Neopterin is a valuable biomarker of cellular immunity associated with various pathological situations such as viral and bacterial infections, autoimmune, cardiovascular, neurodegenerative and malignant disorders. To produce specific antibodies against neopterin for a rapid multi-biomarker-based diagnosis, a novel hapten derivative was synthesized and attached to carrier proteins. The thoroughly characterized conjugates were used for immunization of BALB/c mice and rabbits. The produced monoclonal antibody reached in both direct and indirect enzyme-linked immunosorbent assay (ELISA) format LoD of 0.18 and 0.45 μg L−1, respectively, and was a superior immunoreagent for further biosensor developments with regard to assay sensitivity and material availability. The best polyclonal antibody was somewhat more sensitive in direct ELISA with LoD of 0.05 μg L−1. The optimized ELISA method was evaluated with blood samples collected from patients with renal insufficiency, patients with sepsis, patients without confirmed clinical diagnosis, and healthy volunteers. In plasma samples, neopterin concentrations ranging from 3.2 to 103 μg L−1 could be determined with the monoclonal ELISA whereas twofold lower results were obtained with the polyclonal ELISA. A satisfactory correlation of results was found between the polyclonal ELISA and IBL Neopterin ELISA kit within the concentration range 0.5–16 μg L−1 (R = 0.874; n = 40), and slightly lower correlation was found for monoclonal-based ELISA (R = 0.819; n = 40). These data show that the generated antibodies may be used as functional analytical reagents for the integration into multianalyte biochip detection systems.  相似文献

A piezoelectric quartz crystal (PQC) sensor based on a molecularly imprinted polymer (MIP) has been developed for enantioselective and quantitative analysis of d-(+)-methamphetamine (d(+)-MA). The sensor was produced by bulk polymerization and the resulting MIP was then coated on the gold electrode of an AT-cut quartz crystal. Conditions such as volume of polymer coating, curing time, type of PQC, baseline solvent, pH, and buffer type were found to affect the sensor response and were therefore optimized. The PQC-MIP gave a stable response to different concentrations of d(+)-MA standard solutions (response time = 10 to 100 s) with good repeatability (RSD = 0.03 to 3.09%; n = 3), good reproducibility (RSD = 3.55%; n = 5), and good reversibility (RSD = 0.36%; n = 3). The linear range of the sensor covered five orders of magnitude of analyte concentration, ranging from 10−5 to 10−1 μg mL−1, and the limit of detection was calculated as 11.9 pg d(+)-MA mL−1 . The sensor had a highly enantioselective response to d(+)-MA compared with its response to l(−)-MA, racemic MA, and phentermine. The developed sensor was validated by applying it to human urine samples from drug-free individuals spiked with standard d(+)-MA and from a confirmed MA user. Use of the standard addition method (SAM) and samples spiked with d(+)-MA at levels ranging from 1 × 10−3 to 1 × 10−2 μg mL−1 showed recovery was good (95.3 to 110.9%).  相似文献

An anodic stripping voltammetric procedure for the determination of Cu(II) at an in situ-plated stannum film electrode (SnFE) was described. The results indicated that the SnFE had an attractive electroanalytical performance, with two distinct voltammetric stripping signals for copper and stannum, and showed the superior advantage for the determination of copper compared with the bismuth film electrode. Several experimental parameters were optimized. The SnFE exhibited highly linear behavior in the concentration range from 1.0 to 100.0 μg L−1 of Cu(II) (r = 0.994) with the detection limit of 0.61 μg L−1 (S/N = 3), and the relative standard deviation for a solution containing 40.0 μg L−1 Cu(II) was 2.2% (n = 8). The procedure has been successfully applied for the determination of Cu(II) in lake water sample.  相似文献

A sensitive and effective method for simultaneous determination of triazolopyrimidine sulfonamide herbicide residues in soil, water, and wheat was developed using ultra-performance liquid chromatography coupled with tandem mass spectrometry. The four herbicides (pyroxsulam, flumetsulam, metosulam, and diclosulam) were cleaned up with an off-line C18 SPE cartridge and detected by tandem mass spectrometry using an electrospray ionization source in positive mode (ESI+). The determination of the target compounds was achieved in <2.0 min. The limits of detection were below 1 μg kg−1, while the limits of quantification did not exceed 3 μg kg−1 in different matrices. Quantitation was determined from calibration curves of standards containing 0.05–100 μg L−1 with r 2 > 0.997. Recovery studies were conducted at three spiked levels (0.2, 1, and 5 μg kg−1 for water; 5, 10, and 100 μg kg−1 for soil and wheat). The overall average recoveries for this method in water, soil, wheat plants, and seeds at three levels ranged from 75.4% to 106.0%, with relative standard deviations in the range of 2.1–12.5% (n = 5) for all analytes.  相似文献

Nickel(II) and lead(II) ionic imprinted 8-hydroxyquinoline polymers were synthesized by a precipitation polymerization technique and were used as selective solid phase extraction supports for the determination of nickel and lead in seawater by flow injection solid phase extraction on-line inductively coupled plasma-optical emission spectrometry. An optimum loading flow rate of 2.25 mL min−1 for 2 min and an elution flow rate of 2.25 mL min−1 for 1 min gave an enrichment factor of 15 for nickel. However, a low dynamic capacity and/or rate for adsorption and desorption was found for lead ionic imprinted polymer and a flow rate of 3.00 mL min−1 for 4-min loading and a flow rate of 2.25 mL min−1 for 1-min elution gave a enrichment factor of 5. The limit of detection was 0.33 μg L−1 for nickel and 1.88 μg L−1 for lead, with a precision (n = 11) of 8% (2.37 μg Ni L−1) for nickel and 11% (8.38 μg Pb L−1) for lead. Accuracy was also assessed by analyzing SLEW-3 (estuarine water) and TM-24 (lake water) certified reference materials, and the values determined were in good agreement with the certified concentrations.  相似文献

This work reports a comparative study of two automated flow-through fluorosensors for the determination of digoxin in serum samples: an immunosensor with an anti-digoxin polyclonal antibody as the reactive phase permanently immobilised on controlled-pore glass and a sensor with a selective reaction system based on a methacrylic molecularly imprinted polymer (MIP) synthesised by bulk polymerisation. The variables affecting the sensitivity and dynamic range of the sensors (e.g. the carrier and elution solutions, flow rates, pH and reagent concentrations) were optimized, and the binding characteristics of their reactive phases were compared in a competitive fluorescent assay. Digoxin was reproducibly determined by both sensors at the milligram per litre level (detection limit = 1.20 × 10−3 mg L−1 and RSD = 4–7% for the immunosensor; detection limit = 1.7 × 10−5 mg L−1 and RSD = 1–2% for the MIP sensor). No cross-reactivity with digoxin-related compounds was seen for either sensor at a digoxin/interferent ratio of 1:100. The lifetime of the immunosensor was about 50 immunoassays; its shelf life, when unused, is about 3 months. The lifetime of the MIP sensor was over 18 months. Both sensors were used to determine the digoxin concentration of human serum samples with satisfactory results.  相似文献

SiO2/ZrO2/C carbon ceramic material with composition (in wt%) SiO2 = 50, ZrO2 = 20, and C = 30 was prepared by the sol–gel-processing method. A high-resolution transmission electron microscopy image showed that ZrO2 and the graphite particles are well dispersed inside the matrix. The electrical conductivity obtained for the pressed disks of the material was 18 S cm−1, indicating that C particles are also well interconnected inside the solid. An electrode modified with flavin adenine dinucleotide (FAD) prepared by immersing the solid SiO2/ZrO2/C, molded as a pressed disk, inside a FAD solution (1.0 × 10−3 mol L−1) was used to investigate the electrocatalytic reduction of bromate and iodate. The reduction of both ions occurred at a peak potential of −0.41 V vs. the saturated calomel reference electrode. The linear response range (lrr) and detection limit (dl) were: BrO3 , lrr = 4.98 × 10−5–1.23 × 10−3 mol L−1 and dl = 2.33 μmol L−1; IO3 , lrr = 4.98 × 10−5 up to 2.42 × 10−3 and dl = 1.46 μmol L−1 for iodate.  相似文献

The development and characterization of one rat monoclonal antibody (mAb) for 2,4-dinitroaniline and of two rat mAbs for 2,6-dinitroaniline are described. With the immunization of rats with 2,4,6-trinitrophenyl-glycylglycine–keyhole limpet hemocyanine (KLH) conjugate one mAb (PK 5H6) has been developed and formatted into a competitive enzyme-linked immunosorbent assay (ELISA). This assay no. 1 is very sensitive for 2,4-dinitroaniline with a test midpoint of 0.24 ± 0.06 μg L−1 (n = 19) in 40 mM phosphate-buffered saline (PBS). A second hapten, 3-(4-amino-2,6-dinitrophenyl)propionic acid, which was also conjugated to KLH and used for the immunization of rats, led to two sensitive ELISAs for 2,6-dinitroaniline in 40 mM PBS with test midpoints of 0.61 ± 0.08 μg L−1 (n = 15; mAb DNT4 3C6; assay no. 2) and 0.94 ± 0.29 μg L−1 (n = 17; mAb DNT4 1A7, assay no. 3). Selectivities of all mAbs were checked with more than 20 compounds, including nitroaromatic compounds, 2,6-dinitroaniline pesticides, and other substituted derivatives of aniline. As very noticeable cross-reactivities, all mAbs recognize 2-chloro-4,6-dinitroaniline, 4-chloro-2,6-dinitroaniline and 2-bromo-4,6-dinitroaniline, the last of these being a major metabolite of the azo dye Disperse Blue 79. As first demonstrations of applications, two ELISAs (assays no. 1 and 2) were used for the analysis of 2,4- or 2,6-dinitroaniline in spiked water and soil samples. Recovery data were determined and the majority of these data were in the range of 90–120%. These assays can contribute to a very cost-effective and environmentally friendly immunochemical surveillance monitoring of environmental samples for contaminations with these compounds. To the best of the authors’ knowledge, these are the first antibodies described for 2,4-dinitroaniline and for 2,6-dinitroaniline.  相似文献

The possibilities of different media formed by lecithin/n-butanol (n-BuOH)/water ternary mixtures for the analysis of all-trans-retinol by fluorescence have been studied. Fluorescence intensity of retinol increases in the presence of different types of aggregates formed in these media. Analytical features are good, the detection limit and quantification limit have micrograms per liter levels, and the linear range and sensitivity are appropriate to determine retinol in cosmetic samples. The analysis of retinol in anti-wrinkle creams can be achieved directly without any pretreatment of the sample. The vesicles built up from a biocompatible surfactant (lecithin) in aqueous solution with a low amount of n-BuOH permit an appropriated media for a simple, rapid, and sensitive analytical method. This method has a linear range between 64.1 and 800 μg L−1, a sensitivity of 202.3 L mg−1, and a low detection and quantification limit at 19.2 and 64.1 μg L−1, respectively.  相似文献

A simple and efficient method, based on ultrasound-enhanced surfactant-assisted dispersive liquid–liquid microextraction (UESA-DLLME) followed by high-performance liquid chromatography (HPLC) has been developed for extraction and determination of ketoconazole and econazole nitrate in human blood samples. In this method, a common cationic surfactant, cetyltrimethylammonium bromide (CTAB), was used as dispersant. Chloroform (40 μL) as extraction solvent was added rapidly to 5 mL blood containing 0.068 mg mL−1 CTAB. The mixture was then sonicated for 2 min to disperse the organic chloroform phase. After the extraction procedure, the mixture was centrifuged to sediment the organic chloroform phase, which was collected for HPLC analysis. Several conditions, including type and volume of extraction solvent, type and concentration of the surfactant, ultrasound time, extraction temperature, pH, and ionic strength were studied and optimized. Under the optimum conditions, linear calibration curves were obtained in the ranges 4–5000 μg L−1 for ketoconazole and 8–5000 μg L−1 for econazole nitrate, with linear correlation coefficients for both >0.99. The limits of detection (LODs, S/N = 3) and enrichment factors (EFs) were 1.1 and 2.3 μg L−1, and 129 and 140 for ketoconazole and econazole nitrate, respectively. Reproducibility and recovery were good. The method was successfully applied to the determination of ketoconazole and econazole nitrate in human blood samples.  相似文献

A novel, simple, and rapid reversed-phase liquid chromatography–tandem mass spectrometric methodology was developed for the analysis of natamycin in wine samples. Natamycin was protonated to form singly charged ions in an electrospray positive ion mode. Data acquisition under MS/MS was achieved by applying multiple reaction monitoring (MRM) of three fragment ion transitions (666.3 → 648.2, 666.3 → 503.3, and 666.3 → 485.2) to provide a high degree of sensitivity and specificity. Chromatographic separation was performed on a rapid resolution column using a mobile phase consisting of an acetonitrile/water mixture with a total run time of 5.0 min. After only filtration as pretreatment, the sample was injected into the chromatographic system. The proposed method was validated in terms of selectivity, trueness, precision, decision limit (CCα), and detection capability (CCβ) according to 2002/657/EC Commission decision. The values for trueness, reported as bias (%), agreed with those established by the aforementioned document. Repeatability (intraday variability) values were 12.37% at a concentration of 1.0 μg L−1 and 8.99–4.19% at concentrations between 2.5 and 10 μg L−1. The overall within-laboratory (interday variability) reproducibility was 15.47% at a concentration of 1.0 μg L−1, which was significantly lower than the indicative value reported in the EU decision. The results indicated that the proposed approach is a sensitive, fast, reproducible, and robust methodology suitable for the analysis of natamycin levels in wine samples.  相似文献

The worldwide contamination of winery by-products by mycotoxins may present a serious hazard to human and animal health. Mycotoxins are secondary metabolites of fungi with possible adverse effects on humans, animals, and crops that result in illnesses and economic losses. Mycotoxins are under continuous survey in Europe, but the regulatory aspects still need to be set up for winery by-products, which may be used in animal feed. The aim of this study was to implement a simple but reliable analytical methodology for ochratoxin A (OTA) quantification in grape pomaces in order to perform a survey of samples from the Douro Demarcated Region, Portugal. The method involved a unique preparation step, solvent extraction, followed by high-performance liquid chromatography (HPLC) with fluorescence (FL) detection. A comparative study was performed with two extraction solvents (ethyl acetate and methanol) as well as using extraction on an immunoaffinity column. The linearity range for OTA analysis was 0.05–23.5 μg L−1 with a detection limit of 0.05 μg L−1 and a precision (expressed by the coefficient of variation under repeatability conditions) of 0.4–14.7%. The percentage of recovery was on average 23.5 ± 3.6% (extraction with ethyl acetate) or 70.1 ± 2.5% (extraction with 70% methanol). Accounting for the recovery factor and the chromatographic detection limit, as well as the preconcentration factor, the limit of detection in grape pomaces is 0.04 μg kg−1 (ethyl acetate extraction) and 0.33 μg kg−1 (methanol extraction). Samples from 12 out of 13 sites in the Douro Demarcated Region showed OTA presence with concentrations not exceeding 0.4 μg kg−1. A mid-infrared enzymatic assay for label-free monitoring of the enzymatic reaction of fructose-1,6-bisphosphatase with fructose 1,6-bisphosphate has been proposed. The whole procedure was done in an automated way operating in the stopped flow mode by incorporating a temperature-controlled flow cell in a sequential injection manifold. Fourier transform infrared difference spectra were evaluated for kinetic parameters, like the Michaelis–Menten constant (K M) of the enzyme and V max of the reaction. The obtained K M of the reaction was 14 ± 3 g L−1 (41 μM). Furthermore, inhibition by adenosine 5′-monophosphate (AMP) was evaluated, and the K MApp value was determined to be 12 ± 2 g L−1 (35 μM) for 7.5 and 15 μM AMP, respectively, with V max decreasing from 0.1 ± 0.03 to 0.05 ± 0.01 g L−1 min−1. Therefore, AMP exerted a non-competitive inhibition.  相似文献

A novel and environmentally friendly microextraction method, termed ionic liquid dispersive liquid-phase microextraction (IL-DLPME), has been developed for rapid enrichment of triclosan and triclocarban before analysis by high-performance liquid phase chromatography–electrospray tandem mass spectrometry (HPLC–ESI-MS–MS). Instead of using toxic organic solvents, an ionic liquid was used as a green extraction solvent. This also avoided the instability of the suspending drop in single-drop liquid-phase microextraction, and the heating and cooling step in temperature-controlled ionic liquid dispersive liquid phase microextraction. Factors that may affect the enrichment efficiency, for example volume of ionic liquid, type and volume of dispersive solvent, pH, extraction time, and NaCl content were investigated in detail and optimized. Under optimum conditions, linearity of the method was observed over the range 0.2–12 μg L−1 for triclocarban and 1–60 μg L−1 for triclosan with correlation coefficients ranging from 0.9980 to 0.9990, respectively. The sensitivity of the proposed method was found to be excellent, with limits of detection in the range 0.040–0.58 μg L−1 and precision in the range 7.0–8.8% (RSD, n = 5). This method has been successfully used to analyze real environmental water samples and satisfactory results were achieved. Average recoveries of spiked compounds were in the range 70.0–103.5%. All these results indicated that the developed method would be a green method for rapid determination of triclosan and triclocarban at trace levels in environmental water samples.  相似文献

 Simple, rapid, sensitive and selective methods for the determination of Cr(III) and W(VI) with flavonol derivatives in the presence of surface-active agents are proposed. In the pH ranges 3.4–4.2 and 1.9–2.5, the molar absorptivities of Cr(III)-morin-emulsifier S (EFA) and W(VI)-morin-polyvinylpyrrolidone (PVP) systems are 1.13×105 and 2.13×104 L mol−1 cm−1 at 435 and 415 nm, respectively. The Cr(III)-quercetin-PVP and W(VI)-quercetin-cetylpyridinium bromide (CPB) systems are formed in the pH ranges 4–4.6 and 2.2–2.8 with molar absorptivities 1.02×105 and 9.02×104 L. mol−1 cm−1 at 441 and 419 nm, respectively. The linear dynamic ranges for the determination of Cr(III) and W(VI) with morin in the presence of EFA and PVP are 0.03–0.46 and 0.71–8.1 μg mL−1, respectively. The corresponding ranges with quercetin are 0.04–0.54 and 0.14–2.1 μg mL−1 of Cr(III) and W(VI), respectively. The r.s.d (n = 10) for the determination of 0.25 and 3.7 μg mL−1 of Cr(III) and W(VI) with morin and their detection limits are 0.88 and 0.99% and 0.016 and 0.63 μg mL−1, respectively. Using quercetin, the r.s.d (n = 10) for 0.22 and 1.2 μg mL−1 of Cr(III) and W(VI) and their detection limits are 0.92 and 0.91% and 0.015 and 0.08 μg mL−1, respectively. The critical evaluation of the proposed methods is performed by statistical analysis of the experimental data. The proposed methods are applied to determine Cr in steel, non-ferrous alloys, wastewater and mud filtrate and to the determination of W in steel. Received March 8, 1999.  A simple, rapid, selective, sensitive and economical method has been developed for the simultaneous determination of trace amounts of palladium and nickel in aqueous methanolic medium using 2-(2-thiazolylazo)-5-dimethylam inobenzoic acid as an analytical reagent by first derivative spectrophotometr y. Palladium is determined by measuring base to peak distance at λ=695.0 nm while nickel is estimated by zero crossing method in the mixture. The linearity is maintained between 0.12–1.75 μg mL−1 for palladium and 0.07–1.60 μg mL−1 for nickel in the pH range 2.8–7.2 and 3.4–8.8 respectively. Seven replicate determinations of 1.0 μ g mL−1 of palladium and 0.8 μg mL−1 of nickel in a mixture give a mean signal height of 0.391 for Pd and 0.541 for Ni with relative standard deviations of 0.9% and 1.2%, respectively. The sensitivity of the proposed method is 0.391 (dA/dλ)/(μg mL−1) for palladium and 0.685 (dA/dλ)/(μg mL−1) for nickel. This article describes a robust methodology using the combination of instrumental design (high matrix interface—HMI), sample dilution and internal standardization for the quantification of beryllium (Be) in various digested autopsy tissues using inductively coupled plasma mass spectrometry. The applicability of rhodium as a proper internal standard for Be was demonstrated in three types of biological matrices (i.e., femur, hair, lung tissues). Using HMI, it was possible to achieve instrumental detection limits and sensitivity of 0.6 ng L−1 and 157 cps L ng−1, respectively. Resilience to high salt matrices of the HMI setup was also highlighted using bone mimicking solution ([Ca2+] = 26 to 1,400 mg L−1), providing a 14-fold increase in tolerance and a 2.7-fold decrease in method detection limit compared to optimized experimental conditions obtained without the HMI configuration. Precision of the methodology to detect low levels of Be in autopsy samples was demonstrated using hair and blood certified reference materials. Be concentration ranging from 0.015 to 255 μg kg−1 in autopsy samples obtained from the U.S. Transuranium and Uranium Registries were measured using the methodology presented.  相似文献

The electrochemical solid phase micro-extraction of salicylic acid (SA) at graphite-epoxy-composed solid electrode surface was studied by cyclic voltammetry. SA was oxidized electrochemically in pH 12.0 aqueous solution at 0.70 V (vs. saturated calomel electrode) for 7 s. The oxidized product shows two surface-controlled reversible redox couples with two proton transferred in the pH range of 1.0∼6.0 and one proton transferred in the pH range of 10.0∼13.0 and is extracted on the electrode surface with a kinetic Boltzman function of i p = 3.473–4.499/[1 + e(t − 7.332)/6.123] (χ 2 = 0.00285 μA). The anodic peak current of the extracted specie in differential pulse voltammograms is proportional to the concentration of SA with regression equation of i p = −5.913 + 0.4843 c (R = 0.995, SD = 1.6 μA) in the range of 5.00∼200 μM. The detection limit is 5.00 μM with RSD of 1.59% at 60 μM. The method is sensitive and convenient and was applied to the detection of SA in mouse blood samples with satisfactory results.  相似文献

The effect of substrate (glucose) concentration on the stability and yield of a continuous fermentative process that produces hydrogen was studied. Four anaerobic fluidized bed reactors (AFBRs) were operated with a hydraulic retention time (HRT) from 1 to 8 h and an influent glucose concentration from 2 to 25 g L−1. The reactors were inoculated with thermally pre-treated anaerobic sludge and operated at a temperature of 30 °C with an influent pH around 5.5 and an effluent pH of about 3.5. The AFBRs with a HRT of 2 h and a feed strength of 2, 4, and 10 g L−1 showed satisfactory H2 production performance, but the reactor fed with 25 g L−1 of glucose did not. The highest hydrogen yield value was obtained in the reactor with a glucose concentration of 2 g L−1 when it was operated at a HRT of 2 h. The maximum hydrogen production rate value was achieved in the reactor with a HRT of 1 h and a feed strength of 10 g L−1. The AFBRs operated with glucose concentrations of 2 and 4 g L−1 produced greater amounts of acetic and butyric acids, while AFBRs with higher glucose concentrations produced a greater amount of solvents.  相似文献

A fully automated method has been developed for analysis of eighteen antibacterial compounds, including penicillins, cephalosporins and sulfonamides, in animal feed with limits of quantification in the range 0.25–5.79 μg kg−1. The method is based on pressurized liquid extraction of 3 g homogenized feed with water and online clean-up of 500 μL of the extract with C18HD cartridges. The purified sample was directly analysed by liquid chromatography–electrospray tandem mass spectrometry (SPE–LC–ESI-MS–MS). Chromatographic separation was achieved within 10 min by use of a C12 Phenomenex Hydro-RP reversed-phase analytical column and a mobile phase gradient (water + 0.1% formic acid–methanol + 0.1% formic acid). The method was validated, revealing capability for detection of concentrations as low as 0.09 μg kg−1, decision limits (CCα) and detection capabilities (CCβ) in the range 10–174 μg kg−1 and 22–182 μg kg−1, respectively, and inter-day precision ranging from 0.7 to 8.3%. Recovery, with internal standard correction, was in the range 93–134% for all analytes. The method was then applied to analysis of fifteen feed samples, nine of which contained at least one antimicrobial at concentrations between 0.006 and 1.526 mg kg−1. The performance data and results from the method were compared with those from a previous method developed by our group, using offline SPE, by analyzing the same set of samples by both methods. The online SPE approach resulted in slightly improved sensitivity, with LODs of 0.09–2.12 μg kg−1 compared with 0.12–3.94 μg kg−1 by the offline approach. In general, better recovery was achieved by use of online purification (for 72% of the analytes) and the correlation between the two methods was good. The main advantages of the new online method are rapid and automated sample pre-treatment, and reduction of sample manipulation, enabling high-throughput analysis and highly accurate results. Because of all these characteristics, the proposed method is applicable and could be deemed necessary within the field of food control and safety.  相似文献
