Ethyl 7‐amino‐3‐(3‐methyl‐5‐oxo‐1‐phenyl‐2‐pyrazolin‐4‐yl)‐5‐aryl‐5H‐thiazolo[3,2‐a]pyrimidine‐6‐carboxylate was hydrolyzed with an ethanolic sodium hydroxide and the sodium salt thus formed underwent cyclization with acetic anhydride to afford 2‐methyl‐7‐(3‐methyl‐5‐oxo‐1‐phenyl‐2‐pyrazolin‐4‐yl)‐5‐arylthiazolo[3,2‐a]pyrimido[4,5‐d]oxazin‐4(5H)‐one. This compound was transformed to related heterocyclic systems via its reaction with various reagents. The biological activity of the prepared compounds was tested against Gram positive and Gram negative bacteria as well as yeast‐like and filamentous fungi. They revealed in some cases excellent biocidal properties.  相似文献

Benzoylacetone has been reacted with phenyl isothiocyanate to afford two different thiocarbamoyl derivatives (α-phenylthiocarbamoyl benzoylacetone and benzoyl thioacetanilide) depending upon the base used to perform the reaction. Several new 2-substituted-4-sulfamoylphenylazo-thiophene and/or thiazole derivatives were synthesized by heterocyclization of the thiocarbamoyl derivatives with various halogenated reagents. The synthesized compounds were screened for their antibacterial activities; they showed accepted activities with respect to the control drugs.  相似文献

Ethyl 7‐amino‐3‐(3‐methyl‐5‐oxo‐1‐phenyl‐2‐pyrazolin‐4‐yl)‐5‐aryl‐5H‐thiazolo[3,2‐a]pyrimidine‐6‐carboxylate was synthesized by the reaction of 4‐(2‐aminothiazol‐4‐yl)‐3‐methyl‐5‐oxo‐1‐phenyl‐2‐pyrazoline with arylidene ethyl cyanoacetate and it transformed to related fused heterocyclic systems via reaction with various reagents. A series of new thiazolyl chalcones, 1‐[2‐amino‐4‐methyl‐1, 3‐thiazol‐5‐yl]‐3‐aryl‐prop‐2‐en‐1‐one were prepared by piperidine mediated Claisen‐Schmidt condensation of thiazolyl ketone with substituted aromatic aldehyde. These chalcones on cyclization gave 2‐amino‐6‐(2‐amino‐4‐methyl‐1,3‐thiazol‐5‐yl)‐4‐aryl‐4H‐pyridine‐3‐carbonitrile and 2‐amino‐6‐(2‐amino‐4‐methyl‐1,3‐thiazol‐5‐yl)‐4‐aryl‐4H‐pyran‐3‐carbonitrile. The results showed that this skeletal framework exhibited marked potency as antimicrobial agents. The most active antibacterial agent was 2‐amino‐6‐(2‐amino‐4‐methyl‐1,3‐thiazol‐5‐yl)‐4‐(4‐chlorophenyl)‐4H‐pyran‐3‐carbonitrile while 2‐amino‐6‐(2‐amino‐4‐methyl‐1,3‐thiazol‐5‐yl)‐4‐(4‐methoyphenyl)‐4H‐pyran‐3‐carbonitrile appeared to be the most active antifungal agent. 3‐(Methylthio)‐5‐oxo‐2‐(2‐phenyl‐1,3‐thiazol‐4‐yl)‐4,5‐substituted‐pent‐2‐enenitriles 3a–j were obtained in good yields by condensation of (2‐phenyl‐1,3‐thiazol‐4‐yl)‐acetonitrile (1) with various α‐oxoketene dithioacetals 2aj in the presence of sodium hydride. The intermediates 3aj underwent smooth cyclization in the presence of PTSA to afford the corresponding benzothiazoles 4aj in moderate yields.  相似文献

3‐Acetyl‐4‐hydroxy‐chromen‐2‐one ( 1 ) was brominated with phenyltrimethylammonium tribromide to afford 3‐(2‐bromoacetyl)‐4‐hydroxy‐chromen‐2‐one ( 2 ) whose reactions with thiourea, thioacetamide and ammonium dithiocarbamate gave respectively 3‐(2‐amino‐thiazol‐4‐yl)‐4‐hydroxy‐, 4‐hydroxy‐3‐(2‐phenyl‐thiazol‐4‐yl)‐ and 4‐hydroxy‐3‐(2‐mercapto‐thiazol‐4‐yl)chromen‐2‐one. In a similar manner, com pound 2 was treated with four 1‐substituted‐2‐thioureas and thiobenzamide to give the corresponding 4‐hydroxy‐3‐(thiazol‐4‐yl)‐chromen‐2‐one derivatives.  相似文献

2‐(4,5‐Dihydropyrazol‐1‐yl)‐thiazol‐4‐ones ( 2–5 ) have been synthesized starting from 3‐phenyl‐5‐aryl‐1‐thiocarbamoyl‐2‐pyrazolines via [2+3]‐cyclization with 2‐bromopropionic acid, maleic anhydride, N‐arylmaleimides, and aroylacrylic acids. The in vitro anticancer activity of 2a , 3a , 4a , 5b , and 5c were tested by the National Cancer Institute. Compounds 4a , 5b , and 5c demonstrated selective inhibition of leukemia cell lines growth at a single concentration (10?5 M). The screening of antiviral activity for a broad panel of viruses revealed that N‐(4‐methoxyphenyl)‐2‐{2‐[5‐(4‐methoxyphenyl)‐3‐phenyl‐4,5‐dihydropyrazol‐1‐yl]‐4‐oxo‐4,5‐dihydrothiazol‐5‐yl}‐acetamide 4a was highly active against Tacaribe TRVL 11 573 virus strain (EC50 = 0.71 μg/mL, selectivity index = 130).  相似文献

2‐Amino‐6‐(3‐methyl‐5‐oxo‐1‐phenyl‐2‐pyrazolin‐4‐yl)‐4‐phenylpyridine‐3‐carbonitrile (1) obtained by the reaction of 4‐(1‐iminoethyl)‐3‐methyl‐1‐phenyl‐2‐pyrazolin‐5‐one with benzylidenemalononitrile, was reacted with triethyl orthoformate followed by hydrazine hydrate, acetic anhydride, acetyl chloride, alkyl halides, benzoyl chloride, sulphuric acid followed by formamide, phenyl isothiocyanate, carbon disulphide followed by ethyl iodide, formamide, trichloroacetonitrile, nitrous acid, giving new oxopyrazolinylpyridines ( 2,3,5,6,8,9,10 ) and related pyridopyrimidines ( 11‐17 ) and pyridotriazine ( 18 ).  相似文献

4‐(1H‐benzo[d]imidazol‐2‐yl)thiazol‐2‐amine and its 1‐methyl derivative ( 1 ) were reacted with different reagents such as acid anhydrides, malononitrile, chloroacetyl chloride, and aromatic aldehydes to produce the corresponding benzimidazole products 2 , 3 , 4 , 5 , 6 , 7 , 8 , 9 , respectively. Also, 2‐chloro‐N‐(4‐(1‐methyl‐1H‐benzo[d]imidazol‐2‐yl)thiazol‐2‐yl) acetamide ( 6 ) was reacted with diaminoethane, ortho‐substituted aniline, thioglycolic acid, thiosemicarbazide derivatives, secondary amines, and potassium isothiocyanate to afford the corresponding derivatives 10 , 11 , 12 , 13 , 14 , 15 , 16 , 17 , respectively. The cytotoxic activity of some newly synthesized derivatives was studied against two different cell lines HepG2 and PC12. Compounds 9 and 15b showed promising anticancer activity against both types of the tested cancerous cell lines.  相似文献

A new series of 3‐substituted‐thiazolyl‐2‐iminothiazolidin‐4‐ones were synthesized by nucleophilic substitution of p‐substituted‐thiazol‐2‐yl‐chloroacetamides with potassium thiocyanide by cyclization. The starting material p‐substituted‐thiazol‐2‐yl‐chloroacetamides were synthesized from p‐substituted‐thiazol‐2‐yl‐amines with chloroacetyl chloride, which in turn was prepared from one pot reaction of substituted aryl acetophenone and amino group of thiourea. The title compounds were investigated for their anticonvulsant activity. Among the tested compounds, compound 3‐(4‐(4‐fluorophenyl)thiazol‐2‐yl)‐2‐iminothiazolidin‐4‐one ( 16 ) emerged as the most active compound of the series, and it is moderately more potent than the reference standard diazepam.  相似文献

The reaction of 1H‐indol‐2,3‐diones with 1,6‐dibromohexane has resulted in the formation of new 1H‐indol‐2,3‐diones‐1,1′‐(1,6‐hexanediyl)bis in quantitative yields. These compounds have been used for the synthesis of novel [3′‐(2,3‐dimethyl‐5‐oxo‐1‐phenyl‐3‐pyrazolin‐4‐yl)spiro[3H‐indol‐3,2′‐thiazolidine]‐2,4′‐dione]‐1,1′‐(1,6‐hexanediyl)bis via bis Schiff's bases, [3‐(2,3‐dimethyl‐5‐oxo‐1‐phenyl‐3‐pyrazolin‐4‐yl) imino‐1H‐indol‐2‐one]‐1,1′‐(1,6‐hexanediyl)bis.  相似文献

Reaction of 3‐(2‐bromo‐acetyl)‐chromen‐2‐one with thiosemicarbazide and 2‐acetylbutyro lactone in anhydrous ethanol gave 3‐{2‐[5‐hydroxy‐4‐(2‐hydroxy‐ethyl)‐3‐methyl‐pyrazol‐1‐yl]‐thiazol‐4‐yl}‐chromen‐2‐one in good yields.  相似文献

The 2‐ethoxy carbonyl methylene thiazol‐4‐one ( 3 ) reacts with acetophenone ( 4 ) to give the ethyl 2‐(4‐oxo‐4,5‐dihydro‐thiazol‐2‐yl)‐3‐phenyl‐2‐butenoate ( 5 ). The reactivity of the latter product towards aromatic aldehydes 6a‐d , cyanomethylene reagents 9a,b , aromatic aldehydes 13a‐d , phenylisothiocyanate ( 16 ), elemental sulfur and aromatic amines ( 20a‐c ) was studied to give arylidene, pyridine, thiophene and anilide derivatives. Some of the newly synthesized derivatives were used to synthesize fused derivatives. The antimicrobial activities of the newly synthesized products were tested in vitro for antimicrobial activity against two bacterial isolates, one saprophytic (Escherichia coli) and the other parasitic (Xanthomonas citri) and for antifungal activity against one saprophytic (Aspergillus fumigatus) and two phytopathogenics (Rhizoctonia solani and Fusarium oxysporum).  相似文献

Thiazole 1 , when reacted with chloroacetyl chloride, afforded N‐(5‐acetyl‐4‐methylthiazol‐2‐yl) chloroacetamide 2 . It has been found that compound 2 reacted with α‐cyanocinnamonitrile derivatives 6a–c to afford reaction products 8a–c . Also, compound 2 coupled smoothly with benzenediazonium chloride afforded the phenylhydrazone 14 . Coupling of the sulfonium bromide 17 with diazotized aromatic amines or N‐nitrosoacetanilides afforded the arylhydrazones 20a,b . Eight new 2‐methyl‐4(3H)‐quinazolinones (8a‐8d, 9c, 9d, 10c, 10d) with one or two chlorine atoms in the benzene ring and a 5‐methyl‐1,3‐thiazol‐2‐yl, 4‐methyl‐1,3‐thiazol‐2‐yl, and 5‐ethyl‐1,3,4‐thiadiazol‐2‐yl substituent in position 3 of the heterocyclic ring were synthesized and characterized. The two step procedure (Scheme 1) utilizes chlorosubstituted anthranilic acids (3a‐3d) and acetic anhydride as the starting materials, with the respective chlorosubstituted 2‐methyl‐4H‐3,1‐benzoxazin‐4‐ones (4a‐4d) as the intermediates. The quinazoline derivatives were characterized by their melting points, elemental analyses and the mass, ultraviolet, infrared, and 1H and 13C nmr spectra. Efficient and suitable methods for the synthesis of novel class of simple and fused heterocyclic compounds were prepared starting with 1‐naphthyl‐2‐cyanoacetamide and commercially available reagents. The cyclocondensation of 1‐naphthyl‐2‐cyanoacetamide with sulfanylacetic acid furnished phenylthiazolinone derivative. Stirring of the starting compound with PhNCS afforded thiocarbamoyl derivative which underwent heterocyclization with chloroacetyl chloride to give thiazolinone derivative. 5‐Aminopyrazole derivative was prepared by following mild procedures via refluxing the last thiocarbamoyl with hydrazine hydrate. Different synthetic approaches were discussed to obtain the novel fused pyrazolo[1,5‐a ]pyrimidine, 4H‐pyrazolo[3,4‐d ]pyrimidin‐4‐one moieties involving the reaction of the prepared 5‐aminopyrazole with a ) 1, 3‐dielectrophilic centers (acetylacetone, acetoacetanilide), b ) arylidines of malononitrile, and c ) isothiocyanate derivatives. The action of iced sodium nitrite solution in acidic medium on the last 5‐aminopyrazole gave pyrazolo[3,4‐d ][1,2,3]triazine. All novel structure were elucidated by different spectroscopic data (IR, MS, 1H, and 13C NMR) and elemental analysis.  相似文献

In the present investigation, a novel series of 3‐(4‐(2‐substituted thiazol‐4‐yl)phenyl)‐2‐(4‐methyl‐2‐substituted thiazol‐5‐yl)thiazolidin‐4‐one derivatives were synthesized by condensation of 2‐substituted‐4‐methylthiazole‐5‐carbaldehyde with 4‐(2‐substituted thiazol‐4‐yl)benzenamine followed by cyclo‐condensation with thioglycolic acid in toluene. All the newly synthesized compounds were characterized by spectral (IR, 1H NMR, 13C NMR, and Mass) methods. The title compounds were screened for quantitative antibacterial activity (minimal inhibitory concentration). All compounds 7a , 7b , 7c , 7d , 7e , 7f , 7g , 7h and 8a , 8b , 8c , 8d , 8e , 8f , 8g , 8h show moderate to good antimicrobial activity, whereas compounds ( 7a , 7b , 7c , 7d , 7e , 7f , 7g , 7h ) also show moderate antifungal activity.  相似文献

In the present investigation, we have utilized 3‐[3′‐(5″‐aryl‐2″‐pyrazolin‐3″‐yl)]phenylsydnones 1a–c to couple two biodynamic moieties in amide derivatives 7 , 8 , 9 , 10 . The compounds 1a–c were brominated to 4‐bromo‐3‐[3′‐(5″‐aryl‐2″‐pyrazolin‐3″‐yl)]phenylsydnone 2 , which were further reacted with secondary amines to form the title compounds. These novel molecules were subjected to antitubercular activity against Mycobacterium tuberculae (H37Rv) and antioxidant activity assay.  相似文献
