Gamma-irradiation was found to affect the physicochemical properties of dry red kidney beans. The highest dose used (8 kGy) significantly (P0.05) modified the extent of deamidation, the number of sulfhydryl groups, as well as the solubility and the hydrophobicity of the protein. Deamidation, protein solubility and hydrophobicity all increased with the irradiation dose while the number of sulfhydryl groups was reduced by the treatment. Furthermore, irradiation also affected the outgrowth of natural filamentous fungi contaminants present on the dry beans. A dose of 1.5 kGy reduced the number of filamentous fungi by 2 log cycles immediately after treatment. However, the highest dose used (3 kGy) did not eliminate the filamentous fungi completely. Moreover, the filamentous fungi population was a lot less diversified on the irradiated samples. Species of Aspergillus sp. and Penicillium sp. were more abundant on the unirradiated beans while the beans irradiated at 3 kGy contained were predominantly infected by species of Rhizopus sp. , Cladosporium sp. and Alternaria sp.  相似文献

Influence of gamma irradiation and storage on the microbial load, chemical and sensory quality of chicken kabab was investigated. Chicken kabab was treated with 0, 2, 4 or 6 kGy doses of gamma irradiation. Treated and untreated samples were kept in a refrigerator (1–4 °C). Microbiological, chemical and sensory characteristics of chicken kabab were evaluated at 0–5 months of storage. Gamma irradiation decreased the microbial load and increased the shelf-life of chicken kabab. Irradiation did not influence the major constituents of chicken kabab (moisture, protein and fats). No significant differences (p>0.05) were observed for total acidity between non-irradiated (control) and irradiated chicken kabab. Thiobarbitric acid (TBA) values (expressed as mg malonaldehyde (MDA)/kg chicken kabab) and volatile basic nitrogen (VBN) in chicken kabab were not affected by the irradiation. Sensory evaluation showed no significant differences between irradiated and non-irradiated samples.  相似文献

Multi-walled carbon nanotubes(MWCNTs) were grafted with poly(N-vinyl-2-pyrrolidone) and with poly(2-acrylamido-2-methyl-1-propanesulfonic acid) at different compositions by using γ-rays technique as initiator. The MWCNTs, MWCNT-graft-PNVP, MWCNT-graft-PAMPS and MWCNTs-graft-P(NVP-co-AMPS) were characterized by Fourier transform infra red(FTIR) spectroscopy, X-ray diffraction(XRD), thermal gravimetric analysis(TGA), elemental analysis and high-resolution transmission electron microscopy(HR-TEM). The results indicated that the grafting processes of PNVP, PAMPS and P(NVP-co-AMPS) occurred on to the surfaces of MWCNTs without destroying the framework of MWCNTs. Tramadol hydrochloride(TH) was loaded as model drug and its release behavior was analyzed via various kinetic models. Release of the loaded TH was studied in simulated gastric fluid(SGF, p H = 1.2) and simulated intestinal fluid(SIF, p H = 7.4) at 37 °C. Controlled release of TH from grafted MWCNTs was investigated. The outcome results suggest that the grafted MWCNTs could be used as a promising matrix candidate for oral drug delivery system by harmonization between the composition and p H level of the simulated biological fluids.  相似文献

Gamma irradiation is highly effective in inactivating microorganisms in various foods and offers a safe alternative method of food decontamination. In the present study, soybeans (Glycine max L. Merrill) were treated with 0, 1.0, 3.0, 5.0 and 10.0 KGy of gamma irradiation. Microbial populations on soybeans, isoflavone, tocopherol contents, raffinose family oligosaccharides, color and sensory properties were evaluated as a function of irradiation dose. The results indicated that gamma irradiation reduced aerobic bacterial and fungal load. Irradiation at the doses applied did not cause any significant change (p>0.05) in the contents of isoflavone of soybeans, but decreased tocopherol contents. The content of key flatulence-producing raffinose family oligosaccharides in irradiated soybeans (10.0 kGy) decreased by 82.1% compared to the control. Sensory analysis showed that the odor of the soybeans was organoleptically acceptable at doses up to 5.0 kGy and no significant differences were observed between irradiated and nonirradiated samples in flavor, texture and color after irradiation.  相似文献

The Brazilian bean varieties Phaseolus vulgaris L. var. Carioca and Vigna unguiculata (L.) Walp var. Macaçar were irradiated with doses of 0.5, 1.0, 2.5, 5.0 and 10 kGy and subsequently stored at ambient temperature for 6 months. The anti-nutrients phenolic compounds, tannins and phytate were determined to be 0.48 mg g−1 dry basis, 1.8 mg g−1 dry basis and 13.5 μmol g−1 dry basis in the raw non-irradiated Carioca beans and 0.30 mg g−1 dry basis, 0.42 mg g−1 dry basis and 7.5 μmol g−1 dry basis in the raw non-irradiated Macaçar beans. After soaking and cooking a higher content of phenolic compounds and a lower phytate content was observed in both bean varieties. Tannin content was not affected by soaking and cooking of Carioca beans, but higher after soaking and cooking of Macaçar beans. Using radiation doses relevant for food did not effect the content of the anti-nutrients under investigation in both bean varieties.  相似文献

Two lots of mangoes of the Nahng Glahng Wahn variety from Thailand were irradiated at the Thai Irradiation Center (TIC) at 0.49 to 0.77 kGy. Following this, one batch was retained in Thailand (TIC) while the other was shipped to the Canadian Irradiation Center (CIC) for investigation of the same variables during storage. This way, it was hoped to compare the effects of gamma radiation with hot water dip (HWI) or without (I) before and after transportation, on the respiratory activity, rotting, texture, ph, titratable acidity and on the total soluble solids in mangoes. Irradiation stimulated carbon dioxide (CO2) production early in the study but delayed ethylene (C2H4) production which peaked later. The production of CO2 and C2H4 occurred later at CIC than at TIC. The I and particularly HWI treatments also delayed the rotting of the mangoes. The texture of the mangoes declined earlier and more rapidly during storage in Thailand than in Canada. The irradiated samples developped a softer texture early during storage and the rise in pH was less marked than the controls. The control showed lower values of total soluble solids during the first 10 days of storage but increased later.  相似文献

The Amatungula fruit, from Carissa macrocarpa, is commonly consumed by the local people of KwaZulu-Natal (KZN), South Africa. Levels of elements in the fruit were determined to assess if they conform to recommended dietary allowances (RDAs) and to assess for potential toxicities. Soils and fruit samples from nine sites in eastern KZN were investigated. Concentrations of elements in the fruit were found to be in the order of Ca > Mg > Fe > Mn ≈ Cu ≈ Pb > Se > Cr > Ni > Zn. For the elements in focus, except for Pb, all of the elements found in the fruit contribute significantly towards the RDAs. Lipid profiling was also done to determine the fruits potential as a source of essential fatty acids. The fruit was rich in monounsaturated and essential fatty acids with the linoleic acid to α-linolenic acid ratio conforming to the recommended range for cardiac health. Concentrations of elements in soil had no significant effect on plant concentrations, but competition between elements in soil influenced their availability. Total soil concentrations of most metals studied have significantly correlated Pb availability, indicating the impact of these metals on Pb availability. The Amatungula fruit showed tendency to accumulate Pb, with Pb levels in fruit at all sites being toxic to human health. Site location had a major effect on plant concentrations however uptake and distribution was primarily dependent on the plants inherent controls, as evidenced by the accumulation and exclusion of elements, to meet its physiological requirements.  相似文献

A simple and sensitive method for the analysis of sterols, tocopherols and triterpenic dialcohols from the unsaponifiable fraction from oil samples in a single analytical run using liquid chromatography coupled to mass spectrometry was developed. With this method, the compounds could be detected directly after dissolving the unsaponifiable fraction in acetonitrile without necessity of time-consuming sample pre-treatment or derivatization. Separation of the analytes was carried out at room temperature, by using a C18 column (5 μm i.d. 3.0 mm × 250 mm) with a linear gradient of acetonitrile/water (0.01% acetic acid) at a flow rate of 1.5 mL/min. The full scan mass spectra of the investigated compounds were measured by an ion trap mass spectrometer equipped with an APCI ion source. The optimized methodology was suitable for the identification of 23 compounds belonging to different families present in olive oil and other kinds of oils, as well as for the quantification of 15 analytes (vs. their commercial standards).  相似文献

Novel 4‐ethynylphthaloyl amino acid esters carrying different terminal groups, 4‐ethynylphthaloyl glycine (1S,2R,5S)‐menthyl ester ( 1 ), 4‐ethynylphthaloyl glycine (1R,2S,5R)‐menthyl ester ( 2 ), 4‐ethynylphthaloyl L ‐leucine methyl ester ( 3 ), 4‐ethynylphthaloyl L ‐leucine (1S,2R,5S)‐menthyl ester ( 4 ), 4‐ethynylphthaloyl L ‐leucine (1R,2S,5R)‐menthyl ester ( 5 ) were synthesized and polymerized with a rhodium catalyst. Polymers with high molecular weights were obtained in 71–92% yields. The helical conformation of the polymers could be tuned by the chirality of the amino acid connected to the backbone, together with the chirality and bulkiness of the terminal pendent groups. © 2008 Wiley Periodicals, Inc. Amino alcohol alkaloids are the active components in the lateral root of Aconitum carmichaelii Debx. (Fuzi), and they have a variety of pharmacological activities. However, the chemical fingerprints of the ester alkaloids reported to date were mainly obtained from high‐performance liquid chromatography coupled with ultraviolet detection, and it is difficult to obtain information about amino alcohol alkaloids in Fuzi from such chromatograms. In this paper, a comprehensive fingerprinting method was established using high‐performance liquid chromatography coupled with an evaporative light‐scattering detector for the simultaneous quantitative analysis of both the amino alcohol alkaloids and ester alkaloids. A total of 42 samples of Fuzi from four production areas were analyzed by constructing high‐performance liquid chromatography fingerprints. Then, the quantitative results of the chemical fingerprints combined with chemometrics methods were employed to reveal the factors affecting the geo‐authentic Fuzi and to determine characteristic components that can be used to identify these samples. The results indicated distinct differences in the alkaloid contents among samples from the four regions; the geographical origin may be the primary factor affecting the geo‐authentic Fuzi, and 15 major components (including songorine, neoline, and hypaconitine, which were quantitatively determined) were found to be characteristic components for the discrimination of Fuzi samples from various regions. Neoline might be a critical component for identifying geo‐authentic Fuzi. This approach is convenient, reproducible and provides a promising method for the quality evaluation of Fuzi.  相似文献

The irradiated groups appeared to have a slightly higher content of ascorbic acid on the first day after irradiation than their corresponding controls. The sensory evaluation indicates that the panelists mostly favoured the I and HWI irradiated groups for the overall appearance and texture of whole mangoes and pulp and in taste and palatability of the pulp alone. Transportation stress did not appear to have much of an effect on the ascorbic acid and dehydroascorbic acid levels and the sensorial tests. The mangoes stored and studied in Canada appeared to have a lower vitamin A content.  相似文献

A great number of analyses is performed every year, the results of which are used for many purposes, e.g. the quality of goods and food, the status of quality of the environment or the health of patients. The accuracy of these results is a prerequisite for a good interpretation of the data obtained. One of the most powerful tool for achieving quality control of chemical analysis is to use reference materials (RMs) and certified reference materials (CRMs). These materials are necessary for one or more of the following items: method validation (CRMs), monitoring of the state of statistical control (RMs), samples in inter-comparisons (RMs), etc. The requirements and use of RMs and CRMs in chemical analysis are described, with special emphasis on environmental analysis, and some examples of environmental materials currently in production within the Standards, Measurements and Testing Programme (formerly BCR) of the European Commission are given.  相似文献

High hydrostatic pressure (HHP) treatment is a non-thermal processing technology, which is widely used in the food processing field at present. In this study, the effects of HHP treatment (100~500 MPa for 5 min) on the physicochemical properties, texture parameters, and volatile flavor compounds of oysters were investigated. The results showed that HHP treatment increased the water content while reducing the crude protein and ash content of the oyster. Texture parameters showed that HHP treatment improved the hardness, springiness, chewiness, and cohesiveness of oysters, compared with the control group. In addition, the saturated fatty acid (SFA) content was slightly increased after HHP treatment, while the difference in monounsaturated fatty acid (MUFA) and polyunsaturated fatty acid (PUFA) content was not significant. Furthermore, HHP increased hexenoic aldehyde, 2,4-heptadienal, 1-octene-3-ol, and 2-octen-1-ol and decreased the contents of 3. 6-nadien-1-ol, 3-octanone, and 2-undecanone, suggesting that HHP might inhibit the fishiness of oyster and showed a positive effect on its flavor. Based on the above results, HHP improved the edible qualities such as texture properties and volatile flavor of oysters. This meets the requirements of consumers on the edible quality of seafood and provides new ideas for the development of seafood.  相似文献

New Pb(II), Mn(II), Hg(II), and Zn(II) complexes, derived from 4-(4-chlorophenyl)-1-(2-(phenylamino)acetyl)thiosemicarbazone, were synthesized. The compounds with general formulas, [Pb(H2L)2(OAc)2]ETOH.H2O, [Mn(H2L)(HL)]Cl, [Hg2(H2L)(OH)SO4], and [Zn(H2L)(HL)]Cl, were characterized by physicochemical and theoretical studies. X-ray diffraction studies showed a decrease in the crystalline size of compounds that were exposed to gamma irradiation (γ-irradiation). Thermal studies of the synthesized complexes showed thermal stability of the Mn(II) and Pb(II) complexes after γ-irradiation compared to those before γ–irradiation, while no changes in the Zn(II) and Hg(II) complexes were observed. The optimized geometric structures of the ligand and metal complexes are discussed regarding density functional theory calculations (DFT). The antimicrobial activities of the ligand and metal complexes against several bacterial and fungal stains were screened before and after irradiation. The Hg(II) complex has shown excellent antibacterial activity before and after γ-irradiation. In vitro cytotoxicity screening of the ligand and the Mn(II) and Zn(II) complexes before and after γ-irradiation disclosed that both the ligand and Mn(II) complex exhibited higher activity against human liver (Hep-G2) than Zn(II). Molecular docking was performed on the active site of MK-2 and showed good results.  相似文献

Surface treatment procedures such as grinding and polishing are needed to provide the ceramic dental restorative materials with proper fitting and occlusion. The treated surfaces are customarily glazed to improve the strength and smoothness. Though smoothness and wetting of the dental surfaces are important to minimize bacterial plaque retention, influence of the surface treatment and glazing procedures on the final surface roughness and its correlation to wettability are overlooked.

In this work, effect of various treatment (diamond fraising, stoning, sanding and aluminum oxide and rubber polishing) and glazing (auto and overglazing) techniques on the final roughness and the resulting wettability of dental ceramic surfaces were investigated using scanning electron microscopy (SEM) observations and atomic force microscopy (AFM) scans, 75 scans per sample. The surfaces were characterized and assigned an average roughness measure, Ra. The wettability of the same surfaces was evaluated using micro-contact angle measurements (25 micro-bubbles placed on a grid on each surface) to correlate the final surface roughness and wettability.

The results show that overglazing prevails over surface irregularities from different treatment procedures and provides homegeneously smooth surfaces with mean Ra < 10 nm. It also produces uniformly wetted surfaces with low contact angles around 20°. The autoglazed surfaces are less smooth (mean Ra around 50 nm) and displays sporadic topographic irregularities. They display larger and less uniform contact angles ranging between 35° and 50°. The results suggest that overglazing should be preferred after surface treatment to obtain a smooth and well-wetted dental ceramic surface.  相似文献

The effects of the electrolyte composition, deposition potential, temperature, and the salt-solvent cation on the oxygen factor (the [O]/[U] atomic ratio) of uranium oxides obtained by potentiostatic electrolysis of molybdate melts are studied. It is shown that the oxygen factor of the cathodic product increases with the melt temperature. The shift of the deposition potential toward more electronegative values, an increase in the Mo2O2? 7 ion concentration, and a decrease in the UO2MoO4 concentration favor the cathodic formation of uranium oxides with lower oxygen factors. All other conditions being the same, the oxygen factor decreases with increasing the salt-solvent cation radius. The observed anomalous behavior of the Li2MoO4-based electrolytes can be caused by a lower activity in the melts of the O2? anions that are capable of forming stable complexes Li3O+ with lithium cations. The obtained results can be reasonably explained in terms of the model of ionic composition of the uranyl-containing tungstate melts, which is based on the concept of involvement of the uranyl ions in the complexes’ formation and stepwise solvolysis.  相似文献

The effect of substituents on the strength of N-X (X = H, F, and Cl) bonds has been investigated using the high-level W2w thermochemical protocol. The substituents have been selected to be representative of the key functional groups that are likely to be of biological, synthetic, or industrial importance for these systems. We interpreted the effects through the calculation of relative N-X bond dissociation energies (BDE) or radical stabilization energies (RSE(NX)). The BDE and RSE(NX) values depend on stabilizing/destabilizing effects in both the reactant molecule and the product radical of the dissociation reactions. To assist us in the analysis of the substituent effects, a number of additional thermochemical quantities have been introduced, including molecule stabilization energies (MSE(NX)). We find that the RSE(NH) values are (a) increased by electron-donating alkyl substituents or the vinyl substituent, (b) increased in imines, and (c) decreased by electron-withdrawing substituents such as CF(3) and carbonyl moieties or through protonation. A different picture emerges when considering the RSE(NF) and RSE(NCl) values because of the electronegativities of the halogen atoms. The RSE(NX)s differ from the RSE(NH) values by an amount related to the stabilization of the N-halogenated molecules and given by MSE(NX). We find that substituents that stabilize/destabilize the radicals also tend to stabilize/destabilize the N-halogenated molecules. As a result, N-F- and N-Cl-containing molecules that include alkyl substituents or correspond to imines are generally associated with RSE(NF) and RSE(NCl) values that are less positive or more negative than the corresponding RSE(NH). In contrast, N-F- and N-Cl-containing molecules that include electron-withdrawing substituents or are protonated are generally associated with RSE(NF) and RSE(NCl) values that are more positive or less negative than the corresponding RSE(NH).  相似文献

The effect of different light qualities (white, blue, green, yellow and red light) on photosynthesis, measured as chlorophyll fluorescence, and the accumulation of photosynthetic pigments, proteins and the UV-absorbing mycosporine-like amino acids (MAAs) was studied in the red alga Porphyra leucosticta. Blue light promoted the highest accumulation of nitrogen metabolism derived compounds i.e., MAAs, phycoerythrin and proteins in previously N-starved algae after seven days culture in ammonium enriched medium. Similar results were observed in the culture under white light. In contrast, the lowest photosynthetic capacity i.e., lowest electron transport rate and lowest photosynthetic efficiency as well as the growth rate were found under blue light, while higher values were found in red and white lights. Blue light favored the accumulation of the MAAs porphyra-334, palythine and asterina-330 in P. leucosticta. However, white, green, yellow and red lights favored the accumulation of shinorine. The increase of porphyra-334, palythine and asterina-330 occurred in blue light simultaneous to a decrease in shinorine. The accumulation of MAAs and other nitrogenous compounds in P. leucosticta under blue light could not be attributed to photosynthesis and the action of a non-photosynthetic blue light photoreceptor is suggested. A non-photosynthetic photoreceptor could be also involved in the MAAs interconversion pathways in P. leucosticta.  相似文献
