A multi-residue method using liquid chromatography coupled to triple quadrupole tandem mass spectrometry (LC-MS/MS), associated with solid-phase extraction (SPE), was developed for the determination of 21 pesticides in water samples. The compounds investigated are used for the maintenance of golf courses and ordinarily measured by gas chromatography-mass spectrometry (GC-MS). Electrospray ionisation (ESI) was applied to all compounds, and LC and MS conditions were optimised to measure them under SRM mode. This method showed excellent linearity ranges for all pesticides, with correlation coefficients greater than 0.996. Two kinds of extraction cartridges, namely, styrene divinylbenzene polymer (Sep-Pak PS-2) and divinylbenzene-N-vinylpyrrolidone copolymer (Oasis HLB), were tested and the extraction conditions were optimised. All the pesticides were determined using acetonitrile and ethyl acetate as eluents in both cartridges, and good recoveries (>77%) and repeatability with low relative standard deviations (RSDs, <12%) were achieved from ultra-pure water. In addition, satisfactory recoveries (>76%) and low intra-day and inter-day RSDs (<15%) of all pesticides were also obtained with the Sep-Pak PS-2 cartridge when using river water. The method limits of detection (LODs) ranged between 0.068 (diazinon) and 3.9 (triclopyrbutoxyethyl)?ng?L?1. The analytical method was successfully applied for the determination of pesticides in surface river water.  相似文献

In the present study a multi-residue analytical method was developed for monitoring some polar pesticides such as acephate, methamidophos, carbofuran, isoproturon, dimethoate in water with SPE (solid-phase extraction) and LC–MS–MS. Acetochlor was taken as surrogate, and alachlor as internal standard. SPE with different types of columns was compared with LLE (liquid-liquid extraction). Further, the breakthrough volume for different pesticides was determined. The results showed that the selected pesticides can be determined very sensitively with LC–MS–MS. The minimum detectable quantity (MDQ) for each pesticide was about 1.0 ng. To date, SPE cartridge studies showed that the Oasis HLB cartridges were suitable for further studies. However, for Oasis HLB cartridge, different pesticide showed different breakthrough volume. The results showed that for acephate and methamidophos, the breakthrough volume was about 30 mL of water sample, much less than the breakthrough volume of other pesticides studied. Because of the higher vapor pressure and higher Henry's constant of methamidophos, dimethoate and carbofuran, much attention should be paid on their losses in the evaporation step of the experiment. The results of validation of a method for the analysis of free and bound phenolics in wine and grapes are presented. Wine and grape extracts are fractionated by automated solid-phase extraction on Bond Elut PPL cartridges to give free and bound phenolic fractions. Bound fractions are subjected to acid hydrolysis, and the phenolics released are recovered by solid-phase extraction on Bond Elut PPL cartridges. The fractions are further purified by automated solid-phase extraction on Bond Elut silica cartridges. After derivatisation to form trimethylsilyl ethers, the phenolics are determined by gas chromatography–mass spectrometry with selected ion monitoring. The method is suitable for robust, high-throughput monitoring of the concentrations of phenolics that can affect the palatability of wine.  相似文献

Bisphenol A (BPA) is a synthetic industrial reactant used in the production of polycarbonate plastics, and genistein is a natural phytoestrogen abundant in the soybean. Current studies investigating the endocrine-disrupting effects of concomitant exposures to BPA and genistein have warranted the development of an analytical method for the simultaneous measurement of BPA and genistein, as well as their primary metabolites, bisphenol A ?-d-glucuronide (BPA gluc) and genistein 4′-?-d-glucuronide (genistein gluc), respectively. All four analytes were extracted from rat plasma via solid phase extraction (SPE). Three SPE cartridges and four elution schemes were tested. Plasma extraction using Bond Elut Plexa cartridges with sequential addition of ethyl acetate, methanol, and acetonitrile yielded optimal average recoveries of 98.1 ± 1.8% BPA, 94.9 ± 8.0% genistein, 91.4 ± 6.1% BPA gluc, and 103 ± 6.1% genistein gluc. Identification and quantification of the four analytes were performed by a validated HPLC-MS/MS method using electrospray ionization and selective reaction monitoring. This novel analytical method should be applicable to the measurement of BPA, genistein, BPA gluc, and genistein gluc in urine, cultures, and tissue following in vivo exposures. While reports of the determination of BPA and genistein independently exist, the simultaneous optimized extraction and detection of BPA, genistein, BPA gluc, and genistein gluc have not previously been reported.  相似文献

Summary The eleven priority, EPA phenolic pollutants were determined by liquid chromatography followed by two detectors in series; UV and electrochemical. Three different adsorbents, Envi-Carb (a carbon black) and two functionalized polymeric resins, Bond Elut PPL and another synthesized in our laboratory with an ocarboxybenzoyl moiety, were compared for solid-phase extraction (SPE) to detect lower concentrations of the eleven phenolics in natural waters. Higher recoveries were obtained using the functionalized polymeric adsorbents compared with Envi-Carb. When real samples were analysed, the synthetic adsorbent gave lower interference than Bond Elut PPL and phenol was determined at low levels with no humic and fulvic acid inter-ference when Na2SO3 was added. The linearity range for most compounds in tap water was 0.05–20 μg L−1 and the limits of detection were <35 ng L−1. Repeatability and reproducibility between days for real samples spiked at 0.1 μg L−1, expressed as relative standard deviation, were <8% and 10%, respectively.  相似文献

A simple and sensitive method was developed and validated for the simultaneous determination of 103 pesticide residues in tea by LC‐MS/MS. For the analysis of the pesticide with polarity, thermal lability or low volatility, this LC‐MS/MS method has an advantage over GC. In this work, residual pesticides were extracted from the tea sample with ACN and then purified using Carb‐NH2 SPE cartridges. Using the multiple reaction monitoring mode, the pesticides were quantified and identified by the most abundant and characteristic fragment ions. The recoveries obtained for each pesticide ranged between 65 and 114% at three spiked concentration levels. The intra‐day precisions were lower than 19.6%. Good linear relationships were observed with the correlation coefficients r2 >0.996 for all analytes. The established method was successfully applied to the determination of pesticide residues in real tea samples.  相似文献

Liquid chromatography with electrospray mass spectrometry (LC–ESI-MS) instrumentation equipped with a single quadrupole mass filter has been used to determine several benzoylphenylurea insecticides (diflubenzuron, triflumuron, hexaflumuron, lufenuron and flufenoxuron). Chromatographic and MS parameters were optimised to obtain the best sensitivity and selectivity for all pesticides. Solid-phase extraction (SPE) using C18 cartridges was applied for preconcentration of pesticide trace levels in river water samples. Recoveries of benzoylphenylurea pesticides from spiked river water (0.01 and 0.025 μg L−1) were between 73 and 110% and detection limits were between 3.5 and 7.5 ng L−1. The applicability of the method to the determination of benzoylphenylurea insecticides in spiked cucumber, green beans, tomatoes and aubergines was evaluated. Samples were extracted into dichloromethane without any clean-up step. The limits of detection ranged from 1.0 to 3.2 ng mL−1 (0.68 and 2.13 μg kg−1 in the vegetable samples). Mean recoveries ranged from 79 to 114% at spiking levels of 0.01 and 0.03 mg kg−1. The method was applied to determine traces of benzoylphenylureas in both river water and vegetable samples with precision values lower than 10%. Interferences due to the matrix effect were overcome using matrix-matched standards.  相似文献

Effluent from wastewater treatment plants have been identified as an important source of micro-organic contaminants in the environment. An online high-performance liquid chromatography–heated electrospray ionization tandem mass spectrometric method was developed and validated for the determination of basic pesticides in effluent wastewaters. Most available methods for pesticide analysis of wastewater samples are time-consuming, require complex clean-up steps and are difficult to automate. The method developed used a simple solid-phase extraction clean-up for salt and lipid reduction. On-line sample pre-concentration was performed using a reversed phase (C18) column, and analytes were separated by back-flushing onto an analytical column (C8) with detection using QqQ MS. An option to increase MS resolution was exploited to minimize interference from endogenous compounds in the matrix. A better than unit mass resolution was used (Q1 full width half maximum (FWHM) = 0.2 Da and Q3 FWHM = 0.7 Da), which was as rugged as a unit resolution method, and improved signal/noise and better detection limits were achieved for the targeted basic pesticides. This method was applied to the determination of 11 pesticides, including methoxytriazine, chlorotriazines, chloroacetanilides, phenylurea and carbamate pesticides. The percentage recovery values for these pesticides using the online trapping column were within the range, 73–95%, with relative standard deviation (RSD) values <8.9%. The highest concentrations of these pesticides in wastewater effluents in County Cork, Ireland, were simazine (0.51 μg/L), prometon (0.14 μg/L), diuron (0.21 μg/L) and atrazine (0.19 μg/L).  相似文献

Gas chromatography-mass spectrometry (GC/MS) was employed for the determination of 30 widely used pesticides including various transformation products and alkylphenols in water and agricultural soils with the aim of assessing the impact of these compounds in agricultural soils and the underlying aquifer. The extraction, clean-up, and analytical procedures were optimized for both water and soil samples to provide a highly robust method capable of determining target analytes at the ppb–ppt level with high precision. For water samples, different solid-phase extraction cartridges and conditions were optimized; similarly, pressurized liquid extraction conditions were tested to provide interference-free extracts and high sensitivity. Instrumental LODs of 3–4 pg were obtained. The multi-residue extraction procedures were applied to the analysis of groundwaters and agricultural soils from the Ebro river basin (NE Spain). Most ubiquitous herbicides detected were triazines but some acetanilides and organophosphorus pesticides were also found; the pesticide additive tributylphosphate was found in all water samples. Levels varied between 0.57 and 5.37 μg/L in groundwater, whereas nonylphenol was the sole compound detected in soil. Alkylphenols are used as adjuvants in pesticide formulations and are present in sludges employed as soil fertilizers. Occurrence was found to be similar to other environmental studies.  相似文献

Summary A method has been developed for determination of twenty-four polar pesticides—nine organophosphorus pesticides, thirteen organonitrogen compounds, and two triazine degradation products—in surface water. It entails extraction of the target pesticides from 1-L water samples by solid-phase extraction (SPE), then gas chromatography (GC) with large-volume (40 μL) injection. Filtered surface water, from the St Lawrence River in Canada and the River Loire and its tributaries in France, was extracted on cartridges filled with 500 mg Carbopack B (120–400 mesh). Analysis was performed by gas chromatography with a thermionic specific detector (GC-TSD) and a mass spectrometric (MS) detector. Overall percentage recoveries were satisfactory (>70%) for all target pesticides, with precision below 10%. Detection limits were between 0.5 and 4 ng L−1.  相似文献

A novel and simple method has been developed for the simultaneous determination of beta-lactam antibiotics (BLAs) (penicillin G, amoxicillin, ampicillin, penicillin V, oxacillin, cloxacillin, dicloxacillin and nafcillin) in wastewater. The method is based on solid-phase extraction (SPE) and high performance liquid chromatography with UV-diode array detection (UV-DAD). Two SPE cartridges have been compared for sample clean up and preconcentration: a reversed-phase silica-based cartridge (Bond Elut C18, Varian Inc.) and a strong polymeric mixed mode anion exchanger (Oasis MAX, Waters). The penicillins have been separated using a LUNA™ C18 (2) (150 mm × 4.6 mm, 5 μm) HPLC column and gradient elution with mobile phases consisting of aqueous trifluoroacetic acid and acetonitrile. The analytical wavelength was set at 220 nm. Under optimised conditions it was possible to preconcentrate up to 1000 mL of Milli-Q water in the Oasis MAX cartridges with recoveries in the range 82-97% (R.S.D. 2-9%) for all the antibiotic tested, except amoxicillin (52%, R.S.D. 8%), and limits of detection in the range of 8-24 ng L−1. The matrix components in industrial and urban wastewater samples reduce the preconcentration efficiency in both sorbents, especially for the Bond Elut C18. The use of the Oasis MAX allowed detection limits between 2.9-25.6, 2.5-12.4 and 2.2-12.7 μg L−1, when processing 250 mL of industrial, influent and effluent sewage treatment plant (STP) samples. Recoveries ranged between 46-91, 28-91 and 39-114% (industrial, influent and effluent STP, respectively) for samples spiked with all the antibiotics at 25 and 75 μg L−1 (n = 3 for each level).  相似文献

An efficient and sensitive method for simultaneous determination of 118 pesticide residues in teas has been established and validated. A multi-residue analysis of pesticides in tea involved extraction with ethyl acetate-hexane, clean-up using gel permeation chromatography (GPC) and solidphase extraction (SPE), and subsequent identification and quantification of the selected pesticides by gas chromatography-mass spectrometry (GC-MS). For most of the target analytes, optimized pretreatment processes led to no significant interference with analysis of sample matrix, and the determination of 118 compounds was achieved in about 60 min. In the linear range of each pesticide, the correlation coefficient was R 2 ≥ 0.99. At the low, medium and high three fortification levels of 0.05–2.5 mg kg−1, 118 pesticides average recoveries range from 61 % to 121 % and relative standard deviations (RSD) were in the range of 0.6–9.2 % for all analytes. The limits of detection for the method were 0.00030-0.36 mg kg−1, depending on each pesticide.  相似文献

The presence of pesticide residues in water is a huge worldwide concern. In this paper we described the development and validation of a new liquid chromatography tandem mass spectrometric (LC-MS/MS) method for both screening and quantification of pesticides in water samples. In the sample preparation stage, the samples were buffered to pH 7.0 and pre-concentrated on polymeric-based cartridges via solid-phase extraction (SPE). Highly sensitive detection was carried out with mobile phases containing only 5 mM ammonium formate (pH of 6.8) as an eluent additive and using only positive ionization mode in MS/MS instrument. Hence, only 200-fold sample enrichment was required to set a screening detection limit (SDL) and reporting limit (RL) of 10 ng/L. The confirmatory method was validated at 10 and 100 ng/L spiking levels. The apparent recoveries obtained from the matrix-matched calibration (5–500 ng/L) were within the acceptable range (60–120%), also the precision (relative standard deviation, RSD) was not higher than 20%. During the development, 480 pesticides were tested and 330 compounds fulfilled the requirements of validation. The method was successfully applied to proficiency test samples to evaluate its accuracy. Moreover, the method robustness test was carried out using higher sample volume (500 mL) followed by automated SPE enrichment. Finally, the method was used to analyze 20 real samples, in which some compounds were detected around 10 ng/L, but never exceeded the assay maximum level.  相似文献

Personal care products (PCPs) are widely used emerging contaminants which can cause adverse environmental effects. This paper reports the development and validation of a method based on solid-phase extraction (SPE) and ultra-high-performance liquid chromatography–electrospray ionisation–tandem mass spectrometry (UHPLC–(ESI)MS–MS) for simultaneously determining eleven PCPs: 4 preservatives (methylparaben; ethylparaben; benzylparaben; propylparaben); 2 antimicrobial agents (triclocarban and triclosan) and 5 UV filters (2,4-dihydroxybenzophenone; 2,2-dihydroxy-4-methoxybenzophenone; benzophenone-3; octocrylene and octyldimethyl-p-aminobenzoic acid) in environmental waters in only 9 run minutes of chromatographic separation. The SPE was carried out with two polymeric cartridges (Oasis HLB and Bond Elut Plexa). The recoveries obtained with Bond Elut Plexa were between 69% and 101% for 500 mL of river waters, with the exception of octyldimethyl-p-aminobenzoic acid (46%). Limits of detection for 500 mL of river water were in the range of 1–5 ng/L. Oasis HLB was chosen for wastewater samples with recoveries between 38% and 92% (250 mL of effluents) and 36–89% (100 mL of influents). In both wastewater samples, octyldimethyl-p-aminobenzoic acid and methylparaben showed the lowest recoveries (20% and 27%). The method revealed benzophenone-3 as having the highest concentration levels (7 ng/L) in river waters. Most of PCPs determined were found in influent waters being methylparaben and propylparaben the ones found at highest concentration with values of 5613 and 1945 ng/L, respectively. In effluent waters, significant lower levels of some PCPs were found, being benzophenone-3 the one found at the highest concentration (100 ng/L).  相似文献

A multiresidue method for the determination of more than 80 pesticides in water has been developed and validated. The proposed method is based on SPE followed by GC coupled to MS/MS. Different variables affecting SPE procedure, such as cartridges, sample volume and solvents were studied, and mass spectrometric conditions were optimised in order to increase selectivity and sensitivity. Calibration curves were linear over the range of 0.03-0.5 microg/L. Recoveries were in the range of 70-110% and repeatability was below 20% for the lowest calibration point. LODs ranged from 0.001 to 0.025 microg/L and LOQs from 0.003 to 0.076 microg/L. Finally, the method was successfully applied to the analysis of water samples from southeast of Spain.  相似文献

There is considerable interest in maximising the amount of information obtained from animal product analyses, when screening for the presence of veterinary drug residues. One of the barriers to effective multi-residue analysis to date has been a lack of effective clean up procedures to isolate a wide range of residues from the potential interferents, which may be present in both simple and complex (including processed) foods. A cation-exchange clean up has, therefore, been developed for use with acetonitrile extracts of foods, when analysing for several basic drug groups (sulfonamides, benzimidazoles, levamisole, nitroimidazoles, tranquillisers and fluroquinolones). The clean up procedure has also been shown to be effective using a modified extraction solvent for malachite green and leucomalachite green in fish.Several of the key parameters that influence analyte recovery have been investigated and in an optimised procedure, tissue/biofluid samples containing sulfonamides, benzimidazoles, levamisole, nitroimidazoles, tranquillisers and fluoroquinolones are first extracted with acetonitrile. The extract is then dried with sodium sulfate and acidified with glacial acetic acid before loading onto a Bond Elut, strong cation-exchange (SCX) solid phase extraction (SPE) cartridge. Extracts from fish containing malachite green and leucomalachite green can be cleaned up using the same SCX SPE procedure following extraction with citrate buffer/acetonitrile. Typical recoveries of drugs from low level fortified tissues using the optimised procedure lie in the range 53-104% with the exception of carazolol from pig kidney (31%), malachite green from trout (42-51%) and ciprofloxacin from chicken muscle (44%) and from egg (21%).  相似文献

A reliable multi-residue method for determining gaseous and particulate phase pesticides in atmospheric samples has been developed. This method, based on full scan gas chromatography–mass spectrometry (GC–MS), allowed the proper determination of sixteen relevant pesticides, in a wide range of concentrations and without the influence of interferences. The pesticides were benfluralin, bitertanol, buprofezin, chlorfenvinphos, chlorpyrifos, chlorpyrifos-methyl, ethalfluralin, fenthion, lindane, malathion, methidathion, propachlor, propanil, pyriproxifen, tebuconazol and trifluralin. Comparisons of two types of sampling filters (quartz and glass fibre) and four types of solid-phase cartridges (XAD-2, XAD-4, Florisil and Orbo-49P) showed that the most suitable supports were glass fibre filter for particulate pesticides and XAD-2 and XAD-4 cartridges for gaseous pesticides (>95% recovery). Evaluations of elution solvents for ultrasonic-assisted extraction demonstrated that isooctane is better than ethylacetate, dichloromethane, methanol or a mixture of acetone:hexane (1:1).Recovery assays and the standard addition method were performed to validate the proposed methodology. Moreover, large simulator chamber experiments allowed the best study of the gas-particle partitioning of pesticides for testing the sampling efficiency for the validation of an analytical multiresidue method for pesticides in air. Satisfactory analytical parameters were obtained, with a repeatability of 5 ± 1%, a reproducibility of 13 ± 3% and detection limits of 0.05–0.18 pg m−3 for the particulate phase and 26–88 pg m−3 for the gaseous phase. Finally, the methodology was successfully applied to rural and agricultural samples in the Mediterranean area.  相似文献

A multiresidue method using gas chromatography coupled to ion-trap tandem mass spectrometry (MS/MS) was developed for the analysis of 27 pesticides, commonly used in Alsace, in atmospheric samples (particle and gas phases). As pesticides are expected to be present at very low concentrations and in a complex matrix, the analytical method used was both highly selective and sensitive. These two properties were obtained by associating chromatography with ion-trap MS/MS. To develop this method, analysis of electron impact in single MS was first conducted to choose the parent ions of the pesticides studied. Among the 27 pesticides analysed, seven of them require a derivatisation step. This was the case of some ureas (chlorotoluron, diuron and isoproturon), phenoxy acids (2,4-dichlorophenoxyacetic acid, 4-chloro-2-methylphenoxyacetic acid and mecoprop) and of bromoxynil. The derivatisation was performed with success with pentafluorobenzylbromide. Then, a MS/MS method was optimised by parameters such as the radio frequency storage level and the collision-induced dissociation excitation voltage. Finally, a last step enabled the development of two calibrating programs based on the quantification of daughter ions for the 20 pesticides analysed directly (run 1) and for the seven pesticides which needed derivatisation (run 2). With this analytical procedure, the detection limits varied between 2.5 and 1,250 pg m–3 depending on the compounds studied. This method was tested with success for atmospheric samples collected in Strasbourg (France) during intensive pesticide treatment in 2002.  相似文献

Headspace solid phase microextraction (HS-SPME) was optimized for the analysis of pesticides with gas chromatography electron capture detection (GC-ECD) and high-resolution mass spectrometry. Factors influencing the extraction efficiency such as fiber type, extraction mode and temperature, effect of ionic strength, stirring and extraction time were evaluated. The lowest pesticide concentrations that could be detected in spiked aliquots after HS-SPME–GC-ECD ranged from 0.0005 to 0.0032 μg L− 1. Consequently hexachlorobenzene, trans-chlordane, 4,4′-DDD and 4,4′-DDE were detected in water samples after HS-SPME at concentrations ranging from 2.4 to 61.4 μg L− 1 that are much higher than the 0.1 μg L− 1 maximum limit of individual organochlorine pesticides in drinking water set by the European Community Directive. The same samples were cleaned with ISOLUTE C18 SPE sorbent with an optimal acetone/n-hexane (1:1 v/v) mixture for the elution of analytes. No pesticides were detected after SPE clean-up and pre-concentration. Precision for both methods was satisfactory with relative standard deviations less than 20%. This work demonstrated the superiority of HS-SPME as a sample clean-up and pre-concentration technique for pesticides in water samples as well as the need to identify and control point sources of pesticides.  相似文献

A fast multi-residue method based on dispersive solid-phase extraction (DSPE) followed by liquid chromatography–tandem mass spectrometry was developed for the simultaneous determination of 44 pesticides in raw bovine milk. Raw bovine milk samples did not percolate through SPE cartridges usually applied for pesticide extraction from homogenized pasteurized milk samples. Therefore, a DSPE technique was implemented and validated for the first time in this work. Graphitized non-porous carbon and C18 modified silica materials were tested both in combination with magnesium sulfate and bonded silica with ethylenediamine-N-propyl phase. The efficiency of the DSPE process was studied at several concentration levels obtaining the higher recoveries with C18 material. The method performance was also assessed and the limits of quantification reached the ng g−1 level, complying with the most recent maximum residue levels. The DSPE method was also shown to be suited to both the fatty and skimmed fractions issued from raw milk. Finally, the extraction method was successfully applied to the analysis of raw milk samples collected in 23 farms of dairy cattle from NW Spain (Galicia).  相似文献
