The methanolic extract of Languas galanga rhizomes was investigated for antimalarial activity against Plasmodium berghei (NK65) infections in mice. The median lethal dose was determined to ascertain the safety of the extract in ICR mice of both sexes. The antimalarial activities during early and established infections, as well as the prophylactic activity were evaluated. Phytochemical screening and radical scavenging activity of the extract were also investigated to elucidate the possible mechanism of the antimalarial properties. The acute oral toxicity (LD??) of Languas galanga extract in mice was established to be 4.998 mg/kg. The extract of Languas galanga rhizomes demonstrated significant antiplasmodial activity in all the three models of the antimalarial evaluations. Phytochemical screening revealed the presence of some vital antiplasmodial constituents such as terpenoids and flavonoids. The extract also exhibited a moderate capacity to scavenge the free radicals. The rhizome extract of Languas galanga thus possesses antimalarial activity, which explains the rational usage of this plant in traditional Malaysian medicine.  相似文献

In this study, the rachis extracts of eight date palm Phoenix dactylifera L. cultivars were analysed by phytochemical screening and bioautography on Fusarium oxysporum f. sp. albedinis (Foa). The choice of cultivars was based on their reaction to Foa (resistant, tolerant and sensitive). Phytochemical screening was realised for flavonoids, tannins, alkaloids and coumarins. Antifungal effects were mostly represented by dichloromethanic extracts (seven out of nine inhibition zones). The best results were represented by the dichloromethanic extract of the cultivar 'Bent-Cherk' rachis (6.50?±?1.41?mm) and the ethyl acetate extract of the cultivar 'Rotbi' rachis (6.00?±?1.41?mm). The date palm cultivars presented some similarities concerning phytochemical screening results related to their resistance or sensibility to Foa. From the correlation between phytochemical screening and bioautography, it was observed that the majority of bioactive compounds against Foa seem to be polyphenols. Thus, the natural defence mechanism in vivo against Foa is probably related to the action of polyphenols. The difference between resistant, tolerant and sensitive cultivars is related to their mechanism of action.  相似文献

In this study, antimicrobial activities of water and methanol extract, and three phenolic fractions of the roots of Arbutus unedo L. were investigated. Poor antibacterial activity against both Staphylococcus aureus and Pseudomonas aeruginosa bacteria was shown with water and methanol extract. However moderate antibacterial activity was shown by water extract and phenolic fractions against Escherichia coli and S. aureus, respectively. The phytochemical screening of roots of A. unedo revealed the presence of quinones, anthraquinones reducteurs compounds, anthocyanins, tannins and flavonoids. Quantitative analysis showed that the roots were strongly dominated by anthocyanins compounds (3.65 mg g?1) followed by total flavonoids (0.56 mg?1) and flavones & flavonols (0.17 mg g?1).  相似文献

This study was designed to investigate in vitro biological activities and phytochemical composition of aqueous and ethanolic extracts from Achillea sintenisii Hub- Mor. (AS). To determine the chemical composition of AS extracts, phytochemical analyses were performed by using HPLC–ESI-Q-TOF-MS-MS. Afterwards, both extracts were investigated in terms of their effect on fibroblast proliferation, collagen synthesis, and hydrogen peroxide-induced damage. In addition to cell culture analysis, antibacterial, antioxidant, hyaluronidase inhibitory activities and total phenolic contents of the extracts were analyzed in cell-free systems. Our results demonstrated that the aqueous and ethanolic extracts did not show cytotoxic activity on fibroblasts, on the contrary, promoted fibroblast proliferation. Both AS extracts potently inhibited hyaluronidase activity and the inhibitory effect of ethanolic extract was comparable with the positive control, especially at high concentrations. The aqueous extract was the potent stimulator of collagen synthesis at 200 µg/mL concentration. Although the ethanolic extract showed antibacterial activity against all gram-positive bacteria, the aqueous extract was only effective against K. pneumoniae and B. subtilis. The ability of AS extracts, which have a rich phenolic compound content (≥50 mg GAE/g), to scavenge free radicals and protect fibroblasts against hydrogen peroxide-induced damage can be considered as a result of their antioxidant potential. Our findings scientifically support the widespread use of this plant, by demonstrating the pharmacological properties of the extracts.  相似文献

The current investigation aimed to shed light in the volatile and non-volatile secondary metabolites of Ajuga orientalis L. from Jordan. GC/MS and GC/FID analysis of the hydrodistilled essential oil obtained from aerial parts of the plant revealed tiglic acid (18.90 %) as main constituent. Each of the methanol and butanol fractions of A. orientalis were screened for their total phenol content (TPC), total flavonoid content (TFC), and antioxidant activity determined by DDPH and ABTS methods. The extracts were then analyzed by LC-ESI-MS/MS to unveil their chemical constituents, especially phenols and flavonoids. Results showed that the AO-B extract had the highest TPC (217.63 ± 2.65 mg gallic acid/g dry extract), TFC (944.41 ± 4.77 mg quercetin /g dry extract), highest DPPH and ABTS antioxidant activity ((4.00 ± 0.20) × 10-2; (3.00 ± 0.20) × 10-2 mg/mL, respectively) as compared to the AO-M extract. LC-ESI-MS/MS analysis of both extracts revealed the presence of several phenolics, flavonoids and nonphenolic acids.  相似文献

Abstract

Long-term hyperglycemia maintenance is responsible for increased protein glycation and formation of advanced glycation end products (AGEs), both are associated with the onset of diabetes mellitus complications. Efforts have been made to discover new agents having antiglycation potential. The aim of this study was to investigate the effects of the hydroethanolic extract and the ethyl acetate and methanolic fractions of Simaba trichilioides roots on the formation of AGEs. In an in vitro model system of protein glycation, incubations with hydroethanolic extract, ethyl acetate or methanolic fractions of S. trichilioides decreased the fluorescent AGEs, and markers of tyrosine and tryptophan oxidation. Protein crosslinking was reduced in the presence of the ethyl acetate fraction of S. trichilioides. Simaba trichilioides roots seem to be a promising source of compounds having ability to prevent glycoxidation changes, with potential applications in complementary therapies for management of diabetic complications.  相似文献

We evaluated the in vitro antioxidant property and phytochemical constituents of the crude ethyl acetate and methanol extract of the three genders of carob tree barks (spontaneous male, spontaneous female, and grafted female). The scavenging activity on DPPH (1,1-diphenyl-2-picrylhydrazyl) was determined, as well as the phenolic contents (Folin–Ciocalteu method) of both the extracts. The highest antioxidant activity and the higher amounts of total phenols were shown in methanol crude bark extract for the three genders. Variety significantly affected the phenol content and the antioxidant activity, with the spontaneous male variety globally showed a higher polyphenol concentration and antioxidant activity than the grafted female and spontaneous female.  相似文献

Alangium salviifolium root was screened for phytochemical and anti-inflammatory properties. The percentage inhibition of carrageenan induced paw oedema was studied in rats. Alangium salvifolium gave maximum extractive values with Ethanol and the Loss on Drying value, total ash value and acid-insoluble ash and water soluble ash values were within limits. The extract gave positive tests for phytosterols, triterpenes, flavonoids, carbohydrates and alkaloids. The extract was free from glycosides, saponins, tannins, proteins and amino acids. In acute toxicity studies, Alangium salviifolium root extract was found to be safe up to 3000?mg?kg?1, p.o. in the albino rats. The Alangium salviifolium root gave significant per cent inhibition of the maximal paw oedema and very highly significant per cent inhibition of total paw oedema during 6?h. This study revealed that Alangium salviifolium root has good anti-inflammatory actions when compared with Diclofenac sodium.  相似文献

(±)-Crataegusnorin A (1a/1b) and B (2a/2b), two pairs of rare 8,9′-epoxy-type norlignan enantiomers featuring a γ-butyrolactone ring, were isolated from the fruit of Crataegus pinnatifida. Their structures were determined via extensive spectroscopic analyses. Gauge-independent atomic orbital (GIAO) NMR chemical shift calculations, combined with the advanced statistical method DP4+ were employed to establish the relative configurations of four compounds. Next, chiral separation was accomplished by chiral chromatographic column and the absolute configurations of the four compounds were unambiguously assigned by comparison between their experimental electronic circular dichroism curves with the quantum-mechanically calculated curves based on time-dependent density functional theory (TDDFT). All the isolates were evaluated for their neuroprotective activities against H2O2-induced cell injury in human neuroblastoma SH-SY5Y cells. The results showed that two pairs of enantiomers 1a/1b and 2a/2b displayed diff ;erent effect on neuroprotective activity. Among them, compound 2a displayed the most potent neuroprotective effect. Further flow cytometry analysis indicated that 2a could protect SH-SY5Y cells from oxidative damage through inhibiting cell apoptosis.  相似文献

The effects of black tea polyphenol extract (BTPE) on the retrogradation of starches from different plant sources were studied using differential scanning calorimetry (DSC) and X-ray diffraction (XRD). DSC analysis shows that the gelatinization temperature of maize starch and starches from different rice varieties increased with increasing BTPE level. After storage at 4 °C, BTPE at a concentration of 15% markedly retarded the retrogradation of maize starch and starches from different rice varieties. Native maize starch and starches from different rice varieties showed typical A-type X-ray diffraction patterns, while native potato starch showed a typical B-type X-ray diffraction pattern. Adding BTPE significantly affected the crystalline region and intensities of X-ray diffraction peaks of maize and rice starch granules. It is concluded that adding BTPE markedly inhibits the retrogradation of maize starch and starches from different rice varieties, but has no significant influence on the gelatinization and retrogradation characteristics of potato starch.  相似文献

Fast analysis in LC can be performed with sub-2 microm particles at very high pressures (up to 1000 bar) known as ultra performance LC (UPLC). With this configuration, it is possible to obtain fast and/or highly efficient separations compared to conventional LC. For the analysis of compounds without chromophores, the evaporative light scattering detector (ELSD) is an attractive alternative because of its quasi-universality, versatility, low-cost and good sensitivity. The UPLC-ELSD was investigated in terms of sensitivity and apparent efficiency, with a conventional ELSD instrument, for two types of commercially available nebulisers, using different mobile phase flow rates and column ids. Results were finally compared with the UPLC-UV configuration. Three applications with phytochemical compounds were selected to highlight the potential of this approach (i.e. the isocratic separations of artemisinin and its derivatives, of calystegines and the gradient separation of several tropane alkaloids). Depending on the used column length, baseline separations were obtained in 3-10 min, with an average apparent efficiency ranging from 7000 to 30,000 plates.  相似文献

Liquid chromatography coupled to high‐resolution mass spectrometry is the technique of choice for the untargeted profiling of food matrices. Despite the high potential of high‐resolution mass spectrometry, when dealing with complex mixtures, an efficient separation technique is also needed. The novel core‐shell chromatographic columns packed with sub‐2 μm sized particles are claimed to show very good resolution. However, the analytes retention can be significantly altered when working under ultra‐high performance chromatographic conditions. In this work, an evaluation of four chromatographic systems, with either a single or two in‐series Kinetex™ C18 columns, either packed with 2.6 or 1.7 μm particles, is presented for the targeted analysis of a standard mixture and the untargeted analysis of a strawberry extract. An ultra‐high performance chromatographic system coupled via an electrospray source to a hybrid quadrupole‐Orbitrap mass spectrometer was used. From the extensive comparison, a surprising result was obtained, namely, that the system identifying the largest number of features was the one with two in‐series connected columns with the larger particle size. The inconsistency among the theoretical assumptions and the applicative findings points out the importance of an extensive chromatographic evaluation for the comprehensive untargeted profiling of complex real samples.  相似文献

Polygonum cognatum Meisn. (Polygonaceae) is used both as food and as a folk medicine to treat diabetes. This study aimed to evaluate the effect of the extracts, along with isolated compounds, from P. cognatum aerial parts on diabetes. In vitro studies were conducted using an α-glucosidase inhibitory assay, while in vivo antidiabetic studies were carried out on streptozotocin-induced diabetic rats. Effective extracts were subjected to isolation studies, and structures of the compounds were elucidated by spectroscopic methods. The ethyl acetate and n-butanol extracts had the highest effect in both in vitro and in vivo experiments. They also decreased aspartate transaminase, alanine transaminase and malondialdehyde levels, while increasing glutathione and superoxide dismutase activity in rats. From the active extracts, 11 phenolic compounds were isolated and characterized. Among the isolated compounds, quercetin was found to be the most active according to α-glucosidase inhibitory activity studies. This study provided scientific evidence for the traditional use of P. cognatum as a folk medicine for treating diabetes. The findings suggest that the ethyl acetate and n-butanol extracts, as well as quercetin, have the potential for development as antidiabetic agents.  相似文献

A new method was suggested for estimating the hydrophobic effect of contributions to the Gibbs energies and enthalpies of hydration of hydrocarbons, inorganic gases and rare gases. In accordance with this method the hydrophobic effect contribution to the Gibbs energy was evaluated from the difference between the hydration Gibbs energy of a solute and the non hydrophobic contribution. To estimate the latter value, the known dependence connecting the Gibbs energies of solvation of a solute in a number of aprotic solvents to the Hildebrand solubility parameter for these solvents was used. The non hydrophobic contribution to the Gibbs energy of hydration was calculated for various solutes from such dependences extended to water as solvent. The Hildebrand solubility parameter for water used in the calculation was corrected for the effect of association through hydrogen bonding. This correction was made by subtraction of the water self-association enthalpy from the enthalpy of vaporization of water. The evaluated Gibbs energies of the hydrophobic effect are positive for saturated hydrocarbons, inorganic gases and rare gases and linearly depend on the solute molecular refraction. The hydrophobic contribution to the hydration enthalpies of the solutes was calculated in the same manner as was made to calculate the hydrophobic contribution to Gibbs energies of hydration. Enthalpies of the hydrophobic effect for the solutes under study are negative.  相似文献

The Total Phenolic Content (TPC), antioxidant and antibacterial activities of methanolic extract of Marigold flower (MF) (Tagetes erecta) and Beet root (BR) (Beta vulgaris) were examined. The present work reveals that MF contained greater amount of TPC (42.5 mg/g GAE) as compared to BR (39.4 mg/g GAE). Methanolic extract of MF exhibited excellent DPPH free radical scavenging power (IC50 0.0716 mg/mL) and reducing power at 1 mg/mL concentration. Similar results have been obtained in FTC and TBA method. The results of antibacterial test indicated that the methanolic extract of MF and BR is significantly effective against both type of Gram-negative (Escherichia coli, Shigella dysenteriae) and Gram-positive (Bacillus subtilis, Staphylococcus aureus) bacterial strains. Therefore, the present study suggests that the Marigold and BR are promising source of herbal medicinal products with noteworthy antioxidant and antibacterial activity.  相似文献

We aimed to establish a phytochemical analysis of the crude extracts and performed GC-MS of the essential oils (EOs) of Eugenia uniflora L. (Myrtaceae) and Asteraceae species Baccharis dracunculifolia DC, Matricaria chamomilla L. and Vernonia polyanthes Less, as well as determining their antimicrobial activity. Establishment of the minimal inhibitory concentrations of the crude extracts and EOs against 16 Staphylococcus aureus and 16 Escherichia coli strains from human specimens was carried out using the dilution method in Mueller-Hinton agar. Some phenolic compounds with antimicrobial properties were established, and all EOs had a higher antimicrobial activity than the extracts. Matricaria chamomilla extract and E. uniflora EO were efficient against S. aureus strains, while E. uniflora and V. polyanthes extracts and V. polyanthes EO showed the best antimicrobial activity against E. coli strains. Staphylococcus aureus strains were more susceptible to the tested plant products than E. coli, but all natural products promoted antimicrobial growth inhibition.  相似文献
