As determined by both 1H NMR and UV/Vis spectroscopic titration, ESI‐MS, isothermal titration calorimetry, and DFT molecular modeling, advanced glycation end products (AGE) breaker alagebrium (ALA) formed 1:1 guest–host inclusion complexes with cucurbit[7]uril (CB[7]), with a binding affinity, Ka, in the order of magnitude of 105 m ?1, thermodynamically driven by both enthalpy (ΔH=?6.79 kcal mol?1) and entropy (TΔS=1.21 kcal mol?1). For the first time, a dramatic inhibition of keto–enol tautomerism of the carbonyl α‐hydrogen of ALA has been observed, as evidenced by over an order of magnitude decrease of both the first step rate constant, k1, and the second step rate constant, k2, during hydrogen/deuterium exchange in D2O. Meanwhile, as expected, the reactivity of C2‐hydrogen was also inhibited significantly, with an upshift of 2.09 pKa units. This discovery will not only provide an emerging host molecule to modulate keto–enol tautomerism, but also potentially lead to a novel supramolecular formulation of AGE‐breaker ALA for improved stability and therapeutic efficacy.  相似文献

The effect of the macrocyclic host, cucurbit[7]uril (CB7), on the photophysical properties of the 2‐(2′‐hydroxyphenyl)benzimidazole (HPBI) dye have been investigated in aqueous solution by using ground‐state absorption and steady‐state and time‐resolved fluorescence measurements. All three prototropic forms of the dye (cationic, neutral, and anionic) form inclusion complexes with CB7, with the largest binding constant found for the cationic form (K≈2.4×106 M ?1). At pH≈4, the appearance of a blue emission band upon excitation of the HPBI cation in the presence of CB7 indicates that encapsulation into the CB7 cavity retards the deprotonation process of the excited cation, and hence reduces its subsequent conversion to the keto form. Excitation of the neutral form (pH≈8.5), however, leads to an increase in the keto form fluorescence, indicating an enhanced excited‐state intramolecular proton‐transfer process for the encapsulated dye. In both the ground and excited states, the two pKa values of the HPBI dye show upward shifts in the presence of CB7. The prototropic equilibrium of the CB7‐complexed dye is represented by a six‐state model, and the pH‐dependent changes in the binding constants have been analyzed accordingly. It has been observed that the calculated pKa values using this six‐state model match well with the values obtained experimentally. The changes in the pKa values in the presence of CB7 have been corroborated with the modulation of the proton‐transfer process of the dye within the host cavity.  相似文献

A novel side‐chain polypseudorotaxanes P4VBVBu/CB[7] was synthesized from poly‐Nn‐butyl‐N′‐(4‐vinylbenzyl)‐4,4′‐bipyridinium bromide chloride (P4VBVBu) and cucurbit [7]uril (CB[7]) in water by simple stirring at room temperature. CB[7] beads are localized on viologen units in side chains of polypseudorotaxanes as shown by 1H NMR, IR, XRD, and UV–vis studies, and it is considered that the hydrophobic and charge‐dipole interactions are the driving forces. TGA data show that thermal stability of the polypseudorotaxanes increases with the adding of CB[7] threaded. DLS data show that P4VBVBu and CB[7] could form polypseudorotaxanes, and the average hydrodynamic radius of the polypseudorotaxanes increases with increasing the concentration of CB[7]. The typical cyclic voltammograms indicate that the oxidation reduction characteristic of P4VBVBu is remarkably affected by the addition of CB[7] because of the formation of polypseudorotaxanes and the shielding effects of CB[7] threaded on the viologen units of polypseudorotaxanes. With the increase of the concentration of KBr or K2SO4, the formation of the polypseudorotaxanes was inhibited due to the shielding effects of both Br? or SO to viologen ion and K+ to CB[7] by UV–vis. © 2010 Wiley Periodicals, Inc. A stereoregular 2‐amino‐glycan composed of a mannosamine residue was prepared by ring‐opening polymerization of anhydro sugars. Two different monomers, 1,6‐anhydro‐2‐azido‐mannose derivative ( 3 ) and 1,6‐anhydro‐2‐(N, N‐dibenzylamino)‐mannose derivative ( 6 ), were synthesized and polymerized. Although 3 gave merely oligomers, 6 was promptly polymerized into high polymers of the number‐average molecular weight (Mn) of 2.3 × 104 to 2.9 × 104 with 1,6‐α stereoregularity. The differences of polymerizability of 3 and 6 from those of the corresponding glucose homologs were discussed. It was found that an N‐benzyl group is exceedingly suitable for protecting an amino group in the polymerization of anhydro sugars of a mannosamine type. The simultaneous removal of O‐ and N‐benzyl groups of the resulting polymers was achieved by using sodium in liquid ammonia to produce the first 2‐amino‐glycan, poly‐(1→6)‐α‐D ‐mannosamine, having high molecular weight through ring‐opening polymerization of anhydro sugars.© 2012 Wiley Periodicals, Inc. Catalytic enantioselective α‐fluorination reactions of carbonyl compounds are among the most powerful and efficient synthetic methods for constructing optically active α‐fluorinated carbonyl compounds. Nevertheless, α‐fluorination of α‐nonbranched carboxylic acid derivatives is still a big challenge because of relatively high pKa values of their α‐hydrogen atoms and difficulty of subsequent synthetic transformation without epimerization. Herein we show that chiral copper(II) complexes of 3‐(2‐naphthyl)‐l ‐alanine‐derived amides are highly effective catalysts for the enantio‐ and site‐selective α‐fluorination of N‐(α‐arylacetyl) and N‐(α‐alkylacetyl) 3,5‐dimethylpyrazoles. The substrate scope of the transformation is very broad (25 examples including a quaternary α‐fluorinated α‐amino acid derivative). α‐Fluorinated products were converted into the corresponding esters, secondary amides, tertiary amides, ketones, and alcohols with almost no epimerization in high yield.  相似文献

Five cationic complexes of the general formula [Cp′2Ti(A)2]2+ [Cl?]2 [Cp′ = η5‐(CH3)C5H4 and A = glycine, 1 ; 2‐methylalanine, 2 ; N‐methylglycine, 3 ; L ‐alanine, 4 ; and D ‐alanine 5 ] were prepared by the reaction of Cp′2TiCl2 and the appropriate α‐amino acid in 1:2 molar ratio from methanol–water solution in high yield. Air‐stable crystalline solids, highly soluble in water, were characterized by means of elemental analysis, IR, Raman, 1H, 13C and 14N NMR spectroscopy. The structure of compound 3 was determined by single crystal X‐ray crystallography: orthorhombic Pbca No. 61, a = 9.5310(3), b = 18.2980(5), c = 26.6350(5) Å, V = 4654 Å3, Z = 8. Hydrolytic stability of all compounds in D2O was investigated using 1H NMR spectroscopy within the pD interval of 2.9–6.5. All compounds slowly decomposed during 24 h at pD = 2.94, forming a mixture of hydrolytic products [Cp′2Ti(A)(D2O)]2+, [Cp′2Ti(D2O)2]2+ and respective α‐amino acids. By elevating pD to 4.0 and up to 6.5, a yellowish precipitate was formed, which indicates decomposition of the complexes. These compounds were characterized using elemental analyses, IR and Raman spectroscopy and attributed to oligomeric and/or polymeric structures described empirically by the formula Ti(Cp′)xOy(OH)z (x = 0.65; y = 0.3, z = 1.9). The reaction of 2,4‐pentanedione ( 1 ) with (R)‐(—)‐2‐phenylglycine methyl ester ( 2 ), (R)‐(—)‐2‐phenylglycinol ( 3 ) and the proteinogenic amino acids (2S,3R)‐(—)‐2‐amino‐3‐hydroxybutyric acid (L ‐threonine) ( 4 ) and (R)‐(—)‐2‐amino‐3‐mercaptopropionic acid (L ‐cysteine) ( 5 ) methyl esters was investigated. The corresponding enamines 6 , 7 and 8 were isolated and characterized spectroscopically whereas 9 , which is unstable, was transformed in situ into 13 . Treatment of 7 , 8 and 9 with boron trifluoride etherate afforded the new [1,4]oxazepines 10 , 11 and [1,4]thiazepine ( 12 ) as their BF3O? salts. The structures of the enamines and their corresponding seven‐membered heterocycles were assessed by 1D and 2D NMR spectroscopy. The modulation of the hierarchical nucleated self‐assembly of tri‐β3‐peptides has been studied. β3‐Tyrosine provided a handle to control the assembly process through host‐guest interactions with CB[7] and CB[8]. By varying the cavity size from CB[7] to CB[8] distinct phases of assembling tri‐β3‐peptides were arrested. Given the limited size of the CB[7] cavity, only one aromatic β3‐tyrosine can be simultaneously hosted and, hence, CB[7] was primarily acting as an inhibitor of self‐assembly. In strong contrast, the larger CB[8] can form a ternary complex with two aromatic amino acids and hence CB[8] was acting primarily as cross‐linker of multiple fibers and promoting the formation of larger aggregates. General insights on modulating supramolecular assembly can lead to new ways to introduce functionality in supramolecular polymers.  相似文献

Nucleobase‐anion glycosylation of 2‐[(2‐methyl‐1‐oxopropyl)amino]imidazo[1,2‐a]‐1,3,5‐triazin‐4(8H)‐one ( 6 ) with 3,5‐di‐O‐benzoyl‐2‐deoxy‐2‐fluoro‐α‐D ‐arabinofuranosyl bromide ( 8 ) furnishes a mixture of the benzoyl‐protected anomeric 2‐amino‐8‐(2‐deoxy‐2‐fluoro‐D ‐arabinofuranosyl)imidazo[1,2‐a]‐1,3,5‐triazin‐4(8H)‐ones 9 / 10 in a ratio of ca. 1 : 1. After deprotection, the inseparable anomeric mixture 3 / 4 was silylated. The obtained 5‐O‐[(1,1‐dimethylethyl)diphenylsilyl] derivatives 11 and 12 were separated and desilylated affording the nucleoside 3 and its α‐D anomer 4 . Similar to 2′‐deoxy‐2′‐fluoroarabinoguanosine, the conformation of the sugar moiety is shifted from S towards N by the fluoro substituent in arabino configuration.  相似文献

The X‐ray analyses of 2,3,4,6‐tetra‐O‐acetyl‐α‐d ‐glucopyranosyl fluoride, C14H19FO9, (I), and the corresponding maltose derivative 2,3,4,6‐tetra‐O‐acetyl‐α‐d ‐glucopyranosyl‐(1→4)‐2,3,6‐tri‐O‐acetyl‐α‐d ‐glucopyranosyl fluoride, C26H35FO17, (II), are reported. These add to the series of published α‐glycosyl halide structures; those of the peracetylated α‐glucosyl chloride [James & Hall (1969). Acta Cryst. A 25 , S196] and bromide [Takai, Watanabe, Hayashi & Watanabe (1976). Bull. Fac. Eng. Hokkaido Univ. 79 , 101–109] have been reported already. In our structures, which have been determined at 140 K, the glycopyranosyl ring appears in a regular 4C1 chair conformation with all the substituents, except for the anomeric fluoride (which adopts an axial orientation), in equatorial positions. The observed bond lengths are consistent with a strong anomeric effect, viz. the C1—O5 (carbohydrate numbering) bond lengths are 1.381 (2) and 1.381 (3) Å in (I) and (II), respectively, both significantly shorter than the C5—O5 bond lengths, viz. 1.448 (2) Å in (I) and 1.444 (3) Å in (II).  相似文献

Polyol Metal Complexes.471) Crystalline D ‐Mannose‐Copper Complexes from Fehling Solutions Blue, unstable crystals of K3[Cu5(β‐D ‐Manp)4H—13] · α‐D ‐Manp · 16.5 H2O ( 1 ), which contain a pentanuclear cupric complex of the reducing sugar D ‐mannose in its β‐pyranose form (β‐D ‐Manp), have been obtained from ice‐cold aqueous alkaline solutions. The homoleptic pentacuprate contains bridging mannopyranose ligands, which are charged 4— and 2.5—. Addition of ethylenediamine (en) to such Fehling solutions yields N, N′‐Bis(β‐D ‐mannopyranosyl)‐ethylenediamine (L) as a condensation product of the diamine and mannopyranose. Crystals of [(en)2Cu7(β‐D ‐Manp1, 2, 3, 4H—4)2(L2, 3, 4H—3)2] · 26.6 H2O ( 2 ) could be isolated. The heptanuclear cupric complex is a structural derivative of the homoleptic mannose complex.  相似文献

As part of our studies on the structure of yeast tRNAfMet, we investigated the incorporation of N‐{[9‐(β‐D ‐ribofuranosyl)‐9H‐purin‐6‐yl]carbamoyl}‐L ‐threonine (t6A) in the loop of a RNA 17‐mer hairpin. The carboxylic function of the L ‐threonine moiety of t6A was protected with a 2‐(4‐nitrophenyl)ethyl group, and a (tert‐butyl)dimethylsilyl group was used for the protection of its secondary OH group. The 2′‐OH function of the standard ribonucleotide building blocks was protected with a [(triisopropylsilyl)oxy]methyl group. Removal of the base‐labile protecting groups of the final RNA with 1,8‐diazabicyclo[5.4.0]undec‐7‐ene (DBU) and then with MeNH2 was done under carefully controlled conditions to prevent hydrolysis of the carbamate function, leading to loss of the L ‐threonine moiety.  相似文献

The α‐arylation between cyanoacetate and 2‐(2‐bromophenyl)‐1H‐perimidine took place first catalyzed by CuI/L‐proline in the presence of NaHCO3, and then the amino on perimidine chemoselectively attacked the cyano group to undergo a nucleophilic addition, giving a series of 13‐aminoisoquinolino[2,1‐a]perimidine‐12‐carboxylate derivatives in good yields.  相似文献

The endofungal metabolite epicoccamide D was synthesised in eighteen steps and 17 % yield as the first member of the family of natural glycotetramic acids. The modular character of the synthesis opens access also to analogues featuring different sugars and spacers. It comprises several high‐yielding key steps. The β‐D ‐mannosyl group was introduced by using an α‐D ‐glucosyl imidate donor with subsequent oxidative‐reductive epimerisation at C‐2′. The pyrrolidine ring was closed quantitatively by a Lacey‐Dieckmann condensation of an N‐(β‐ketoacyl)‐N‐methyl alaninate. The resulting 3‐[ω‐(β‐D ‐mannosyl)octadec‐2‐enoyl]tetramic acid was hydrogenated in the presence of the rhodium catalyst (R,R)‐[Rh(Et‐DUPHOS)][BF4] to establish the (7S)‐stereocentre. This was possible only after blocking the acyltetramic acid as a BF2‐chelate to prevent capture of the metal catalyst. We also assigned the hitherto unknown configuration of the natural product as being 5S,7S by comparison of its 13C NMR spectroscopic and optical rotation data with those of our two synthetic 5S,7R/S‐diasteromers.  相似文献

The construction of supramolecular systems in aqueous media is still a great challenge owing to the limited sources of building blocks. In this study, a series of 4‐aryl‐N‐methylpyridinium derivatives have been synthesized. They formed very stable host–guest (1:2) complexes with CB[8] in water (binding constants up to 1014 M ?2) with the two guest molecules arranged in a head‐to‐tail manner and the complexes showed high thermostability, which was revealed by 1H NMR and UV/Vis spectroscopic studies, ITC, and crystallographic analysis.  相似文献

Complex formation between N‐butylboronic acid and D ‐(+)‐glucose, D ‐(+)‐mannose, methyl‐α‐D ‐glucopyranoside, methyl‐β‐D ‐galactopyranoside and methyl α‐D ‐mannopyranoside under neutral conditions was investigated by 1H, 13C and 11B NMR spectroscopy and gas chromatography–mass spectrometry (GC–MS) D ‐(+)‐Glucose and D ‐(+)‐mannose formed complexes where the boronates are attached to the 1,2:4,6‐ and 2,3:5,6‐positions of the furanose forms, respectively. On the other hand, the boronic acid binds to the 4,6‐positions of the two methyl derivatives of glucose and galactose. Methyl α‐D ‐mannopyranoside binds two boronates at the 2,3:4,6‐positions. 11B NMR was used to show the ring size of the complexed sugars and the boronate. GC–MS confirmed the assignments. Molecular clip 1 remains monomeric in water and engages in host–guest recognition processes with suitable guests. We report the Ka values for 32 1? guest complexes measured by 1H NMR, UV/Vis, and fluorescence titrations. The cavity of 1 is shaped by aromatic surfaces of negative electrostatic potential and therefore displays high affinity and selectivity for planar and cationic aromatic guests that distinguishes it from CB[n] receptors that prefer aliphatic over aromatic guests. Electrostatic effects play a dominant role in the recognition process whereby ion–dipole interactions may occur between ammonium ions and the C=O groups of 1 , between the SO3? groups of 1 and pendant cationic groups on the guest, and within the cavity of 1 by cation–π interactions. Host 1 displays a high affinity toward dicationic guests with large planar aromatic surfaces (e.g. naphthalene diimide NDI+ and perylene diimide PDI+) and cationic dyes derived from acridine (e.g. methylene blue and azure A). The critical importance of cation–π interactions was ascertained by a comparison of analogous neutral and cationic guests (e.g. methylene violet vs. methylene blue; quinoline vs. N‐methylquinolinium; acridine vs. N‐methylacridinium; neutral red vs. neutral red H+) the affinities of which differ by up to 380‐fold. We demonstrate that the high affinity of 1 toward methylene blue (Ka=3.92×107 m ?1; Kd=25 nm ) allows for the selective sequestration and destaining of U87 cells stained with methylene blue.  相似文献

Two water‐soluble 6‐(pyrazin‐2‐yl)‐1,3,5‐triazine‐2,4‐diamino (pzta)‐based Cu(II) complexes, namely [Cu(l ‐Val)(pzta)(H2O)]ClO4 ( 1 ) and [Cu(l ‐Thr)(pzta)(H2O)]ClO4 ( 2 ) (l ‐Val: l ‐valinate; l ‐Thr: l ‐threoninate), were synthesized and characterized using elemental analyses, molar conductance measurements, spectroscopic methods and single‐crystal X‐ray diffraction. The results indicated that the molecular structures of the complexes are five‐coordinated and show a distorted square‐pyramidal geometry, in which the central copper ions are coordinated to N,N atoms of pzta and N,O atoms of amino acids. The interactions of the complexes with DNA were investigated using electronic absorption, competitive fluorescence titration, circular dichroism and viscosity measurements. These studies confirmed that the complexes bind to DNA through a groove binding mode with certain affinities (Kb = 4.71 × 103 and 1.98 × 103 M?1 for 1 and 2 , respectively). The human serum albumin (HSA) binding properties of the complexes were also evaluated using fluorescence and synchronous fluorescence spectroscopies, indicating that the complexes could quench the intrinsic fluorescence of HSA in a static quenching process. The relevant thermodynamic parameters revealed the involvement of van der Waals forces and hydrogen bonds in the formation of complex–HSA systems. Finally, molecular docking technology was also used to further verify the interactions of the complexes with DNA/HSA.  相似文献

Ethylene–vinyl alcohol copolymers, EVOH, with two different vinyl alcohol compositions have been functionalized with carboxylic acid groups by reaction with phthalic anhydride. Later on, the coupling reaction of three aminosaccharides (D ‐(+)‐glucosamine, D ‐(+)‐galactosamine, and D ‐(+)‐mannosamine) to functionalized EVOH copolymers has been carried out in dimethyl sulfoxide at 70 °C to achieve water soluble glycopolymers. The structure of the resulting functionalized copolymers and the new glycopolymers was confirmed by 1H and 13C NMR. Likewise, the thermal behavior of glycopolymers has been performed by differential scanning calorimetry and thermal gravimetric analysis. In addition, their affinity to lectins, specifically to Concanavalin A and Ricinus Communis Agglutinin, has been evaluated. © Wiley Periodicals, Inc. A diversity of new 7 ‐substituted[1,2,4]triazolo[1,5‐a]pyrimidine and 6‐substituted[1,2,4]triazolo[1,5‐a]pyrimidine‐7‐amine derivatives has been synthesized via reaction of 3‐amino‐[1,2,4]triazole with enaminonitriles and enaminones. The regio orientation and the structure of the products were confirmed by spectral and analytical data and synthesis via an alternative route. The procedure proved to be simple, efficient, and high yielding, and diversities of [1,2,4]triazolo[1,5‐a]pyrimidines were obtained.  相似文献
