Extracellular poly[(R)-3-hydroxybutyrate] (PHB) depolymerase (PhaZRpiT1) from Ralstonia pickettii T1 adsorbs to the PHB surface via its substrate-binding domain (SBD) and cleaves the PHB chain using its catalytic domain. Our previous study (Biomacromolecules 2010; 11: 113-119) has suggested that the hydrophobic interaction between the amino acid residues at positions 441, 443, and 445 in the SBD and the PHB surface plays a crucial role in facilitating the association phase of the enzyme adsorption process. In the present study, in order to improve PhaZRpiT1 for effective PHB degradation, we targeted Tyr at position 443 for substitution with a more highly hydrophobic amino acid residue because its hydrophobicity shows medium to high degree compared to those of general naturally occurring amino acid residues. We designed a mutant enzyme with an amino acid substitution at this position, taking the following factors into consideration: (1) to achieve higher hydrophobicity than the original residue, (2) to retain the β-sheet structure, and (3) to change as little as possible the volume of the amino acid residue after the substitution. As a result, the substitution of Tyr443 with Phe (Y443F) was considered to be appropriate. The purified Y443F enzyme showed identical CD spectrum and hydrolysis activity for a water-soluble substrate with the wild type, indicating that the mutation had no influence on the structure and the ester bond cleavage activity. In contrast, the Y443F enzyme had higher PHB degradation activity than the wild type. Kinetic analysis of PHB degradation suggests that this amino acid substitution promoted not only the adsorption of the mutant enzyme to PHB, but also the disruption of the PHB surface to enhance the hydrolysis of the PHB polymer chain.  相似文献

The 3-dimensional structure of the Paucimonas lemoignei poly(3-hydroxybutyrate) (PHB) depolymerase PhaZ7 has significant similarity to Bacillus subtilis lipase LipA but differs from the latter by the presence of an additional domain. Analysis of this lid-like domain revealed the presence of many hydrophobic amino acid residues including Tyr105. In this study we constructed His-tag fusions of PhaZ7 for simplified purification and investigated the effect of amino acid exchange of eight tyrosine codons of the lid-like domain. Exchanges of Tyr103, Tyr172, Tyr173, Tyr203 or Tyr204 to alanine or serine had no phenotype but muteins with substitution of Tyr189, Tyr190 and Tyr105 to alanine showed a lag phase of the in vitro PHB depolymerase reaction. Replacement of Tyr105 by glutamate further increased the lag phase. Binding assays of the purified PHB depolymerase proteins with the natural substrate, native PHB granules, revealed a significantly reduced binding ability of the Tyr105Glu mutant compared to the wild type protein and confirmed that Tyr105 is involved in interaction with the polymeric substrate.  相似文献

Type I polyhydroxyalkanoate (PHA) synthases, as represented by Ralstonia eutropha enzyme (PhaC(Re)), have narrow substrate specificity toward (R)-3-hydroxyacyl-coenzyme A with acyl chain length of C3-C5 to yield PHA polyesters. In this study, saturation point mutagenesis of a highly conserved alanine at position 510 (A510) in PhaC(Re) was carried out to investigate the effects on the polymerization activity and the substrate specificity for in vivo PHA biosynthesis in bacterial cells. A series of saturation mutants were first applied for poly[(R)-3-hydroxybutyrate] homopolymer synthesis in Escherichia coli and R. eutropha PHB(-)4 (PHA negative mutant) cells to assess the polymerization activity. All mutants showed quantitatively similar polymerization activities when R. eutropha PHB(-)4 was used for assay, whereas several mutants such as A510P showed low activities in E. coli. Further analysis has revealed that majority of mutants synthesize polyesters with higher molecular weights than the wild-type. In particular, substitution by acidic amino acids, A510D(E), led to remarkable increases in molecular weights. Subsequently, PHA copolymer synthesis from dodecanoate (C12 fatty acid) was examined. The copolymer compositions were varied depending on the mutants used. Significant increased fractions of long monomer units (C6 and C8) in PHA copolymers were observed for three mutants [A510M(Q,C)]. From these results, the mutations at this potion are beneficial to change the molecular weight of polyesters and the substrate specificity of PhaC(Re). Electrospinning has been extensively used to construct tissue‐engineered scaffolds because of its ability to provide the fibrous scaffold with structurally analogous to the naturally occurring protein in the extracellular matrix of native tissues. In addition, the modification of scaffolds with bioactive molecules is beneficial as this can create an environment that consists of biochemical cues to further promote cell adhesion, proliferation, and differentiation. In the present contribution, we prepared and investigated the potential used of aligned electrospun poly(3‐hydroxybutyrate) (PHB) scaffold immobilized with bioactive molecule to serve as nervous scaffold. Laminin was successfully immobilized on the surface using covalent binding between functional groups of modified scaffolds and protein. The ability to use for neural regeneration was evaluated in vitro towards murine neuroblastoma Neuro2a cell line and mouse brain‐derived neural stem cells. The surface modification with laminin immobilized on the PHB fibrous scaffolds supported the attachment and promoted the proliferation of Neuro2a very wells. Despite the good attachment and proliferation of Neuro2a and mouse brain‐derived neural stem cells were not able to proliferate on the neat PHB, hydrolyzed PHB and laminin immobilized on hydrolyzed PHB fibrous scaffold.  相似文献

The adsorption behavior of PHB depolymerase from R. pickettii T1 on a silicon wafer and on P(3HB) single crystals has been studied by real-time and AFM in air and a buffer solution. First, the morphology of PHB depolymerase adsorbed on a silicon wafer was characterized to show that one molecule of PHB depolymerase has dimensions of 2.2 +/- 0.7 nm height and 16 +/- 5 nm width. The observation of PHB depolymerase adsorbed on a P(3HB) single crystal indicated that the dimensions of enzyme on the crystalline surface in air were 1.2 +/- 0.5 nm high and 28 +/- 7 nm wide, while enzyme molecules with dimensions of 2.1 +/- 0.6 nm height and 16 +/- 7 nm width were detected in a buffer solution. Comparison of the dimensions of PHB depolymerase in air with those in a buffer solution showed that the enzyme was squashed in air, but not in a buffer solution. In addition, the influence of enzymatic adsorption on the molecular state of the P(3HB) crystalline surface was investigated. The AFM images of P(3HB) single crystals after enzymatic adsorption and washing with ethanol indicated that the adhesion of PHB depolymerase changed the molecular state and generated holes on the crystalline surface.  相似文献

Summary: Chemical modification of polymer surface may potentially be used to create smart materials that can guide cellular adhesion, proliferation and maintenance of specific expression of molecules. The microbial polyester poly (3-hydroxybutyrate) (PHB) has been attracted attention as promising material for applications in tissue engineering. In this work, a wet-chemical method, base ethylenediamine aminolysis, was performed to improve the adhesion of chondrocytes isolated from human articular cartilage to PHB films. The effects of chemical treatment on PHB films was evaluated by following changes in morphology and surface chemical composition using atomic force microscopy (AFM) and X-ray photoelectron spectroscopy (XPS), respectively. While the effect on cells morphology was studied by scanning electron microscopy (SEM). The treatment with ethylenediamine did not change significantly the morphology of the structures of PHB films surface. However, the roughness of the aminolyzed films was slightly higher. The introduction of nitrogen-containing groups was confirmed by XPS. In vitro experiments indicated that the surface modification did not have toxic effects in cells, since they could adhere and proliferate on modified PHB films. It was observed that long-time treatment improved ability of PHB films to support cell growth, which could be accounted to physicochemical and topological effects.  相似文献

Polyhydroxyalkanoates (PHAs) are a family of aliphatic polyesters produced by a variety of microorganisms as a reserve of carbon and energy. Enzymes involved in the synthesis of PHAs can be utilized to produce polymers in vitro, both in bulk and on solid surfaces. Here, site-specific attachment of the key catalytic enzyme, PHA synthase, on lithographically patterned surfaces and subsequent addition of (R)-3-hydroxybutyryl-CoA substrate allowed us to fabricate spatially ordered polyhydroxybutyrate (PHB) polymeric structures via an in situ enzymatic surface-initiated polymerization (ESIP). By varying the reaction conditions, we enhanced the growth of PHB on solid surfaces and analyzed the resulting structures by fluorescence microscopy, atomic force microscopy (AFM), attenuated total reflectance Fourier transform infrared (ATR-FTIR) spectroscopy, and gel permeation chromatography (GPC). We found that stabilization of smaller PHB granule structures by an addition of bovine serum albumin (BSA) was the most important factor for a successful synthesis of a PHB layer up to 1mum in thickness, consisting mainly of larger cluster assemblies of PHB granules that cover the entire patterned area. Immunofluorescence detection and surface contact angle analysis revealed that BSA was physically bound to the PHB polymer all through the cluster, and reduced the overall hydrophobicity of the polymer surface. Based on information obtained from AFM, kinetic measurements and various polymer characterization methods, a plausible model for roles of BSA in the enhancement of PHB formation on surfaces is discussed. Furthermore, by using biotinylated BSA conjugates, we were able to incorporate biotin groups into the PHB polymer matrix, thus generating a bioactive surface that can be used for displaying other functional biomolecules through streptavidin-biotin interaction on the PHB structures. Because of its versatility, our fabrication strategy is expected to be a useful surface modification tool for numerous biomedical and biotechnological applications.  相似文献

Fully-biodegradable bacterial poly(3-hydroxybutyrate) (PHB)/chemosynthetic poly(vinyl alcohol) (PVA) blend films with compositional gradient from one surface to the other surface of the films were prepared by a dissolution-diffusion technique. Three kinds of PVA samples, high- and low-molecular weight atactic PVA and highly syndiotactic PVA (s-PVA), were used in order to investigate the effects of molecular weight and tactic structure on the generation of compositional gradient. The solution of PHB in 1,1,1,3,3,3-hexafluoro-2-propanol (HFIP), which is also a good solvent for PVA, was cast on the PVA film and then the solvent HFIP was evaporated. By selecting the optimum volume of solvent and the evaporation rate, the PHB/PVA blend film with compositional gradient was obtained. The formation of compositional gradient was confirmed by FT-IR microscopy and ATR-FT-IR analysis. The 50%/50% PHB/s-PVA blend film with a nearly ideal compositional gradient, that is, the composition of PHB (or PVA) in the film changing gradually from 100% at one surface to 0% at the other surface of the film was obtained by casting PHB/HFIP solution on to the s-PVA film. Elasticity of various poly(hydroxybutyrate) (PHB) molecules of regular and irregular conformational structure was examined by the molecular mechanics (MM) calculations. Force - distance functions and the Young's moduli E were computed by stretching of PHB molecules. Unwinding of the 2(1) helical conformation H is characterized at small deformations by the Young's modulus E = 1.8 GPa. The H form is transformed on stretching into the highly extended twisted form E, similar to the beta-structure observed earlier by X-ray fiber diffraction. The computations revealed that in contrast to paraffins, the planar all-trans structure of undeformed PHB is bent. Hence, a PHB molecule attains the maximum contour length in highly straightened, but slightly twisted conformations. A dependence of the single-chain moduli of regular and disordered conformations on the chain extension ratio x was found. The computed data were used to analyze elastic response of tie (bridging) molecules in the interlamellar (IL) region of a semi-crystalline PHB. A modification of the chain length distribution function of tie molecules tau(N) due to secondary crystallization of PHB was conjectured. The resulting narrow distribution tau(N) comprises the taut tie molecules of higher chain moduli prone to overstressing. The molecular model outlined is in line with the macroscopically observed increase in the modulus and brittleness of PHB with storage time.  相似文献

Adsorption of PHB depolymerase from Ralstonia pickettii T1 to biodegradable polyesters such as poly[(R)-3-hydroxybutyrate] (PHB) and poly(l-lactic acid) (PLLA) was investigated by atomic force microscopy (AFM). The substrate-binding domain (SBD) with histidines within the N-terminus was prepared and immobilized on the AFM tip surface via a self-assembled monolayer with a nitrilotriacetic acid group. Using the functionalized AFM tips, the force-distance measurements for polyesters were carried out at room temperature in a buffer solution. In the case of AFM tips with immobilized SBD and their interaction with polyesters, multiple pull-off events were frequently recognized in the retraction curves. The single rupture force was estimated at approximately 100 pN for both PLLA and PHB. The multiple pull-off events were recognized even in the presence of a surfactant, which will prevent nonspecific interactions, but reduced when using polyethylene instead of polyesters as a substrate. The present results provide that the PHB depolymerase adsorbs specifically to the surfaces of polyesters and that the single unbinding event evaluated here is mainly associated with the interaction between one molecule of SBD and the polymer surface.  相似文献

Two series of segmented poly(ester‐urethane)s were synthesized from bacterial poly[(R)‐3‐hydroxybutyrate]‐diol (PHB‐diol), as hard segments, and either poly(ε‐caprolactone)‐diol (PCL‐diol) or poly(butylene adipate)‐diol (PBA‐diol), as soft segments, using 1,6‐hexamethylene diisocyanate as a chain extender. The hard‐segment content varied from 0 to 50 wt.‐%. These materials were characterized using 1H NMR spectroscopy and GPC. The polymers obtained were investigated calorimetrically and dielectrically. DSC showed that the Tg of either the PCL or PBA soft segments are shifted to higher temperatures with increasing PHB hard‐segment content, revealing that either the PCL or PBA are mixed with small amounts of PHB in the amorphous domains. The results also showed that the crystallization of soft or hard segments was physically constrained by the microstructure of the other crystalline phase, which results in a decrease in the degree of crystallinity of either the soft or hard segments upon increase of the other component. The dielectric spectra of poly(ester‐urethane)s, based on PCL and PHB, showed two primary relaxation processes, designated as αS and αH, which correspond to glass–rubber transitions of PCL soft and PHB hard segments, respectively. Whereas in the case of other poly(ester‐urethane)s, derived from PBA and PHB, only one relaxation process was observed, which broadens and shifts to higher temperature with increasing PHB hard‐segment content. It was concluded from these results that our investigated materials exhibit micro‐phase separation of the hard and soft segments in the amorphous domains.  相似文献

E. coli and Salmonella are two of the most common bacterial pathogens involved in foodborne and waterborne related deaths. Hence, it is critical to develop rapid and sensitive detection strategies for near-outbreak applications. Reported is a simple and specific assay to detect as low as 1 CFU mL−1 of E. coli in water within 6 hours by targeting the bacteria's surface protease activity. The assay relies on polythiophene acetic acid (PTAA) as an optical reporter and a short unlabeled peptide (LL37FRRV) previously optimized as a substrate for OmpT, an outer-membrane protease on E. coli. LL37FRRV interacts with PTAA to enhance its fluorescence while also inducing the formation of a helical PTAA-LL37FRRV construct, as confirmed by circular dichroism. However, in the presence of E. coli LL37FRRV is cleaved and can no longer affect the conformations and optical properties of PTAA. This ability to distinguish between an intact and cleaved peptide was investigated in detail using LL37FRRV sequence variants.  相似文献

Our daily life needs depend on plastics, as they are cheap and durable, so they become the most commonly used synthetic chemical products. But from an environmentalist's point of view, a major concern related to these plastics is their non‐biodegradable nature. Driven by growing demand to search for sustainable solutions to dispose off generating huge volume of synthetic plastic wastes, shifted the mind of researcher towards the use of biodegradable plastics which can be completely disposed‐off by microbial enzymatic degradation. These biodegradable plastics or “bioplastics” are also synthesized by microbes under certain stressed environmental conditions out of which poly(R‐3‐hydroxybutyrate) (PHB) is the most ubiquitous and best known representatives of polyhydroxyalkanoate family. The PHB is most intensively used for the innovative biomedical applications owing to suitable combination of biocompatibility, transport characteristics, and mechanical properties. These challenging aspects of PHB can be used for designing of novel medical devices, in tissue engineering, and for systematic sustained drug delivery. Lots of research reports on PHB degrading enzymes and their producing microorganisms including biochemical aspects are available but in scattered form. So this review highlighted all the relevant information of PHB and PHB‐degrading enzymes starting with basic classification, synthesis, mechanism, and applications that are environment friendly and are of public interest.  相似文献

This study aims to evaluate the photocatalytic activity and biodegradation of polyhydroxybutyrate (PHB) films containing titanium dioxide (TiO2). Nanosized TiO2 photocatalysts were immobilized onto PHB film to overcome the difficulty of the recovery process. PHB is a suitable base material as it is naturally biodegradable and is produced from renewable resources. The photocatalytic degradation of organic compounds, photocatalytic sterilization activity and biodegradation rate in garden soil of PHB-TiO2 composite films were investigated. After an hour under solar illumination, 96% of methylene blue solution was decolorized. The antibacterial activity against Escherichia coli (E. coli) using PHB-TiO2 composite film exhibited enhanced photocatalytic sterilization activity over time. As for the ability to biodegrade, PHB-TiO2 composite films placed on soil surface with no direct solar illumination showed slower degradation rate compared to those receiving direct solar illumination. Interestingly, the latter composite films showed faster degradation rates compared to pure PHB films indicating that the degradation is mainly due to photocatalytic activity. PHB-TiO2 composite films buried in soil generally showed slower degradation rates compared to pure PHB films and were dependent on the soil microbial activity.  相似文献

The poly[(R)-3-hydroxybutyrate] (PHB) is a highly crystalline, biosourced polymer. The advantages of the PHB are its biodegradability and biocompatibility; however, the brittleness caused by its high crystallinity decreases the application ability of the PHB in comparison with the polyolefins. Excellent results were observed for the reactive extrusion of PHB in the presence of peroxides in many investigations of the modifications of PHB mechanical properties. The disadvantage must be considered to be the thermal degradation of PHB during extended extrusion and its limitation in natural composite preparation. The peroxides are highly reactive with natural fillers, and this causes a decrease of the filler's mechanical properties. Consequently, the reactive extrusion may be a useful tool for the production of additives only. The results we present of this investigation is based on a different material preparation strategy. The two-stage method incorporated additives preparation via reactive extrusion of PHB and the blending of the obtained product with neat PHB. Theself-reinforced composite material obtained in this way revealed significantly higher values of stress and strain compared to neat PHB. The thermal degradation of the PHB matrix was retarded and total crystallinity of the composite was decreased. The materials were characterized using DSC, SEM and SEC techniques. The samples were also investigated employing tensile and impact strength tests.  相似文献

Poly(3-hydroxybutyrate) (PHB) is a highly crystalline, biodegradable and biocompatible thermoplastic. However, its limited utilization as a commodity plastic is associated to both high cost and very poor mechanical properties. Blending PHB with a natural polymer, such as starch, is one way to improve its properties and to get low price raw materials, though they are not miscible since there are no strong interactions between the hydrophilic starch and the hydrophobic PHB. In this study binary blends of PHB were prepared with natural starch, starch-adipate and grafted starch-urethane derivatives. The PHB blends were characterized in terms of their mechanical and thermal properties. For all blends a decrease of the Young modulus was observed as compared to the pure PHB. However, blends containing natural starches and starch adipate resulted in brittle materials. A significant decrease of both glass transition temperature (Tg) and melting point (Tm) was observed for all formulations. The best results, lower modulus and Tg were obtained with grafted starch-urethane blends using poly(propylene glycol).  相似文献

 The specular and off-specular X-ray reflectivities were efficiently employed to study the evolution of surface morphology as a function of temperature in a single layer of poly(3-hydroxybutyrate) (PHB) and a bilayer of PHB/poly(vinyl phenol) (PVPh) on Si substrates. The results indicate that the changes of thickness and surface roughness caused by pre-melting of PHB crystals are not obvious for the single layer, whereas the surface roughness of the PHB layer and the intensity of the off-specular X-ray reflectivity for the bilayer exhibit a remarkable non-monotonic change in the temperature range of 100-150℃; the roughness parameter evaluated by the specular X-ray reflectivity reaches its maximum at 120℃. The interaction at the interface between PVPh and PHB certainly contributes to the non-monotonic changes. Such interaction also affects the crystallization and melting behavior of PHB thin film greatly. The crystallization of PHB thin film is inhibited even on the glassy surface of PVPh sublayer. In the melting process, the PHB crystals on PVPh sublayer feature a three-section melting curve separated by a plateau region of 120-140℃.  相似文献

考察了一种表面接枝季铵盐型高分子材料对水溶液中大肠杆菌的抗菌过程特性,发现此材料可以在短时间内迅速降低溶液中的大肠杆菌的活菌量.用菌体耗氧测定法与扫描电镜观察法研究了其抗菌过程.结果表明,其抗菌过程由吸附和杀灭两个步骤构成,其中吸附过程由纤维与大肠杆菌表面静电作用所控制,是一个快速过程,而杀灭过程则伴随着细胞壁在纤维表面的溶解,是一个慢过程.菌体耗氧测定法与扫描电镜观察法所获得的结果具有一致性,并可广泛应用于研究抗菌纤维的抗菌作用机理及过程特性.  相似文献

Bioplastic production from microbial sources is an emerging area which provides opportunities even to convert the wastes into bioplastics. Poly (3-hydroxybutyric acid), commonly called as PHB, is a bioplastic, which is stored as intracellular cytoplasmic inclusions in microorganisms. The objectives of this study are to calorimetrically monitor the PHB production and evaluate the thermokinetic data in a bioreaction calorimeter (BioRC1e). Thus, a well-known PHB-producing bacteria Ralstonia eutropha was selected for batch process in a bioreaction calorimeter. The metabolic heat generated was found to be correlated with the biomass, substrate consumption, oxygen uptake rate (OUR), carbon dioxide evolution rate (CER) and PHB production. The OUR pattern explained the oxidative metabolism of the strain R. eutropha. The heat yields due to biomass and glucose consumption during PHB production were found to be 12.56 and 13.56 kJ/g, respectively. The oxycalorific value obtained for the PHB production was 443.80 kJ/mol of O2. The concentration of PHB obtained in BioRC1e was 4.33 g/L with a production rate of 0.09 g/L/h. The chemical structure of the extracted PHB by R. eutropha was confirmed using fourier transform infrared spectroscopy (FT-IR) and 1H and 13C nuclear magnetic resonance (NMR) analysis.  相似文献
