido-4,5,6,7-tetrahydro-1-benzothiophen-3-yl(phenyl)methanone] were synthesized and used in anionic domino reactions with activated acetonitriles to yield thieno[3,2-e][1,2,3]triazolo[1,5-a]pyrimidines and/or 2-(5-amino-1H-1,2,3-triazol-1-yl)thiophenes. Finally, a new ring system of thieno[3,2-e]pyrazolo[1,5-a]pyrimidine was synthesized via a domino reaction of ethyl 2-[(2Z)-2-(1-chloro-2-ethoxy-2-oxoethylidene)hydrazino]-4,5,6,7-tetrahydro-1-benzothiophene-3-carboxylate with activated acetonitriles.     

Synthesis of 1-hydroxybenzotriazoles angularly annulated by furazan or furoxan rings

Synthesis of 6-hydroxy-6H-[1,2,3]triazolo[4,5-e][2,1,3]benzoxadiazole and a mixture of isomeric 6-hydroxy-6H-[1,2,3]triazolo[4,5-e][2,1,3]benzoxadiazole-1(3)-oxides is carried out starting from 2,4-dinitrosoresorcinol. Total assignment of the signals in the ¹³C NMR spectra of O-methylated products of these compounds is performed.     

Synthesis of 1‐substituted [1,2,3]triazolo[4,5‐d]pyridazines as precursors for novel tetraazapentalene derivatives

A series of 1‐substituted 4,5‐diformyl‐[1,2,3]triazole derivatives were prepared by 1,3‐dipolar cyclo‐addition of aryl azides with acetylene dicarboxaldehyde mono‐diethylacetal. The triazoles were readily converted into 1‐substituted [1,2,3]triazolo[4,5‐d]pyridazines in good yields. The 1‐(2‐nitrophenyl)‐[1,2,3]triazolo[4,5‐d]pyridazine was found to be a useful intermediate for the generation of the novel 5H‐benzo[1,2,3]triazolo[1′,2′:1,2]triazolo[4,5‐d]pyridazin‐6‐ium inner salt ring system.     

Synthesis and characterization of mono- and bicyclic heterocyclic derivatives containing 1,2,4-triazole, 1,3,4-thia-, and -oxadiazole rings

A series of new N- and S-substituted 1,3,4-oxadiazole derivatives were synthesized. 5-Pyridin-3-yl-3-[2-(5-thioxo-4,5-dihydro-l,3,4-thiadiazol-2-yl)ethyl]-1,3,4-oxadiazole-2(3H)-thione and 5-[(5-(pyridin-3-yl)-1,3,4-oxadiazol-2-ylthio)methyl]-N-phenyl-1,3,4-thiadiazol-2-amine were formed by cyclization of 3-(5-pyridin-3-yl-2-thioxo-1,3,4-oxadiazol-3(2H)-ylpropanimidohydrazide and 2-[(5-pyridin-3-yl-1,3,4-oxadiazol-2-yl)thio]thiosemicarbazide with CS₂ and H₂SO₄. On the other hand, a number of new bicyclic 1,2,4-triazolo[3,4-b][1,3,4]thiadiazole derivatives were synthesized. 6-Pyridin-3-ylbis[1,2,4]‐triazolo[3,4-b:4′,3′-d][1,3,4]thiadiazole-3(2H)-thione was synthesized by reaction of 6-(hydrazino)-3-pyridine-3-yl[1,2,4]triazolo[3,4-b][1,3,4]thiadiazole with CS₂/KOH/EtOH. Reaction of 2′-hydroxy-1,1′: 3′,1″-terphenyl-5′-carbaldehyde with naphthalen-1-amine,quinolin-8-amine,and 1,3-diketones

Condensation of 2′-hydroxy-1,1′: 3′,1″-terphenyl-5′-carbaldehyde with naphthalen-1-amine and cyclohexane-1,3-dione, methyl 2,2-dimethyl-4,6-dioxocyclohexane-1-carboxylate, or dimedone gave the corresponding 7-(2′-hydroxy-1,1′: 3′,1″-terphenyl-5′-yl)-7,8,9,10,11,12-hexahydro-12H-benzo[c]acridin-8-ones. The reaction of 2′-hydroxy-1,1′: 3′,1″-terphenyl-5′-carbaldehyde with naphthalen-1-amine and indan-1,3-dione produced 7-(2′-hydroxy-1,1′: 3′,1″-terphenyl-5′-yl)-8H-benzo[h]indeno[1,2-b]quinolin-8-one. 7-(2′-Hydroxy-1,1′: 3′,1″-terphenyl-5′-yl)-7,8,9,10,11,12-hexahydrobenzo[b][1,10]phenanthrolin-8-ones were obtained by three-component condensation of 2′-hydroxy-1,1′: 3′,1″-terphenyl-5′-carbaldehyde with quinolin-8-amine and cyclohexane-1,3-dione, methyl 2,2-dimethyl-4,6-dioxocyclohexane-1-carboxylate, or dimedone.     

Synthesis of 1-methyl-7-(trifluoromethyl)-1H-pyrido[2,3-c][1,2]thiazin-4(3H)-one 2,2-dioxide

The reaction of methyl 2-bromo-6-(trifluoromethyl)-3-pyridinecarboxylate ( 1 ) with methanesulfonamide gave methyl 2-[(methylsulfonyl)amino]-6-(trifluoromethyl)-3-pyridine-carboxylate ( 2 ). Alkylation of compound 2 with methyl iodide followed by cyclization of the resulting methyl 2-[methyl(methylsulfonyl)amino]-6-(trifluoromethyl)-3-pyridinecarboxylate ( 3 ) yielded 1-methyl-7-(trifluoromethyl)-1H-pyrido[2,3-c][1,2]thiazin-4(3H)-one 2,2-dioxide ( 4 ). The reaction of compound 4 with α,2,4-trichlorotoluene, methyl bromopropionate, methyl iodide, 3-trifluoromethylphenyl isocyanate, phenyl isocyanate and 2,4-dichloro-5-(2-propynyloxy)phenyl isothiocyanate gave, respectively, 4-[(2,4-dichlorophenyl)methoxy]-1-methyl-7-(trifluoromethyl)-1H-pyrido[2,3-c][1,2]thiazine 2,2-dioxide ( 5 ), methyl 2-[[1-methyl-2,2-dioxido-7-(trifluoromethyl)-1H-pyrido[2,3-c][1,2]thiazin-4-yl]oxy]propanoate ( 6 ), 1,3,3-trimethyl-7-(trifluoromethyl)-1H-pyrido[2,3-c][1,2]thiazin-4(3H)-one 2,2-dioxide ( 7 ), 4-hydroxy-1-methyl-7-(trifluoromethyl)-N-[3-(trifluoromethyl)phenyl]-1H-pyrido[2,3-c][1,2]thiazine-3-carboxamide 2,2-dioxide ( 8 ), 4-hydroxy-1-methyl-7-(trifluoromethyl)-N-phenyl-1H-pyrido[2,3-c][1,2]thiazine-3-carboxamide 2,2-dioxide ( 9 ) and N-[2,4-dichloro-5-(2-propynyloxy)phenyl]-4-hydroxy-1-methyl-7-(trifluoromethyl)-1H-pyrido[2,3-c][1,2] thiazine-3-carboxamide 2,2-dioxide ( 10 ).     

α-Thioureidoalkylation of urea heteroanalogs

α-Thioureidoalkylation of urea heteroanalogs such as thiosemicarbazide, amino-guanidine, sulfamide, and sulfonamides with 4,5-dihydroxyimidazolidine-2-thiones has been studied. Previously unknown 4,5-bis[thiosemicarbazido(guanidinoamino)]imidazolidine-2-thiones, 5,7-dialkylperhydroimidazo[4,5- e][1,2,4]triazine-3,6-dithiones, 4,6-diethyl-5(3H)-thioxotetrahydro-1 H-imidazo[4,5- c][1,2,5]thiadiazole 2,2-dioxide, and 1,3-dialkyl-4-[guanidinoimino(arylsulfonylimino)]imidazolidine-2-thiones have been synthesized.     

Annulated benzotetrazine 1,3-dioxides 2.* [1,2,5]Oxadiazolo[3,4-f ][1,2,3,4]benzotetrazine 1,3,7-trioxide

Thermolysis of 8-azido-7-nitrobenzotetrazine 1,3-dioxide led to benzotetrazine 1,3-dioxide annulated with the furoxan ring at the C(7)—C(8) bond. Complete assignment of the signals in the ¹³C NMR spectrum of the compound obtained was performed. Attempted syntheses of benzotetrazine 1,3-dioxides annulated with a furoxan ring at the C(6)—C(7) bond or two furoxan rings at the C(5)—C(6) and C(7)—C(8) bonds were unsuccessful. Three-component condensation of 4-aryl-1,4-dihydro-benzo[4,5]imidazo[1,2-<Emphasis Type="Italic">a</Emphasis>][1,3,5]triazin-2-amines with formaldehyde and primary amines

Three-component cyclization of 4-aryl-1,4-dihydrobenzo[4,5]imidazo[1,2-a][1,3,5]triazin-2-amines with formaldehyde and primary amines gave the corresponding 6-unsubstituted or 6-aryl-2-alkyl-2,3,4,6-tetrahydro-1H-[1,3,5]triazino[1′,2′:3,4][1,3,5]triazino[1,2-a]benzimidazoles.     

Synthesis of 1H-1,2,3-triazole derivatives by the cyclization of aryl azides with 2-benzothiazolylacetonone, 1,3-benzo-thiazol-2-ylacetonitrile,and (4-aryl-1,3-thiazol-2-yl)acetonitriles

The cyclization of aryl azides with 2-benzothiazolylacetone, 1,3-benzothiazol-2-ylacetonitrile, and (4-aryl-1,3-thiazol-2-yl)acetonitriles in methanol in the presence of sodium methylate gives high yields of new products, 2-(5-methyl(amino)-1-aryl-1H-1,2,3-triazol-4-yl)-1,3-benzothiazoles and 1-aryl-(4-aryl-1,3-thiazol-2-yl)-1H-1,2,3-triazole-5-amines. 1,3-Benzothiazol-2-ylacetonitrile undergoes an anionic domino reaction with methyl 2-azidobenzoate or 2-azidobenzonitrile to give [1,2,3]triazolo[1,5-a]quinazoline derivatives.
