In this study, biological properties of the essential oil isolated from seeds of Foeniculum vulgare (F. vulgare) were evaluated. GC-MS analysis revealed Trans-Anethole (80.63%), L-Fenchone (11.57%), Estragole (3.67%) and Limonene (2.68%) were the major compounds of the essential oil. Antibacterial activity of the essential oil against nine Gram-positive and Gram-negative strains was studied using disc diffusion and micro-well dilution assays. Essential oil exhibited the antibacterial activity against three Gram-negative strains of Pseudomonas aeruginosa, Escherichia coli, and Shigella dysenteriae. The preliminary study on toxicity of seed oil was performed using Brine Shrimp lethality test (BSLT). Results indicated the high toxicity effect of essential oil (LC50 = 10 μg/mL). In vitro anticancer activity of seed oil was investigated against human breast cancer (MDA-Mb) and cervical epithelioid carcinoma (Hela) cell lines by MTT assay. Results showed the seed oil behave as a very potent anticancer agent with IC50 of lower than 10 μg/mL in both cases.  相似文献

The copper complexes of 2‐furaldehyde and furan oximes have previously demonstrated potent cytotoxicity, L1210 DNA synthesis inhibition, DNA topoisomerase II inhibition and DNA fragmentation. Currently a series of cobalt metal complexes of 2‐furaldehyde oximes were compared with copper complexes of furan oximes to determine whether the type of metal is important to the cytotoxicity and mode of action of the complexes. The cobalt complexes of furan oximes, like the copper complexes, were shown to be cytotoxic to suspended tumor cell lines, e.g. leukemias, lymphomas, acute monocytic leukemia and HeLa‐S3 uterine carcinoma. The cobalt complexes did not demonstrate dramatic cytotoxicity against the growth of tumors derived from solid human tumor lines. The cobalt complexes preferentially inhibited L1210 DNA synthesis, followed by inhibition of RNA and protein synthesis from 25 to 100 µM over 60 min. These agents, like the copper complexes of 2‐furaldehyde and furan oximes, were inhibitors of DNA polymerase α activity and de novo purine synthesis with marginal inhibition of ribonucleoside reductase and dihydrofolate reductase activities with DNA fragmentation. Unlike the copper complexes, the cobalt complexes did not inhibit L1210 DNA topoisomerase II activity but did reduce thymidylate synthetase activity. Thus, varying the type of metal within the complexes of 2‐furaldehyde and furan oximes produces differences in both cytotoxicity and mode of action. Copyright © 1999 John Wiley & Sons, Ltd.  相似文献

Acetone:chloroform (1:2) extract of the aerial parts of Euphorbia connata Boiss. (Euphorbiaceae) was investigated for its diterpenoids. This led to the isolation of one known and two new diterpenes, belonging to the pentahydroxy-13(17)-epoxy-8,10(18)-myrsinadiene and tetrahydroxy-5,6-epoxy-14-oxo-jatropha-11(E)-ene classes. The structures were elucidated based on 13C and 1H NMR as well as 2D NMR, IR and MS spectra and the cytotoxicity for compounds 13 were evaluated by using MTT assay against two human breast cancer cell lines. Myrsinane-type compounds – 3,7,14,15-tetraacetyl-5-propanoyl-13(17)-epoxy-8,10(18)-myrsinadiene (1) and 3,7,10,14,15-pentaacetyl-5-butanoyl-13,17-epoxy-8-myrsinene (2) – exhibited moderate inhibitory effects, with IC50 values of 24.53 ± 3.39 and 26.67 ± 1.41 μM against the MDA-MB cell line, and 37.73 ± 3.41 and 34.57 ± 2.12 μM against the MCF-7 cell line, respectively. Jatrophane-type diterpene – 5,6-epoxy-8,9,15-triacetyl-3-benzoyl-14-oxo-jatropha-11(E)-ene (3) – showed weak cytotoxicity, with IC50 values of 55.67 ± 7.09 μM against MDA-MB, and moderate cytotoxicity with IC50 values of 24.33 ± 3.21 μM against MCF-7 cell line.  相似文献

醇脱氢酶(ADH)用于食品工业,还是酶法测定乙醇或乙醛含量的工具酶。沉淀法和层析法至今仍然广泛用于分离蛋白质和酶。用离子交换层析、亲和层析等方法经过4~5个纯化步骤后,分离纯化ADH和其它蛋白质,纯化倍数可达100多倍。我们从酵母细胞中分离出醇脱氢酶,并通过硫酸铵分段盐析、凝胶层析以及离子交换层析对该酶进行纯化,用较少的步骤得到了相对高的纯化倍数。  相似文献

The synthesis of nanoparticles is most important in the context of cancer therapy, particularly copper nanoparticles, which are widely used. In this work, copper(II)-tyrosinase was isolated from potato peel powder. Copper nanoparticles (Tyr-Cu(II)-AEEA NPs) were synthesized via the reaction of tyrosinase with N-aminoethylethanolamine to produce Cu(II)-NPs and these were characterized by means of FT-IR, UV-Spectroscopy, XRD, SEM, TEM and a particle size analyzer. These Tyr-Cu(II)-AEEA NPs were tested as anticancer agents against MCF-7 breast cancer cells. Fluorescence microscopy and DNA fragmentation were also performed, which revealed the inhibiting potentials of Cu(II)-AEEA NPs and consequent cell death; Tyr-Cu(II)-AEEA NPs show potential cytotoxicity activity and this nano material could be contemplated as an anticancer medicament in future investigations.  相似文献

A trimetallic CuII derivative, [Cu3(L)2(CF3COO)2] (1) (where H2L = N,N′-bis(salicylidene)-1,3-propanediamine), was prepared and characterized. In 1, the two terminal CuII ions are linked to the central CuII by trifluoroacetato and doubly bridging phenoxido. Both the square-pyramidal and octahedral geometries are observed among two different CuII centers in the linear arrangement of the trimetallic unit. Compound 1 is characterized by IR and UV-Vis spectra. Compound 1 has high cytotoxic activity in breast adenocarcinoma (MCF-7), colorectal carcinoma (HCT116) and particularly, in ovarian carcinoma (A2780) cell line compared to a lung adenocarcinoma cell line. The IC50 in A2780 cells is 25 times lower than the respective value for normal human primary fibroblasts demonstrating 1 has higher cytotoxicity towards cancer cells. Additionally, combination of DOX with 1 induces a higher loss of HCT116 cell viability compared with each drug alone.  相似文献

A new diterpenoid glycoside, 6E,10E,14Z-(3S)-17-hydroxygeranyllinalool-17-O-β-d-glucopyranosyl-(1?→?2)-[α-l-rhamnopyranosyl-(1?→?6)]-β-d-glucopyranoside (1) together with the known diterpenoid glycoside (2) and two known flavonoid glycosides (3, 4) were isolated from the methanol extract of Blumea lacera leaves. The structures were determined by the interpretation of their spectroscopic data and comparison with the literature. All compounds were isolated for the first time from B. lacera and evaluated for their cytotoxic activity. Only the new compound (1) showed strong cytotoxic activity with the lowest IC50 value (8.3 μM) being displayed against MCF-7 breast cancer cells. In apoptosis and cell cycle analysis, 1 revealed strong apoptotic activity against MCF-7 cells (45.5% AV+/PI?) after 24 h, but showed no arresting of any of the cell cycle phases in MCF-7.  相似文献

Previously, the authors conducted phytochemical investigations of the aerial parts of Larrea tridentata and reported triterpene glycosides and lignan derivatives. In continuation of the preceding studies, 17 lignans and lignan glycosides (1–17) were isolated, including seven new compounds (1–7). Herein, the structure of the new compounds was determined based on spectroscopic analysis and enzymatic hydrolysis. The cytotoxicity of 1–17 against HL-60 human promyelocytic leukemia cells was examined. Compounds 4–11 and 14–16 were cytotoxic to HL-60 cells, with IC50 values in the range of 2.7–17 μM. Compound 6, which was the most cytotoxic among the unprecedented compounds, was shown to induce apoptotic cell death in HL-60 cells.  相似文献

Cells have evolved elaborate mechanisms to regulate DNA replication machinery and cell cycles in response to DNA damage and replication stress in order to prevent genomic instability and cancer. The E3 ubiquitin ligase SCFDia2 in S. cerevisiae is involved in the DNA replication and DNA damage stress response, but its effect on cell growth is still unclear. Here, we demonstrate that the absence of Dia2 prolongs the cell cycle by extending both S- and G2/M-phases while, at the same time, activating the S-phase checkpoint. In these conditions, Ctf4—an essential DNA replication protein and substrate of Dia2—prolongs its binding to the chromatin during the extended S- and G2/M-phases. Notably, the prolonged cell cycle when Dia2 is absent is accompanied by a marked increase in cell size. We found that while both DNA replication inhibition and an absence of Dia2 exerts effects on cell cycle duration and cell size, Dia2 deficiency leads to a much more profound increase in cell size and a substantially lesser effect on cell cycle duration compared to DNA replication inhibition. Our results suggest that the increased cell size in dia2∆ involves a complex mechanism in which the prolonged cell cycle is one of the driving forces.  相似文献

The reaction of α,α′‐dimercapto‐o‐xylene (H2dmox) with different precursors such as SnMe2Cl2, [Ti(η5‐C5H5)2Cl2] and [Ti(η5‐C5H4Me)2Cl2] (1:1) in the presence of two equivalents of NEt3 yielded the complexes [SnMe2(dmox)] (1), [Ti(η5‐C5H5)2(dmox)] (2) and [Ti(η5‐C5H4Me)2(dmox)] (3), respectively. 1–3 have been characterized by spectroscopic methods; in addition, complex 3 has been determined by X‐ray diffraction studies. Furthermore, structural studies based on density functional theory calculations of 1 and 2 have been carried out. The cytotoxic activity of 1–3 was tested against the tumour cell lines human adenocarcinoma HeLa, human myelogenous leukaemia K562 and human malignant melanoma Fem‐x. The results of this study show a higher cytotoxicity of the tin(IV) complex (1) in comparison to their titanium(IV) counterparts (2 and 3) as well as an improvement in the cytotoxic activity of compounds 2 and 3 compared to their titanocene(IV) dichloride analogues [Ti(η5‐C5H5)2Cl2] and [Ti(η5‐C5H4Me)2Cl2]. In view of the relatively high cytotoxicity of compound 1, a detailed study on the effects of the in vitro treatment of cancer cell lines using this compound was carried out. Thus cell cycle experiments on all the studied cell lines treated with 1 show that this complex seems to cause disturbances in the G1 phase of HeLa, and in the G1 and G2/M phases of Fem‐x cell line, while almost no disturbances were observed in the cycle of K562 cells treated with 1. Finally, DNA laddering method showed that treatment of the HeLa and Fem‐x cell lines with IC90 doses of 1 resulted in the induction of apoptosis. Copyright © 2012 John Wiley & Sons, Ltd.  相似文献

Four two-armed chiral calix[4]arenes (1a1d) functionalised at the lower ring with amino acid units have been synthesised and the structures of these compounds were characterised by IR, MS, 1H NMR, 13C NMR spectra and elemental analysis. Their molecular recognition abilities towards amino alcohol were examined by fluorescence titration experiment in three kinds of solution. The results indicated that these receptors exhibited excellent fluorescent response to phenylglycinol and could distinguish phenylglycinol from phenylalaninol rapidly through the obvious difference in the fluorescent response. Solvent comparative experiments also indicated that acetonitrile was the best solvent to detect these phenomena.  相似文献

Reliable compounds with low toxicity are tempting potential chemotherapeutics. With an aim of achieving less toxic but more potent metallodrugs, four new‐generation hydrophilic Cu(II) and Zn(II) complexes with DNA‐targeting properties were synthesized and characterized using various physicochemical data. The excellent DNA binding and cleavage results confirmed the mode of binding of DNA with the complexes and their ability to denature it. The profound in vitro cytotoxicity exhibited by complex 3 against a panel of cell lines (HeLa, MCF‐7 and HepG‐2) along with NHDF (normal human dermal fibroblasts) with distinct activity towards HepG‐2 and low toxicity to NHDF prompted in vivo studies of induced hepatocellular carcinoma‐affected Swiss albino rats. On evaluating various serum hepatic, biological and histopathological parameters, complex 3 showed excellent activity in restoring the damaged liver to normal. As a means of identifying the pathway of DNA damage, flow cytometric evaluation of cell cycle analysis was performed, which revealed S phase arrest‐induced apoptosis in HepG‐2 cells by complex 3 , making it a cell cycle‐specific drug.  相似文献

Four transition metal complexes, namely Cu2Cl2(HL1)2 ( 1 ), Co(HL1)2 ( 2 ), Cu(HL2)2 ( 3 ), Co(HL2)3 ( 4 ) {H2L1 = 2‐{[2‐(2‐hydroxy‐ethoxy)‐ethylimino]‐methyl}‐phenol, H2L2 = 3‐{[2‐(2‐hydroxy‐ethoxy)‐ethylimino]‐methyl}‐naphthalen‐2‐ol} were synthesized and characterized by single‐crystal X‐ray diffraction, IR spectroscopy, and elemental analysis. The interactions of these copper and cobalt complexes with calf thymus DNA (CT‐DNA) were studied by absorption and emission titration spectroscopic methods. The interactions of these copper and cobalt complexes with bovine serum albumin (BSA) were also investigated using fluorescence, UV/Vis, and synchronous fluorescence spectroscopy. Further, the in vitro cytotoxic effect of the complexes are examined on human colon carcinoma cell lines (HCT‐116) and human lung carcinoma cell line (A549).  相似文献

A new series of hybrid molecules containing cinnamic acid and 2-quinolinone derivatives were designed and synthesized. Their structures were confirmed by 1H-NMR, 13C-NMR and mass analyses. All the synthesized hybrid molecules were assessed for their in vitro antiproliferative activity against more than one cancer cell lines. Compound 3-(3,5-dibromo-7,8-dihydroxy-4-methyl-2-oxoquinolin-1(2H)-ylamino)-3-phenylacrylic acid (5a) with IC50 = 1.89 μM against HCT-116 was proved to the most potent compound in this study, as compared to standard drug staurosporin. DNA flow cytometry assay of compound 5a revealed G2/M phase arrest and pre-G1 apoptosis. Annexin V-FITC showed that the percentage of early and late apoptosis was increased. The results of topoisomerase enzyme inhibition activity showed that the hybrid molecule 5a displays potent inhibitory activity compared with control.  相似文献

1,4-Naphthoquinones (1,4-NQ) have been reported to possess a variety of pharma-cological properties including antibacterial, antifungal, antiviral, anti-inflammatory, anti-artherosclerotic, and anticancer effects. In this study, new N- and S,S-substituted-1,4-NQ derivatives were synthesized in excellent yields and were completely characterized by spectroscopic analysis IR, NMR (1H and 13C), MS and microanalysis. The cytotoxic activities of 1,4-NQ derivatives were examined against to A-549, DU145, HCT-116 and MDA-MB-231 cancer cells. Among these compounds, 2-[4-(2-furoyl)piperazine-1-yl]-3-chloro-1,4-NQ 5 and 2,3-bis(cyclobuthylsulfanyl)-1,4-NQ 17 were identified as the most potent anticancer agents with cytotoxic activity against three cell lines (breast (MDA-MB-231), prostate (DU145), colorectal (HCT-116).  相似文献

Although several molecular markers for human breast cancer exist, their versatility is limited. Here we demonstrate, through a differential proteome analysis utilizing the fluorogenic derivatization-liquid chromatography/tandem mass spectrometry (FD-LC-MS/MS) method between seven cancer cells and one normal cell, that the presence of cooperatively expressed annexin-2 and galectin-1 without tropomyosin-1 in a tissue could be used to diagnose metastatic breast cancer. Interestingly, in a metastatic cancer cell, the expression of the former two together with highly expressed cofilin-1 activates the Rho signal pathway to aggressively form disorganized actin filaments. Despite the excess expression of annexin-2 and galectin-1 in the normal cell, the highly expressed tropomyosin-1 counteracted the activity of cofilin-1 and stabilized the filaments, resulting in the restoration of the disorganization. This phenomenon suggests that enhancement of tropomyosin-1 should be used as therapy for metastatic breast cancer.  相似文献

In this study, rosemary leaf extract was effectively used to synthesize cobalt oxide nanoparticles (Co3O4 NPs) using a rapid, low-cost, and environmentally friendly approach. The prepared Co3O4 NPs were examined using various analytical techniques. However, UV spectrum analysis displayed two sharp absorption peaks at ~350 and 745 nm. The dynamic light scattering and zeta potential measurements were used to evaluate the particle size and the effective stabilization of the synthetic nanoparticles in the suspensions. A semi-triangular pyramidal shape of the Co3O4 NPs with a wide particle-size distribution could be observed in the scanning electron microscopy images. The energy-dispersive X-ray spectrum confirmed their successful synthesis, as the experimental atomic percentages agreed with the theoretical values. Moreover, X-ray diffraction analysis revealed that the synthesized Co3O4 NPs had a cubic crystalline structure corroborating that of theoretical Co3O4. Additionally, the Co3O4 NPs were not toxic at ≤62.5 µg/mL for Hep G2 and at ≤31.25 µg/mL for Mcf7. Therefore, these unique environmentally friendly Co3O4 NPs at this safe concentration could be studied in the future for their therapeutic activity.  相似文献

The study was undertaken to investigate the antioxidant, genotoxic, and cytotoxic potentialities of phyto-fabricated zinc oxide nanoparticles (ZnO-NPs) from Ipomoea obscura (L.) Ker Gawl. aqueous leaf extract. The UV-visible spectral analysis of the ZnO-NPs showed an absorption peak at 304 nm with a bandgap energy of 3.54 eV, which are characteristics of zinc nanoparticles. Moreover, the particles were of nano-size (~24.26 nm) with 88.11% purity and were agglomerated as observed through Scanning Electron Microscopy (SEM). The phyto-fabricated ZnO-NPs offered radical scavenging activity (RSA) in a dose-dependent manner with an IC50 of 0.45 mg mL−1. In addition, the genotoxicity studies of ZnO-NPs carried out on onion root tips revealed that the particles were able to significantly inhibit the cell division at the mitotic stage with a mitotic index of 39.49%. Further, the cytotoxic studies on HT-29 cells showed that the phyto-fabricated ZnO-NPs could arrest the cell division as early as in the G0/G1 phase (with 92.14%) with 73.14% cells showing early apoptotic symptoms after 24 h of incubation. The results of the study affirm the ability of phyto-fabricated ZnO-NPs from aqueous leaf extract of I. obscura is beneficial in the cytotoxic application.  相似文献
