Color variants of Ca2+‐regulated photoprotein obelin were shown to be an important tool for dual‐analyte binding assay. To provide site‐directed conjugation with biospecific molecules, several obelin color mutants carrying unique cysteine residues were obtained and characterized for their novel properties. A pair of obelins Y138F,A5C and W92F,H22E,D12C was found to be most suitable (in terms of high bioluminescent activity and stability) as reporters in simultaneous assay of two targets in a sample. Availability of SH‐groups, accessible for chemical modification, essentially simplifies the synthesis of biospecific conjugates, increases their yield and conserves obelins' bioluminescence activity. Conjugates with immunoglobulin and oligonucleotide were produced and successfully applied in single nucleotide polymorphism genotyping.  相似文献

The active ingredient of ecstasy, N-methyl-3,4-methyldioxyphenylisopropylamine (MDMA) can be manufactured by a number of easy routes from simple precursors. We have synthesised 45 samples of MDMA following the five most common routes using N-precursors from 12 different origins and three different precursors for the aromatic moiety. The 13C and 15N contents of both the precursors and the MDMA samples derived therefrom were measured by isotope ratio mass spectrometry coupled to an elemental analyser (EA-IRMS). We show that within-pathway correlation between the 15N content of the precursor and that of the derived MDMA can be strong but that no general pattern of correlation can be defined. Rather, it is evident that the δ15N values of MDMA are strongly influenced by a combination of the δ15N values of the source of nitrogen used, the route by which the MDMA is synthesised, and the experimental conditions employed. Multivariate analysis (PCA) based on the δ15N values of the synthetic MDMA and of the δ15N and δ13C values of the N-precursors leads to good discrimination between the majority of the reaction conditions tested.  相似文献

In order to evaluate alternative analytical methodologies to study the geographical origin of ciders, both multi-elemental analysis and Sr isotope abundance ratios in combination with multivariate statistical analysis were estimated in 67 samples from England, Switzerland, France and two Spanish regions (Asturias and the Basque Country). A methodology for the precise and accurate determination of the 87Sr/86Sr isotope abundance ratio in ciders by multicollector inductively coupled plasma mass spectrometry (MC-ICP-MS) was developed. Major elements (Na, K, Ca and Mg) were measured by ICP-AES and minor and trace elements (Li, Be, B, Al, Sc, Ti, V, Cr, Mn, Fe, Co, Ni, Cu, Zn, Ga, As, Se, Rb, Sr, Y, Mo, Cd, Sn, Sb, Cs, Ba, La, Ce, W, Tl, Pb, Bi, Th and U) were measured by ICP-MS using a collision cell instrument operated in multitune mode. An analysis of variance (ANOVA test) indicated that group means for B, Cr, Fe, Ni, Cu, Se, Cd, Cs, Ce, W, Pb, Bi and U did not show any significant differences at the 95% confidence level, so these elements were rejected for further statistical analysis. Another group of elements (Li, Be, Sc, Co, Ga, Y, Sn, Sb, La, Tl, Th) was removed from the data set because concentrations were close to the limits of detection for many samples. Therefore, the remaining elements (Na, Mg, Al, K, Ca, Ti, V, Mn, Zn, As, Rb, Sr, Mo, Ba) together with 87Sr/86Sr isotope abundance ratio were considered for principal component analysis (PCA) and linear discriminant analysis (LDA). Finally, LDA was able to classify correctly 100% of cider samples coming from different Spanish regions, France, England and Switzerland when considering Na, Mg, Al, K, Ca, Ti, V, Mn, Zn, As, Rb, Sr, Mo, Ba and 87Sr/86Sr isotope abundance ratio as original variables.  相似文献

Studying ecosystem processes in the context of carbon cycling and climate change has never been more important. Stable carbon isotope studies of gas exchange within terrestrial ecosystems are commonly undertaken to determine sources and rates of carbon cycling. To this end, septum‐capped vials (‘Exetainers’) are often used to store samples of CO2 prior to mass spectrometric analysis. To evaluate the performance of such vials for preserving the isotopic integrity (δ13C) and concentration of stored CO2 we performed a rigorous suite of tests. Septum‐capped vials were filled with standard gases of varying CO2 concentrations (~700 to 4000 ppm), δ13C values (approx. ?26.5 to +1.8‰V‐PDB) and pressures (33 and 67% above ambient), and analysed after a storage period of between 7 and 28 days. The vials performed well, with the vast majority of both isotope and CO2 concentration results falling within the analytical uncertainty of chamber standard gas values. Although the study supports the use of septum‐capped vials for storing samples prior to mass spectrometric analysis, it does highlight the need to ensure that sampling chamber construction is robust (air‐tight). The application of isotope ratio methods in authenticity and traceability relies on the accuracy and robustness of the methodology employed. An unexpected source of error has now been identified, which can introduce major and variable inaccuracies into the determination of site-specific isotope ratio measurement by quantitative 13C NMR spectrometry if not correctly controlled. This is the isotope chemical shift effect, which comes into play when hydrogen atoms in the target molecule enter into exchange with deuterated water present at trace levels in the deuterated solvent used as the frequency lock. Even at a level of contamination as low as 0.02%, an error of 5‰ can be introduced, fivefold the required accuracy of 1‰. How to avoid this source of error is discussed.  相似文献

1,2,3,4,6‐Penta‐O‐acetyl‐α‐d ‐glucopyranose and the corresponding [1‐2H], [2‐2H], [3‐2H], [4‐2H], [5‐2H], and [6,6‐2H2]‐labeled compounds were prepared for measuring deuterium/hydrogen‐induced effects on 13C chemical shift nΔ (DHIECS) values. A conformational analysis of the nondeuterated compound was achieved using density functional theory (DFT) molecular models that allowed calculation of several structural properties as well as Boltzmann‐averaged 13C NMR chemical shifts by using the gauge‐including atomic orbital method. Ni2+‐complexed poly(2‐acetamidoacrylic acid) (PAAA) hydrogel beads were developed for the site‐specific reversible immobilization and purification of the histidine‐tagged green fluorescent protein (His‐tagged GFP). PAAA hydrogel beads were prepared by photopolymerization, and significantly improved mechanical properties of PAAA hydrogel beads were observed in comparison with PAAA hydrogel from our previous study. Confocal laser scanning microscopy was used to determine the binding of His‐tagged GFP to the hydrogel beads in three‐dimensional space. Photoluminescence spectroscopy revealed 89% of binding efficiency of His‐tagged GFP to the Ni2+‐PAAA hydrogel beads, 51% of yielding recovery. The maximum binding capacity of His‐tagged GFP was estimated to be 0.45 µg/mg of Ni2+‐PAAA hydrogel beads. The recombinant His‐tagged GFP from the soluble fraction of E. coli BL21(DE3) cell lysates was purified with Ni2+‐PAAA hydrogel beads. The major advantage of the Ni2+‐PAAA hydrogel beads system was simple preparation procedures of producing the matrix, because PAAA hydrogel beads had relatively enhanced mechanical strength than soft hydrogels. Herein, we describe an environmentally benign and cost‐effective protocol for the synthesis of valuable bifuranyl dicarboxylates, starting with α‐bromination of readily accessible furan‐2‐carboxylates by LiBr and K2S2O8. Furthermore, the bromination intermediate product 5‐bromofuran‐2‐carboxylates were then conducted in a palladium‐catalyzed reductive homocoupling reactions in the presence of alcohols to afford bifuranyl dicarboxylates. One of the final products in this protocol, [2,2’]bifuran‐5,5’‐dicarboxylic acid esters, are essential monomers of poly(ethylene bifuranoate), which can be served as an green and versatile alternative polymer for traditional poly(ethylene terephthalate) that is currently common in technical plastics.  相似文献

Novel dual molecular‐ and ion‐recognition responsive poly(N‐isopropylacrylamide‐co‐benzo‐12‐crown‐4‐acrylamide) (PNB12C4) linear copolymers with benzo‐12‐crown‐4 (B12C4) as both guest and host units are prepared. The copolymers exhibit highly selective sensitivities toward γ‐cyclodextrin (γ‐CD) and Na+. The presence of γ‐CD induces the lower critical solution temperature (LCST) of PNB12C4 copolymer to shift to a higher value due to the formation of 1:1 γ‐CD/B12C4 host‐guest inclusion complexes, while Na+ causes a negative shift in LCST due to the formation of 2:1 “sandwich” B12C4/Na+ host‐guest complexes. Regardless of the complexation order, when γ‐CD and Na+ coexist with PNB12C4, competitive complexation actions of B12C4 as both guest and host units toward γ‐CD and Na+ finally form equilibrium 2:2:1 γ‐CD/B12C4/Na+ composite complexes, and the final LCST values of PNB12C4 copolymer reach almost the same level. The results provide valuable guidance for designing and applying PNB12C4‐based smart materials in various applications.
