By the condensation of 3,4-diaminothiocoumarins containing primary or secondary amino groups in position 4, with formic and acetic acids (or acetic anhydrides), a number of 1H-[1]benzothiopyrano[3,4-d]imidazol-4-ones and their 2-methyl derivatives have been synthesized; the latter have also been obtained from the corresponding 3-monoacetylamino-4-aminothiocoumarins. By the acetylation of 3-4-diaminothiocoumarins with tertiary amino groups in positions 4 their 3-diacetyl derivative has been obtained, and these, under the action of bases, have been converted into 3-N-acetyl derivatives. The diazotization of 3-aminocoumarin followed by its reduction gave the cournarin-3-yl-hydrazine which, without isolation, reacted with various carbonyl compounds in a Fisher's indohzation reaction to give derivatives (Ib-Ih) of the yet unreported system 3H,4H[1]benzopyrano[3,4-b]pyrrol-4-one.  相似文献

Acylation of 4-alkoxycarbonyl-3-amino-6-hydroxy-2H-1-benzopyran-2-one derivatives 3 and 4 gave under mild conditions the O-substituted derivatives 5–10, N,O -disubstituted derivative 11 and N,N-disubstituted derivative 12 . The compound 4 was transformed with benzoyl chloride under more drastic conditions into 13 , a derivative of a new heterocyclic system 2-benzopyrano[3,4-d][1,3]oxazine. The derivatives of 1-benzopyrano-[3,4-d]pyrimidine 19 and 20 were prepared either from 3 and 4 through the corresponding N-heteroarylformamidines 14 and 15 and N-heteroarylformamide oximes 17 and 18 or by cyclization of thiourea derivative 20 .  相似文献

By the reaction of 6-aryl(alkyl)amino-5-cyano-2,3-dihydro-1,3-thiazin-4(10)-ones with α-halogenoketones in the presence of triethylamine, 2,3-dihydrothieno[2,3-d]pyrimidin-4(1H)-ones have been synthesized and their acid-catalyzed recyclization to 2,3-dihydrothieno[3,4-d]pyrimidin-4(1H)-ones has been found and studied. © 2006 Wiley Periodicals, Inc. A series of pyrazolo[3,4-d]pyrimidine-4-one derivatives was conveniently synthesized via tandem aza-Wittig and annulation reactions of the corresponding iminophosphoranes, arylisocyanate, and substituted thiophenols. The structures of the target compounds were confirmed by IR, 1H NMR, 13C NMR, LC-MS, and elemental analysis. The preliminary bioassay demonstrated that some title compounds such as 6-(3-chlorophenylthio)-1-phenyl-3-methylthio-5-(4-chlorophenyl)-1H-pyrazolo[3,4-d]-pyrimidin-4(5H)-one and 6-(4-fluorophenylthio)-1-phenyl-3-methylthio-5-(3-chlorophenyl)-1H-pyrazolo[3,4-d]pyrimidin-4(5H)-one showed good inhibition activities against the root of Brassica napus (rape) and Echinochloa crusgalli (barnyard grass) at a dosage of 100 mg/L.  相似文献

The reaction of 2H-thiopyran-3,5(4H,6H)-dione with N,N-dimethylformamide dimethyl acetal gave in good yield 4-dimethylaminomethylene-2H-thiopyran-3,5(4H,6H)dione (II), which afforded 1-substituted 5,7-dihydrothiopyrano[3,4-c]pyrazol-4(1H)-ones with aliphatic and aromatic hydrazines, 5H-thiopyrano[4,3-d]isoxazol-4(7H)-one (IV) with hydroxylamine hydrochloride and 2-substituted 6H-thiopyrano[3,4-d]pyrimidin-5(8H)-ones with amidines and guanidines, generally in satisfactory yields. 4-(t-Butylhydrazonoformyl)-2H-thiopyran-3,5(4H,6H)-dione was isolated as an intermediate in the reaction of II with t-butylhydrazine, whereas formamidine gave with II 4-iminoformyl-2H-thiopyran-3,5(4H,6H)-dione as the sole product. The isoxazole IV isomerized easily with sodium methoxide to 3,4,5,6-tetrahydro-5,5-dihydroxy-3-oxo-2H-thiopyrano-4-carbonitrile.  相似文献

The synthesis of 1,3-disubstituted and 1,3,5-trisubstituted 1H-pyrazolo[3,4-d]pyridazin-4(5H)-ones is conveniently performed by photochemical cyclization. Functionalization of the former compounds leading to the formation of 1,4-disubstituted and 1,3,4-trisubstituted 1H-pyrazolo[3,4-d]pyridazines is smoothly effected through pertinent nucleophilic substitutions.  相似文献

Regioselective 1,3-dipolar cycloaddition of nitrile oxides 5a-c to ethyl o-hydroxycinnamate (3) gave the corresponding ethyl trans-3-aryl-4,5-dihydro-5-(2-hydroxyphenyl)-4-isoxazolecarboxylates 6a-c . Their structure was confirmed by reductive cleavage to 1 and compounds 9a-c . Compounds 6a-c afforded upon heating in the presence of pyridine the 3-aryl-4H-[1]benzopyrano[3,4-d]isoxazol-4-ones 11a-c .
