New hybrid materials based on Pt, Fe, and Pt–Fe nanoparticles stabilized in a matrix of polymethylsilsesquioxane nanogel and ultrahigh molecular weight polyethylene (UHMWPE) were prepared. Metal vapor synthesis was used to produce mono- and bimetallic nanoparticles. It was shown that organosilicon nanogel effectively stabilizes Pt nanoparticles with an average size of 0.9 nm. Using the nanogel results in the formation of superparamagnetic Fe particles 3–5 nm in size that consist of ferromagnetic Fe0 core and antiferromagnetic shells of Fe oxides. It is established that using an organosilicon matrix in the formation of Pt-Fe/UHMWPE systems helps reduce the average particle size of Fe in the material from 6.5 to 4.5 nm and narrow their particle size distribution. Hyaluronic acid nanogel (HyA‐AT) is a redox sensitive crosslinkable nanogel, obtained through the conjugation of a thiolated hydrophobic molecule to the hyaluronic acid chain. Engineered nanogel was studied for its biocompatibility, including immunocompatibility and hemocompatability. The nanogel did not compromise the metabolic activity or cellular membrane integrity of 3T3, microvascular endothelial cells, and RAW 264.7 cell lines, as determined by the 3‐[4,5‐dimethylthiazol‐2‐yl]‐2,5‐diphenyl tetrazolium bromide and lactate dehydrogenase release assays. Also, we didn't observe any apoptotic effect on these cell lines through the Annexin V‐FITC test. Furthermore, the nanogel cell internalization was analyzed using murine bone marrow derived macrophages, and the in vivo and ex vivo biodistribution of the Cy5.5 labeled nanogel was monitored using a non‐invasive near‐infrared fluorescence imaging system. The HyA‐AT nanogel exhibits fairly a long half‐live in the blood stream, thus showing potential for drug delivery applications.
Protein pharmaceuticals show great therapeutic promise, but effective intracellular delivery remains challenging. To address the need for efficient protein transduction systems, we used a magnetic nanogel chaperone (MC): a hybrid of a polysaccharide nanogel, a protein carrier with molecular chaperone‐like properties, and iron oxide nanoparticles, enabling magnetically guided delivery. The MC complexed with model proteins, such as BSA and insulin, and was not cytotoxic. Cargo proteins were delivered to the target HeLa cell cytosol using a magnetic field to promote movement of the protein complex toward the cells. Delivery was confirmed by fluorescence microscopy and flow cytometry. Delivered β‐galactosidase, inactive within the MC complex, became enzymatically active within cells to convert a prodrug. Thus, cargo proteins were released from MC complexes through exchange interactions with cytosolic proteins. The MC is a promising tool for realizing the therapeutic potential of proteins. 相似文献
Flexible multivalent 3D nanosystems that can deform and adapt onto the virus surface via specific ligand–receptor multivalent interactions can efficiently block virus adhesion onto the cell. We here report on the synthesis of a 250 nm sized flexible sialylated nanogel that adapts onto the influenza A virus (IAV) surface via multivalent binding of its sialic acid (SA) residues with hemagglutinin spike proteins on the virus surface. We could demonstrate that the high flexibility of sialylated nanogel improves IAV inhibition by 400 times as compared to a rigid sialylated nanogel in the hemagglutination inhibition assay. The flexible sialylated nanogel efficiently inhibits the influenza A/X31 (H3N2) infection with IC50 values in low picomolar concentrations and also blocks the virus entry into MDCK‐II cells. 相似文献
