A sensitive fluorescence liquid chromatographic analytical method was developed for the simultaneous determination of carnosine enantiomers in rat plasma. The method was applied to pharmacokinetic studies. Chiral separation of carnosine enantiomers was achieved by pre-column derivatization with o-phthaldialdehyde and the thiol N-acety-l-cysteine as derivating reagents. They were separated on an ODS column and detected by fluorescence detection (λex = 350 nm, λem = 450 nm). γ-Aminobutyric acid was used as internal standard. The method was linear up to 6,000 ng mL?1 for l-carnosine, 4,000 ng mL?1 for d-carnosine. Low limit of quantitation (LLOQ) was 40 ng mL?1 for each isomer. The relative standard deviations obtained for intra- and inter-day precision were lower than 12% and the recoveries were higher than 75% for both enantiomers. The method was applied to a stereoselective study on the pharmacokinetics of carnosine after oral administration with a single dose (carnosine, 75 mg kg?1 for each isomer) to a rat. The initial data indicated that l-carnosine had a larger value of the highest plasma concentration than d-carnosine (C max 5,344 vs. 1,914 ng mL?1), and that of l-carnosine had a lower value of AUC(0?∞) and t 1/2(h) (AUC(0?∞) 5,306 vs. 6,321 ng h mL?1, t 1/2 1.43 vs. 3.37 h). Our results indicated that the pharmacokinetic of l-carnosine and d-carnosine revealed enantioselective properties significantly.  相似文献

A simple, rapid and sensitive column liquid chromatographic method was developed and validated to measure simultaneously the amount of ascorbic acid and phenolic acids at single wavelength (240 nm) in order to assess drug release profiles and drug-excipients compatibility studies for a new sustained release tablet formulation and its subsequent stability studies. A combined isocratic and linear gradient reversed-phase LC method was carried out at 240 nm. Quantification was achieved with reference to the external standards. The linearity for concentrations between 0.042 and 0.150 mg mL?1 for ascorbic acid, 0.084–0.250 mg mL?1 for chlorogenic acid, 0.053–0.360 mg mL?1 for caffeic acid, and 0.016–0.250 mg mL?1 for ferulic acid (r > 0.99 for all analytes) were established. The recovery of the active ingredients from the samples was at the range of 92.3–102.9%. Intra- and inter-day precisions were less than 2.5%. The limits of detection and quantification were 8 and 24 μg mL?1 for ascorbic acid, 18 and 54 μg mL?1 for chlorogenic acid, 37 and 112 μg mL?1 for caffeic acid, and 11 and 34 μg mL?1 for ferulic acid. The determination of the four active ingredients was not interfered by the excipients of the products. Samples were stable in the release mediums (37 °C) at least for 12 h.  相似文献

A rapid and simple high-performance liquid chromatographic tandem mass spectrometric method has been developed and validated for analysis of strictosamide in rat plasma. Chromatographic separation was achieved on a C18 column by gradient elution with mixtures of methanol, water, and acetonitrile containing 0.05% acetic acid. Digoxin was used as internal standard. Selected reaction monitoring (SRM) was used for MS quantitation. Linearity was good in the range 0.05–20 ng mL?1 in rat plasma. The lower limit of quantitation was 0.04 ng mL?1. The method is precise and reliable and can be applied to pharmacokinetic studies.  相似文献

The objective of this study was to develop a simple, efficient and reliable method for routine quantitative analysis for Psoralea corylifolia L. An ultra performance liquid chromatography with DAD detector system was employed for simultaneous quantification of ten compounds. The chromatographic analysis was performed by UPLC with C18 column and gradient elution of 0.05% formic acid aqueous solution and acetonitrile in 16 min. All calibration curves were linear (R 2 ≥ 0.9990) over the tested ranges. The LOD and LOQ were lower to 13.07 and 39.22 ng mL?1 with 2 μL of injection volume, respectively. The intra- and inter-day precisions as determined from sample solutions were below 4.1 and 4.2%. The average recoveries were ranged from 94.2 to 108.8% with RSDs ≤ 4.6%. This validated method was applied for the analysis of ten analytes in P. corylifolia L. from different origins. The variation of the content of ten compounds was remarkable among the tested samples: psoralenoside increased from 7.42 to 17.04 mg g?1, isopsoralenoside from 6.05 to 14.34 mg g?1, psoralen from 2.37 to 3.90 mg g?1, isopsoralen from 2.53 to 3.65 mg g?1, neobavaisoflavone from 1.59 to 2.96 mg g?1, bavachin from 1.02 to 2.35 mg g?1, psoralidin from 0.45 to 1.91 mg g?1, isobavachalcone from 1.33 to 4.71 mg g?1, corylifol A from 1.02 to 2.40 mg g?1 and bakuchiol from 28.10 to 63.89 mg g?1.  相似文献

A sensitive and accurate liquid chromatographic method for the determination of AR-42 enantiomeric purity has been developed and validated. Baseline separation with a resolution higher than 1.9 was accomplished within 10 min using a CHIRALPAK AD column (250 mm × 4.6 mm; particle size 5 μm) and n-hexane/2-propanol/diethylamine (75:25:0.1 v/v/v) as mobile phase at a flow rate of 1 mL min?1. Eluted analytes were monitored by UV absorption at 260 nm. The effects of mobile phase components, temperature and flow rate on enantiomeric selectivity and resolution of enantiomers were investigated. Calibration curves were plotted within the concentration range between 0.001 and 0.5 mg mL?1 (n = 10), and the recoveries between 98.23 and 101.87% were obtained, with relative standard deviation lower than 1.31%. Limit of detection and limit of quantitation for AR-42 were 0.39 and 1.28 μg mL?1 and for its enantiomer were 0.36 and 1.19 μg mL?1, respectively. It was demonstrated that the developed method was accurate, robust and sensitive for the determination of enantiomeric purity of AR-42, especially for the analysis of bulk samples.  相似文献

A stability-indicating LC method was developed for the simultaneous determination of ibuprofen and diphenhydramine citrate in pharmaceutical dosage forms. The chromatographic separation was achieved on an Inertsil ODS 3V, 150 × 4.6 mm, 5 μm, column. The mobile phase contained a mixture of 50 mM potassium dihydrogen phosphate buffer:acetonitrile:triethylamine:glacial acetic acid (55:45:0.2:0.2, v/v/v/v). This method allowed the determination of 2.85–9.14 mg mL?1 of ibuprofen and 0.54–1.73 mg mL?1 of diphenhydramine citrate, in a diluent consisting of pH 7.2, 50 mM potassium dihydrogen phosphate buffer:acetonitrile (40:60, v/v). The flow rate was 1.2 mL min?1 and the detection wavelength was 260 nm. The limit of detection for ibuprofen and diphenhydramine citrate was 1.72 and 0.54 μg mL?1 and the limit of quantification was 5.73 and 1.64 μg mL?1, respectively. This method was validated for accuracy, precision and linearity. The method was also found to be stability indicating.  相似文献

A new, sensitive and stability indicating liquid chromatographic method has been developed for the determination of imatinib mesylate (IM). Efficient chromatographic separation was achieved using a C18 column with simple mobile phase combination delivered in an isocratic mode and quantitation was carried out using ultraviolet detection. For the first time, a novel microwave assisted degradation procedure was employed for stress testing studies. In addition, orthogonal separation technique was applied to demonstrate selectivity of the proposed method. The method has demonstrated excellent linearity over the range of 25–1,600 ng mL?1. Moreover, the method was found to be sensitive with a low limit of detection (3.35 ng mL?1) and limit of quantitation (10.16 ng mL?1). The method has shown good and consistent recoveries (99.35–100.69%) with low intra- and inter-day relative standard deviation (RSD) (<2.5%). Experimental design confirmed that peak area was unaffected by small changes in critical factors, in robustness study. The validated method was successfully applied for determination of IM in pharmaceutical formulations.  相似文献

A simple, rapid, and stability-indicating reversed-phase high-performance liquid chromatographic (LC) method for analysis for dutasteride has been successfully developed. Chromatography was performed on a 150 mm × 4.6 mm C18 column with acetonitrile–water 60:40 (v/v) as isocratic mobile phase at 1.0 mL min?1. Ultraviolet detection of dutasteride was at 210 nm. Its retention time was approximately 10 min and its peak was symmetrical. Response was a linear function of concentration over the range 0.2–1 μg mL?1 (R 2 = 0.997) and the limits of detection and quantitation were was 0.05 and 0.10 μg mL?1, respectively. The method was validated for linearity, precision, repeatability, sensitivity, and selectivity. Selectivity was validated by subjecting dutasteride stock solution to photolytic, acidic, basic, oxidative, and thermal degradation. The peaks from the degradation products did not interfere with that from dutasteride. The method was used to quantify dutasteride in pharmaceutical preparations.  相似文献

A simple, sensitive, precise and accurate reversed phase liquid chromatographic method has been developed for the simultaneous estimation of atorvastatin (AT) calcium, ramipril (RA) and aspirin (AS) from capsule dosage form. The method was developed using a Phenomenex Luna C18 (250 mm, 4.6 mm i.d., 5 µm) column with a mobile phase consisting of 0.1%, orthophosphoric acid buffer:acetonitrile:methanol (45:50:5 v/v/v), pH 3.3, at a flow rate of 1 mL min?1. Detection was carried out with ultra-violet detection at 210 nm. The retention times were about 12.19, 2.35, and 3.95 min for AT calcium, RA and AS, respectively. The developed method was validated for linearity, accuracy, precision, limit of detection, limit of quantitation and robustness. The linearity ranges were 1–6 µg mL?1 for AT calcium, 0.5–3 µg mL?1 for RA and 7.5–45 µg mL?1 for AS with mean recoveries of 100.59 ± 0.68, 100.62 ± 0.83 and 100.49 ± 0.73% for AT calcium, RA and AS, respectively. Limit of detection obtained were 29.85 ng mL?1 for AT calcium, 4.71 ng mL?1 for RA and 85.13 ng mL?1 for AS. Impurity of salicylic acid was found in capsule dosage form at the retention time of about 4.84 min. The proposed method can be used for the estimation of these drugs in combined dosage forms.  相似文献

A rapid and sensitive method for determination of methyl nonyl ketone in rat plasma was developed based on GC–MS. The analyte and internal standard, propyl p-hydroxybenaoate, were extracted from plasma with methyl tert-butyl ether and then separated by an HP-5MS capillary analytical column (30 m × 0.25 mm, 0.25 µm) and determined by a quadrupole mass spectrometer detector operated under EI ionization and selected ion monitoring mode. Excellent linearity was found to be from 5.0 to 1,000 ng mL?1 with a lower limit of quantitation of 5.0 ng mL?1. The accuracy was between 96.0 and 106.3%, and the intra- and inter-day precision and accuracy values were found to be within the assay variability criteria limits according to the FDA guidelines. The developed method was successfully applied to the pharmacokinetic study of methyl nonyl ketone, a typical component in the traditional Chinese medicine Houttuynia cordata injection, in rats after a single oral dose of 100 mg kg?1 and peritoneal injection dose of 5 mg kg?1, respectively.  相似文献

This work reports the first investigation of Remersonia thermophila hemicellulosic hydrolytic enzyme production, with subsequent purification of an extracellular endo-β-1,4-xylanase (RtXyl) and its application in bread making. The research describes RtXyl purification from sorghum-induced submerged liquid cultures of this moderately thermophilic, aerobic, ascomycete fungus. The purified enzyme is a single subunit protein with a molecular mass of 42 kDa and exhibits glycosyl hydrolase family-10-like activity over a broad pH and temperature range. Optimal activity was measured at pH 6.0 and 65 °C respectively, which is suitable for bread making applications. Substrate specificity studies revealed that RtXyl is purely xylanolytic with no side-activities against other plant polysaccharides. The RtXyl catalytic efficiency (K cat/K m) was highest with oats spelt xylan (810.90 mg mL?1 s?1), wheat arabinoxylan (809.52 mg mL?1 s?1) and beechwood xylan (417.40 mg mL?1 s?1) with less efficiency towards insoluble oats spelt xylan (236.40 mg mL?1 s?1). Hydrolysis products analysed by thin layer chromatography yielded a range of xylosaccharides, predominantly xylotriose and xylobiose. RtXyl application in a basic wheat bread recipe at low dosages (0.297 XU/g) showed its suitability to increase loaf volume by 8.0 % compared with the control bread. RtXyl increased loaf softness by 19.6 % while reducing bread staling by 20.4 % up to 4 days of storage.  相似文献

A rapid, sensitive and specific reversed-phase liquid chromatographic method was developed and validated for the quantification of p-hydroxyphenethyl anisate (HPA), which is one of the main constituents of Notopterygium Radix (underground parts of Notopterygium incisum and N. forbesii), in rat plasma, and study its pharmacokinetics after the intravenous administration of 40 mg kg?1 HPA to rats. The method involves a plasma clear-up step using liquid–liquid extraction by ethyl acetate, followed by RP-LC separation and detection. Separation of HPA was performed on an analytical Diamonsil ODS C18 column equipped with a Dikma ODS C18 EasyGuard column using a mobile phase consisting of MeOH–H2O (75:25, v/v) at a flow-rate of 1.0 mL min?1. The UV detection was performed at a wavelength of 256 nm. The linear calibration curves were obtained in the concentration range of 0.05–5.0 μg mL?1 (r = 0.9992, n = 5) in rat plasma with the lower limit of detection of 0.01 μg mL?1 and the lower limit of quantification of 0.04 μg mL?1, and the extraction recovery of HPA was calculated to be the range of 82.01–86.66%. The intra- and inter-day precisions in terms of % relative standard deviation were lower than 2.33 and 3.99% in rat plasma, respectively, with accuracies ranging from 91.22 to 110.5%. The developed method was suitable for the determination and pharmacokinetic study of HPA in rat plasma.  相似文献

A sensitive, specific and rapid high-performance liquid chromatography method was developed in an effort to quantify extremely low curcuminoid levels for the future transdermal experiments where the curcuminoids are incorporated with excipients such as microemulsion, liposomes, and micelles. The chromatographic separation was performed using a Symmetry® C18, 250 × 4.6 mm, 5-μm column, with a mobile phase composed of 5 mM acetonitrile:phosphoric acid (45:55, v/v) at a flow rate of 1.0 mL min?1, it was sensitive with a low limit of quantitation for curcuminoids (0.626 ng mL?1 for curcumin) and good linearity (r 2 ≥ 0.999) over the range 1–100 ng mL?1. All the validation data, such as accuracy and precision, were within the required limits from the ICH guideline. The assay method was successfully applied during forced degradation of curcuminoid solutions. The method retained its accuracy and precision when the standard addition technique was applied.  相似文献

A rapid, sensitive and accurate high performance liquid chromatography method using tandem mass spectrometry detection for hydralazine in BALB/C mouse plasma and brain was developed and validated. The method involved a derivatization with 2,4-pentanedione at 50 °C for 1 h, and a step of solid phase extraction to purify and concentrate hydralazine derivative. Chromatographic separation was carried out on an Agilent ZORBAX SB-C18 column by elution with methanol?C0.01 mol L?1 ammonium acetate (60:40, v/v). The multiple reaction monitoring transition used for quantification was m/z 225.2 ?? 129.5 in the electrospray positive ionization mode. Good linearity was obtained over the concentration range of 10?C200 ng mL?1. The limits of detection were 0.49 and 1.05 ng mL?1 for hydralazine in mouse plasma and brain, respectively. The limits of quantitation were 1.5 and 3.18 ng mL?1 for hydralazine in mouse plasma and brain, respectively. Sample analysis time was 6 min including sample separation. The method was successfully applied to a pharmacokinetic study following intraperitoneal injection of hydralazine in BALB/C mice at the dose of 20 mg kg?1.  相似文献

A simple and rapid HPLC method using phenacetin (PHN) as internal standard has been developed for simultaneous determination of acetaminophen, caffeine, and chlorphenamine maleate in the product compound paracetamol and chlorphenamine maleate granules. Separation and quantitation were achieved on a 250 mm × 4.6 mm, 5 μm particle, C18 column. The mobile phase was methanol 0.05 mol L?1 aqueous KH2PO4 solution, 45:55 (v/v), containing 0.1% triethylamine and adjusted to pH 3.6 by addition of phosphoric acid; the flow rate was 1.0 mL min?1. Detection of all compounds was by UV absorbance at 260 nm and elution of the analytes was achieved in less than 12 min. The linearity, accuracy, and precision of the method were acceptable to good over the concentration ranges 6.4–153.6 μg mL?1 for acetaminophen, 5.0–120.0 μg mL?1 for caffeine, and 9.6–230.4 μg mL?1 for chlorphenamine maleate.  相似文献

The aim of this research was to develop a sensitive liquid chromatographic–electrospray ionization–mass spectrometric (LC–MS) method for direct measurement of the concentration of Atorvastatin in human plasma. Plasma samples (1 mL) were extracted with 3 mL ethyl acetate, and by a simple reversed-phase chromatography. Pitavastatin was used as internal standard (IS). The LOQ was 0.25 ng mL?1 (RSD 4.24%). The assay was linear from 0.25–20 ng mL?1. And the correlation coefficient for the calibration regression line was 0.9996 or better. Intra-day and inter-day accuracy were better than 15%. The method has been successfully used for a pharmacokinetic study with human subjects. A two-period crossover designed bioequivalence research was also progressed in healthy Chinese volunteers. Among the pharmacokinetic data obtained, T max was 1.36 ± 0.68 h for reference formulation and 0.81 ± 0.54 h for test formulation. C max was 8.54 ± 5.06 ng mL?1 for reference formulation and 9.54 ± 3.68 ng mL?1 for test formulation. t 1/2 was 8.50 ± 2.74 h for reference formulation and 9.24 ± 3.17 h for test formulation. AUC 0?48h was 54.77 ± 21.82 h ng mL?1 for reference formulation and 55.66 ± 20.91 h ng mL?1 for test formulation. The method was successfully applied to the study of pharmacokinetics of Atorvastatin in healthy Chinese volunteers.  相似文献

A specific and accurate high-performance liquid chromatographic method for analysis of cinnamic acid (CA) and paeonol (PN) in rat plasma has been developed and validated. Plasma samples were pretreated by protein precipitation with methanol, and the supernatant was injected for reversed-phase separation on a C18 column with acetonitrile–0.1% phosphoric acid 24:76 (v/v) as mobile phase at a flow-rate of 1.0 mL min?1. Phenylbutyric acid was used as the internal standard. Good linear relationships were obtained between response and concentration in the range 0.130–52.0 μg mL?1 (r = 0.9980) and 0.1785–71.4 μg mL?1 (r = 0.9950) for CA and PN, respectively. Intra-day and inter-day assay precision (RSD, n = 6) at three concentrations was not above 15.1% for either CA and PN, and accuracy was from 94.3 to 104.7% and from 103.3 to 113.1% for CA and PN, respectively. Mean recovery of CA and PN from plasma samples was 87.5 and 86.8%, respectively, and recovery of the internal standard at a concentration of 1.00 mg mL?1 was 88.5%. Results from a stability study suggested CA and PN were stable under the experimental conditions used. Finally, the validated method was successfully applied to a pharmacokinetic study of CA and PN in rats after intragastric administration of Guizhi–Fuling capsule. The results obtained would be very useful for evaluation of the clinical efficacy of GFC.  相似文献

A liquid chromatography–electrospray ionization tandem mass spectrometry method has been developed to perform the determination of 5-hydroxytryptamine (5-HT), norepinephrine (NE), dopamine (DA) and their metabolites, i.e., 5-hydroxyindole-3-acetic acid (5-HIAA), 4-hydroxy-3-methoxyphenylglycol (MHPG) sulfate, 3,4-dihydroxyphenylacetic acid (DOPAC) and homovanillic acid (HVA) in rat brain tissue. Analytes were separated on a Thermo C18 column (4.6 mm × 250 mm, 5 μm, SN: 1245575T, Thermo electron corporation, USA) with a mobile phase of 0.05% formic acid/acetonitrile (92:8 for ESI+, 82:18 for ESI?, v/v) at the flow-rate of 0.8 mL min?1. The LC system was coupled to a Waters Micromass Quattro Premier XE tandem quadruple mass spectrometer. MS acquisition of 5-HT, NE and DA was performed in positive electrospray ionization multiple reaction monitoring (MRM) mode, while negative electrospray ionization MRM mode was used to monitor their metabolites. The calibration curves were linear within the concentration range of 4–4,450 ng mL?1 for 5-HT, 4–4,110 ng mL?1 for NE and 4–4,100 ng mL?1 for DA (≥ 0.999). The limit of quantitation was 4 ng mL?1. 5-HIAA, MHPG, DOPAC and HVA have good linearity within the range of 12–1,000 ng mL?1(≥ 0.998) and the limit of quantitation was 12 ng mL?1. The intra- and inter-day RSD were lower than 8.45%. The method is sensitive, fast, accurate and usable for quantity determination of monoamine neurotransmitters and their metabolites in neuropsychiatric diseases.  相似文献

A rapid and selective liquid chromatographic/tandem mass spectrometric method for the determination of JBP485 was developed and validated. Following protein precipitation, the analyte and internal standard (JBP923) were separated from human plasma using an isocratic mobile phase on an Elite Kromasil C18 column. An API 3200 tandem mass spectrometer equipped with a Turbo ionSpray ionization source was used as the detector and operated in the positive ion mode. Multiple reaction monitoring using the precursor to product ion combinations of m/z 201.2 ?? 86.2 and m/z 219.2 ?? 86.2 was performed to quantify JBP485 and JBP923, respectively. The method was linear in the concentration range of 0.10?C50.00 ??g mL?1 using 100 ??L of plasma. The lower limit of quantification was 0.10 ??g mL?1. The intra- and inter-day relative standard deviations over the entire concentration range were less than 6.65%. Accuracy determined at three concentrations (0.25, 4.00 and 25.00 ??g mL?1 for JBP485) ranged from ?0.78 to 2.74% in terms of relative error. Each plasma sample was chromatographed within 2.0 min. The method was successfully applied to characterize the pharmacokinetic profiles of JBP485 in rats after an intravenous injection of 6.25 mg kg?1 JBP485.  相似文献

An accurate LC method was developed and validated for simultaneous determination of irinotecan (CPT-11) and its active metabolite SN-38 in rat plasma. Plasma samples were pretreated with 0.4 g mL?1 sodium dodecyl sulfate to inactive the carboxylesterase and avoid the conversion of CPT-11 to SN-38. Chromatographic separation was achieved on a Diamaonsil C18 column using acetonitrile–50 mM phosphate buffered solution (30:70, v/v) at pH 4.0 as the mobile phase with the flow rate of 1 mL min?1. The linear quantitation ranges for CPT-11 and SN-38 were 5.05–3,030 and 3.15–315 ng mL?1 with r 2 > 0.99, respectively. The lower limit of quantification (LLOQ) was 2.33 ng mL?1 for CPT-11 and 0.26 ng mL?1 for SN-38 with intra- and inter-day relative standard deviation of <12% and the accuracy values of >90%. The method was proved to be accurate and sensitive enough and was successfully applied to a pharmacokinetic study of CPT-11 in rats.  相似文献
