The effect of hydrophobic alkylated gold nanoparticles (Au NPs) on the phase behavior and structure of Langmuir monolayers of dipalmitoylphosphatidylcholine (DPPC) and Survanta, a naturally derived commercial pulmonary surfactant that contains DPPC as the main lipid component and hydrophobic surfactant proteins SP-B and SP-C, has been investigated in connection with the potential implication of inorganic NPs in pulmonary surfactant dysfunction. Hexadecanethiolate-capped Au NPs (C(16)SAu NPs) with an average core diameter of 2 nm have been incorporated into DPPC monolayers in concentrations ranging from 0.1 to 0.5 mol %. Concentrations of up to 0.2 mol % in DPPC and 16 wt % in Survanta do not affect the monolayer phase behavior at 20 °C, as evidenced by surface pressure-area (π-A) and ellipsometric isotherms. The monolayer structure at the air/water interface was imaged as a function of the surface pressure by Brewster angle microscopy (BAM). In the liquid-expanded/liquid-condensed phase coexistence region of DPPC, the presence of 0.2 mol % C(16)SAu NPs causes the formation of many small, circular, condensed lipid domains, in contrast to the characteristic larger multilobes formed by pure lipid. Condensed domains of similar size and shape to those of DPPC with 0.2 mol % C(16)SAu NPs are formed by compressing Survanta, and these are not affected by the C(16)SAu NPs. Atomic force microscopy images of Langmuir-Schaefer-deposited films support the BAM observations and reveal, moreover, that at high surface pressures (i.e., 35 and 45 mN m(-1)) the C(16)SAu NPs form honeycomb-like aggregates around the polygonal condensed DPPC domains. In the Survanta monolayers, the C(16)SAu NPs were found to accumulate together with the proteins in the liquid-expanded phase around the circular condensed lipid domains. In conclusion, the presence of hydrophobic C(16)SAu NPs in amounts that do not influence the π-A isotherm alters the nucleation, growth, and morphology of the condensed domains in monolayers of DPPC but not of those of Survanta. Systematic investigations of the effect of the interaction of chemically defined NPs with the lipid and protein components of lung surfactant on the physicochemical properties of surfactant films are pertinent to understanding how inhaled NPs impact pulmonary function.  相似文献

By screening uronic acid-based surfactant interfacial properties, the effect of the hydroxyl group stereochemistry (OH-4) on the conformation of bicatenary (disubstituted) derivatives at the air-water interface has been evidenced by experimental and computational approaches. Physical and optical properties of a monolayer characterized by Langmuir film balance, Brewster angle microscopy, and ellipsometry at 20 °C reveal that the derivative of glucuronate (C(14/14)-GlcA) forms a more expanded monolayer, and shows a transition state under compression, in the opposite to that of galacturonate (C(14/14)-GalA). Both films are very mechanically resistant (compression modulus > 300 mN m(-1)) and stable (collapse pressure exceeding 60 mN m(-1)), while that of C(14/14)-GalA exhibits a very high compression modulus up to 600 mN m(-1) like films in the solid state. Computational approaches provide single and assembly molecular models that corroborate the molecule expansion degree and interactions data from experimental results. Differences in the molecular conformation and film behaviours of uronic acid bicatenary derivatives at the air-water interface are attributed to the intra-H-bonding formation, which is more favourable with an OH-4 in the axial (C(14/14)-GalA) than in the equatorial position (C(14/14)-GlcA).  相似文献

We have studied the structure of films made by low density lipoproteins (LDL) from hen egg yolk, which are composed of apoproteins, neutral lipids and phospholipids. These LDL have been deposited on air–water interface to form a monolayer which has been compressed to measure an isotherm using Langmuir balance. This isotherm presented three transitions (neutral lipid (surface pressure, π = 19 mN/m), apoprotein–lipid (π = 41 mN/m) and phospholipid (π = 51 mN/m) transitions). We have studied only the apoprotein–lipid transition. In order to observe the LDL film structure before (π = 30 mN/m) and after (π = 45 mN/m) the apoprotein–lipid transition, the formed films were transferred and visualised by atomic force microscopy (AFM). Our results have shown that the structures observed in the LDL film were different depending on the surface pressure. The apoproteins and neutral lipids appeared to be miscible up to the apoprotein–lipid transition, when demixing occured. The structures observed after the apoprotein–lipid transition should be due to the demixing between apoproteins and neutral lipids. On the other hand, apoproteins and phospholipids seemed miscible whatever the surface pressure. Hence, the first transition (π = 19 mN/m) should be attributed to the free neutral lipid collapse; the second transition (π = 41 mN/m) should be attributed to the demixing of apoprotein–neutral lipid complexes; and the last transition (π = 51 mN/m) should be attributed to phospholipid collapse or to demixing of apoprotein–phospholipid complexes.  相似文献

Natural lung surfactant contains less than 40% disaturated phospholipids, mainly dipalmitoylphosphatidylcholine (DPPC). The mechanism by which lung surfactant achieves very low near-zero surface tensions, well below its equilibrium value, is not fully understood. To date, the low surface tension of lung surfactant is usually explained by a squeeze-out model which predicts that upon film compression non-DPPC components are gradually excluded from the air-water interface into a surface-associated surfactant reservoir. However, detailed experimental evidence of the squeeze-out within the physiologically relevant high surface pressure range is still lacking. In the present work, we studied four animal-derived clinical surfactant preparations, including Survanta, Curosurf, Infasurf, and BLES. By comparing compression isotherms and lateral structures of these surfactant films obtained by atomic force microscopy within the physiologically relevant high surface pressure range, we have derived an updated squeeze-out model. Our model suggests that the squeeze-out originates from fluid phases of a phase-separated monolayer. The squeeze-out process follows a nucleation-growth model and only occurs within a narrow surface pressure range around the equilibrium spreading pressure of lung surfactant. After the squeeze-out, three-dimensional nuclei stop growing, thereby resulting in a DPPC-enriched interfacial monolayer to reduce the air-water surface tension to very low values.  相似文献

Fluid monolayers of 1-palmitoyl-2-oleoyl-phosphatidylcholine collapse from an air/water interface to form a three-dimensional bulk phase at the equilibrium spreading pressure (pie) of approximately 47 mN/m. This phase transition limits access to higher surface pressures under equilibrium conditions or during slow continuous compressions. We have shown previously that these films avoid collapse and become metastable when compressed on a captive bubble to surface pressures above 60 mN/m and that the metastability persists during expansion at least to pie. Here, we first documented the extent of this persistent metastability. Rates of isobaric collapse during expansion of the metastable films were up to 3 orders of magnitude slower than those during the initial compression to high surface pressures. Recovery of the ability to collapse depended on the surface pressure to which the films were expanded and how long they were held there. Films reverted after brief exposure to 20 mN/m and after 1 h at 35 mN/m. At pie, films remained capable of reaching high surface pressures during slow compressions after 65 h, although an increase in compressibility above 55 mN/m suggested somewhat increased rates of collapse. We also determined if the films remained metastable when they acquired sufficient free area to allow reinsertion of collapsed material. Faster isobaric expansion in the presence of more collapsed material and with further deviation below pie supported the existence of reinsertion. The persistence of metastability to pie shows that films with sufficient free area to allow reinsertion remain resistant to collapse. Observations that suggest heterogeneous reinsertion, however, argue that free area may be distributed heterogeneously and leave open the possibility that metastability persists because significant regions retain a restricted free area.  相似文献

The 2D complex formed at the air-water interface between the dialkyl chain cationic surfactant, dihexadecyldimethylammonium bromide, and the anionic porphyrin, tetrakis-(4-sulfonatophenyl) porphine, was studied using surface pressure-area isotherms as well as X-ray and neutron reflection measurements. The surface structure of these films was determined by the use of simultaneously constrained analysis of the neutron and X-ray reflectometry data and BAM images. Isotopic contrast variation methods were employed to enhance the information content of the neutron reflection data. The rigid complex forms at the interface due to the electrostatic interaction between the cationic headgroups of the surfactant and the anionic functional groups at the meso position of the porphyrin. The surface pressure-area isotherms show three distinct regions on compression: an initial condensed phase that ends with a pressure peak at 36 mN m-1, a second plateau region of high compressibility, and a final condensed phase. BAM images show that at the beginning of the plateau region in the isotherm there is complete surface coverage by a monolayer. The constrained simultaneous fitting of neutron and X-ray data measured just prior to and after the pressure peak shows a structurally similar 2D complex at the interface. Modeling of X-ray reflectometry data also reveals that in the final high-pressure phase the film has folded to form a trilayer. The conclusion is that the plateau region of the isotherm is due to the formation of trilayer surface coverage through localized buckling or folding, and that after this is complete there is some condensation before final film collapse.  相似文献

A comparison of the collapse of Langmuir monolayers of docosyl trifluoroethyl ether (DFEE) and docosyl ethyl ether (DEE) on water shows that in both films the 3D phase is formed layer-by-layer. The substitution of CH3 by a CF3 group in the hydrophilic head yields a more stable bilayer exhibiting lower equilibrium spreading pressure, pi(esp)(DFEE) < pi(esp)(DEE). Upon lateral compression, the DFEE bilayer fractures abruptly as a compact solid body whereas the DEE bilayer breaks down gradually as a polycrystalline material. A comparison of the collapse kinetics of the two films at the same constant supersaturation pi-pi(esp) = 7 mN/m shows that the fluorinated DFEE monolayer transforms more quickly, yielding a stable bilayer of closely packed upright molecules, whereas the DEE film undergoes a continuous monolayer-bilayer-multilayer transition. Brewster angle microscopy allows us to visualize different collapse mechanisms of the DFEE and DEE films; the domains of the fluorinated DFEE bilayer grow laterally at constant thickness and density, and the collapse of the nonfluorinated DEE monolayer occurs through a sequence of disordered stripelike and broken elongated textures. The characteristic molecular areas of the monolayer and bilayer collapse suggest that the 2D-3D transition in the DFEE and DEE films is accompanied by at least partial dehydration of their headgroups. The faster collapse of the fluorinated monolayer could result from a lower energy barrier due to the more hydrophobic CF3 group in the heads. The increased stability of the DFEE bilayer could be associated with the electrostatic attraction between the -C(F delta-)3 versus (H delta+)3C- terminals at the heads-to-tails contact plane of the top and the bottom layer, contrasting with the repulsion between the -C(H delta+)3 versus (H delta+)3C- terminals of the top-layer heads and the bottom-layer tails in the DEE bilayer.  相似文献

In this work we have analyzed the topography by atomic force microscopy (AFM) of dipalmitoyl-phosphatidyl-choline (DPPC) monolayers previously spread at the air–water interface and penetrated by β-casein. AFM images of β-casein–DPPC monolayers were taken from Langmuir–Blodgett films deposited onto hydrophilic mica substrates at different initial surface pressures (πi) and after the compression of the mixed films. The monolayer topography depends on the initial structure of the phospholipid:liquid expanded (LE) at 3 mN/m, coexistence between LE and liquid condensed (LC) structures at 7 mN/m, at the end of the LE–LC transition at 10 mN/m, and with a LC structure at 15 mN/m. The area occupied by DPPC domains in the mixed film increases with the πi value, especially for DPPC with a LC structure at 15 mN/m. At this surface pressure the thickness of the film is at a maximum. After the film compression at 25 mN/m, which is above the equilibrium spreading pressure of β-casein (), this protein is displaced from the interface by DPPC and the topography of the mixed monolayer depends on the initial structure of the DPPC monolayer. A notable feature of the topography of these mixed monolayers is the presence of multilayers of β-casein and DPPC of high thickness (50–70 nm) at the lower πi values. Although the film is dominated by DPPC at the highest surface pressures (at 25 mN/m), β-casein is not displaced totally from the interface and coexists as β-casein collapsed domains within the network of the DPPC structure.  相似文献

In this work we have analyzed the penetration of betalactoglobulin into a monoglyceride monolayer (monopalmitin or monoolein) spread at the air-water interface and its effects on the structural, dilatational, and topographical characteristics of mixed films. Dynamic tensiometry, surface film balance, Brewster angle microscopy (BAM), and surface dilatational rheology have been used, maintaining the temperature constant at 20 degrees C and the pH and ionic strength at 7 and 0.05 M, respectively. The initial surface pressure (mN/m) of the spread monoglyceride monolayer (pii(MONOGLYCERIDE)) at 10, 20, and the collapse point is the variable studied. Beta-lactoglobulin can penetrate into a spread monoglyceride monolayer at every surface pressure. The penetration of beta-lactoglobulin into the monoglyceride monolayer with a more condensed structure, at the collapse point of the monoglyceride, requires monoglyceride molecular loss by collapse and/or desorption. However, the structural, topographical, and dilatational characteristics of monoglyceride penetrated by beta-lactoglobulin mixed monolayers are essentially dominated by the presence of monoglyceride (either monopalmitin or monoolein) in the mixed film. In fact, monoglyceride molecules have the capacity to re-enter the monolayer after expansion and recompression of the mixed monolayer. Thus, monoglyceride molecular loss by collapse and/or desorption is reversible. The topography of the monolayer under dynamic conditions corroborates these conclusions.  相似文献

Multilayer films of 2,3-di-O-acetyl-6-O-(4-stearyloxytrityl)cellulose (ASTC) were prepared and investigated. The fairly high surface pressure (π)–area (A) isotherms (36– 47 mN/m) suggest that the balky and hydrophobic trityl group contribute to form a condensed monolayer. The cellulose derivative formed a homogeneous monolayer at 10 mN/m. The monolayer on the water surface was transferred successfully at 10 mN/m onto various substrates by modifying the preparation methods, to form Z-type multilayer films. The ultraviolet–visible (UV–Vis) absorbance was independent of the number of layers, indicating that each layer is made of definite number of anhydroglucose unit (AGU). The monolayer thickness determined from atomic force microscope (AFM) and ellipsometry was calculated to be about 1.9 nm, suggesting that the long alkyl chain in the film has a so-called hairy-rod type structure. This is the first paper about the Z-type LB film prepared from cellulose derivatives having long mono-alkyl chain only at 6-O-position.  相似文献

The collapse of Langmuir monolayers of poly(vinyl stearate) (PVS) at the air-water interface has been investigated by combined measurements of the surface pressure-area isotherms and Brewster angle microscopy (BAM). Atomic force microscopy (AFM) has been used to gain out-of-plane structural information on collapsed films transferred onto a solid substrate by a modified version of the inverse Langmuir-Schaefer deposition method. At high areas per monomer repeat unit, BAM imaging revealed that the films are heterogeneous, with large solidlike domains (25-200 mum in diameter) coexisting with liquidlike domains. Upon film compression, the domains coalesced to form a homogeneous monolayer before the film collapsed at constant pressure, forming irreversible three-dimensional (3D) structures. BAM images showed that two 3D structures coexisted: buckles of varying width extending across the surface and perpendicular to the direction of the compression and dotted islandlike structures. Upon expansion, the film fractured and both 3D protrusions persisted, explaining the marked hysteresis recorded in the Langmuir isotherms. Experiments with AFM confirmed the 3D nature of both protrusions and revealed that many buckles contain substructures corresponding to narrow buckles whose heights are a multiple of a single bilayer. Additionally, many multilayer islands with diameters spanning from 0.2 mum to over 3.5 mum were characterized by varying heights between 2 nm and up to over 50 nm. The key to the formation of the irreversible 3D structures is the presence of large inhomogeneities in the PVS monolayer, and a generalized phenomenological model is proposed to explain the collapse observed.  相似文献

Microcin J25 forms stable monolayers at the air-water interface showing a collapse at a surface pressure of 5 mN/m, 220 mV of surface potential, and 6 fV per squared centimeter of surface potential per unit of molecular surface density. The adsorption of microcin J25 from the subphase at clean interfaces leads to a rise of 10 mN/m in surface pressure and a surface potential of 220 mV. From these data microcin appears to be a poor surfactant per se. Nevertheless, the interaction with the lipid monolayer further increase the stability of the peptide at the interface depending on the mode in which the monolayer is formed. Spreading with egg PC leads to nonideal mixing up to 7 mN/m, with hyperpolarization and expansion of components at the interface, with a small excess free energy of mixing caused by favorable contributions to entropy due to molecular area expansion compensating for the unfavorable enthalpy changes arising from repulsive dipolar interactions. Above 7 mN/m microcin is squeezed out, leaving a film of pure phospholipid. Nevertheless, the presence of lipid at 10 and 20 mN/m stabilize further microcin at the interface and adsorption from the subphase proceeds up to 30 mN/m, equivalent to surface pressure in bilayers.  相似文献

Fullerene derivative C60TT, which is substituted with the low-molecular-weight organogelator tris(dodecyloxy)benzamide, formed nanowire structures on application of the Langmuir-Blodgett (LB) method. The surface morphology of the C60TT LB film was dependent on the holding time before deposition at a surface pressure of 5 mN m(-1); it changed from a homogeneous monolayer to a bilayer fibrous structure via a fibrous monolayer structure, which was estimated to have dimensions of 1.2 nm in height, 8 nm in width, and 5-10 microm in length. From the structural and spectroscopic data, it is inferred that close packing of the fullerene moiety occurs along with intermolecular hydrogen bonding within the monolayer fibrous structure. The morphological changes in the LB film are explained kinetically by the Avrami theory, based on the decrease in the surface area of the monolayer at the air/water interface. The growth of the quasi-one-dimensional fibrous monolayer structures at holding times from 0 to 0.2 h is considered to be an interface-controlled process, whereas the growth of the quasi-one-dimensional bilayer fibrous structures from 0.2 to 18 h is thought to be a diffusion-controlled process.  相似文献

Asymmetrically substituted poly(paraphenylene) (PhPPP) with hydrophilic and hydrophobic side chains was investigated. The polymer behavior at the air-water interface was studied on the basis of surface pressure-area (pi-A) isotherms and compression/expansion hysteresis measurements. PhPPP can form stable monolayers with an area per repeat unit of A=0.20+/-0.02 nm2 and a collapse pressure in the range of pi=25 mN/m. Then, Langmuir-Blodgett-Kuhn (LBK) films of PhPPP were prepared by horizontally and vertically transferring the Langmuir monolayers onto hydrophilic solid substrates at pi=12 mN/m. Cross-section analysis of the AFM tapping-mode topography images of a single transferred monolayer reveals a thickness of d0=0.9+/-0.1 nm. Taking into account the obtained monolayer thickness, curve-fitting calculations of angular scan data of LB monolayers measured using surface plasmon resonance (SPR) spectroscopy lead to a value for the refractive index of n=1.78+/-0.02 at lambda=632.8 nm. Next, the spontaneous formation of a PhPPP monolayer by adsorption from solution was studied ex situ by atomic force microscopy and UV-vis spectroscopy and in situ by using SPR spectroscopy. Stable self-assembled monolayers of PhPPP can be formed on hydrophilic surfaces with a thickness similar to that of the monolayer obtained using the LB method. The characterization results confirmed the amphiphilic character and the self-assembly properties of PhPPP, as well as the possibility of preparing homogeneous monolayer and multilayer films.  相似文献

The behaviour of monolayers and bilayers formed by the dialkyl chain non-ionic surfactant, 1,2-di-O-octadecyl-rac-glycerol-3-omega-methoxydodecaethylene glycol (2C(18)E(12)) in water at 297 K has been investigated. Using a surface film balance (or Langmuir trough) the compression-expansion cycle of the 2C(18)E(12) monolayer was found to be reversible when compressed to surface pressures (pi) less than 42 mN m(-1). Compression of 2C(18)E(12) monolayer to pi greater than 42 mN m(-1) above this resulted in a considerable hysteresis upon expansion with the pi remaining high relative to that obtained upon compression, suggesting a time/pressure dependent re-arrangement of 2C(18)E(12) molecules in the film. Morphology of the 2C(18)E(12) monolayer, investigated using Brewster angle microscopy, was also found to depend upon monolayer history. Bright, randomly dispersed domains of 2C(18)E(12) of approximately 5 mum in size were observed during compression of the monolayer to pi less than 42 mN m(-1). At pi of 42 mN m(-1) and above, the surfactant film appeared to be almost completely 'solid-like.' Regardless of the extent of compression of the monolayer film, expansion of the film caused formation of chains or 'necklaces' of individual surfactant domains, with the extent of chain formation dependent upon pressure of compression of the monolayer and the length of time held at that pressure. Irreversible effects on 2C(18)E(12) vesicle size were also seen upon temperature cycling the vesicles through their liquid-crystalline phase transition temperature with vesicles shrinking in size and not returning to their original size upon standing at 298 K for periods of more than 24 h. No comparable hysteresis, time, pressure or temperature effects were observed with the monolayer or vesicles formed by the corresponding phospholipid, disteaorylphosphatidylcholine, under identical conditions. The effects observed with 2C(18)E(12) are attributed to the ability of the polyoxyethylene head group to dehydrate and intrude into the hydrophobic chain region of the mono- and bilayers. These studies have important implications for the use of the vesicles formed by 2C(18)E(12) as drug delivery vehicles.  相似文献

Neutral tris-chelated chromium complex [Cr(L(a))(3)] (1a), and its surfactant derivatives [Cr(L(b))(3)] (1b), [Cr(L(c))(3)] (1c), and [Cr(L(d))(3)] (1d) (where L(a)=2-(4'-methoxyphenylazo)pyridine, L(b)=2-(4'-butyloxyphenylazo)pyridine, L(c =2-(4'-octyloxyphenylazo)pyridine, and L(d)=2-(4'-dodecyloxyphenylazo)pyridine) were synthesized. The molecular structure of compound 1a, determined by X-ray diffraction, showed that the local geometry around the metal center is a distorted octahedral with meridional coordination of the ligands. The structural parameters, spectroscopic data, and density functional theory (DFT) calculations on representative complex 1a suggest that ligand L(a) is predominantly an azo-anion-radical-type, and so the complex can be represented as [Cr(III)(L(a.-))(3)]. An assessment of their physicochemical and surface properties was performed with the aim of using these triple-tailed metallosurfactants as precursors for redox-responsive films. The surface-pressure-molecular-area isotherm measurement for compound 1d shows that the complex forms a stable Langmuir film at the air/water interface. The monolayer and multilayers were successfully transferred onto the quartz substrate and the platinum working electrode at a surface pressure of 10 mN m(-1) by the Langmuir-Schaefer (LS) technique. The LS films were studied by UV/Vis spectrometry, infrared spectroscopy, field-emission scanning electron microscopy, and atomic force microscopy. A good linear relationship between the absorbance at 370 nm and the thickness of the layers against the number of deposited layers indicated the uniformity and reproducibility of this transfer process. Voltammograms for platinum-surface-bound LS film of compound 1d showed that the redox response owing to the first oxidation is stable and reproducible after many cycles (>300 cycles). Spectroscopic studies and electrochemical measurements of compound 1d on the LS films revealed that these complexes are potential candidates for molecular devices.  相似文献
