The synthesis of some N,N-disubstituted benzhydrazidoyl chlorides and their reaction with cyano compounds in the presence of a Lewis acid are described. Whereas N-methyl-N-phenylbenzhydrazidoyl chloride (1a) gave 1H-1,2,4-triazoles, N,N-diphenylbenzhydrazidoyl chloride (1b) afforded 1H-1,2,4-benzotriazepines (3b) and N,N-(2,2′-biphenylen)benzhydrazidoyl chloride (10) yielded the corresponding 1H-1,2,4-benzotriazepine (12) or a hydrolysis product (13). Properties of compounds 3b, specially their near-quantative acid hydrolysis to 1-phenylindazoles, are reported.  相似文献

The synthesis of layered donor/acceptor-materials based on perylenes (1ac) and ter(thiophen)es (2a, 2b), ordered by hydrogen bonding moieties is reported. Based on the successful (selective) chlorination of 3,4:9,10-perylene tetracarboxylic dianhydride (3) to obtain a perylene derivative with only four chlorine atoms, subsequent functionalization with different hydrogen-bonding moieties is achieved via the azide/alkyne click reaction as proven by extensive ESI-TOF measurements. The perylene- (1ac) and terthiophene- (2a, 2b) compounds are useful as acceptor and donor parts, respectively, in organic solar cells as proven via UV–vis and fluorescence measurements. Charge transfer between donor and acceptor parts (2a/1b) was determined as 41% via fluorescence resonance energy transfer (FRET), proving the association of the two components via the attached hydrogen bonding moieties. These measurements indicate that the mixture 2a/1b displays large potential for use as a layered ordered material with controlled spacings for organic solar cells based on a thereby facilitated charge-transfer.  相似文献

A new heterocyclic derivative of 2-mercaptobenzothiazole (MBT) comprising 4-oxothiazolidines, tetrazole and triazole, through the reaction of (MBT) with hydrazine hydrate obtained 2-hydrazobenothiazol (1), which was condensed with various aromatic aldehydes. Azomethine derivatives (2ac) are converted into a number of 4-oxothiazolidines (3ac) and tetrazole derivatives (4ac), through the reaction of azomethine derivatives (2ac) with mercaptoacetic acid and sodium azide, respectively. Also the reaction of compound (1) with triethylorthoformate and nitrous acid to produce the corresponding (triazole and tetrazole) benzothiazole (5,6) was reported.Triazole moieties reported condensation (MBT) with ethylbromo acetate and potassium hydroxide by the fusion method and resulted in ester-2-mercaptobenzothiazole (7), which was treated with hydrazine hydrate to give a hydrazine derivative (8), then converting these compounds (8) to phenyl semicarbazide (9) and phenyl thiosemicarbazide (10) derivatives. Cyclization compounds (9,10) in alkaline media (4 N·NaOH) gave triazoles compounds (11,12). Furthermore the compound (8) was converted to the dithiocarbazate salt (13) which was then cyclized with hydrazine hydrate to give substituted triazole (14). The prepared compounds were identified by spectral methods (FTIR, 1H NMR, 13C NMR) and some of its physical properties were measured and furthermore the effects of the preparing compounds on some strains of bacteria were studied.  相似文献

A convenient, multigram synthesis of a novel α-amino acid (2S)-2-amino-3-(1H-4-indolyl)propanoic acid (1a), is reported. An Fmoc–t-Boc derivative of this novel regioisomer of the natural aromatic amino acid tryptophan could be readily incorporated into bioactive synthetic peptides using standard solid phase synthesis. The synthesis featured the use of Schöllkopf chiral auxiliary reagents for chirality induction during a key step.  相似文献

The first total synthesis of ankorine (4), an Alangium lamarckii alkaloid, has been accomplished in the form of a recemic modification by means of an initial condensation of 2-benzyloxy-3,4-dimethoxyphenacyl bromide with the lactim ether 6, derived from ethyl (±)-trans-5-ethyl-2-oxo-4-piperidineacetate (5), and succeeding steps proceeding through the intemediates 7a, 8a, 9a, 10a (X=Cl), 11a, and 12a. A parallel synthetic route starting with 3,4,5-trimethoxyphenacyl bromide and 6 gave (±)-11-methoxyprotoemetinol (12c) via the intermediates 7c, 8c, 9c, 10c (X =I,ClO4), and 11c. The trimethyl ether 12c did not match the O-Me derivative (type 12e) of natural ankorine. Thus, the formula 4 defines the structure and relative stereochemistry of ankorine.  相似文献

In our series of nucleophilic substitution reactions on N,N-disulfonylimides we hereby report the preparation and the nucleophilic substitution of the N,N-1,2-naphthalenedisulfonylimide derivative 1a of the chiral amine 1. The disulfonimide was prepared by using the disulfonyl chloride reagent. Nucleophilic substitution of 1a by KNO2 and azide afforded the corresponding alcohol 2 and the azide product 3 with, respectively, 63 and 70% inversion of configuration. The stereochemical results are compared with previously reported results for a series of N,N-disulfonylimides showing that the degree of inversion of 1a is lower than for the other N,N-disulfonylimides.  相似文献

Reaction of 3β - hydroxy - 14α,15α - epoxy - 5β - bufa - 20,22 - dienolide (1a) with 72% perchloric acid was found to yield 14β - artebufogenin (2a), 14α - artebufogenin (2b), 15α - hydroxy-bufalin (3a), and a new substance 3β,15ξ - dihydroxy - 5β,14ξ - bufa - 8,20,22 - trienolide (4a). Similar results were realized by antimony (III) chloride and an iron(III) chloride catalyzed ring opening of epoxide 1a. Selective acetylation of the new bufadienolide (4a) followed by oxidation completed a route to the title substance.  相似文献

A facile three-step synthetic route toward 1,3-diaryl-1H-benzo[g]indazoles 1a1n starting with 3,4-dimethoxy-2-allylbenzaldehyde (6) in modest total yield is described. The facile route was carried by aldol condensation of aldehyde 6 with aryl methyl ketones 5a5d in alkaline MeOH at reflux, Knorr pyrazole synthesis of the resulting chalcones 4a4d with aryl hydrazines 3a3e in EtOH at reflux followed by DDQ-mediated aromatization in toluene at reflux, and oxidative cleavage annulation of olefins 2a2n with the one-pot combination of OsO4/NaIO4/HOAc in the aqueous THF at reflux.  相似文献

A diastereoselective and enantioselective synthesis of (Z)-1-benzoylamino-2-tritylsulfanylcyclopropanecarboxylic acid derivatives 8a,b and 9a,b was achieved starting from (−)- or (+)-menthyl 2-benzoylamino-3-tritylsulfanylacrylates 3a,b. Compounds 3 were reacted with diazomethane giving the corresponding pyrazolines 4a,b and 5a,b. These compounds, on melting, were transformed, under steric control, into the cyclopropaneamino acid derivatives (R,R)-8a,b and (S,S)-9a,b. The synthesis of a large class of chiral 2-S-alkyl-1-aminocyclopropanecarboxylic acid derivatives is possible after removing the trityl protecting group and subsequent alkylation reactions.  相似文献

3-Methylindol-2-yl methyl ketone reacted with phosphoryl chloride in dimethylformamide to give 3-chloro-3(3-methylindol-2-yl), propenal (4). 2-Carbomethoxy- and 2-carbethoxy-1-chloro- 9H-pyrrolo[1,2-a]indol-9-ylidene acetaldehyde (10a and b) were formed by reaction with the same reagent of 3(3-methylindol-2-yl) propenoate (7) and 3(3-methylindol-2-yl)-3-oxopropanoate (2) respectively. In the latter case, 2-carbethoxy-1-chloro-3-dimethylamino-9-methyl-3H-pyrrolo[1,2-a]indole (9b) was isolated as an intermediate. The structure of the pyrroloindolylidene acetaldehydes was proved by synthesis of the chromophore from 4-acetyl-3-methyl-1-phenyl-pyrrole-2-carboxylate (26). The preparation and behaviour of 1-phenylpyrrole-2,4- and 3,4-dicarboxylates, monomethylated in the pyrrole ring, is described. These compounds were prepared during a search for a satisfactory route to a starting material for the synthesis of compounds related to 10a and b. The saponification of such diesters is remarkably selective, and in the case of the 2,4-dicarboxylates, contradicts accepted generalisations concerning the lability of pyrrole α,β-diesters to selective hydrolysis.  相似文献

Nucleophilic attack of azide on 2-bromo-3-fluoro-3-(1-trityl-1H-imidazol-4-yl)-propan-1-ol (1a) in aprotic solvent occurs on the 2-position to give the 2-azido derivative (2a). Reduction of azide and removal of the trityl group produces β-fluorohistidinol (6a). Elimination of HBr from 1a followed by “FBr” addition to the resulting double bond gives 2-bromo-3,3-difluoro-3-(1-trityl-1H-imidazol-4-yl)-propan-1-ol (1b). Nucleophilic attack of azide followed by reduction and removal of the trityl group, as for the preparation of 6a, gives β,β-difluorohistidinol (6b). Initial attempts, under a variety of conditions, to oxidize the fluorinated histidinol precursors to carboxylic acids have not been successful.  相似文献

A novel three-step synthesis of 4-substituted-(1-benzylpiperazin-2-yl)methanols 5a5c is described starting from the methyl ester of the proteinogenic amino acid (S)-serine, which involves the diastereomeric oxazolidine derivatives 11a–11c as key intermediates. 5b and 5c were transformed into the chiral, non-racemic bicyclic lactams 14b and 14c via Swern oxidation followed by Wittig reaction and subsequent hydrogenation. In the screening for central nervous system receptor affinity the butyl derivative 14b was found to have promising interaction with σ1-receptors (Ki=345 nM).  相似文献

A general route for the stereoselective synthesis of 3-amino-4,5-dihydroxyaldehydes, with almost any desired configuration at the three stereogenic centers, is described by applying a combination of enzymatic and chemical steps. l-Daunosamine 1, for example, the glycosidic fragment of many important anthracycline antibiotics has been prepared by this route starting from O-allyl-l-lactaldehyde (S)-6a. (R)-Hydroxynitrile lyase (HNL) catalyzed addition of HCN to (S)-6a yields the 2,3-dihydroxynitrile (2S,3S)-7a with high stereoselectivity (91% de) in 75% yield. The addition of allyl Grignard to the O-protected 2,3-dihydroxynitrile (2S,3S)-9a and subsequent hydrogenation of the imino intermediate leads to 4-amino-2,3-dihydroxy-1-heptene (4S,5S,6S)-12a, which after ozonization and deprotection gives N-acetylated l-daunosamine 14a in a total yield of 15% referring to (S)-6a. The general applicability of this chemoenzymatic multistep procedure is demonstrated in the stereoselective synthesis of the unnatural aminodeoxy sugar (2S,3S,4S)-14b, starting from isovaleraldehyde 3.  相似文献

The reaction of 4-methyl-2-phenyl-1,2-dihydro-6-oxo-5-pyridine- carbonitrile (1) with arylidene malononitrile afforded isoquinoline derivatives 2a,b. 6-Chloro-4-methyl-2-phenyl-5-pyridinecarbonitile (3) obtained by chlorination of compound 1 with phosphoryl chloride was converted into 6-amino-4-methyl-2-phenyl-5-pyridinecarbonitrile (4) and 6-hydrazido-4-methyl-2-phenyl-5-pyridinecarbonitrile (5) in good yield, through reactions with ammonium acetate and hydrazine hydrate, respectively. Treatment of 4 with ethyl acetoacetate, acetic anhydride, formic acid, urea and thiourea gave the corresponding pyrido [2,3-d] pyrimidine derivatives 7-10a,b. A new series of 6-substituted-4-methyl-2-phenyl-5-pyridine carbonitriles 11-13 has been synthesized via reaction of 4 with phenyl isothiocyanate, benzenesulphonyl chloride and acetic anhydride. Treatment of 4 with malononitrile gave 1,8-naphthyridine derivative 14. The reactivity of hydrazide 5 towards acetic acid, phenylisothiocyanate and methylacrylate to give pyrazolo-[3,4-b]-pyridine derivatives 15-17 was studied. Treatment of 5 with acetic anhydride, phthalic anhydride and carbon disulphide gave pyridine derivatives 18,19 and 1,2,4-triazolo-[3,4-a]-pyridine derivative 20.  相似文献

Purpurosamine C (1) is a component of the aminoglycoside antibiotic gentamicin C1a. A derivative of 1 was synthesized from d-galactose via its 2-acetoxy-3,4,6-tri-O-acetyl glycal (3). Compound 3 undergoes glycosylation with 2-propanol in the presence of SnCl4, with two succesive allylic rearrangements of the double bond to give isopropyl 6-O-acetyl-3,4-dideoxy-α-d-glycero-hex-3-enopyranosid-2-ulose (7). Compound 7 was hydrogenated, and O-deacetylated to afford 8. The free OH group of 8 was tosylated and substituted by azide, and the carbonyl function of the resulting ulose 10 reacted with hydroxylamine to give the E,Z-oximes (11,12). Highly diastereoselective reduction of the oxime acetate (13) by borane, which also reduced the azide function, led to the purpurosamine C derivative 14 (∼40% yield from 3).  相似文献

The structure of the abnormal product 1a formed in the Knoevenagel condensation of 2-carbethoxycyclohexanone and malononitrile has been further confirmed. Oxidation of the tetrahydroisoquinoline 3b using Na2Cr2O-AcOH-H2SO4 gave the keto isoquinoline 3d and the isoquinoline-1-carboxylic acid 5a. The acid chloride of 5a was condensed with diethyl ethoxymagnesiomalonate to afford after decarbethoxylation the methyl ketone 5d which on Baeyer-Villiger oxidation gave a mixture of the acetate 1g and the title compound 1b. The unambiguous synthesis of 1b confirms the structure assigned earlier to the title compound also formed during the partial hydrolysis of the diethoxy compound 1c. Condensation of 2-acetylcyclohexane-1,3-dione with malononitrile gave the quinoline derivative 4c which on ethylation yielded the ketoquinoline 4d. The present studies have confirmed that the quinoline compound 4a is also formed in the condensation of 2-acetylcyclohexanone and cyanoacetamide.  相似文献

Novel 3-(4,6-dimethoxybenzofuran-5-yl)-1-phenyl-1H-pyrazole-4-carboxaldehyde (3) and 3-chloro-3-(4,6-dimethoxybenzofuran-5-yl)propenal (4) were prepared via Vilsmeier–Haack reaction of 1-(4,6-dimethoxybenzofuran-5-yl)ethanone (1) and its hydrazone derivative 2. Reaction of compound 4 with some hydrazine derivatives, namely hydrazine hydrate, phenylhydrazine and benzylhydrazine hydrochloride led to the formation of pyrazole derivatives 58, respectively. On the other hand, reaction of compound 4 with thiourea, urea or guanidine gave the pyrimidine derivatives 911, respectively. Reaction of amino compound 11 with acetic anhydride, benzoyl chloride and benzenesulphonyl chloride yielded N-substituted pyrimidine derivatives 12–14, respectively. Reaction of diazonium salt of compound 11 with sodium azide afforded azidopyrimidine derivative 15, which upon reaction with ethyl acetoacetate gave 1,2,3-triazole derivative 16. Acid catalyzed reaction of 11 with p-nitrobenzaldehyde gave Schiff base 17, which cyclized upon reaction with thioglycolic acid or chloroacetyl chloride to give thiazolidin-4-one 18 and azetidin-2-one 19, respectively. The newly synthesized compounds were tested for their anti-inflammatory, analgesic and anticonvulsant activities. Depending on the obtained results, the newly synthesized compounds possess significant anti-inflammatory, analgesic and anticonvulsant activities.  相似文献

2H-1,2,3-Triazoles (2) were synthesized by [3+2] cycloaddition of (Z)-2,3-diaryl substituted acrylonitriles (1) with sodium azide and ammonium chloride in DMF/water. This method represents a facile and efficient reaction procedure for the synthesis of 4,5-diaryl-2H-1,2,3-triazoles in modest to good yields.  相似文献
