The stereoselective construction of the DE fragment (2) of everninomicin 13,384-1 (1) is reported. From the two possible ways of inserting the DE fragment between the A1B(A)C and FGHA2 domains of the natural product, the sequence involving the DEFGHA2 segment was found to be the most viable. This coupling was followed by attachment of a suitably protected and activated A1B(A)C fragment which led, after orthoester construction and final deprotection to the targeted everninomicin 13,384-1 (1), completing the total synthesis of this complex naturally occurring substance.  相似文献

基于沸石咪唑骨架材料在离子液体和低共熔溶剂中冷却结晶的析出方式,开发了一种离子热法合成沸石咪唑骨架材料的新途径,采用X射线衍射、扫描电镜、核磁共振光谱、红外光谱以及热重分析等方法对制备的产物进行了表征,研究了合成条件对产物结晶度、尺寸和形貌的影响,探讨了沸石咪唑骨架材料在离子热合成体系中的溶解-结晶析出机理.研究发现,冷却速率能够影响产物形貌,急速冷却时,sod(RCSR代码)型产物的形貌为球形,zni(RCSR代码)型产物为棒状或平板状;程序控制冷却时,sod型产物的形貌为多面体,zni型产物呈团簇状.  相似文献

The total synthesis of apoptolidin (1) is reported together with the design, synthesis, and biological evaluation of a number of analogues. The assembly of key fragments 6 and 7 to vinyl iodide 3 via dithiane coupling technology was supplemented by a second generation route to this advanced intermediate involving a Horner-Wadsworth-Emmons coupling of fragments 22 and 25. The final stages of the synthesis featured a Stille coupling between vinyl iodide 3 and vinylstannane 2, a Yamaguchi lactonization, a number of glycosidations, and final deprotection. The developed synthetic technology was applied to the construction of several analogues including 74, 75, and 77 which exhibit significant bioactivity against tumor cells.  相似文献

Addition of Et(2)AlCN and i-PrOH to ketosulfinimines (N-sulfinyl imines) affords corresponding alpha-alkyl alpha-amino nitriles in moderate to good yields. The diastereoselectivity is largely dependent on the E/Z isomer ratio of the ketosulfinimine. Hydrolysis of the diastereomerically pure amino nitriles affords enantiopure alpha-alkyl alpha-amino acids in moderate to good yields.  相似文献

The first total synthesis of the ristocetin aglycon is described employing a modular and highly convergent strategy. An effective 12-step (12% overall) synthesis of the ABCD ring system 3 from its amino acid subunits sequentially features an intramolecular aromatic nucleophilic substitution reaction for formation of the diaryl ether and closure of the 16-membered CD ring system (65%), a respectively diastereoselective (3:1, 86%) Suzuki coupling for installation of the AB biaryl linkage on which the atropisomer stereochemistry can be further thermally adjusted, and an effective macrolactamization (51%) for closure of the 12-membered AB ring system. A similarly effective 13-step (14% overall) synthesis of the 14-membered EFG ring system 4 was implemented employing a room-temperature intermolecular S(N)Ar reaction of an o-fluoronitroaromatic for formation of the FG diaryl ether (69%) and a key macrolactamization (92%) with formation of the amide linking residues 1 and 2. The two key fragments 3 and 4 were coupled, and the remaining 16-membered DE ring system was closed via diaryl ether formation to provide the ristocetin tetracyclic ring system (15 steps, 8% overall) enlisting an unusually facile (25 degrees C, 8 h, DMF, >/=95%) and diastereoselective (>/=15:1) aromatic nucleophilic substitution reaction that benefits from substrate preorganization.  相似文献

The synthesis of the antibiotic anisomycin ( 1 ) is described. In the key step 3, 4-dimethoxy-thiopyrrolidone ( 16 ), prepared from diethyl L -( + )-tartarate ( 4 ) was converted to the thioiminoester 18 , which upon treatment with trimethylphosphite yielded the benzylidene derivative 19 . Cleavage of the ester group of 19 and hydrogenation of the decarboxylation product gave the two isomers 21 and 22 . 22 was converted into desacetylanisomycin ( 3 ) and anisomycin ( 1 ).  相似文献

The total synthesis of the linear trihydroxamate siderophore, Danoxamine, is described. Danoxamine is a siderophore component of the naturally occurring siderophore-drug conjugates Salmycin A-D. The synthesis of Danoxamine features a series of coupling reactions involving N-(5-benzyloxypentyl)-O-benzylhydroxylamine being linked by a succinoyl linker to N-(benzyloxy)-1,5-pentanediamine. Two more succinoyl linkers and another N-(benzyloxy)-1,5-pentanediamine were used in coupling reactions to afford the fully protected siderophore. The linear tetrabenzyl-protected trihydroxamate was deprotected to afford the natural product Danoxamine.  相似文献

Mathematical and numerical models of the yttrium aluminium garnet (YAG) synthesis are presented in the article. The models allow the effective computer simulation of the YAG synthesis. The synthesis by sol–gel and solid-state reaction methods is considered in the article. The question concerning the reasons for the observed changes in the preparation temperature by changing synthesis method is answered. The inverse modelling problem is solved: using known experimental data (synthesis time, dimensions of reactants) the unknown input parameters of the model (diffusion and reaction rate coefficients) are calculated.  相似文献

A concise and efficient synthesis of the tetracyclic CDEF ring system of lactonamycin (1) is described. The key step involved the Lewis acid mediated, intramolecular Friedel-Crafts acylation of carboxylic acid 6 to produce the tetracyclic CDEF core structure of target 1. The synthesis of 6 was carried out using a high-yielding Negishi coupling of benzyl bromide 7 with triflate 8, which was accessible in 11 steps and 31% overall yield on a multigram scale starting from trihydroxy acid 9.  相似文献

[reaction: see text] In an effort to develop a synthesis of the VAPOL ligand that avoids the use of a chromium carbene complex, a route was examined that involved the annulation of a naphthalene carboxamide via the method of Snieckus. The latter derivatives could be converted in a two-step sequence to 2-phenyl-4-phenanthrols in 60-72% overall yields. Two syntheses of the Amaryllidaceae alkaloid clivonine (1) are described. Both employ previously reported 7-arylhydrindane 6 as an intermediate but differ in the method employed for subsequent introduction of what becomes the ring-B lactone carbonyl carbon (C7). The synthesis featuring a Bischler-Napieralski reaction for this transformation constitutes the first asymmetric synthesis of natural (+)-clivonine. The first method is chiral auxiliary-based and utilizes aldol reactions of N-acetyl thiazolidinethiones followed by a malonate displacement/decarboxylation reaction. The oligomerization of (guanosine 5'-phosphor)-2-methylimidazolide on a poly(C) template proceeds efficiently at temp. as high as 37 degrees, although the helical complex formed by the template and substrate melts at a much lower temp. At higher temp. oligomerization depends on the template-independent formation of short oligomeric initiators, which then elongate on the template.
