A sensitive gas chromatographic-electron-capture detection method for the simultaneous determination of the antianginal drug nitroglycerin (GTN) and its dinitrate metabolites (1,2-GDN and 1,3-GDN) was developed. Human plasma samples (1 ml) spiked with 2,6-dinitrotoluene as the internal standard were extracted once with 10 ml of a methylene chloride-pentane mixture (3:7, v/v). Using this solvent system, less contaminants are extracted into the organic phase from plasma, resulting in cleaner chromatograms and prolonged column life. A break point was observed on the standard curves of GTN and GDNs. The two linear regions for the detectable concentrations of GTN are 0.025-0.3 and 0.3-3 ng/ml and for 1,2-GDN and 1,3-GDN they are 0.1-1 and 1-10 ng/ml. The limits of detection by this method for GTN, 1,2-GDN and 1,3-GDN in plasma are 0.025, 0.1 and 0.1 ng/ml, respectively.  相似文献

Trimipramine is a tricyclic antidepressant drug often assayed by gas chromatographic or gas chromatographic-mass spectrometry techniques. A high-performance liquid chromatographic method with electrochemical detection is described for the assay of trimipramine and its major metabolites, monodesmethyltrimipramine and 2-hydroxytrimipramine, in plasma. The method is sensitive, accurate and robust and thus suitable for routinely assaying samples following single doses of trimipramine to man. The assay was applied to plasma samples obtained following a single 50-mg dose of trimipramine to healthy volunteers.  相似文献

A specific, sensitive, and precise assay for simultaneous quantitation of glyceryl trinitrate (GTN) and its metabolites 1,2-and 1,3-glyceryl dinitrate (GDN) has been developed. The method involves one single extraction step with benzene, followed by evaporation at 55°C. One μl of the extract was injected onto a capillary fused silica column with trifluoropropyl as the stationary phase. The column separated all the nitrates within 17 minutes using temperature programming from 90 to 155°C. The lower limits of detection were 0.05, 0.5, and 0.1 nmol/I for GTN, 1,2-GDN and 1,3-GDN respectively. The calibration curves were linear in the concentration ranges 0.05–10.0 nmol/I (GTN), 0.5–10.0 nmol/I (1,2-GDN) and 0.1–10.0 nmol/I (1,3-GDN). Plasma concentration data from two patients receiving intravenous GTN infusion are presented.  相似文献

A gas chromatographic method using nitrogen-selective detection for the quantitative determination of nanogram amounts of chlorimipramine, chlorpromazine and their nor1 and nor2 derivatives in plasma is described. Derivatization with trifluoroacetic anhydride of nor1 and nor2 metabolites allowed the chromatographic separation of these compounds. A three-step solvent extraction procedure was performed using n-heptane containing 1% isoamyl alcohol and n-hexane and compared with a plasma clean-up procedure using C18 Sep-Pak cartridges. The two procedures were characterized by similar degrees of precision. The use of C18 Sep-Pak cartridges, however, produced a significant time and material saving over the conventional extraction method.  相似文献

A rapid and sensitive method for the simultaneous determination of perhexiline and its cis-4-axial and trans-4-equatorial monohydroxy metabolites (M1 and M3, respectively) in human plasma, urine and bile is described. The assay utilises a single diethyl ether extraction, heptafluorobutyric acid anhydride derivatisation and separation and detection by gas chromatography-electron-capture detection. The limits of detection are 0.1 microgram/ml for perhexiline and 0.025 microgram/ml for the M1 and M3 metabolites. This method has been used in a five-day kinetic study of three healthy adult males who ingested a single 300-mg dose of perhexiline maleate. One of these volunteer subjects exhibited elevated plasma perhexiline and markedly reduced plasma and urinary M1 concentrations together with profoundly prolonged plasma and urinary M1 elimination times when compared with the other two subjects. These differences are thought to be of genetic origin. There were also obvious differences in urinary M3 concentrations which were discussed.  相似文献

Propafenone, a class Ic antiarrhythmic agent, is metabolized to 5‐hydroxypropafeone (5‐OHP) and N‐depropylpropafenone (NDPP). Simultaneous determination of serum propafenone and its metabolites was performed using HPLC equipped with a conventional octadecylsilyl silica column and ultraviolet detector. The wavelength was set at 250 nm. Propafenone and its metabolites in the serum were extracted using diethyl ether. The mobile phase solution, comprising 1‐pentanesulfonic acid sodium salt (0.1 m ), acetonitrile and acetic acid (280:185:2.5, v/v/v), was pumped at a flow rate of 1 mL/min. The recoveries of propafenone, 5‐OHP and NDPP were greater than 85, 82 and 60%, respectively, with the coefficients of variation (CVs) less than 5.4, 1.9 and 2.9%, respectively. The calibration curves were linear for a concentration range of 12.5–1500 ng/mL for propafenone and 2–500 ng/mL for 5‐OHP and NDPP (r > 0.999). CVs in the intraday assays were 1.0–3.8% for propafenone, 0.6–2.0% for 5‐OHP and 0.6–1.7% for NDPP. CVs in interday assays were 1.3–7.7% for propafenone, 1.1–6.5% for 5‐OHP and 5.4–8.0% for NDPP. The present HPLC method can be used to assess the disposition of propafenone and its metabolites for pharmacokinetic studies and therapeutic drug monitoring of propafenone.  相似文献

A simple method is described for the simultaneous determination of the three monoamine metabolites, 4-hydroxy-3-methoxyphenylacetic acid, 4-hydroxy-3-methoxyphenylethyleneglycol and 5-hydroxyindole-3-acetic acid, in cerebrospinal fluid by high-performance liquid chromatography with electrochemical detection. Quantitation is accomplished by the standard addition technique. Chromatographic peak heights are corrected for volume effects by comparison with the signal obtained for an added auxiliary reference substance. Sample preparation is kept to a minimum, involving precipitation of proteins by means of perchloric acid and subsequent neutralization. The reproducibility was estimated to be 10%. For one of the metabolites, 4-hydroxy-3-methoxyphenylethyleneglycol, a correlation between the results obtained by this method and a mass fragmentographic method was made, and a satisfactory correlation (r = 0.904, slope = 0.914, intercept 3.26 ng/ml) found. The sensitivity of the method is in the picogram range. The methodology has been applied to measure biogenic amine metabolites in both rabbit and human cerebrospinal fluid. The levels found are in agreement with previously reported values.  相似文献

A hydrophilic interaction chromatographic (HILIC) system interfaced with atmospheric pressure ionization (API) sources and a tandem mass spectrometer (MS/MS) was developed for the simultaneous determination of nicotinic acid (NiAc) and its metabolites in dog plasma in support of a pharmacokinetic study. A silica column was adapted for separation of NiAc and its two metabolites, nicotinamide (NiNH2) and nicotinuric acid (NiUAc), under HILIC conditions. The influence of experimental factors such as the composition of mobile phase on ionization efficiency and chromatographic performance of all analytes was investigated. The feasibility of the proposed HILIC/MS/MS methods was explored by comparing the plasma levels of NiAc, NiNH2, and NiUAc in dog obtained by using either electrospray ionization or atmospheric pressure chemical ionization interfaces in positive ion mode. The methods were partially validated in terms of inter-day accuracy and precision, extraction recovery, benchtop and freeze/thaw stability. Further, the potential of ionization suppression resulting from endogenous components of the biological matrixes on the HILIC/API-MS/MS methods were investigated using the post-column infusion technique.  相似文献

A reversed-phase high-performance liquid chromatographic assay was developed to simultaneously quantitate nefiracetam (NEF), a novel nootropic agent, and its three known oxidized metabolites (N-[(2,6-dimethylphenylcarbamoyl)methyl]succinamic acid (5-COOH-NEF), 4-hydroxy-NEF and 5-hydroxy-NEF) in human serum and urine. The quantitative procedure was based on solid-phase extraction with Sep-Pak C18 and ultraviolet detection at 210 nm. The calibration curves of NEF and the metabolites were linear over a wide range of concentrations (0.5-21.5 nmol/ml for NEF and 0.4-9.5 nmol/ml for metabolites in serum and 4-86 nmol/ml for NEF and 8-190 nmol/ml for metabolites in urine). Intra- and inter-day assay coefficients of variation for the compounds were less than 10%. The limit of detection was 0.1 nmol/ml for NEF, 5-COOH-NEF and 4-hydroxy-NEF, and 0.2 nmol/ml for 5-hydroxy-NEF in both serum and urine. This method is applicable for the determination of NEF and its metabolites in human serum and urine with satisfactory accuracy and precision.  相似文献

A method was developed for the simultaneous determination of imazalil and its major metabolite, R 14821, in citrus fruit. This method (designated the SPE method) is based on solid-phase extraction (SPE) and capillary gas chromatography (GC) with electron-capture detection (ECD). The SPE method is highly sensitive for both compounds (limit of detection for each: 0.001 microg/g) and is less time consuming than the previously reported method based on repeated liquid-liquid partitioning and GC-ECD. Recoveries for 3 levels of fortification (0.02, 0.2, and 2.5 microg/g) from satsuma mandarins ranged from 94.3 to 96.5% for imazalil and from 93.9 to 96.3 % for R 14821, with coefficients of variation (CVs) ranging from 3.1 to 6.3% for imazalil and from 4.5 to 5.6% for R 14821. Average residual levels found in grapefruit, oranges, and lemons by the SPE method and the previously reported method, and the corresponding CVs, were similar.  相似文献

A sensitive high-performance liquid chromatographic assay for isbufylline and its major metabolites in rabbit blood and urine is described. After extraction, samples were eluted by a linear reversed-phase gradient. Specimens obtained after intravenous administration of isbufylline to rabbits were analysed to identify and subsequently quantify the potential metabolites. Using the ultraviolet absorption trace on the recorder as a reference, elution fractions were collected and analysed by mass spectrometry with the direct inlet system and gas chromatography-mass spectrometry after derivatization. Seven metabolites were identified and another five quantified. The method is specific, accurate, reproducible and recommended for pharmacokinetic studies.  相似文献

Electron-capture gas chromatography was carried out to determine midazolam and its three hydroxy metabolites (1-hydroxymethylmidazolam, 4-hydroxymidazolam and 1-hydroxymethyl-4-hydroxymidazolam) in human plasma. The assay involves extraction from plasma, buffered to pH 9.3, into cyclohexane-dichloromethane (6:4) and analysis by gas chromatography. The use of an HP-17 cross-linked, capillary column makes derivatization unnecessary. The sensitivity of the method was 2-3 ng/ml for midazolam, 1-hydroxymethylmidazolam and 4-hydroxymidazolam, and 20 ng/ml for 1-hydroxymethyl-4-hydroxymidazolam. The extraction recovery of midazolam, 1-hydroxymethylmidazolam, 4-hydroxymidazolam and 1-hydroxymethyl-4-hydroxymidazolam was 99.3 +/- 2.4, 67.0 +/- 4.6, 92.7 +/- 4.7 and 28.7 +/- 6.3%, respectively. This gas chromatographic assay was used to assess the concentration-time profiles of midazolam and its metabolites in human plasma after rectal and intravenous administration of midazolam.  相似文献

Oral prochlorperazine (PCZ), an antiemetic, undergoes extensive first-pass metabolism. The study developed a simultaneous analytical method for PCZ and its major metabolites, prochlorperazine sulfoxide (PCZSO), N-demethylprochlorperazine (NDPCZ) and 7-hydroxyprochlorperazine (PCZOH), in human plasma using an isocratic liquid chromatography-tandem mass spectrometry (LC-MS/MS) method. Deproteinized plasma specimens were separated using a 3 μm particle size octadecylsilyl column, and the run time was 10 min. The calibration curves were linear over the concentration ranges of 0.01-40 μg/L for PCZ, NDPCZ and PCZOH, and 0.05-80 μg/L for PCZSO. The intra- and inter-assay precisions and accuracies were within 7.0 and 99-104% and within 9.0 and 99-105%, respectively. The lower limits of quantification in human plasma were 10 ng/L for PCZ, NDPCZ and PCZOH, and 50 ng/L for PCZSO. The validated method was applied to the determination of plasma samples in 37 cancer patients receiving PCZ. Large interindividual variations were observed in plasma concentrations of PCZ, PCZSO, NDPCZ and PCZOH (relative standard deviation, 89.4, 88.7, 86.4 and 78.2%, respectively). In conclusion, this simultaneous LC-MS/MS method with acceptable analytical performance can be helpful for evaluating the pharmacokinetics of PCZ, including the determination of its metabolites in cancer patients and in clinical research.  相似文献
