A series of new 4‐aryl‐4H‐chromene‐3‐carbonitrile derivatives were obtained by one‐pot synthesis using substituted benzaldehydes, malononitrile, and substituted phenols. All the synthesized compounds ( 1a , 1b , 1c , 1d , 1e ) were screened in vitro for antioxidant and anticancer activities. Compounds 1c , 1d , 1e showed significant antioxidant activity in nitric oxide free radical scavenging method while compounds 1c and 1e showed significant activity in hydrogen peroxide free radical scavenging method. The other compounds showed significant to moderate activities in both the methods in comparison with ascorbic acid and butylated hydroxytoluene as standards. Compounds 1c , 1d , 1e exhibited good anticancer activity, using Michigan Cancer Foundation‐7 (MCF‐7) cell line, compared with those of other synthesized compounds.  相似文献

A series of novel substituted (E)‐N′‐benzylidene‐4‐methyl‐2‐(2‐propylpyridin‐4‐yl)thiazole‐5‐carbohydrazide derivatives ( 6a‐l ) have been synthesized by following the multistep synthetic route starting from prothionamide. The resulting compounds were characterized via 1H, 13C NMR, and HRMS spectral data. The synthesized carbohydrazides were evaluated for their in vitro antimicrobial and antioxidant activities. Tested molecules have displayed moderate to good growth inhibition activity. Among the screened compounds, 6b , 6e , 6j, and 6k are found to be the more promising antimicrobial agents. A 2,2‐diphenyl‐1‐picrylhydrazyl assay was used to test the antioxidant activity of the carbohydrazides. The carbohydrazide derivatives 6b and 6i have shown better free‐radical scavenging ability than the other investigated compounds.  相似文献

A simple and efficient synthesis of aryl‐5H‐dibenzo[b,i]xanthene‐5,7,12,14‐(13H)‐tetraones and 3,3‐(arylmethylene)bis(2‐hydroxynaphthalene‐1,4‐diones) by the condensation of aromatic aldehydes and 2‐hydroxy‐1,4‐naphthoquinone under extremely mild conditions using catalytic amount of H2SO4 or in the presence of acidic ionic liquid 1‐butyl‐3‐methylimidazolium hydrogen sulphate, which could be recycled, has been reported. The radical scavenging capacity of the synthesized compounds has been examined towards the stable free radical 2,2‐diphenyl‐1‐picrylhydrazyl (DPPH), and the compounds 2 were found to scavenge DPPH free radical efficiently.  相似文献

Some novel [1,2,4]triazolo[3,4‐b][1,3,4]thiadiazole derivatives were synthesized from aryl acetic acids. All the synthesized derivatives were selected for the screening of antibacterial potential against Gram‐positive bacteria [Staphylococcus aureus (MTCC 3160) and Micrococcus luteus (MTCC 1538)] and Gram‐negative bacteria [Escherichia coli (MTCC 1652) and Pseudomonas aeruginosa (MTCC 424)] and antifungal potential against Aspergillus niger (MTCC 8652) and Candida albicans (MTCC 227), and free radical scavenging activity through 2,2‐diphenyl‐2‐picrylhydrazyl hydrate method. The compounds TH‐4 , TH‐13 , and TH‐19 were found to be more potent antimicrobial agents compared to standard drugs. The compounds TH‐3 , TH‐9 , and TH‐18 also showed significant antimicrobial activity. The compound TH‐13 showed antioxidant activity with IC50 value better than the standard compound. The structures of all the synthesized compounds were confirmed by Fourier transform infrared, 1H‐NMR, liquid chromatography–mass spectrometry, and CHN analyzer.  相似文献

3‐Methyl‐1‐phenyl‐4‐((2‐phenyl‐1H‐indol‐3‐yl)methylene)‐1H‐pyrazol‐5(4H)‐ones (5a‐i) was prepared by the condensation reaction of different 3‐formyl‐2‐phenylindole derivatives (2a‐i) and 3‐methyl‐1‐phenyl‐2‐pyrazoline‐5‐one in quantitative yield by applying various green synthetic methods as grinding, microwave irradiation using different catalysts under solvent‐free mild reaction conditions with high product yields. The structures of the synthesized compounds were characterized on the basis of elemental analysis, infrared, 1HNMR, 13C NMR, and mass spectral data. The synthesized compounds were screened for free radical scavenging, antimicrobial, and DNA cleavage activities. Most of the tested compounds belonging to the 3‐methyl‐1‐phenyl‐4‐((2‐phenyl‐1H‐indol‐3‐yl)methylene)‐1H‐pyrazol‐5(4H)‐ones series exhibited promising activities.  相似文献

A series of novel bispyrazoles joined by arylmethylene at C‐4 position were synthesized with aromatic aldehydes obtained from lignin and screened for their in vitro antioxidant activities by N,N‐diphenyl‐N′‐picrylhydrazyl (DPPH) and 2,2′‐azino‐bis(3‐ethylenzothiazoline‐sulphonic acid) diammonium salt (ABTS+) radical scavenging assays. All of these compounds exhibited good DPPH and ABST+ radical scavenging activities as compared to the standard, Trolox, which suggested their potential as promising agents for curing tumors or other free radical‐related diseases.  相似文献

A series of new α‐aminophosphonates containing 1,3,4‐thiadiazole moiety (4a–l) were synthesized via a simple, efficient, and one‐pot three‐component Kabachnik–Fields reaction of 2‐amino‐5‐ethyl‐1,3,4‐thiadiazole with various aryl/heteroaryl aldehydes and diethylphosphite under solvent‐free microwave irradiation conditions using phosphosulfonic acid, as a reusable and heterogeneous solid acid catalyst. All the title compounds were screened for radical scavenging activity by DPPH and H2O2 methods, and antimicrobial activity against bacteria (Gram‐positive and Gram‐negative) and fungi using the disc diffusion technique. They exhibited potent in vitro antioxidant and moderate antimicrobial activity.  相似文献

Novel N‐aryl (and N‐alkyl) γ‐ and δ‐imino esters 2a–g ( 3a–g ) and N‐aryl (and N‐alkyl) ketimines 2h–j ( 3h–j ) were synthesized in high yields (80–99%) from their corresponding γ‐ and δ‐keto esters and ketones in this study. The structures of the synthesized compounds were clarified by Fourier transform infrared (FT‐IR), NMR (1H and 13C), mass spectrometry, and elemental analyses. Isomerizations [E/Z] were also determined by their 1H NMR spectra. The free‐radical scavenging activity of imines was evaluated using the 1,1‐diphenyl‐2‐picryl‐hydrazyl (DPPH) method. The relationships between the structure and antioxidant activity of these compounds are discussed. Among these compounds, 2a–c (at the concentration 1000 μg/mL) exhibit high antioxidant activity similar to those of the standards (butylated hydroxyanisole [ BHA], butylated hydroxytoluene [ BHT], and ascorbic acid).  相似文献

Fourteen novel [1,2,4]triazolo[3,4-b][1,3,4]thiadiazine derivatives bearing benzimidazole moiety ( 7a-n ) have been synthesized using the one-pot nitro reductive cyclization method. All the synthesized compounds were confirmed by 1H nuclear magnetic resonance (1H NMR), 13C NMR, fourier-transform infrared (FT-IR), mass spectrum, and elemental analyses. All the title compounds were subjected to in vitro antioxidant activity. The free radical scavenging activity of the compounds was examined using DPPH, nitric oxide, and superoxide radical scavenging methods. The results demonstrated that compound 3-(2-(3,4-dimethoxyphenyl)-1-propyl-1H-benzo[d]imidazol-5-yl)-6-4-tolyl-7H-[1,2,4]triazolo[3,4-b][1,3,4]thiadiazine ( 7c ) was potent in scavenging both DPPH and nitric oxide radical with IC50 values of 13.57 and 18.55 μg/ml when compared to the standard with IC50 values of 23.75 and 23.14 μg/ml, respectively, which was due to the presence of electron-donating groups. The activity was found to decline when electron-donating groups were replaced by electron-withdrawing groups. Moderate scavenging activity was observed for the superoxide radical. Structure activity relationship and physiochemical properties were studied for all the derivatives.  相似文献

The antioxidant activity of two primary antioxidants, 2,2′‐methylenebis(4‐methyl‐6‐tert ‐butylphenol) (MMBPH2) and 2,2′‐methylenebis(4,6‐di‐tert ‐butylphenol) (MDBPH2), has been studied using the 1,1‐diphenyl‐2‐picrylhydrazyl (DPPH) method. The synthesized compounds have been successfully characterized systematically using elemental analyses, infrared, 1H NMR and 13C NMR spectra and GC–MS. Importantly, it has been found that the compound MMBPH2 in particular is more active in DPPH radical scavenging. In addition, density functional theory calculations (B3LYP) have been used to predict the antioxidant activity and predict structural geometries of the compounds in the gas phase.  相似文献

Previously unknown 3′‐R1‐5‐R2‐spiro[indoline‐3,6′‐[1,2,4]triazino[2,3‐c]quinazoline]‐2,2′‐(7′H)‐diones and their N‐substituted analogues were obtained via reaction of 6‐R1‐3‐(2‐aminophenyl)‐1,2,4‐triazin‐5‐ones with isatin and its substituted derivatives. It was shown that alkylation of 3′‐R1‐5‐R2‐spiro[indoline‐3,6′‐[1,2,4]triazino[2,3‐c]quinazolin]‐2,2′‐(7′H)‐diones by N‐R3‐chloroacetamides or chloroacetonitrile in the presence of а base proceeds by N‐1 atom of isatin fragment. The spectral properties (1H and 13C NMR spectra) of synthesized compounds were studied, and features of spectral patterns were discussed. The high‐effective anticonvulsant and radical scavenging agents among 3′‐R1‐5‐R2‐spiro[indoline‐3,6′‐[1,2,4]triazino[2,3‐c]quinazolin]‐2,2′(7′H)‐diones and their N‐substituted derivatives were detected. It was shown that compounds 2.2 , 2.8 , and 3.1 exceed or compete the activity of the most widely used in modern neurology drug—lamotrigine on the pentylenetetrazole‐induced seizures model. The aforementioned fact may be considered as a reason for further profound study of synthesized compounds using other pathology models.  相似文献

Clinacanthus nutans has been used in traditional herbal medicine for cancer prevention, but the specific bioactive compounds responsible for the observed activities have not been explored. Different polar solvents such as methanol, chloroform, ethyl acetate, and hexane were used for the extraction. The extracts, fractions, and isolated compounds were subjected to DPPH and ferric reducing antioxidant potential (FRAP) assays. Methanol extracts show significant free‐radical scavenging activity of 69.09% in DPPH and 56.49% FRAP. Purification of MeOH extracts afforded the fraction FB28 and two new sulfur‐containing compounds, named clinamide D and E ( 1 , 2 ). Compound ( 1 ) proved to be more active with an IC50 value for DPPH radical scavenging of 118.27 ± 0.01 µg/mL and reduction of Fe3+–TPTZ complex of 386.24 ± 0.02, higher than that of the standard ascorbic acid. Sulfur‐containing compounds isolated from C. nutans is a potential natural antioxidant.  相似文献

Bis(N‐ethylbenzimidazol‐2‐ylmethyl)aniline (Etbba) and its transition metal complexes, [Cu(Etbba)(Cl)2]?DMF ( 1 ), [Mn(Etbba)(Cl)2] ( 2 ) and [Ni(Etbba)(Cl)2] ( 3 ), have been synthesized and characterized on the basis of elemental analysis, molar conductivity, UV–visible, infrared and NMR spectroscopies and X‐ray crystallography. The coordination environment of complex 1 can be described as distorted square‐based pyramidal, while complexes 2 and 3 each have a distorted trigonal bipyramidal geometry. Cyclic voltammograms of complex 1 indicate an electrochemically quasi‐reversible Cu2+/Cu+ couple. In addition, the antioxidant activities of the free ligand and its complexes were investigated using the superoxide and hydroxyl radical scavenging methods in vitro. Complexes 1 , 2 , 3 are found to possess potent hydroxyl radical scavenging activity and to be better than standard antioxidants like vitamin C and mannitol. A novel series of α‐(benzoylamino)‐β‐substituted acrylic amide derivatives of pyrazolo[1,5‐a]pyrimidine has been synthesized using a convergent multistep synthesis. The synthesized compounds were characterized by 1H NMR, 13C NMR, ESI‐MS, and IR analyses. Those new compounds were screened for their in vitro antiproliferative activity using an MTT assay analysis. Out of nine derivatives synthesized in the current study, compounds 13g , 13d , 13h , and 13i exhibited the greatest anticancer activities in HeLa and HepG2 cell lines. The in vitro anticancer activity of compound 13g against HeLa, HepG2, and MCF‐7 cell lines is superior to the marketed drugs paclitaxel and SAHA.  相似文献

A novel series of (Z)‐1‐(arylhydrazono)‐1‐(benzothiazol‐2′‐ylthio)propan‐2‐ones have been prepared and their utility as building blocks in the synthesis of novel derivatives of Pyridazin‐6(1H)‐ones, pyridazin‐6(1H)‐imines, and phthalazine incorporating a benzothiazole residue are investigated. All of the title compounds were confirmed by elemental analysis, IR, 1H NMR, 13C NMR, NOE, and mass spectrometry. The synthesized compounds are screened for their antibacterial as well as antifungal activity. All the tested compounds showed high to moderate activity against the selected microorganisms. J. The radical‐scavenging ability of synthesized C4‐phenolic‐substituted 1,4‐dihydropyridines (1,4‐DHPs) toward 2,2‐diphenyl‐1‐picrylhydrazyl (DPPH?) and alkyl/alkylperoxyl ABAP‐derived radicals at pH 7.4 was assessed by UV–visible spectroscopy. Reactivity of 1,4‐DHPs toward DPPH? was measured by following the decay of the absorption corresponding to the radical λmax at 525 nm, permitting the calculation of EC50, tEC50, and antiradical efficiency values. Pseudo–first‐order kinetic rate constants for the reactivity between the C4‐phenolic‐substituted 1,4‐DHP compounds and alkyl/alkylperoxyl ABAP‐derived radicals were followed by the decrease in λmax at 356 nm corresponding to 1,4‐DHP moiety. C4‐phenolic‐substituted 1,4‐DHPs were more reactive toward alkyl free radicals than the other tested radicals. The 3,4,5‐trihydroxyphenyl‐1,4‐DHP was the most reactive derivative toward this radical with a kinetic rate constant value of 513.2 s?1. Also, this derivative was the most effective toward the DPPH? radical with the lowest EC50 value (5.08 µM). Comparative studies revealed that synthesized 1,4‐DHPs were more reactive than commercial 1,4‐DHPs. The scavenging mechanism involves the contribution of both pharmacophores, that is, hydroxyphenyl and 1,4‐DHP rings, which was supported by the identification of the reaction products. © 2012 Wiley Periodicals, Inc. A new 4‐hydroxy‐3‐carboxycoumarin ligand and its ruthenium(II) complexes ( 1 – 5 ) have been synthesized, characterized and screened for their in vitro antibacterial activity against a range of Gram‐positive and Gram‐negative bacteria. In addition, compounds 1 – 5 were investigated for antioxidant activities using superoxide radical, 2,2‐diphenyl‐1‐picrylhydrazyl radical and hydroxyl radical scavenging assays, in which most of them displayed significant antioxidant activities. Furthermore, compounds 1 – 5 were evaluated for anti‐inflammatory activity using indirect haemolytic and lipoxygenase inhibition assays and revealed good activity. The new complexes were characterized using spectroscopic methods in addition to elemental analysis.  相似文献

The synthesis of the title compounds was accomplished in four steps. The synthetic route involves the preparation of Schiff's base by reacting salicylaldehyde with m‐chloroaniline in EtOH. The Schiff's base was then reduced with NaBH4/MeOH. In the second step, PCl3 was reacted with p‐chlorophenol/p‐bromophenol in THF in the presence of Et3N to obtain P(III) dichloride derivatives. The reduced Schiff's base and dichloride derivatives were reacted in equimolar quantities in the presence of Et3N in THF to get the cyclized product. Alkyl azides were prepared by reacting alkyl bromides with sodium azide, and then alkyl azides were treated with the cyclized product to obtain the title compounds. The structure of these novel compounds was elucidated by elemental analysis, IR, 1H, 13C, 31P NMR, and mass spectroscopy. © 2010 Wiley Periodicals, Inc. Synthesis of novel 4-(4-methylsulfonylphenyl)-3-phenyl-2(3H)-thiazole thione derivatives with functionalized diarylheterocycle pharmacophore as potential COX-2 inhibitors was described. The title compounds were synthesized by cyclocondensation of corresponding dithiocarbamate and 2-bromo-1-(4-methylsulfonylphenyl)ethanone, followed by dehydration with H2SO4. On the basis of the principle of combination of active groups, a series of novel N‐(4‐([2,2′:5′,2′′‐terthiophen]‐5‐yl)‐2‐methylbut‐3‐yn‐2‐yl) benzamide derivatives were designed, synthesized and systematically evaluated for their antiviral activity against tobacco mosaic virus (TMV). The bioassay results showed that most of these compounds displayed good anti‐TMV activity, and some of them exhibited higher antiviral activity than commercial Ningnanmycin. Especially, compound 8e with excellent anti‐TMV activity (inactivation activity, 92.3%/500 µg·mL?1; curative activity, 85.7%/500 µg·mL?1 and protection activity, 64.7%/500 µg·mL?1) emerged as a potential inhibitor of plant virus TMV. Quantitative structure‐activity relationship studies proved that the van der Waals volume (V) and electronic parameter (∑(∑σo+σp) and ∑σm) for the substituent R1 were very important for antiviral activities in this class of compounds.  相似文献
