A single‐drop microextraction (SDME) method followed by in‐syringe derivatization and GC‐MS determination has been developed for analysis of five parabens, including methyl, ethyl, isopropyl, n‐propyl and n‐butyl paraben in water samples and cosmetic products. N,O‐Bis(trimethylsilyl)acetamide (BSA) was used as derivatization reagent. Derivatization reaction was performed inside the syringe barrel using 0.4 μL of BSA. Parameters that affect the derivatization yield such as temperature and time of the reaction were studied. In addition, experimental SDME parameters such as selection of organic solvent, addition of salt, extraction time and extraction temperature were investigated and optimized. The RSD of the method for aqueous samples varied from 8.1 to 13%. The LODs ranged from 0.001 (n‐butyl paraben) to 0.015 (methyl paraben) μg/L, and the enrichment factors were between 23 and 150.  相似文献

In this work, for the first time, headspace (HS) single‐drop microextraction and simultaneous derivatization followed by GC‐MS was developed to determine the aliphatic amines in tobacco samples. In the HS extraction procedure, the mixture of derivatization reagent and organic solvent was employed as the extraction solvent for HS single‐drop microextraction and in situ derivatization of aliphatic amine in the samples. Fast extraction and simultaneous derivatization of the analytes were performed in a single step, and the obtained derivatives in the microdrop extraction solvent were analyzed by GC‐MS. The optimized experiment conditions were: sample preparation temperature of 80°C and time of 30 min, HS extraction solvent (the mixture of benzyl alcohol and 2,3,4,5,6‐pentafluorobenzaldehyde) volume of 2.0 μL, extraction time of 90 s. With the optimal conditions, the method validations were also studied. The method has good linearity (R2 more than 0.99), accepted precision (RSD less than 13%), good recovery (98–104%) and low limit of detection (0.11–0.97 μg/g). Finally, the proposed technique was successfully applied to the analyses of aliphatic amines in tobacco samples of seven different brands. It was further demonstrated that the proposed method offered a simple, low‐cost and reliable approach to determine aliphatic amines in tobacco samples.  相似文献

Methylation is an essential metabolic process in the biological systems, and it is significant for several biological reactions in living organisms. Methylated compounds are known to be involved in most of the bodily functions, and some of them serve as biomarkers. Theoretically, all α‐amino acids can be methylated, and it is possible to encounter them in most animal/plant samples. But the analytical data, especially the mass spectral data, are available only for a few of the methylated amino acids. Thus, it is essential to generate mass spectral data and to develop mass spectrometry methods for the identification of all possible methylated amino acids for future metabolomic studies. In this study, all N‐methyl and N,N‐dimethyl amino acids were synthesized by the methylation of α‐amino acids and characterized by a GC‐MS method. The methylated amino acids were derivatized with ethyl chloroformate and analyzed by GC‐MS under EI and methane/CI conditions. The EI mass spectra of ethyl chloroformate derivatives of N‐methyl ( 1–18 ) and N,N‐dimethyl amino acids ( 19–35 ) showed abundant [M‐COOC2H5]+ ions. The fragment ions due to loss of C2H4, CO2, (CO2 + C2H4) from [M‐COOC2H5]+ were of structure indicative for 1–18 . The EI spectra of 19–35 showed less number of fragment ions when compared with those of 1–18 . The side chain group (R) caused specific fragment ions characteristic to its structure. The methane/CI spectra of the studied compounds showed [M + H]+ ions to substantiate their molecular weights. The detected EI fragment ions were characteristic of the structure that made easy identification of the studied compounds, including isomeric/isobaric compounds. Fragmentation patterns of the studied compounds ( 1–35 ) were confirmed by high‐resolution mass spectra data and further substantiated by the data obtained from 13C2‐labeled glycines and N‐ethoxycarbonyl methoxy esters. The method was applied to human plasma samples for the identification of amino acids and methylated amino acids. Copyright © 2016 John Wiley & Sons, Ltd.  相似文献

A method for the quantitative determination of ten musk fragrances extensively used in personal care products from sewage sludge was developed by using a pressurized liquid extraction (PLE) followed by an automated ionic liquid‐based headspace single‐drop microextraction and gas chromatography‐tandem mass spectrometry. The influence of main factors on the efficiency of PLE was studied. For all musks, the highest recovery values were achieved using 1 g of pretreated sewage sludge, H2O/methanol (1:1) as an extraction solvent, a temperature of 80°C, a pressure of 1500 psi, an extraction time of 5 min, 2 cycles, a 100% flush volume, a purge time of 120 s, and 1 g Florisil as in‐cell clean‐up extraction sorbent. The use and optimization of an in‐cell clean‐up sorbent was necessary to remove fatty interferents of the PLE extract that make the subsequent ionic liquid‐based headspace single‐drop microextraction difficult. Validation parameters, namely LODs and LOQs, ranged from 0.5–1.5 to 2.5–5 ng/g, respectively. Good levels of intra‐ and interday repeatabilities were obtained analyzing sewage sludge samples spiked at 10 ng/g (n = 3, RSDs < 10%). The method applicability was tested with sewage sludge from different wastewater treatment plants. The analysis revealed the presence of all the polycyclic musks studied at concentrations higher than the LOQs, ranging from 6 to 530 ng/g. However, the nitro musk concentrations were below the LOQs or, in the case of musk xylene, was not detected.  相似文献

Cup horn boosters are miniaturized ultrasound baths that maximize efficiency and precision. The optimization of an ultrasonic‐assisted derivatization step by means of a cup horn booster and the determination of estrone, 17β‐estradiol, estriol, 17α‐ethynyl estradiol and mestranol was developed by GC‐MS. Different derivatization reagents and solvents were studied for maximizing the di‐derivatization of 17α‐ethynyl estradiol under ultrasound energy. Only N,Obis(trimethylsilyl)trifluoroacetamide with 1% of trimethylchlorosilane in pyridine gave satisfactory results and this mixture was further used in the optimization of the ultrasound assisted derivatization. The experiment designs included sonication time (1–10 min), sonication power (20–80%), sonication cycles (1–9), derivatization reagent volume (25–125 μL) and solvent volume (25–125 μL). Once the optimum conditions were fixed, the effect of organic matter and the frequency of the water bath change were studied. Finally, the validation of the analytical method was carried out using spiked natural and synthetic waters. Recoveries (natural (138–70%) and synthetic (112–89%)), the LODs (0.35–1.66 ng/L), and LOQs (1.16–5.52 ng/L) and the precision (0.2–5.3%) of the method were studied. This is the first work in the literature where a cup horn booster is used with the aim of minimizing derivatization time during the determination of estrogenic compounds.  相似文献

The volatile composition of different apple varieties of Malus domestica Borkh. species from different geographic regions at Madeira Islands, namely Ponta do Pargo (PP), Porto Santo (PS), and Santo da Serra (SS) was established by headspace solid‐phase microextraction (HS‐SPME) procedure followed by GC‐MS (GC‐qMS) analysis. Significant parameters affecting sorption process such as fiber coating, extraction temperature, extraction time, sample amount, dilution factor, ionic strength, and desorption time, were optimized and discussed. The SPME fiber coated with 50/30 μm divinylbenzene/carboxen/PDMS (DVB/CAR/PDMS) afforded highest extraction efficiency of volatile compounds, providing the best sensitivity for the target volatiles, particularly when the samples were extracted at 50°C for 30 min with constant magnetic stirring. A qualitative and semi‐quantitative analysis between the investigated apple species has been established. It was possible to identify about 100 of volatile compounds among pulp (46, 45, and 39), peel (64, 60, and 64), and entire fruit (65, 43, and 50) in PP, PS, and SS apples, respectively. Ethyl esters, terpenes, and higher alcohols were found to be the most representative volatiles. α‐Farnesene, hexan‐1‐ol and hexyl 2‐methylbutyrate were the compounds found in the volatile profile of studied apples with the largest GC area, representing, on average, 24.71, 14.06, and 10.80% of the total volatile fraction from PP, PS, and SS apples. In PP entire apple, the most abundant compounds identified were α‐farnesene (30.49%), the unknown compound m/z (69, 101, 157) (21.82%) and hexyl acetate (6.57%). Regarding PS entire apple the major compounds were α‐farnesene (16.87%), estragole (15.43%), hexan‐1‐ol (10.94), and E‐2‐hexenal (10.67). α‐Farnesene (30.3%), hexan‐1‐ol (18.90%), 2‐methylbutanoic acid (4.7%), and pentan‐1‐ol (4.6%) were also found as SS entire apple volatiles present in a higher relative content. Principal component analysis (PCA) of the results clustered the apples into three groups according to geographic origin. Linear discriminant analysis (LDA) was performed in order to detect the volatile compounds able to differentiate the three kinds of apples investigated. The most important contributions to the differentiation of the PP, PS, and SS apples were ethyl hexanoate, hexyl 2‐methylbutyrate, E,E‐2,4‐heptadienal, p‐ethyl styrene, and E‐2‐hexenal.  相似文献

A novel method using vortex‐assisted surfactant‐enhanced‐emulsification liquid–liquid microextraction has been developed for the extraction of phthalate esters (PAEs) in Chinese liquor samples prior to analysis by GC–MS. In the proposed method, a high‐density extraction solvent (carbon tetrachloride) was dispersed into samples with the aid of a surfactant (Triton X‐100) and vortex agitation, resulting in a short extraction equilibrium (30 s). After centrifugation, a single microdrop of solvent was easily collected for GC–MS analysis. Key factors that affected the extraction efficiency were optimized. Under the optimum conditions, linearity was found in the range from 0.05 to 50 μg/L. Coefficients of determination varied from 0.9938 to 0.9971. LODs, based on an S/N of 3, ranged from 4.9 to 13 ng/L. Enrichment factors varied from 140 to 184. Reproducibility and recoveries were assessed by testing a series of three liquor samples that were spiked with different concentration levels. Finally, the proposed method was successfully applied to the determination of PAEs in 16 Chinese liquor samples. In this work, high‐density‐solvent vortex‐assisted surfactant‐enhanced‐emulsification liquid–liquid microextraction was applied for the first time for the extraction of PAEs in Chinese liquor samples and was proved to be simple, rapid, and sensitive.  相似文献

A sensitive GC–MS method has been established for the determination of acrylamide in surface and drinking water based on derivatization with xanthydrol. Deuterated acrylamide (acrylamide‐d3) was chosen as the internal standard for analyzing the water sample. The derivatization of acrylamide was performed directly in water, and the best reaction conditions (xanthydrol of 1.6 mM, HCl concentration of 0.05 M, reaction for 30 min at ambient temperature) were established by variation of parameters. Under the established conditions, the detection and quantification limits were 3.0 and 9.7 ng/L, respectively, and the interday RSD was less than 8% at concentrations of 20 and 100 ng/L.  相似文献

This paper described a novel approach for the determination of bisphenol A by dispersive liquid‐phase microextraction with in situ acetylation prior to GC‐MS. In this derivatization/extraction method, 500 μL acetone (disperser solvent) containing 30.0 μL chlorobenzene (extraction solvent) and 30.0 μL acetic anhydride (derivatization reagent) was rapidly injected into 5.00 mL aqueous sample containing bisphenol A and K2CO3 (0.5% w/v). Within a few seconds the analyte was derivatized and extracted at the same time. After centrifugation, 1.0 μL of sedimented phase containing enriched analyte was determined by GC‐MS. Some important parameters, such as type and volume of extraction and disperser solvent, volume of acetic anhydride, derivatization and extraction time, amount of K2CO3, and salt addition were studied and optimized. Under the optimum conditions, the LOD and the LOQ were 0.01, 0.1 μg/L, respectively. The experimental results indicated that there was linearity over the range 0.1–50 μg/L with coefficient of correlation 0.9997, and good reproducibility with RSD 3.8% (n = 5). The proposed method has been applied for the analysis of drinking water samples, and satisfactory results were achieved.  相似文献

The volatile components of Cnidium monnieri were obtained by supercritical fluid extraction (SFE) and analyzed by GC‐MS (identification and determination of metabolites). The compounds were identified according to their retention times and mass spectra. The effects of different parameters, such as extraction pressure, temperature, dynamic extraction time, flow rate of CO2, on the SFE of C. monnieri extracts were investigated. A total of 14 compounds of SFE extracts were identified. Osthole (69.52%), bornyl acetate (10.03%), α‐pinene (4.71%), and imperatorin (2.42%) were the major compounds identified in C. monnieri SFE extracts. The quantitation of osthole and imperatorin were then accomplished. The linear calibration ranges were all 5–1000 μg/mL for osthole and imperatorin by GC‐MS analysis. The recovery of osthole and imperatorin were in the range 96.5–101.8%. The LODs for osthole and imperatorin were 1.0 and 0.6 μg/mL, respectively.  相似文献

Three derivatization agents used in GC analysis of amino acids were compared: N,O‐bis(trimethylsilyl)trifluoroacetamide, (BSTFA), N‐methyl‐N‐(tert‐butyldimethylsilyl)trifluoroacetamide (MTBSTFA), and isobutyl chloroformate (iBuCF). It was shown that the analytical characteristics achieved in the case of silylation with MTBSTFA are comparable to those obtained for esterification/acylation. However, since the former approach requires laborious sample preparation to isolate the compounds in question prior to derivatization, determination of amino acids as N(O,S)‐alkoxycarbonyl alkyl esters seems to be preferable in many cases. Application of the esterification/acylation procedure to analysis of lyophilized E. coli microbial culture was demonstrated.  相似文献

Volatile organic compounds (VOCs) are toxic compounds in the air, water and land. In the proposed method, ultrasound-assisted emulsification microextraction (USAEME) combined with gas chromatography-mass spectrometry (GC-MS) has been developed for the extraction and determination of eight VOCs in water samples. The influence of each experimental parameter of this method (the type of extraction solvent, volume of extraction solvent, salt addition, sonication time and extraction temperature) was optimized. The procedure for USAEME was as follows: 15 μL of 1-bromooctane was used as the extraction solvent; 10 mL sample solution in a centrifuge tube with a cover was then placed in an ultrasonic water bath for 3 min. After centrifugation, 2 μL of the settled 1-bromooctane extract was injected into the GC-MS for further analysis. The optimized results indicated that the linear range is 0.1-100.0 μg/L and the limits of detection (LODs) are 0.033-0.092 μg/L for the eight analytes. The relative standard deviations (RSD), enrichment factors (EFs) and relative recoveries (RR) of the method when used on lake water samples were 2.8-9.5, 96-284 and 83-110%. The performance of the proposed method was gauged by analyzing samples of tap water, lake water and river water samples.  相似文献

A headspace solid‐phase microextraction method coupled to GC–MS was successfully developed for the trace determination of formaldehyde in veterinary bacterial and human vaccines, and diphtheria–tetanus antigen. The formaldehyde was derivatized by means of the Hantzsch reaction prior to extraction and subsequent determination. Three different types of solid‐phase microextraction fibers, polar, and nonpolar poly(dimethylsiloxane) and polyethylene glycol were prepared by using a sol–gel technique. The effects of different parameters such as type of fiber coating, extraction time and temperature, desorption conditions, agitation rate, and salt effect were investigated. Under the optimized conditions, the detection limit of the method was 979 ng/L using the selected ion‐monitoring mode. The interday and intraday precisions of the developed method under the optimized conditions were below 13%, and the method shows linearity in the range of 1.75–800 μg/L with a correlation coefficient of 0.9963. The optimized method was applied to the determination of formaldehyde from some biological products. The results were satisfactory compared to the standard method.  相似文献

High performance liquid chromatography (HPLC) and electrospray ionization mass spectrometry (ESI-MS) have been utilized to analyze the synthesized 2-(2-arylaminomethylphenoxy)pyrimidine derivatives, which are a new kind of environmentally benign herbicides and have passed the temporary pesticide registration. The identification of main product and impurities has been achieved according to the UV and mass spectra. Moreover, one impurity, introduced by the raw material in the last step of the synthetic route, was identified by GC-MS analysis. It can be concluded that the combination of chromatography and mass spectrometry, including LC-MS and GC-MS, provided a vital tool of the pesticide science.  相似文献

This study aimed to develop a potential analytical method to discriminate the Chinese winter wheat according to geographical origin and cultivars. A total of 90 wheat samples of 10 different wheat cultivars among three regions were examined by headspace solid phase microextraction coupled with gas chromatography‐mass spectrometry (GC‐MS). The peak areas of 32 main volatile compounds were selected and subjected to statistical analysis, which revealed significant differences among different regions and cultivars. Multivariate analysis of variance showed a significant influence of regions, wheat genotypes, and their interaction on the volatile composition of wheat. Principal component analysis of the aromatic profile showed better visualization for wheat geographical origins. Finally, a classification model based on the linear discriminant analysis was successfully constructed for the discrimination of regions and cultivars with the correct classification percentages of 90 and 100%, respectively.  相似文献

Like many new designer drugs of abuse, synthetic cannabinoids (SC) have structural or positional isomers which may or may not all be regulated under law. Differences in acute toxicity may exist between isomers which impose further burden in the fields of forensic toxicology, medicine and legislation. Isomer differentiation therefore becomes crucial from these standpoints as new designer drugs continuously emerge with just minor positional modifications to their preexisting analogs. The aim of this study was to differentiate the positional isomers of JWH‐081. Purchased standard compounds of JWH‐081 and its positional isomers were analyzed by gas chromatography‐electron ionization‐mass spectrometry (GC‐EI‐MS) first in scan mode to investigate those isomers who could be differentiated by EI scan spectra. Isomers with identical or near‐identical EI spectra were further subjected to GC‐tandem mass spectrometry (MS/MS) analysis with appropriate precursor ions. EI scan was able to distinguish 3 of the 7 isomers: 2‐methoxy, 7‐methoxy and 8‐methoxy. The remaining isomers exhibited near‐identical spectra; hence, MS/MS was performed by selecting m/z 185 and 157 as precursor ions. 3‐Methoxy and 5‐methoxy isomers produced characteristic product ions that enabled the differentiation between them. Product ion spectrum of 6‐methoxy isomer resembled that of JWH‐081; however, the relative ion intensities were clearly different from one another. The combination of EI scan and MS/MS allowed for the regioisomeric differentiation of the targeted compounds in this study. Copyright © 2015 John Wiley & Sons, Ltd.  相似文献

A simple, fast, sensitive and cost-effective method based on headspace solid-phase microextraction (HS-SPME) with on-fiber derivatization coupled with gas chromatography-mass spectrometry was developed for the determination of six typical aldehydes, 2E-hexenal, heptanal, 2E-heptenal, 2E,4E-heptadienal, 2E-decenal and 2E,4E-decadienal in laboratory algae cultures. As derivatization reagent, O-2,3,4,5,6-(pentafluorobenzyl) hydroxylamine hydrochloride, was loaded onto the poly(dimethylsiloxane)/divinylbenzene fiber for aldehydes on-fiber derivatization prior to HS-SPME. Various influence factors of extraction efficiency were systematically investigated. Under optimized extraction conditions, excellent method performances for all the six aldehydes were attained, such as satisfactory extraction recoveries ranging from 67.1 to 117%, with the precision (relative standard deviation) within 5.3-11.1%, and low detection limits in the range of 0.026-0.044 μg/L. The validated method was successfully applied for the analysis of the aldehydes in two diatoms (Skeletonema costatum and Chaetoceros muelleri), two pyrrophytas (Prorocentrum micans and Scrippsiella trochoidea) and Calanus sinicus eggs (feeding on the two diatoms above).  相似文献

A sensitive and accurate method for the determination of two model phenoxy herbicides, 4‐chloro‐2‐methylphenoxy acetic acid and 4‐chloro‐2‐methylphenoxy propanoic acid, in water is explained. This method utilizes a simple phase transfer catalyst‐assisted microextraction with simultaneous derivatization. Factors affecting the performance of this method including pH of the aqueous matrix, temperature, extraction duration, type and amount of derivatization reagents, and type and amount of the phase transfer catalyst are examined. Derivatization and the use of phase transfer catalyst have proven to be especially vital for the resolution of the analytes and their sensitive determination, with an enrichment factor of 288‐fold for catalyzed over noncatalyzed procedure. Good linearity ranging from 0.1 to 80 μg L?1 with correlation of determination (r2) between 0.9890 and 0.9945 were obtained. Previous reported detection limits are compared with our new current method. The low LOD for the two analytes (0.80 ng L?1 for 4‐chloro‐2‐methylphenoxy propanoic acid and 3.04 ng L?1 for 4‐chloro‐2‐methylphenoxy acetic acid) allow for the determination of low concentrations of these analytes in real samples. The absence of matrix effect was confirmed through relative recovery calculations. Application of the method to seawater and tap water samples was tested, but only 4‐chloro‐2‐methylphenoxy propanoic acid at concentrations between 0.27 ± 0.01 and 0.84 ± 0.06 μg L?1 was detected in seawater samples.  相似文献

Ultrasound-assisted dispersive liquid-liquid microextraction (UDLLME) and simultaneous derivatization followed by GC-MS was developed for the analysis of four aldehydes including acetaldehyde (ACE), propionaldehyde (PRO), butyraldehyde (BUT) and valeraldehyde (VAL) in water samples. In the proposed method, the aldehydes were derivatized with O-2,3,4,5,6-(pentafluorobenzyl)hydroxylamine (PFBHA) and extracted by UDLLME in aqueous solution simultaneously; finally, the derivatives were analyzed by GC-MS. The experimental parameters were investigated and the method validations were studied. The optimal conditions were: aqueous sample of 5 mL, PFBHA of 50 μL, 1.0 mL ethanol (disperser solvent) containing 20 μL chlorobenzene (extraction solvent), ultrasound time of 2 min and centrifuging time of 3 min at 6000 rpm. The proposed method provided satisfactory precision (RSD 1.8-10.2%), wide linear range (0.8-160 μg/L), good linearity (R(2) 0.9983-0.9993), good relative recovery (85-105%) and low limit of detection (0.16-0.23 μg/L). The proposed method was successfully applied for the analysis of aldehydes in water samples. The experimental results showed that the proposed method was a very simple, rapid, low-cost, sensitive and efficient analytical method for the determination of trace amount of aldehydes in water samples.  相似文献
