Venom variability in specimens of Tityus serrulatus scorpion was assessed by matrix-assisted laser desorption/ionization time-of-flight mass spectrometry (MALDI-TOFMS) analyses. An expanded time lag venom extraction protocol was carried out using ten scorpions to study individual variations that might occur due to different rates in protein expression and/or processing. The first extraction of venom was made from the animals after 20 days of starvation, which allowed the venom gland to be filled up. The second extraction event was carried out 24 hours after the first one. The third was 8 days after the first extraction. By means of MALDI-TOF analyses, important variations were observed in venoms of a single specimen extracted at different times, especially in latter extraction events. These variations are most probably related to dynamics in cell gland production. Since T. serrulatus is a parthenogenetic species, sexual variations are naturally excluded and we did not expect intra-specific variations, which was confirmed. Knowledge of individual venom variability is extremely important to avoid misunderstandings in the use of venom proteomic analysis as a taxonomic tool. 相似文献

The red scorpion, Mesobuthus tamulus, is found in two distinct biotopes within the Indian state of Maharastra-a tropical, sea-level biotope and a semi-arid biotope, up to 600 m. Scorpions from these two geographical areas show marked differences in toxicity. Using mass spectrometry, we have shown biotope-specific variation in the expression of peptides from scorpions collected from these two distinct areas. Matrix-assisted laser desorption/ionisation time-of-flight mass spectrometry (MALDI-TOFMS) and reversed-phase liquid chromatography/electrospray ionisation mass spectrometry (LC/ESI-MS) were assessed as techniques for obtaining mass fingerprint data. On line LC/ESI-MS was judged to be the method of choice and unique biotope-specific mass fingerprints, with key diagnostic markers, were obtained. 相似文献

Mass spectrometric methods, including matrix-assisted laser desorption/ionisation time-of-flight mass spectrometry (MALDI-TOFMS), on-line liquid chromatography/electrospray ionisation mass spectrometry (LC/ESI-MS), and nanospray ionisation/hybrid quadrupole time-of-flight mass spectrometry (nanoESI-QqTOFMS), were applied to characterize by mass fingerprinting the venom of the French Guyanese tarantula Theraphosa leblondi. Of these techniques direct nanoESI-QqTOFMS, which allowed the detection of 65 protonated molecules with high mass accuracy, appeared to give the best results. Three major peptides, TlTx1, TlTx2 and TlTx3, were sequenced using a combination of nanoESI-MS/MS and enzyme digestion/MS and MS/MS experiments. Each sequence was confirmed by automated Edman sequencing. In patch-clamp experiments these peptides were found to have a specific inhibitory effect on the voltage-dependent potassium channel, Kv4.2. 相似文献

Fingerprinting by mass spectrometry has been increasingly used to study venom variations and for taxonomic analyses based on venom components. Most of these studies have concentrated on components heavier than 3 kDa, but Bothrops snake venoms contain many biologically active peptides, principally C-type natriuretic peptides and bradykinin-potentiating peptides (BPPs). In this work, we have examined the peptide profile of Bothrops venoms (B. alternatus, B. erythromelas, B. insularis, B. jararaca, B. jararacussu, B. leucurus and B. moojeni) using direct infusion nano-electrospray ionization mass spectrometry (nano-ESI-MS) subjecting the data further to principal components analysis (PCA) to assess whether the peptide distributions are reliable in distinguishing the venoms. ESI-MS of a low molar mass fraction obtained by ultrafiltration of each venom (5 kDa nominal cutoff filters) revealed that the venoms have a variety of peptides in common but that each venom also contains taxonomic marker peptides not shared with other venoms. One BPP peptide, QGGWPRPGPEIPP, was found to be common to the seven Bothrops species examined. This peptide may represent a specific marker for this genus since it was not found in the venom of the South American rattlesnake, Crotalus durissus terrificus. PCA on the ESI-MS data reveals a close relationship between B. jararaca, B. jararacussu and B. moojeni venoms, with B. leucurus and B. erythromelas being more distant from these three; B. alternatus and B. insularis were also located distant from these five species, as was C. d. terrificus. These results agree partially with established phylogenetic relationships among these species and suggest that ESI-MS peptide fingerprinting of snake venoms coupled with PCA is a useful tool for identifying venoms and for taxonomic analyses. 相似文献

No universally accepted score is currently available to determine when a matrix-assisted laser desorption ionization (MALDI) peptide mass fingerprint (PMF) experiment has been successfully carried out. We describe a software program (ChemApplex) based on a calculated parameter (Combined Protein Score) that takes into account (1) peak intensity, (2) the mass accuracy of the match, and (3) ChemScore, a theoretical intensity factor that estimates the probability of observing a particular peptide based on a combination of chemical considerations, in particular the amino acid composition of the peptide and the amino acid sequence of the amino acids that span the cleavage site. When these three factors are taken into account both at the level of individual peptides and at the protein level, protein components in mixtures whose peptides contribute less than 1% of the total intensity can often be correctly identified, as is demonstrated for mixtures of standard proteins. Moreover, it is possible to make robust database identifications that are nearly independent of the number of masses submitted and the mass error threshold used for matching. Protein scoring based on Combined Protein Score is orthogonal to many of the commonly used probability-based scoring schemes, and makes it possible to archive a more complete set of parameters that more thoroughly characterize the validity of the database match, which increases the confidence in the identifications. 相似文献

As the new fluorescent stains such as SyproRuby and DeepPurple are getting widespread recognition for proteome analyses by the traditional 2-D gel method, it becomes important to test the feasibility of these stains with respect to staining reproducibility, protein quantitation, and compatibility of the stain with downstream MS. The binding of epicocconone, active ingredient of DeepPurple, to one of the primary cleavage sites of trypsin (lysine residue) raises the possibility of incomplete cleavage and interference with PMF. However, the current study tests and concludes that the DeepPurple stain can result in increased peptide recovery compared to SyproRuby stain and can improve MS-based identification of lower intensity proteins spots. 相似文献

The disulfide bond pattern of Trimeresurus stejnegeri lectin (TSL), a new member of the C-type lectin family, was determined by mass spectrometry. Four intrachain disulfide bonds of TSL, Cys(3)-Cys(14), Cys(31)-Cys(131), Cys(38)-Cys(133) and Cys(106)-Cys(123), and two interchain linkages, Cys(2)-Cys(2) and Cys(86)-Cys(86), were determined. Three strategies were used in this work. One intrachain (Cys(106)-Cys(123)) and one interchain (Cys(86)-Cys(86)) disulfide linkages were detected by standard MS methods. The disulfide bonds Cys(2)-Cys(2) and Cys(3)-Cys(14) were analyzed using a modified partial reduction procedure and MS/MS. The last two disulfide bonds were characterized by a MS/MS/MS technique. The strategies developed in this work could be applied more generally to detection of disulfide bond patterns. 相似文献

The high economic value of catalysts containing the platinum group elements platinum, rhodium and palladium as active components causes the need to be able to measure the precious metal loading with small uncertainty and to have suitable certified reference materials fulfilling high demands on the quality of the certified values. In European Reference Material ERM®-EB504, a used cordierite-based car catalyst material, mass fractions of platinum, palladium and rhodium were certified. The raw material was milled, homogenised and annealed before analysis. Seventeen laboratories experienced in precious metals analysis participated in the certification interlaboratory comparison, most of them analysing with inductively coupled plasma optical emission spectrometry using different sample pretreatment techniques. Homogeneity testing was carried out using X-ray fluorescence spectrometry. The certified mass fractions of Pt, Pd and Rh and their expanded uncertainties (k = 2) in ERM®-EB504 are (1777 ± 15), (279 ± 6) and (338 ± 4) mg/kg respectively. 相似文献

The use of robots has major effects on maximizing the proteomic workflow required in an increasing number of high-throughput projects and on increasing the quality of the data. In peptide mass finger printing (PMF), automation of steps downstream of two-dimensional gel electrophoresis is essential. To achieve this goal, the workflow must be fluid. We have developed tools using macros written in Microsoft Excel and Word to complete the automation of our platform. Additionally, because sample preparation is crucial for identification of proteins by matrix-assisted laser desorption/ionization (MALDI) mass spectrometry, we optimized a sandwich method usable by any robot for spotting digests on a MALDI target. This procedure enables further efficient automated washing steps directly on the MALDI target. The success rate of PMF identification was evaluated for the automated sandwich method, and for the dried-droplet method implemented on the robot as recommended by the manufacturer. Of the two methods, the sandwich method achieved the highest identification success rate and sequence coverage of proteins. 相似文献

Arachnid venoms present a diverse and complex matrix for investigation, with their latent potential for innovative drug and pesticide design largely unrealised. The characterisation and quantification of selected low molecular mass compounds isolated from the crude venom of the Cobalt blue tarantula (Haplopelma lividum) were the objectives of this study. Fractionation of the crude venom was performed using reversed‐phase high‐performance liquid chromatography, with compound identification using both electrospray ionisation ion trap mass spectrometry and quadrupole time‐of‐flight mass spectrometry. Four compounds were identified, and quantification on a percentage dry weight basis was achieved by liquid chromatography/electrospray ionisation tandem mass spectrometry based on the formation of their corresponding product ions. Of these the most abundant component was glutamic acid, present at a level of 0.97%. Histamine and adenosine were detected at 0.14% and 0.10% dry weight, respectively, with the polyamine spermine noted in trace amounts at 0.002%. The limits of detection and quantification were established for each of the identified components. The fragmentation profile for histamine has also been proposed. Dehydroindigo: a new piece into the Maya Blue puzzle from the voltammetry of microparticles approach

Combining a novel technique, the voltammetry of microparticles, with spectrometric, nuclear magnetic resonance, electron microscopy, and atomic force microscopy data, Maya Blue is detected in wall paintings of the Substructures A-3, A-5, and A-6, dated in the Early Classical period (440-450 a.c.), and the Substructure II-C, dated in the Late Preclassical period (150 b.C.), in the archaeological site of Calakmul (Campeche, Mexico), thus providing evidence on the use of the pigment 750 years prior to the date currently accepted. Electrochemical measurements, supported by spectrometric data, indicate that the presence of palygorskite-attached dehydroindigo, the oxidized form of indigo, contributes to the greenish color of Maya Blue. Enthalpy and entropy of attachment of such compounds to palygorskite are calculated from the temperature dependence of electrochemical data. Both attachment processes are endothermic, becoming thermodynamically spontaneous at moderate temperatures. Accordingly, ancient Mayas may modulate the hue of Maya Blue from turquoise to greenish blue by controlling the temperature during the crushing process. 相似文献

We studied the hydrolysis of a neuropeptide kyotorphin (L-Tyr-L-Arg) by an enkephalin-degrading aminopeptidase purified from cytosol of rat brain in vitro. The purified enzyme was homogeneous as judged by sodium dodecyl sulfate (SDS)-polyacrylamide gel electrophoresis (PAGE), gel filtration and isoelectric focusing. The aminopeptidase with an apparent molecular weight (Mr) = 98000 catalyzed the hydrolysis of Leu- and Met-enkephalins with Km values of 125 and 142 microM, respectively. The enzyme activity was inhibited by bestatin, amastatin and puromycin but not by pepstatin, leupeptin and phenylmethanesulfonyl fluoride (PMSF). Kyotorphin was degraded by the aminopeptidase at pH 7.0, and the Vmax and Km values were 9.2 mumol/min/mg protein and 95 microM, respectively. The Km value for kyotorphin was compatible to those for Leu- and Met-enkephalins. Taken together, these results suggest a possible involvement of the enkephalin-degrading aminopeptidase in cytosolic degradation of kyotorphin in neuronal cells of rat brain. 相似文献

Aqueous hydrochloric acid hydrolysis of the mixture of thelothurins A and B, the saponins from the sea cucumber Thelonota ananas Jaeger, gave a complex mixture from which six sapogenins were isolated. Chemical and spectroscopic evidence led to the assignment of structures 3–8 to these genins which are correlated to the known seychellogenin acetate (9). It is also shown that compounds 1 and 2, obtained by mild aqueous acetic acid hydrolysis of the thelothurins, are the genuine aglycones, whereas 3–8 are artifacts. Their formation during the hydrolysis of the saponins is discussed. The aglycone moiety of thelothurins A (20A) and B (20B) was biosynthesized from the acetate. 相似文献

The aim of the present study was to analyze mass spectra of scorpions belonging to the genus Parabuthus (Pocock 1890) by means of matrix-assisted laser desorption time-of-flight mass spectrometry (MALDI-TOFMS) and to construct a species-specific venom code for species identification. The venom compositions of sixteen Parabuthus species, occurring in southern Africa, were characterized using representative peaks in the molecular mass range of 6400-8400 Da. This mass range is characteristic for the typical long-chain neurotoxins influencing sodium channels. Only a few of these peptides have been sequenced up to now. The impetus for development of these species-specific profiles was the observation of unique, highly reproducible mass spectral peaks within a specific species. An identification label for all the different species could be found using a minimum number of peaks. MALDI-TOFMS is therefore proposed as a complementary method to morphological and behavioural characteristics for species and ultimately subspecies discrimination. 相似文献

Summary A polystyrene sample obtained by free radical polymerization has been fractionated by preparative scale GPC and the structure and morphology of the fractions have been studied by13C-NMR spectroscopy, DSC and X-ray measurements. No changes in morphology in the course of fractionation have been obeyed. A distribution of syndiotacticity within the molecular weight distribution does not exist.
