天冬氨酸侧链环己酯Boc Asp(OcHex) OBzl(Ⅰ)较苄基酯对酸稳定10倍以上,适于长链肽的合成;且可用甲磺酸温和脱除(0℃/1h);可承受温和的皂化、肼解及氢解反应;可显著降低含有天冬氨酸序列(如Asp Gly、Asp Ser等)的肽在合成过程中形成天冬酰亚胺(aspartimide)(即β 肽)的可能性[1]。我们从价廉易得的L Asp出发,经环己基化和Boc酰化制得Ⅱ[2],将Ⅱ苄基化,得到目标分子Ⅰ[1~6]。Ⅲ的酰化,用四氢呋喃(THF)、水为反应溶剂;Ⅱ的苄基化,试用过(1)PhCH2Br/THF/TEA/10~19℃(2)hCH2Br/EtOAc/TEA/reflux(3)PhCH2OH/DCC/HOBt/T…  相似文献

以3-氨基-1,2-丙二醇,甲酸二叔丁酯及其合成的醛和甘氨酸甲(乙)酯为原料,经碱性缩合、高碘酸钾氧化、亲核加成一消去和10%Pd/C催化还原,设计合成了几种标题化合物。该合成路线方便,且处理简单,产率为64%-84%。其结构经^1H NMR和^13C NMR确定。  相似文献

报道了取代苯丙酸类化合物1a~1d与N-叔丁氧羰基-L-半胱氨酸甲酯(2)在双(2-氧代-3-噁唑烷基)次磷酰氯(BOP-C1)作用下,以79%~92%收率得到缩合产物S-[2-(叔丁氧羰基氨基)乙基]-3-苯基丙酸硫酯类化合物3a~3d;3a~3d在三氟乙酸(TFA)作用下脱除Boc保护基时,结果不仅得到了正常的脱保护基产物4a~4d,还生成了2-取代噻唑啉类化合物5a~5d,研究表明5a~5d是由4a~4d分子内脱水环合而成.通过优化三氟乙酸用量、反应温度以及反应时间等条件,能够以较高收率分别得到4a~4d和5a~5d(收率85%~91%和86%~89%).而S-[2-(叔丁氧羰基氨基)乙基]-3-苯基丙烯酸硫酯类化合物3e~3f由于双键结构,在三氟乙酸作用下仅生成脱除Boc保护基产物4e~4f.该反应的研究为2-取代噻唑啉类化合物的合成提供了一种简便有效的方法.  相似文献

Enzymatic synthesis of cefaclor from 7-aminodesacetoxymethyl-3-chlorocephalosporainc acid(6-ACCA) and phenylglycine derivatives using penicillin G acylase was studied .Many factors that affect the conversion of 7-ACCA to cefaclor were examined.The immobilized enzyme from Bacillis megaterium gave a better catalytic properties and the higher conversion was obtained using phenylglycine methyl ester(PGME) as acyl donor.And the external mass transfer limitation could be eliminated when the stirring rate was more than 150r/min.Low temperature was beneficial for the synthesis and the results showed that the synthetase activity was hardly influenced by temperature while the amidase activity was affected greatly by temperature.The optimum reaction conditions were determined at pH 6.5 and 10℃,respectively.The best 7-ACCA conversion of 56% was achieved when the intial concentration of 7-ACCA and PGME was at 50 mM and 150mM,respectively.  相似文献

由于头孢卡品酯的理化性质及其颗粒制剂包含很多难溶的辅料,为准确测定盐酸头孢卡品酯颗粒中镉、铅、砷、汞、钴、钒、镍、钼、铬等9种元素杂质的含量,尤其是挥发性元素汞,需要对样品进行络合和稳定性处理。以6 mL硝酸、1.5 mL盐酸、0.1 mL氢氟酸为消解酸,于180℃微波消解50 mg盐酸头孢卡品酯颗粒样品30 min;消解结束后,反复赶酸,再用体积比为0.1∶100的氢氟酸和盐酸-硝酸-水(体积比为1∶4∶95)的混合液定容至50 mL,采用电感耦合等离子体质谱法测定样品中9种元素杂质的含量,内标法定量。结果显示:9种元素杂质的质量浓度在一定范围内与其对应的响应值与内标响应值的比值呈线性关系,检出限(3s/k)为0.001~0.070μg·L^(-1);对6份同一加标样品溶液进行测定,9种元素杂质测定值的相对标准偏差为2.9%~5.1%;对实际样品进行加标回收试验,回收率为82.0%~104%。方法用于实际样品分析,9种元素杂质的含量均低于元素杂质指导原则规定的允许浓度限值。  相似文献

提出了制备头孢类抗生素的中间体α-(2-氨基噻唑-4-基)-α[(叔-丁氧基羰基)异丙氧亚胺基]-乙酸(ATIA)的反相高效液相色谱测定方法,采用SpherigelTM C18色谱柱(250 mm×4.6 mm,5μm),以0.05 g·L-1四丁基溴化铵的甲醇-水(6+4)溶液为流动相,流速为1 mL·min-1,柱温30℃,检测波长260 nm,以外标法定量.在0.001~1.0 mg范围内,ATIA的质量与峰面积呈线性关系,回归方程为A=31 475.6m+1 786.7,相关系数为0.999 7,以质量为0.06 mgATIA按方法测定6次,算得其相对标准偏差为3.7%,检出限(3S)为0.6μg,回收率的试验结果在98.8%~104.0%之间.  相似文献

在最优反应条件[海因2 mmol,n(3-戊酮)∶n(海因)=1.5,乙醇胺为碱,p H 4,于55℃反应6 h]下制得5-(1-乙基亚丙基)海因(4);4用5%Pd/C催化加氢制得5-(3-戊基)海因(5);5先在碱性条件下水解,再通过叔丁氧羰基保护氨基合成了(±)-N-叔丁氧羰基-3-乙基-戊氨酸,总收率43%,其结构经~1H NMR,~(13)C NMR和HR-ESI-MS确证。  相似文献

本文构建了一种基于3-苯丙噻唑基-2-羟基-5-甲基苯甲醛的荧光探针用于检测铝离子。该荧光探针能够灵敏、高选择性地检测铝离子,并显示出颜色和强烈的荧光变化双重响应。研究结果表明,该探针对铝离子表现出非常好的荧光增强效果,检测过程中其它金属离子不会对检测结果产生明显的干扰。其比率荧光强度(I490/I567)与铝离子的浓度(0~20μmol/L)之间具有良好的线性关系,检测限低至0.5μmol/L。由于具有高效的选择性,该探针可以用于检测污染河水中的铝离子含量。  相似文献

为了寻找生物活性良好的噻唑基丙烯腈类化合物, 利用2-[4-(2,6-二氟苯基)噻唑-2-基]乙腈(3)分别与取代氯甲酸酯4和取代苯基异氰酸酯6在碱存在下反应, 合成了8个2-[4-(2,6-二氟苯基)噻唑-2-基]-3-羟基-3-烃氧基丙烯腈化合物5和7个2-[4-(2,6-二氟苯基)噻唑-2-基]-3-羟基-3-取代苯胺基丙烯腈化合物7, 均为首次报道的丙烯腈类化合物. 化合物结构经1H NMR, IR, MS和元素分析表征. 初步生物活性测定结果表明, 在试验浓度下, 目标化合物均具有一定的杀虫和抑菌活性, 其中化合物5f和5h在100 mg/L浓度下对炭疽病菌的抑制率达95%; 化合物5g和7d在250 mg/L浓度下对棉红蜘蛛的致死率达85%.  相似文献

A series of new 2-(benzo[d]thiazol-2-yl) phenyl-4-nitrophenyl alkyl/aryl substituted phosphoramidate derivatives (7a-j) of biological interest are synthesized in two steps via an in situ process without the isolation of the intermediate. 2-(Benzo[d]thiazol-2-yl) phenol (3) was treated with 4-nitrophenyl phosphorodichloridate(4) to obtain the monochloro intermediate, 2-(benzo[d]thiazol-2-yl)phenyl (4-nitrophenyl) phosphorochloridate(5). It was subsequently reacted with various amines, 6(a-j) to afford the desired title products. IR, NMR (1H, 13C and 31P), mass spectral and elemental analysis are used to characterize the structures of the newly synthesized compounds. The antibacterial and antifungal activities are determined by the growth of inhibition of bacteria and fungi of the title compounds at two concentrations, 50 and 100 µg/mL, and minimum inhibitory concentrations to the active compounds. The compounds 7e, 7f and 7j exhibited promising inhibition activity against bacterial strains and 7c, 7e and 7i against A. niger and C. albicans and MIC values are in the range of 10.0–15.0 µg/mL.  相似文献

Abstract

A synthesis of novel hybrid molecules containing thiazole or bis(thiazoles) each bearing benzofuran and/or benzo[d]thiazole moieties by the reaction of the appropriate thioamide derivatives with the corresponding bis-bromoacetyl derivatives is reported. Mono- and bis(triazolothiadiazine) derivatives based on benzofuran or benzo[d]thiazole moieties were also synthesized in good yields by the reaction of the appropriate bis(bromoacetyl) derivatives with each of 4-amino-5-mercapto-1,2,4-triazoles and their corresponding bis-derivatives.  相似文献

A series of tetrazolo[1,5-c]quinazolines were synthesized by [4 + 1]-cyclocondensation of 4-hydrazinoquinazolines with sodium nitrite with good yields. Peculiarities of this reaction were discussed, namely, the influence of halogen on the reaction yield. Hydrolytic cleavage of tetrazolo[1,5-c]quinozoline cycle was studied in order to obtain 2-(1H-tetrazolo-5-yl)anilines. The structures of the compounds were elucidated by 1H NMR, 13C NMR, infrared, mass spectrometry, and elemental analysis.  相似文献

以2-(2-甲基烯丙氧基)苯酚为原料, 经Claisen重排、呋喃环化、醚化、Friedel-Crafts乙酰化、α-溴代和噻唑环化等反应合成了22种4-(7-甲氧基-2,2-二甲基-2,3-二氢苯并呋喃-5-基)-2-芳氨基噻唑化合物. 化合物的结构经质谱、1H NMR和元素分析等确证. 采用单晶X射线衍射仪测定了化合物7j的晶体结构, 该晶体属于单斜晶系, 空间群为C2/c, 晶胞参数a=2.21140(12) nm, b=0.87602(5) nm, c=2.13911(12) nm, β=115.5380(10)°; V=3.7391(4) nm3, Z=8, Dc=1.375 g/cm3, F(000)=1616, S=1.046, μ=0.333 mm-1, 最终偏差因子R1=0.0390, wR2=0.1079. 杀虫活性实验结果表明, 化合物7a, 7b, 7h和7t在浓度为500 mg/L时对蚕豆蚜死亡率分别为95.12%, 62.60%, 57.53%和59.06%.  相似文献

在不加任何催化剂条件下, 2-(12-苯并吖啶酮)-乙酸(BAAA)与N,N′-羰基双咪唑(CDI)缩合生成新型荧光探针5-羰基咪唑苯并吖啶酮(IEBA). IEBA在DMF溶剂中与胺类化合物形成的酰胺类衍生物不仅可发出强烈的荧光, 还具有较高的质谱离子化能力. 该衍生物在乙腈和甲醇-水溶液中的百分离子化δ值分别在0~57.32% 和0~62.14%范围内. 最大激发和发射波长λex/λem=272 nm/505 nm. 12种胺类衍生物的荧光检出限范围为0.15~0.50 ng/mL, 在线APCI-MS检出限范围为1.43~8.51 ng/mL.  相似文献

The effect ofmeta-substituents in the aromatic ring on the route of Claisen rearrangement ofN-(pent-3-en-2-yl)-3-fluoro(or 3-trifluoromethyl, 3,4-difluoro)anilines induced by ZnCl2 was investigated. 在K2CO3存在下利用聚乙二醇-400 (PEG-400)作相转移催化剂, 于固-液相转移催化条件下, 通过5-芳亚甲基氨 基-2-巯基-1,3,4-噻二唑与氯乙酰芳胺的硫烷基化反应, 合成了16个未见文献报道的[5-(芳亚甲基氨基)-1,3,4-噻二唑-2-基]硫乙酰芳胺衍生物. 经元素分析, FT-IR,1H NMR和13C NMR确证了其结构. 生物活性实验结果表明, 部分化合物对小麦幼苗的生长具有明显的植物生长调节活性, 并对枯草杆菌具有一定的抑制活性.  相似文献
