The (2,3)J(CH) dependence on dihedral angle (theta H--C--C--X) for cyclopentane derivatives was investigated. We observed that the combined use of experimentally obtained (2,3)J(CH) values and the theoretically determined dihedral angles between the corresponding nuclei can be used to infer the relative stereochemistry of the ring substituents in cyclopentane derivatives. There is a good correlation between the magnitude of (3)J(CH) and the dihedral angle between the hydrogen and the coupled carbon (R2 = 0.88).  相似文献

A modified version of CPMG-HSQMBC pulse scheme is presented for the measurement of long-range heteronuclear coupling constants. The method implements adiabatic inversion and refocusing pulses on the heteronucleus. Low-power composite 180° XY-16 CPMG pulse train is applied on both proton and X nuclei during the evolution of long-range couplings to eliminate phase distortions due to co-evolution of homonuclear proton-proton couplings. The pulse sequence yields pure absorption antiphase multiplets allowing precise and direct measurement of the (n)J(XH) coupling constants regardless from the size of the proton-proton couplings. The applicability of the method is demonstrated using strychnine as a model compound. The selective 1D version of the method is also presented.  相似文献

Four‐ and five‐bond heteronuclear J‐couplings between the hydrogen H‐8 and carbons C‐6 and C‐2 in a series of 7‐ and 9‐benzyl substituted purine derivaties with variuous substituents in positions 2 and 6 were studied by coupled 13 C NMR and H,C‐HMBC experiments and by DFT calculations. We have found that for some of the derivatives, the five‐bond coupling H8‐C2 is higher than the four‐bond H8‐C6 coupling, which is also evidenced by a stronger crosspeak in the HMBC. This finding contradicts the generally accepted opinion that only strong three‐bond crosspeaks and one weak four‐bond H8‐C6 crosspeak can be observed in the HMBC spectra of purine derivatives. The misinterpretation of HMBC spectra may lead to an incorrect determination of the purine derivatives' structure. Copyright © 2012 John Wiley & Sons, Ltd.  相似文献

Regioselective halogen/metal exchange reactions were carried out on a series of 3-substituted- 1,2-dibromoarenes. Product mixtures were quenched with CO2 to form the corresponding benzoic acid analogs to facilitate HPLC and NMR analysis. Substitution at the 3-position could readily be assigned on the basis of 2D HMBC long-range correlations, while assignment at the 2-position was not as straightforward. The use of three-bond J(CH) coupling constant measurements, extracted from 1-D 1H coupled 13C experiments, were necessary to render unequivocal regio assignments.  相似文献

Conformational study of 2-(2-pyrrolyl)pyridine and 2,6-di(2-pyrrolyl)pyridine was performed on the basis of the experimental measurements and high-level ab initio calculations of the one-bond 13C-13C, 13C-1H and 15N-1H spin-spin coupling constants showing marked stereochemical behavior upon the internal rotation around the pyrrole-pyridine interheterocyclic bonds. Both compounds were established to adopt predominant s-cis conformations with no noticeable out-of-plane deviations.  相似文献

An efficient pulse sequence for measuring long‐range C? H coupling constants (JC? H) named selective J‐resolved HMBC has been developed by replacing a 1H 180° pulse with a selective 1H 180° pulse and the HMBC pulse scheme with the constant time (CT) HMBC employed in the J‐resolved HMBC pulse sequence that we reported previously. The novel pulse sequence providing only long‐range JC? H cross peaks for easy and accurate analysis enables to overcome disadvantages of the previous HMBC‐based pulse sequences (J‐resolved HMBC‐1) along with maintaining high sensitivity. The efficiency of measuring long‐range JC? H using the proposed pulse sequence has been demonstrated in applications to the complicated natural products, portmicin and monazomycin. New spin-state-selective (S3) NMR pulse sequences exclusively applying cross-polarization schemes to achieve optimum homonuclear and heteronuclear 1H-X coherence transfer are reported for the simple and accurate measurement of the magnitude and sign of heteronuclear coupling constants for samples at natural abundance. The proposed spin-edited HCP-TOCSY experiments are based on clean heteronuclear S3 excitation, generated by simultaneous co-addition of two independent in-phase and anti-phase components created during the mixing heteronuclear J-cross-polarization (HCP) step, which is finally transferred to other protons by a conventional homonuclear TOCSY mechanism. Selective 1D and non-selective 2D approaches for the easy determination of long-range proton-carbon and proton-nitrogen coupling constants on any protonated and non-protonated heteronuclei are presented and discussed for several organic molecules.  相似文献

Unambiguous resonance assignments of diastereotopic CH2 protons in the anomeric side chain of nine alkyl‐ and aralkylselenoglycosides have been carried out on the basis of experimental CPMG‐HSQMBC measurements and theoretical second order polarization propagator approach (SOPPA) calculations of geminal 77Se‐1H spin‐spin coupling constants involving diastereotopic pro‐R and pro‐S protons. Theoretical conformational analyses have been performed at the MP2/6‐311G** level. The conformational space of each of the selenoglycosides under study could be adequately described as a mixture of six interconverting conformers with the molar fractions depending on the nature of the side chain substituent at the selenium atom. The good agreement observed between measured and the weighted conformational averaged values of the calculated coupling constants provides a basis for reliable diastereotopic assignments in this type of carbohydrate structures. In this contribution, we show that the magnitude of heteronuclear long‐range coupling constants can be directly extracted from the classical 1D HMBC spectra, as all multiplet lines of a cross‐peak always and exclusively vanish for the condition Δ = k/nJCH. To the best of our knowledge, this feature of the classical HMBC has not yet been noticed and exploited. This condition holds true, irrespective of the magnitude and numbers of additional active and passive homonuclear nJHH′ couplings. Alternatively, the nJCH value may also be evaluated by fitting the peak's intensity in the individual spectra to its simple sin(πnJCHΔ)exp(−Δ/T2eff) dependence. Compared to the previously proposed J‐HMBC sequences that also use the variation of the cross‐peak's intensity for extracting the coupling constants, the classical HMBC pulse sequence is significantly more sensitive.  相似文献

Seventy-three unique spin-spin coupling constants have been analyzed for the ten species in the two series X(CH3)nH(4-n), where the central atom X is 13C or 15N+. Thirty-seven experimental values have been obtained from the literature, and several new coupling constants have been measured for the methyl-substituted ammonium ions. Both DFT with the B3LYP functional and ab initio EOM-CCSD calculations have been carried out on these same systems. Coupling constants computed by these two methods are in agreement with experimental values. Some problems related to coupling constants for the cationic ammonium systems have been resolved when these were recomputed at EOM-CCSD for complexes in which NH4+ is hydrogen-bonded to H2O molecules.  相似文献

Carbon-carbon spin-spin coupling constants were calculated at the SOPPA level for a series of seven classical spiroalkanes, spiro[2.2]pentane, spiro[2.3]hexane, spiro[2.4]heptane, spiro[2.5]octane, spiro[3.3]heptane, spiro[4.4]nonane and spiro[5.5]undecane, with special focus upon couplings involving and/or across spiro carbons. Many interesting structural trends were investigated originating in specific geometries and unusual bonding environments at the spiro carbon.  相似文献

We present calculations of indirect nuclear spin-spin coupling constants in large molecular systems, performed using density functional theory. Such calculations, which have become possible because of the use of linear-scaling techniques in the evaluation of the Coulomb and exchange-correlation contributions to the electronic energy, allow us to study indirect spin-spin couplings in molecules of biological interest, without having to construct artificial model systems. In addition to presenting a statistical analysis of the large number of short-range coupling constants in large molecular systems, we analyse the asymptotic dependence of the indirect nuclear spin-spin coupling constants on the internuclear separation. In particular, we demonstrate that, in a sufficiently large one-electron basis set, the indirect spin-spin coupling constants become proportional to the inverse cube of the internuclear separation, even though the diamagnetic and paramagnetic spin-orbit contributions to the spin-spin coupling constants separately decay as the inverse square of this separation. By contrast, the triplet Fermi contact and spin-dipole contributions to the indirect spin-spin coupling constants decay exponentially and as the inverse cube of the internuclear separation, respectively. Thus, whereas short-range indirect spin-spin coupling constants are usually dominated by the Fermi contact contribution, long-range coupling constants are always dominated by the negative diamagnetic spin-orbit contribution and by the positive paramagnetic spin-orbit contribution, with small spin-dipole and negligible Fermi contact contributions.  相似文献

The SELective INverse detection of carbon–proton CORrelation pulse sequence that yields a 1D spectrum of a proton directly bonded to a selected carbon resonance has been converted into a proton and carbon double‐selective variant that provides a 1H spectrum of a selected proton that is long‐range coupled to a specific carbon resonance. The resulting 1D proton multiplet exhibits a pure absorptive in‐phase lineshape for precise measurement of specific long‐range proton–carbon coupling constants in small organic molecules at natural abundance. An application of 3D version of HSQC-TOCSY experiment for the measurement of heteronuclear coupling constants of organic compounds yielding complex spectra is proposed. The approach presented here is based on the optimized random sampling of the evolution time space followed by Multidimensional Fourier Transform (MFT). In this study, we show that the interpretation of omega(3)-(13)C coupled (1)H-(13)C 3D HSQC-TOCSY spectra with E.COSY-type multiplets allows one to evaluate heteronuclear coupling constants of strychnine with high accuracy, whereas the employment of 2D methods is associated with signal overlap and use of conventionally recorded 3D NMR spectra cannot provide accurate results in an overnight experiment.  相似文献

The measurement of the magnitude and sign of 2J(C,H) couplings offers a reliable way to determine the absolute configuration at a carbon center in a fixed cyclic system. A decrease of the dihedral angle ? in the O—CA—CB—H fragment always leads to a change of the 2J(CA,HB) coupling to more negative values, independent of the type and position of substituents at the two carbon centers. The orientations of the two substituents at C‐3 of the epimeric pair 1 and 2 were determined unambiguously through the measurement of the geminal coupling constants between C‐3 and the hydrogen atoms at C‐2 and C‐4. In particular, 2J(C‐3,H‐2ax) with ?1.5 Hz, ? = 174° in 1 and ?6.6 Hz, ? = 47° in 2 , and 2J(C‐3,H‐4) with +1.5 Hz, ? = 175° in 1 and ?4.7 Hz, ? = 49° in 2 showed the greatest differences between the two epimers. Both couplings therefore allow the determination of the absolute configuration at C‐3. It should be noted, however, that the size of the coupling constants can be different for dihedral angles of nearly identical size, when there are different numbers of electronegative substituents on the two coupling pathways, i.e. no O‐substituent at C‐2, but one axial O‐substituent at C‐4. It becomes clear that it is not sufficient to measure the magnitude of 2J coupling constants only, but that the sign of the geminal coupling is needed to identify the absolute configuration at a chiral center. The coupling of C‐3 with H‐2eq is not useful for the determination of the configuration at C‐3, as the similarity of the dihedral angles ? (O—C‐3—C‐2—H‐2eq) (57° in 1 and 70° in 2 ) leads to identical coupling constants (?6.1 Hz) for both epimers. Copyright © 2003 John Wiley & Sons, Ltd.  相似文献

A comprehensive theoretical study of nine classical caged polycycloalkanes (tetrahedrane, prismane, homoprismane, quadricyclane, cubane, pentaprismane, hexaprismane, adamantane and diamantane) was carried out with special focus on the structural behavior of their J(C,C) values calculated at the SOPPA level. The structural behavior of J(C,C) in small carbocages is dominated by steric strain whereas in medium-sized polycycloalkanes the J(C,C) values show no marked peculiarities and follow several well-defined structural trends typical of other alicyclic compounds.  相似文献

The 243 coupling constants of eight N-R-pyrazoles [R=H, CH3, C6H5, COCH3, NH2, NO2, SO2CF3, Si(CH3)3] have been calculated and compared with 131 experimental values. The agreement is good and can be used to estimate new couplings. The whole collection has been statistically analyzed.  相似文献
