Two series of coumarins possessing the aniline‐ and heterocyclic ring at 4th position have been synthesized and evaluated for their in vitro cytotoxic activity against MCF‐7 cancer cell line in MTT assay. Structure activity relationship (SAR) studies reveal that the electron donor group at position‐8 of coumarin played an important role in cytotoxic activity. Compound VIId showed the potent cytotoxic activity followed by compound Xa with IC50 = 6.25 and 6.50 μM, respectively. A docking study has also been carried out for the most potent compound to get an insight into molecular interactions with p50 subunit of NF‐κB protein.  相似文献

In this study, thiazole derivatives containing Schiff bases ( 7a , 7b , 7c , 7d , 7e , 7f , 8a , 8b , 8c , 8d , 8e , 8f , 9a , 9b , 9c , 9d , 9e , 9f ) were synthesized in moderate to high yields (49–94%) using the Hantzsch reaction with thiosemicarbazone derivatives ( 5a , 5b , 5c ) and 2‐bromo‐1‐phenylethanone derivatives ( 6a , 6b , 6c , 6d , 6e , 6f ). The structures of synthesized compounds were elucidated by IR, 1H NMR, 13C NMR, elemental analyses, mass spectroscopy and X‐ray diffraction analysis techniques. Moreover, the synthesized compounds were tested for their in vitro antifungal activity and most of them exhibited moderate to good activity against Fusariumoxysporumf.sp. lycopersici.   相似文献

Coumarins are the important class of naturally occurring heterocyclic compounds. Activities like antioxidant, antibacterial, anti‐inflammatory, and anticancer have been reported for coumarin derivatives. Present work details the synthesis of substituted coumarin‐4‐pyrrolones as well as coumarin‐4‐acetyl amino acids and their DHODH inhibitory activity, which is a dual target for malaria and cancer. Coumarin‐4‐acetic acids ( 2a – c ) were coupled with different methyl esters of α‐amino acids ( 3 ) giving rise to corresponding coumarin‐4‐acetyl amino acid methyl esters ( 4a – o ), which on hydrolysis under basic condition underwent cyclization forming substituted dihydropyrrole‐2‐ones ( 5a – i ), dihydroindolizine‐3‐ones ( 5j – l ), and dihydropyrrolizin‐3‐one ( 5m – o ). Acidic hydrolysis of the compounds ( 4a – o ) yielded corresponding coumarin‐4‐acetyl amino acids ( 6a – f ). The docking study was performed with the protein 4IGH (obtained from PDB) using Surflex–Dock module. The newly synthesized compounds were tested for DHODH inhibitory activity using Brequinar as the standard. Compound 6b showed remarkable inhibition compared with the standard, and the other compounds with terminal COOH showed moderate inhibition.  相似文献

2‐(4,5‐Dihydropyrazol‐1‐yl)‐thiazol‐4‐ones ( 2–5 ) have been synthesized starting from 3‐phenyl‐5‐aryl‐1‐thiocarbamoyl‐2‐pyrazolines via [2+3]‐cyclization with 2‐bromopropionic acid, maleic anhydride, N‐arylmaleimides, and aroylacrylic acids. The in vitro anticancer activity of 2a , 3a , 4a , 5b , and 5c were tested by the National Cancer Institute. Compounds 4a , 5b , and 5c demonstrated selective inhibition of leukemia cell lines growth at a single concentration (10?5 M). The screening of antiviral activity for a broad panel of viruses revealed that N‐(4‐methoxyphenyl)‐2‐{2‐[5‐(4‐methoxyphenyl)‐3‐phenyl‐4,5‐dihydropyrazol‐1‐yl]‐4‐oxo‐4,5‐dihydrothiazol‐5‐yl}‐acetamide 4a was highly active against Tacaribe TRVL 11 573 virus strain (EC50 = 0.71 μg/mL, selectivity index = 130).  相似文献

The scopes of three synthetic methods reported for the preparation of an array of 3‐pyrazolecarboxylates featuring substituents on position 4 were investigated. The first one is based on the potassium permanganate oxidation of methylpyrazoles. The second starts with the condensation between DMF dimethylacetal and ethyl pyruvate and is followed by the addition of hydrazine hydrochloride. The last one makes use of the cycloaddition of diazomethane on acrylate esters followed by a bromine‐based oxidative rearrangement into 4‐substituted 3‐pyrazole esters.  相似文献

1‐Benzotriazolylacetophenone 1 couples with aromatic diazonium salts to yield the corresponding coupling products 2 . Reaction of 1 with diazotized aminopyrazole afforded the benzotriazolylpyrazolo[5,1‐c][1,2,4]triazine 6 . Compound 1 condensed with DMFDMA to yield the enaminone 7 which reacted with hydrazines to yield the pyrazoles 8a,b . Isomeric pyrazoles 10 were synthesized via condensing 1 with phenyl‐hydrazine and subsequent condensation of the formed phenylhydrazone 9 with DMFDMA. Reaction of 7 with hydroxylamine afforded the isoxazole 11 which was converted into the nitrile 13 on reflux in dioxane in the presence of sodium hydride. Compound 13 was also directly obtained from reaction of 1 with 1‐cyanobenzo‐triazole. The reaction of 1 with hippuric acid and arylidenemalononitriles 18a‐c afforded the pyranone 17 and pyridine derivatives 23a‐c , respectively.  相似文献

To evaluate the effect of substituents on biological activities of electron‐rich N‐containing heterocycles, the variably 2‐substituted 5,6‐dihydro‐5‐oxo‐4H‐1,3,4‐oxadiazine‐4‐propanenitriles 26 – 33 were synthesized and evaluated for antibacterial, antifungal, and enzyme‐inhibition activities. The target compounds were obtained from alkyl 4‐ or 3‐hydroxy benzoates 1 and 2 , respectively, and from methyl indoleacetate 3 . The phenolic OH group of benzoates 1 and 2 were substituted with p‐toluenesulfonyl (→ 4 and 5 ), benzoyl (→ 6 and 7 ), and benzyl groups (→ 8 and 9 ) and then converted to 5,6‐dihydro‐5‐oxo‐4H‐1,3,4‐oxadiazine‐4‐propanenitriles. To establish structure‐activity relationships (SAR), a pharmacological screening of the intervening intermediates was also conducted, which revealed that the intermediate hydrazide 11 possesses significant antimicrobial and MAO‐A inhibiting properties and intermediates 12, 24, 28 , and 29 appreciable antifungal activities. In the present study, new 1,2,4‐triazoles, 1,3,4‐thiadiazoles, and acylthiosemicarbaz‐ides derived from 4‐(4‐chlorophenylsulfonyl)benzoic acid hydrazide were synthesized and screened for their antimicrobial and analgesic activities. Acylthiosemicarbazides 2–4 were synthesized by a reaction of 4‐(4‐chlorophenyl‐sulfonyl)benzoic acid hydrazide 1 with different arylisothiocyanates.4,5‐Disubstituted‐2,4‐dihydro‐3H‐1,2,4‐triazol‐3‐thiones 5–7 and 2,5‐disubstituted‐1,3,4‐thiadiazoles 8–10 were obtained by dehydrative cyclization of corresponding acylthiosemicarbazide derivatives 2–4 in basic media (8% aqueous sodium hydroxide) and in acidic media (sulfuric acid or phosphorous oxychloride), respectively. The structures of the newly synthesized compounds have been confirmed on the basis of elemental analysis and spectral studies (IR, 1H NMR, 13C NMR, MS). Their antimicrobial activities against some bacteria and yeasts were investigated. The analgesic activity of all compounds was performed with two pharmacological tests: the writhing test induced with acetic acid and hot‐plate test. The results showed that triazole 7 had the best antimicrobial activity against Bacillus cereus. In the chemical stimulus test, triazoles 6 and 7 were the most active compounds whereas in the hot‐plate test thiadiazoles 9 and 10 exhibited the highest analgesic activity.  相似文献

The dimethylamine in the adducts [(HNMe2)B(azolyl)3] (azolyl=methimazolyl, pyrazolyl), obtained by reaction of the azole with B(NMe2)3, can readily be substituted with a range of nitrogen donors to provide new charge‐neutral, tripodal ligands in high yield. This observation has led to a revision of an earlier interpretation of the mechanism of the formation of these species. The donor properties of the ligands [(nmi)B(azolyl)3] (nmi=N‐methylimidazole) have been compared with their anionic analogues [HB(azolyl)3]? by synthesis of their manganese(I)–tricarbonyl complexes and comparison of their infrared νCO energies. This comparison indicates that the new neutral ligands are only marginally weaker donors than the corresponding anionic hydrotris(azolyl)borate ligands. This may be explained by the ability of the attached nmi ring to stabilize a positive charge remotely from the coordinated metal, which may also account for the fact that the [(nmi)B(pyrazolyl)3] ligand is a substantially stronger donor than the similarly neutral tris(pyrazolyl)methane ligand.  相似文献

The reactions of 4‐substituted 3‐pyridinesulfonamides 1a , 1b , 1c , 1d , 1e with benzenesulfonyl chlorides 2a , 2b , 2c , 2d , 2e , 2f in acetonitrile were investigated. Depending on the structure of arylsulfonyl chlorides and the reaction conditions the following four types of products were obtained in good yields: 3‐sulfamoyl‐4‐R‐1‐arylpyridinium chlorides 3 , 4 , 5 , 6 , 7 , 8 ; 1,10‐bis(3‐nitrobenzenesulfonylimino)deca‐2,4,6,8‐tetraene‐2,9‐disulfonamides 9 , 10 , 11 , 12 ; 1‐substituted 1,4‐dihydro‐4‐oxo‐3‐pyridinesulfonamides 13 , 14 ; and 4‐methoxy‐3‐[N‐(2,5‐dichlorophenyl)sulfonyl] sulfonamidates 15 and 16 . Synthesis and properties of new imines and bisimines derived from 2‐phenyl‐1H‐imidazole‐4‐carbaldehyde and amines/diamines were studied. (2‐Phenyl‐1H‐imidazole‐4‐yl)methanol was oxidized to 2‐phenyl‐1H‐imidazole‐4‐carbaldehyde with better yield 55% by the modification of literature procedure. This aldehyde was condensed with the following achiral and chiral amines or 1,2‐diamines: ethanamine, propan‐1‐amine, butan‐1‐amine, 2‐methylpropan‐1‐amine, cyclohexanamine, (2R)‐ and (2S)‐3‐methylbutan‐2‐amine, (1R)‐ and (1S)‐1‐cyclohexylethanamine, (S)‐1‐aminopropan‐2‐ol, (S)‐1‐(2‐phenyl‐1H‐imidazol‐4‐yl)ethanamine, (S)‐1‐(2‐phenyl‐1H‐imidazol‐4‐yl)‐2‐methylpropan‐1‐amine, (S)‐1‐(2‐phenyl‐1H‐imidazol‐4‐yl)‐3‐methylbutan‐1‐amine, ethane‐1,2‐diamine, and (1R,2R)‐ and (1S,2S)‐cyclohexane‐1,2‐diamine. Sixteen condensation products, especially chiral imines and bisimines, were prepared by founded procedures in 45–99% of yields and characterized by the 1H NMR spectroscopy in solution, mass spectrometry, and elemental analyses. The optical rotation values in the case of chiral ones were also observed. Stability constants of Cu(II) complexes of selected prepared imines/bisimines were determined.  相似文献

A series of novel Mannich base derivatives (E1–E15) of 5‐aryl‐1,3,4‐oxadiazole‐2‐thione with substituted pyrimidine were synthesized and characterized by elemental analysis, IR, 1H‐NMR. The antifungal activities of these compounds were also originally studied. The results showed that most of the title compounds exhibited relatively good fungicidal activities. Especially compounds E8 and E13 showed better antifungal activity than comparison compound hymexazol. The relationship of structure and activity revealed that the presence of the methyl group at four and six positions of pyrimidine ring remarkably enhanced the antifungal activity of title compounds.  相似文献

A series of (Z)‐methyl‐2‐[(Z)‐3‐substituted‐4‐oxo‐2‐(2‐picolinoyl‐/thiophene‐2‐carbonyl)‐hydrazonothiazolidin‐5‐ylidene]acetates were synthesized by condensation N‐substituted‐(2‐picolinoyl‐, thiophene‐2‐carbonyl)hydrazinecarbothioamides with dimethylacetylenedicarboxylate. Novel substituted derivatives of 3‐aryl‐5‐cyano‐6‐methylthiopyrimidine‐2, 4‐diones were synthesized by the reaction of ethyl 2‐cyano‐3,3′‐dimethylthioacrylate with arylureas in good yields. The structures of all title compounds were evaluated by elemental analyses and 1H NMR spectra and compound 2c was also confirmed by X‐ray diffraction. A new 5‐arylidene‐4‐oxo‐(sulfonamoyl phenyl)‐spiro[thiazolidinone‐2,2′‐steroids] series (7–10) was prepared by condensation of sulfanilamide, sulfapyridine and sulfadiazine sulfa drugs with testosterone, epiandrosterone and progesterone steroids, respectively. The resultant imino derivatives 1–3 upon cycloaddition with thioacetic acid in dry 1,4‐dioxane afforded 3‐sulfo‐namoylphenylspiro[4‐oxo‐thiazolidin‐2,2′steroids] (4–6). The latter compounds (4–6) upon condensation with p‐fluorobenzaldehyde in ethanol‐piperidine yielded the corresponding 4‐fluoroarylidene derivatives 7, 8 & 9, respectively. All the newly synthesized compounds were confirmed by UV, IR, 1H NMR, 13C NMR, mass spectral data, elemental analysis and molecular weight determination. In vitro antimicrobial screening of some of the synthesized compounds showed good antimicrobial activities towards some pathogenic Gram‐positive, Gram‐negative bacteria and fungi vs. piperacillin and mycostatine antibiotics as standard antibacterial and antifungal agents, respectively. The voltammetric behavior of two newly spirothiazolidinone steroids ( 2a & 5a ) was critically studied. Compound 5a physically immobilized polyurethane foam solid sorbent was successfully used for removal and/or separation of bismuth(III) from water.  相似文献
