Action of guanidine or urea on cyclohexanone-, cyclopentanone-, cycloheptanone-and acetonecyanohydrine3 a?3 d generates very different products: 3 a reacts with guanidine inDMF to yield 1,3-diazaspiro[4.5]decane-2,4-diimine (5 a). Heating the components without solvent affords 7,14-diazadispiro[5.1.5.2]pentadecan-15-one(7)15–17, the guanidine not participating in the reaction; similarly3 b is transformed by guanidine to a pentacyclic dispirocompound (possible formulae19 and20), whereas3 d reacts to give 3,3,5,5-tetramethylpiperazine-2,6-dione(21)19. In 3-pentanone guanidine-cyanide condensates itself to give 2,4-diamino-triazine (22)21, 22. Action of urea on3 a?3 d yields the 4-imino-1,3-diazaspiroalkan-2-ones6 a?6 c and the 4-imino-5,5-dimethylimidazolidin-2-one6 d 6–8 resp. If the reaction of urea and3 d is carried out inDMF, however, 5,5-dimethyl-4-ureido-3-imidazolin-2-one (28) (or the tautomeric carbamoyliminoimidazolidinone27) is produced. The structures of the compounds prepared are proved by NMR-, IR- and mass spectra.  相似文献

Robustin (31a) occurring in the roots of Derris robusta has been synthesised from mono-O-methylphloroglucinol (20) as follows. Hoesch condensation with homopiperonitrile (21) gave 2,4-dihydroxy-6-methoxyphenyl 3,4-methylenedioxybenzyl ketone (22) which on reaction with 2-hydroxy-2-methyl-3-butene in the presence of borontrifluoride-etherate afforded 5-C-(24) and 3-C-(23) prenyl derivatives. These on cyclodehydrogenation with DDQ followed by coumarin condensation with ethyl chloroformate yielded robustin (31a) and isorobustin (32) respectively. Model experiments with nuclear prenylation of 2,4-dihydroxy-6-methoxy-phenyl benzyl ketone (1), followed by oxidative cyclisation and either courmarin or isoflavone condensation have yielded new isopentenylated 4-hydroxy-3-phenylcourarins or isoflavones.  相似文献

Zusammenfassung 4-Hydroxy-carbostyrile (1a–c) werden mit Benzylchlorid inDMF bevorzugt zu den 4-Benzyloxy-carbostyrilen (2a–c) alkyliert. In wäßr. NaOH dagegen erhält man aus1a und1c als Hauptprodukt das 3,3-Dibenzyl-2,4-dioxo-1,2,3,4-tetrahydrochinolin (3).
Alkylation of 4-hydroxy-quinol-2-ones (1a–c) with benzyl chloride inDMF in the presence of K2CO3 mainly gives the 4-benzylethers2a–c. However, carbon dialkylation takes place in aqueous NaOH and the main product of the reaction between benzylchloride and1a is 3.3-dibenzyl-2.4-dioxo-1.2.3.4-tetrahydro-quinoline3.

The title compounds 3al have been synthesized by the reaction of thiocarbohydrazide with substituted phenoxy acetic acid to obtained substituted 1,2,4-triazoles (1). Compound 1 was treated with various substituted aromatic aldehydes which results in 4-(substituted benzylideneamino)-5-(substituted phenoxymethyl)-2H-1,2,4-triazol-3(4H)-thiones (2ag), further 2ag is converted to 2-[4-(substituted benzylideneamino)-5-(substituted phenoxymethyl)-4H-1,2,4-triazol-3-yl thio] acetic acid (3al) derivatives by the reaction with chloroacetic acid. All the newly synthesized compounds were evaluated for in vivo anti-inflammatory and analgesic activities. Among the series 2-[4-(2,4-dichlorobenzylideneamino)-5-(phenoxymethyl)-4H-1,2,4-triazol-3-yl thio] acetic acid (3d), 2-[4-(4-dichlorobenzylideneamino)-5-(phenoxymethyl)-4H-1,2,4-triazol-3-yl thio] acetic acid (3e), 2-[4-(2,4-dichlorobenzylideneamino)-5-[(2,4-dichlorophenoxy)methyl]-4H-1,2,4-triazol-3-yl thio] acetic acid (3j) and 2-[5-[(2,4-dichlorophenoxy)methyl)]-4-(4-chlorobenzylideneamino)-4H-1,2,4-triazol-3-yl thio] acetic acid (3k) showed significant anti-inflammatory activity with P < 0.001 (63.4%, 62.0%, 64.1% and 62.5% edema inhibition, respectively), as compared to the standard drug diclofenac (67.0%) after third hour respectively and also compounds 3j, 3k exhibited significant analgesic activity with P < 0.001 (55.9% and 54.9% protection, respectively) and less ulcerogenic activity as compared with standard drug aspirin (57.8%).  相似文献

报道了N-{4-{N-甲基-N-[2-羟基-3-(2,4-二氧代-1,2,3,4-四氢嘧啶-5-基)氨基]丙基}氨基-3-溴}苯甲酰基-L-谷氨酸二乙酯及其衍生物的简便合成方法. 分别以4-氨基苯甲酸乙酯和4-氨基苯甲酰基-L-谷氨酸二乙酯为起始物, 经甲基化、烯丙基化、溴羟基化、环氧化、开环、脱保护等反应首次合成了6个新型5-取代氨基嘧啶类化合物, 并通过1H NMR, 13C NMR 和MS对其化学结构进行了表征. 初步生物活性结果表明, 苯环侧链的L-谷氨酸酯部分是此类化合物抑制人重组二氢叶酸还原酶的必需结构.  相似文献

2,4-Bis(4-methoxyphenyl-1,3,2,4-dithiadiphosphetane-2,4-disulfide, 1, reacts with phenylhydrazones of methylketones to give Δ5-1,2,3-diazaphospholines, 3; in one case 2-methyl-3-phenylindole, 4, was also isolated. Phenylhydrazones of cyclopentanone and cyclohexanone similarly yield the corresponding annulated P-heterocycles, 6a, 6b and as by-products the indoles 7a and 7b, respectively. The sole product from the reaction of 1 with the phenylhydrazone of 1,3-diphenyl-2-propanone is 2-benzyl-3-phenylindole, 9. The addition of 2,4-dinitrobenzenesulfenyl chloride (ArSCl) to a number of phenylmethylenecyclopropanes (2, 3, 4) and to α-cyclopropylstyrene (5), in CH2Cl2 and AcOH, has been investigated. The participation of the cyclopropylcarbinyl-phenyl carbonium ions (19a, 19b, 20, 22) is assumed. The attachment of the electronegative 2,4-dinitrobenzenesulfenyl group on the cyclopropyl ring prevents rearrangement with ring opening in the case of 19a and 19b, thus yielding mainly the normal addition products. On the other hand unsubstituted cyclopropyl rings (20 and 22) undergo rearrangement to the homoallylic adducts. The strong influence of the SAr group is clearly demonstrated in the case of 20, where both, a SAr-substituted and an unsubstituted ring, are present—only the unsubstituted ring undergoes ring cleavage. The stereochemistry of the homoallylic adducts is controlled by the bisected conformation of the intermediate cyclopropylcarbinyl carbonium ions, which is depended on the steric interaction of the cyclopropyl ring with the other substituents.  相似文献

The 1-glyco-2-nitrocyclohexa-2,4-dienes 1a and 5a react with cyclopentadiene to yield, almost exclusively, the 10-glyco-1-nitrotricyclo[5,2,2,02,6]undeca-3,8-dienes 4a and 8a. Formation of these products is explained as the result of a tandem consecutive asymmetric Diels–Alder reaction–Cope rearrangement. Periodate oxidation of deprotected sugar side-chains, followed by sodium borohydride reduction yielded enantiomerically pure 10-formyl- and 10-hydroxymethyl-1-nitrotricyclo[5,2,2,02,6]undeca-3,8-dienes. Structures have been determined by X-ray crystallographic and spectroscopic analyses, and chemical correlation.  相似文献

The product formed from cyclopentadiene and the cyclo-octatetraene dimer 5a has been shown to possess structure 6a, probably resulting from Cope rearrangement of an initial adduct 4a. The reaction of tetraphenylcyclopentadienone with 1 - methoxybuta - 1,3 - diene yields a mixture of the exo-adduct 14 and the rearranged endo-adduct 13 Rearrangement of the norbornenone derivative 27 gives the Cope product 29 Treatment of the 4,5-fused 2,3,4,5-tetrachlorocyclopent-2-enones 29,40 and 41 with powdered sodium hydroxide in tetrahydrofuran results in ring-cleaveage with the formation of 2,3,5-trichlorobuta-2,4-dienoic acid derivatives, 36, 43 and 44 respectively.  相似文献

Herein we report a practical and efficient method for the synthesis of optically active 2,4-disubstituted oxazolines (S)-1ah in good to excellent yields. The target compounds were prepared in good yield through the Horner–Wadsworth–Emmons reaction of β-phosphonoamide 3 bearing l-phenylalaninol with commercially available aryl aldehydes followed by the cyclodehydration of the corresponding N-(cinnamoyl)-(S)-phenylalaninol derivatives (S)-2ah. Additionally, the cyclodehydration of β-phosphonoamide (S)-3 followed by the Horner–Wadsworth–Emmons reaction of β-phosphono-oxazoline (S)-4 with aryl aldehydes also gave the 2,4-disubstituted oxazolines (S)-1ah.  相似文献

2,4-Dihydroxy-6,4′-dimethoxydesoxybenzoin 1a with 2-hydroxy-2-methyl-3-butene in the presence of BF3-etherate yielded 3-C-prenyl- 2a and 5-C-prenyl 3 derivatives. However, reaction with prenyl bromide in the presence of methanolic potash afforded 3-C-prenyl 2a and 4-O-prenyl 1b derivatives. The C-prenyl derivatives 2a and 3 readily underwent cyclodehydrogenation with DDQ to form phenacyl chromenes 7 and 9 respectively. The latter on condensation with ethyl chloroformate gave robustic acid 10 identical in all respects with natural sample. The desoxybenzoins 2b, 3 and 7 have also been converted into the corresponding 4-hydroxy-3-phenyl coumarins 11, 12 and 8.  相似文献

A variety of 4′-(4-R-phenyl)-4-(methylthio)-2′-phenyl-2′H-spiro[chromene-2,5′-[1,2,3]thiadiazole] 5ad and 5′-(4-R-phenyl)-4-(methylthio)-3′-phenyl-3′H-spiro[chromene-2,2′-[1,3,4]thiadiazole] 6ad were synthesized regioselectively through the reaction of 4-(methylthio)-2H-chromene-2-thione 2 with diarylnitrilimines under refluxing dry chloroform. Whilst the reaction of 4-(allylthio)-2H-chromene-2-thione 3 with diarylnitrilimines under similar reaction conditions afforded the corresponding 4-(allylthio)-5′-(4-R-phenyl)-3′-phenyl-3′H-spiro[chromene-2,2′-[1,3,4]thiadiazole] 7ad and 5-(4-R-phenyl)-2′-methyl-3-phenyl-2′,3′-dihydro-3H-spiro[[1,3,4]thiadiazole-2,4′-thieno[3,2-c]chromene] 9ad as two unisolable diastereoisomeric forms. The structures of the obtained spiro cycloadduct thiadiazoles have been assigned by means of spectroscopic measurements.  相似文献

We reported here an efficient regioselective protection strategy for xylose with two-step successive protections on 4- and 2-OH. This method enables the expeditious preparation of various xylose building blocks (3ac, 5, 6). The rapid selective 2,4-protection especially facilitates the synthesis of 3-substituted xyloside structures. By this approach, typical arabino-α-1,3-xyloside disaccharide and trisaccharide, as fragment structure units from arabinoxylans, have been successfully synthesized.  相似文献

Heating a neat 1:2 mixture of 2-picolylamine and 2-cyanopyridine followed by treatment of the resultant red gummy substance with aqueous KOH resulted in the isolation of 2,4,5-tris(2-pyridyl)imidazole (1a) as the major product and N-(3-(2-pyridyl)imidazo[1,5-a]pyridine)picolinamidine (2a) in small amounts. Similarly, by using 3-picolylamine, 2,4,-bis(2-pyridyl)-5-(3-pyridyl)imidazole (1b) and N-(3-(3-pyridyl)imidazo[1,5-a]pyridine)picolinamidine (2b) were isolated, and by using 4-picolylamine, 2,4,-bis(2-pyridyl)-5-(4-pyridyl)imidazole (1c) and N-(3-(4-pyridyl)imidazo[1,5-a]pyridine)picolinamidine (2c) were isolated. The plausible mechanism of the formation of 1a-c and 2a-c is delineated.  相似文献

The aminonitriles3a–f react with guanidine inDMF to yield the 5,5-resp. 5-subtituted imidazolidine-2,4-diimines2a–d resp.2f, whereas2e could not be isolated. 7,14-diazadispiro[5.1.5.2]-pentadecan-15-on (7)1, 4-imino-1,3-diazaspiro[4.5]decan-2-one (8a)1 and the 2,4-diaminotriazine (9)1 were isolated as byproducts.1-Aminocyclohexancarbonitrile (3a) reacts with urea to the 4-imino-1,3-diazaspiro[4.5]decan-2-one (8a)1;8a can be prepared from (1-cyancyclohexyl)urea (11a) as well. The structures of the new compounds are proved by NMR-, IR- and mass spectra and the mechanism of the reaction is discussed.

Treatment of the title compound (1a) with potassium in liquid ammonia at ca ?33° immediately afforded the benzylcyclooctatetraene dianion (4a) which, upon quenching with iodine-pentane, gave benzylcyclooctatetraene (5) as the only isolable product. In contrast, treatment of1a with potassium amide in liquid ammonia at ?69° initially afforded the 9-phenylmethylenecyclooctatrienyl anion (6a) as a short-lived intermediate which was then converted to the α-aminobenzylcyclooctatetraene dianion (7a). These results, coupled with the observation that cis-bicyclo[6.1.0]nona-2,4-diene (12) in potassium amide-liquid ammonia affords the cis-bicyclo[6.1.0]nonadienyl anion (8b) which then slowly opens to the methylcyclooctatetraene dianion (4b) at ?12°, lead to the conclusion that4a is produced by a reductive cleavage of1avia a radical anion or dianion.  相似文献

In this work, two new effective methodologies have been adopted for the preparation of 5-(2′-bromobenzyloxy)pyrimidine-2,4-diones 6(a–k). In the first methodology, 5-hydroxy uracils 4(af) were alkylated with 2-bromobenzyl bromide 5a or 2-bromo-5-methoxy benzyl bromide 5b under phase transfer catalysis condition using lithium hydroxide/tetrabutyl ammonium bromide in N,N-dimethylformamide at 45 °C, and in the second method, the microwave irradiation (MWI) protocol has been exploited by mixing 5-hydroxy uracils 4(a–f) with 30 % excess of 2-bromobenzyl bromide 5a or 2-bromo-5-methoxy benzyl bromide 5b. A catalytic amount of TBAB and potassium carbonate was added and irradiated in an open Erlenmeyer flask in a microwave oven for 3–12 min. The tributyltin hydride-mediated radical cyclisation of 6(a–k) was carried out under MWI to generate 1H,3H,6H[2]benzopyrano[4,3-d]pyrimidine-2,4-diones 7(a–k) in 80–89 % yield, and the reaction time was shortened compared to the previously reported conventional radical cyclisation method.  相似文献

Novel 3-phenyl- and 3-(4-nitrophenyl)cyclohepta[4,5]imidazo[1,2-a]-1,3,5-triazine-2,4(3H)-diones and the corresponding imino derivatives 5a,b and 6a,b were synthesized in modest to moderate yields by the abnormal and normal aza-Wittig reaction of 2-(1,3-diazaazulen-2-ylimino)triphenylphosphorane with aryl isocyanates and subsequent heterocyclization reaction with a second isocyanate. The related cationic compound, 1-methyl-3-phenylcyclohepta[4,5]imidazo[1,2-a]-1,3,5-triazine-2,4(3H)-dionylium tetrafluoroborate 7a, was also prepared. The electrochemical reduction of these compounds exhibited more positive reduction potentials as compared with those of the related compounds of 3,10-disubstituted cyclohepta[4,5]pyrrolo[2,3-d]pyrimidine-2,4(1H,3H)-dione systems. In a search of the oxidizing ability, compounds 5a, 6a, and 7a were demonstrated to oxidize some amines to give the corresponding imines in more than 100% yield under aerobic and photo-irradiation conditions, while even benzylamine was not oxidized under aerobic and thermal conditions at 100 °C. The oxidation reactions by cation 7a are more efficient than that by 5a and 6a. Quenching of the fluorescence of 5a was observed, and thus, the oxidation reaction by 5a probably proceeds via electron-transfer from amine to the excited singlet state of 5a. In the case of cation 7a, the oxidation reaction is proposed to proceed via formation of an amine-adduct of 7a and subsequent photo-induced radical cleavage reaction.  相似文献

Synthesis of a series of novel fused tetracyclic mono- and diazaindolo[2,1-c][1,4]benzoxazine heterocyclic compounds 3a-o has been achieved in a two-step one-pot reaction set up starting from commercially available or easily accessible inputs. For example, reaction of di-lithiated (N-Boc)-2-amino-3-methylpyridine Li2-2a with Weinreb amide of 2-(2,4-difluorophenoxy)-2,2-dimethylacetic acid 1a, followed by TFA treatment furnished the tetracyclic compound 3a, which is essentially a fusion of 7-azaindole and 3,4-dihydro[1,4]benzoxazine, in 70% isolated yield. A competitive elimination by-product 4a was also observed (24% isolated yield) in this case. Based on our initial results, a structural basis and molecular mechanism have been suggested to explain these two parallel reactions. Consequently, with appropriate structural tuning of 1, formation of the individual products can be controlled.  相似文献
