2,4‐Disubstituted 1‐benzoyl‐2,3‐dihydro‐1H‐1,5‐benzodiazepines were oxidized by m‐chloroperbenzoic acid (MCPBA) to produce 1a,3‐disubstituted 4‐benzoyl‐1a,2,3,4‐tetrahydro‐oxazirino[2,3‐a][1,5]benzodiazepines, a novel tricyclic heterocyclic system. However, 2,4‐disubstituted 2,3‐dihydro‐1H‐1,5‐benzodiazepines were oxidized by MCPBA under the same conditions to give a 2,4‐disubstituted 2,3‐dihydro‐2‐hydroxy‐1H‐1,5‐benzodiazepine or a 2,4‐disubstituted 3H‐1,5‐benzodiazepine, respectively. 2a,4‐Disubstituted 5‐benzoyl‐2‐chloro/2,2‐dichloro‐2a,3,4,5‐tetrahydro‐azeto [1,2‐a] [1,5]benzodiazepin‐1 (2H)‐ones ( 3a–h ) were synthesized by cycloaddition reactions of 2,4‐disubstituted 1‐benzoyl‐2,3‐dihydr o‐1H‐1,5‐benzodiazepines ( 2a–h ) and ketenes, generated from chloroacetyl chloride or dichloroacetyl chloride in the presence of triethylamine, in anhydrous benzene. The 2,3‐dihydro‐7‐methyl‐1H,5H‐pyrido[3,2,1‐ij]quinoline‐1,5‐dione derivatives 9 and 10 were prepared from 3‐(5,7‐dimethoxy‐4‐methyl‐2‐oxo‐2H‐quinolin‐1‐yl)propionitrile ( 6 ). Cyclodehydration of the amide 8 gave 1,2‐dihydro‐7,9‐dimethoxy‐6‐methylpyimido[1,2‐a]quinolin‐3‐one ( 11 ).  相似文献

A general and easy method for the synthesis of several 1‐H‐1,5‐benzodiazepines using SiO2/ZnCl2 under solvent‐free conditions is described. This efficient and improved method furnishes selectively and in good yields the corresponding 1‐H‐1,5‐benzodiazepines derivatives starting from o‐phenylenediamine and cyclic or acyclic ketones. The catalytic system was reused up four times, and the use of focused microwaves accelerates the reaction. © 2011 Wiley Periodicals, Inc. The condensation of α,β‐unsaturated ketones with substituted o‐aminothiophenols, obtained by reductive cleavage of the corresponding disulfides in the presence of triphenylphosphine, is an effective method for the synthesis of 2,4‐diaryl‐2,3‐dihydro‐1,5‐benzothiazepines under neutral conditions.  相似文献

A series of novel 2,3‐diaryl‐3,4‐dihydro‐2H‐1,3‐benzoxazines have been prepared in high yields from o‐arylaminomethylphenols and aromatic aldehydes in the presence of SnCl4 for the first time, and their fungicidal activities were investigated too. In the present study, a series of 20 indane‐based 1,5‐benzothiazepines ( 5a – t ) has been prepared derived from 3‐phenyl‐2,3‐dihydro‐1H ‐inden‐1‐one ( 1 ). All the synthesized 1,5‐benzothiazepines ( 5a – t ) were screened for their in vitro antimicrobial activities against four bacteria [Bacillus subtilis (MTCC 441), Staphylococcus epidermidis (MTCC 6880), Escherichia coli (MTCC 1652), and Pseudomonas aeruginosa (MTCC 424)] and two fungi [Candida albicans (MTCC 227) and Aspergillus niger (MTCC 8189)]. Among all the tested derivatives, 5n and 5o against E. coli displayed more inhibitory activity than that of the reference drug, ciprofloxacin, while the derivatives 5c , 5m – o , 5s , and 5t against C. albicans , and 5d , 5e , 5n , 5o , 5s , and 5t against A. niger were found to be more potent than the standard drug, that is, fluconazole.  相似文献

A tandem reduction‐reductive amination reaction has been applied to the synthesis of (±)‐4‐alkyl‐2,3,4,5‐tetrahydro‐1,5‐benzoxazepines and (±)‐4‐alkyl‐1‐benzoyl‐2,3,4,5‐tetrahydro‐1H‐1,5‐benzodiazepines. The nitro aldehydes and ketones required for 1,5‐benzoxazepine ring closures were prepared by nucleophilic aromatic substitution of the alkoxides from several 3‐buten‐1‐ol derivatives with 2‐fluoro‐1‐nitrobenzene followed by ozonolysis. Precursors for the 1,5‐benzodiazepines were prepared by similar addition of N‐(3‐butenyl)benzamide anions to 2‐fluoro‐1‐nitrobenzene followed by ozonolysis. Catalytic hydrogenation of the nitro carbonyl compounds using 5% palladium‐on‐carbon in methanol then gave the target heterocycles by a tandem reduction‐reductive amination sequence. The 1,5‐benzoxazepines were isolated in high yield following chromatographic purification; the 1,5‐benzodiazepines were isolated as solids directly from the hydrogenation mixture and possessed differentiated functionality on the two nitrogen atoms.  相似文献

Functionalized 2,3‐dihydro‐1,3‐thiazin‐4(1H)‐one derivatives have been synthesized by cyclocondensation of 3‐alkyl(aryl)amino‐2‐cyano‐3‐mercaptoacrylamides with aldehydes and ketones under acidic catalysis. 6‐Alkyl(aryl)amino‐5‐cyano‐2,3‐dihy‐ dro‐1,3‐thiazin‐4(1H)‐ones, when treated with a dilute solution of potassium hydroxide, are converted into the potassium salts of isomeric compounds, 1‐alkyl‐ (aryl)‐5‐cyano‐6‐mercapto‐2,3‐dihydropyrimidin‐ 4(1H)‐ones. Alkylation of the latter with dimethyl sulfate in situ furnishes 1‐alkyl(aryl)‐6‐alkylthio‐5‐ cyano‐2,3‐dihydropyrimidin‐4(1H)‐ones, whereas boiling them in ethanol with an excess of hydrochloric acid leads to starting 2,3‐dihydro‐1,3‐thiazin‐4(1H)‐ones. © 2005 Wiley Periodicals, Inc. (±)‐cis‐2‐(4‐Methoxyphenyl)‐3‐hydroxy/methoxy‐6‐ethoxy/phenoxy‐2,3‐dihydro‐1,5‐benzothiazepin‐4‐[5H/5‐chloroacetyl/5‐(4′‐methylpiperazino‐1′)acetyl]‐ones have been synthesized by the condensation of 2‐amino‐3‐ethoxy/phenoxybenzenethiol with methyl‐(±)‐trans‐3‐(4‐methoxyphenyl)glycidate in xylene. Ribofuranosides, viz. (±)‐cis‐2‐(4‐methoxyphenyl)‐3‐methoxy‐6‐ethoxy/phenoxy‐2,3‐dihydro‐1,5‐benzothiazepin‐4‐[5‐(2′,3′,5′‐tri‐O‐benzoyl‐β‐D ‐ribofuranosyl)]‐ones, have been synthesized by the treatment of 3‐methoxy derivatives of 1,5‐benzothiazepines with a derivative, sugar, viz. β‐D ‐ribofuranose‐1‐acetate‐2,3,5‐tribenzoate, in toluene in vacuo. The structures of all the synthesized ribofuranosides and their precursors have been characterized on the basis of elemental analyses and IR, 1H NMR, and 13C NMR spectral data. A series of benzo-fused heteroaromatic compounds with 5-, 6- and 7-membered tings, such as benzimidazole,quinoxaline and 1H-1,5-benzodiazepine derivatives, were synthesized through condensation reaction of o-pheny1-enediamine with ary1 aldehydes or ketones. The experimental conditions were carefully examined, and the products were characterized by ^1H NMR, ^13C NMR, MS, IR and elemental analyses. In addition, the structure of a benzodiazaepine derivative with 7-membered ring was confirmed by single crystal X-ray diffraction analysis.  相似文献

The title compound, Na+·C9H7N4O5S·2H2O, presents a Z configuration around the imine C=N bond and an E configuration around the C(O)NH2 group, stabilized by two intra­molecular hydrogen bonds. The packing is governed by ionic inter­actions between the Na+ cation and the surrounding O atoms. The ionic unit, Na+ and 2‐oxo‐3‐semicarbazono‐2,3‐dihydro‐1H‐indole‐5‐sulfonate, forms layers extending in the bc plane. Orthophthalaldehyde (o‐phthalaldehyde, OPA) is an aromatic dialdehyde bearing two electron‐withdrawing carbonyl groups. The reactions of OPA with primary amines are broadly applied for the synthesis of important heterocyclic compounds with biological relevance. A number of such reactions have been investigated recently and several structures of condensation products have been reported, however, the complex reaction mechanism is still not fully understood and comprises concurrent as well as consecutive reactions. The reaction products depend on the primary amine which reacts with OPA, the reaction environment (solvent) and the proportion of the reactants. The title molecule, C11H13NO, the product of the reaction of OPA with isopropylamine, contains a five‐membered pyrrole C4N ring with a carbonyl substituent, which forms part of the isoindolinone unit. Though this pyrrole ring contains one C atom in the sp3‐hybridized state, it is fairly planar. The title molecule has been compared with similar structures retrieved from the Cambridge Structural Database in order to study this phenomenon. The planarity of this fragment has been explained by the presence of partially delocalized C—C, C—N and C—O bonds, and by an inner angle in the planar pentagonal ring (∼108°), which is close to the ideal tetrahedral value for the sp3‐hybridized state of the constituent C atom. Due to this propitious angle, this C atom can be present in states intermediate between sp3‐ and sp2‐hybridized in different structures, while still maintaining the planarity of the ring. There are only weak intermolecular C—H…O hydrogen bonds and C—H…π‐electron ring interactions in the structure. In particular, it is the pyrrole ring which is involved in these interactions.  相似文献

Novel 2‐alkylcarbamato/thiocarbama‐to‐2,3‐dihydro‐5‐propylthio‐1H‐1,3,2‐benzodiazaphos‐phole 2‐oxides ( 4a–J ) were synthesized by cyclization of 4‐propylthio‐1,2‐phenylenediamine ( 3 ) with the corresponding dichlorophosphoryl carbamates/thiocarbamates ( 2a–J ) that were obtained by the addition of alcohols/thiols to isocyanatophosphoryl dichloride ( 1 ). The structures of the title compounds were confirmed by the 1H, 13C, 31P NMR, and mass spectral studies. A series of novel 4‐(3,3‐dimethylspiro{bicyclo[2.2.1]heptan‐2,5′‐isoxazoline‐2}‐3′‐yl)‐2‐phenyl‐2,3‐dihydro‐1H‐1,5‐benzodiazepines were synthesized. These molecules were screened in vitro for their antifungal and antibacterial activity, and none of the tested compounds showed promising antimicrobial or antifungal activity. J.
