New ratiometric two-photon fluorescent probes are developed from 6-substituted quinolines for biological Zn(2+) detection. They show large red shifts and good ratiometric responses upon Zn(2+) binding. They also exhibit high ion selectivities and large two-photon absorption cross sections at nearly 720 nm. Because the new probes are cell-permeable, they can be used to detect intracellular zinc flux under two-photon excitation.  相似文献

Iron is one of the essential trace elements in the human body. It plays an important role in human biology and pathology. Deregulation of iron levels in cells is associated with disease development. In this work, we synthesized a novel near-infrared intramolecular charge transfer (ICT) based ratiometric fluorescent probe to detect Fe2+, by using naphthalimide and indole moieties as building blocks. Our work showed that the radiometric probe has excellent selectivity, sensitivity and rapid response. Moreover, we could successfully perform real-time monitoring of Fe2+ in HeLa cells and C. elegans.  相似文献

An acedan derivative containing Zn(II)-DPA has been developed as a two-photon probe for nucleoside phosphates, which shows enhanced fluorescence toward ATP and ADP at physiological pH 7.4 among other competing anions including AMP; the probe is permeable to cell membranes and thus can be directly used for two-photon imaging of ATP and ADP in live cells.  相似文献

Autophagy plays a vital role in maintaining the balance of normal physiological state of living cells. In this paper, a polarity-specific two-photon fluorescent probe Lyso-NA based on naphthalimide was synthesized for the purpose of monitoring autophagy during biological research. The results of photophysical properties and theoretical calculation confirmed that different polarities of solvents mainly effected fluorescent intensities of probe. Fluorescent intensity, quantum yield and fluorescence lifetime of probe kept a good linear relationship with polarity respectively. In addition, due to its low toxicity and selective accumulation in lysosomes, Lyso-NA is suitable for detecting changes in lysosomal polarity of living cells. Compare with the imaging results of plasmid transfection, a better performed real-time long-term fluorescent visualization of autophagy in living cells was achieved. Probe Lyso-NA can work as an efficient and cost effective imaging tool for visualizing autophagy in living cells.  相似文献

Quinoline-based fluorescent probe as a recognition unit was designed and synthesized in this study. The probe R1 displayed excellent selectivity and sensitivity for cadmium ions (Cd2+) over a wide range of metal ions in acetonitrile-water (MeCN-H2O) mixed solution. In order to better understand the recognition mechanism between probe and Cd2+, the density functional theory calculations were performed. Finally, the colorimetric experiment result was observed and conveniently monitored by the naked eye, and a visual detection limit of 4 × 10?6 mol L?1 was achieved. These experimental results indicated the promising potential of the probe to detect Cd2+ in biological system. Furthermore, the probe R1 was successfully used for the highly sensitive detection of Cd2+ in living cells.  相似文献

A fluorescent probe, PyCalix, which has two pyrene moieties at the lower rim of a calix[4]arene fixed in the cone conformation was synthesized and its complexation behavior with alkali and alkaline earth cations was studied by fluorescence spectrometry. The compound showed intramolecular excimer emission at approximately 480 nm in the fluorescence spectra. Upon complexation with alkaline earth metal cations, a decrease of excimer emission was observed. The decrease of excimer emission was accompanied by an increase of monomer emission of pyrenes at 397 nm. The order of complexation constants of PyCalix with metal ions was Sr(+ approximately Ca2+ > Ba2+ > Mg2+ > K+ > Na+ > Cs+ for all reagents. PyCalix doped polyvinyl chloride (PVC) membrane was fabricated and our results showed that this membrane can be used for selective detection of Sr2+.  相似文献

A novel donor-acceptor(D-A) type of two-photon(TP) fluorescent probe,i.e.Lyso-OSC,based on the lysosome-targeting morpholine group was developed.The polarity sensing coumarin group was functionalized as the acceptor and the 1-vinyl-4-methoxybenzene group was engineered as the donor.The fluorescence intensity and emission maximum wavelength of Lyso-OSC are highly sensitive to the polarity changes of solvent.The two-photon absorption cross-section and tissue penetration depth are up to 254 GM and 150 μm,respectively.The strong fluorescence,high sensitivity to polarity,low cytotoxicity,and accurate lysosome-targeting ability entail Lyso-OSC the excellent performance in detecting the polarity changes ofcellular environment.To this end,a bright,real-time imaging autophagy of living cells has been achieved.  相似文献

In this work, we design and synthesize the novel probe RC through introduction the 1-aza-4,13-dithia-15-crown-5 ring into the structure of rhodamine 6G hydrazide, where the N atom of crown ring is responsible for quenching of rhodamine fluorescence. The compound obtained behaves as multifunctional cation sensor providing selective fluorescent response to Au3+ and selective colorimetric response to Cu2+ ions in aqueous acetonitrile (1/1, v/v) at pH 7.0. The use of 10?5?M RC solution allowed reliable determination of target cations in the presence of a wide range of environmentally relevant ions with detection limits of 2?×?10?6?M and 5?×?10?7?M for gold and copper, respectively.  相似文献

ABSTRACT

In this work, a new turn-on fluorescent probe 1 for Hg2+ ions detection based on rhodamine B spirolactam was reported. Among tested metal ions, probe 1 shows high selectivity towards Hg2+ in the the mixture solution of methanol and 0.02 M HEPES buffer (V/V = 9:1, pH = 7.2). No absorption and emission band of probe 1 was observed in the range from 450 to 700 nm. While only addition of Hg2+ to probe 1 could lead to appearance of a new absorption band centered at 553 nm and a fluorescence emission band around 577 nm upon excitation at 520 nm. Moreover, it exhibits excellent linear relationship (R2 = 0.9993) between fluorescence intensity at 577 nm and the concentration of Hg2+ from 1.6 to 32 μM. The sensing mechanism was proven to be spirolactam ring open induced by Hg2+ through 1H NMR, MS, absorption and fluorescence spectra. In addition, probe 1 could detect Hg2+ in real water samples and on filter paper, which demonstrates its application in environment science.  相似文献

以4-硝基-1,8-萘酐、N,N-二甲基乙二胺为主要原料合成了一种荧光探针:4-(2′-N,N-二甲基氨基乙基)氨基-N-(2-N,N-二甲基氨基)乙基-1,8-萘酰亚胺(DDN)。研究了其紫外可见吸收和荧光发射光谱。考察了Fe3+,Cr3+,Mn2+,Ni2+,Zn2+,Cu2+和Co2+离子对该化合物荧光发射光谱的影响,结果表明Cr3+离子能使DDN荧光显著增强。滴定实验结果推测DDN与Cr3+的结合比为1∶2。在DDN浓度为1.0×10-5mol/L时,Cr3+浓度在5.0×10-7~2.0×10-5mol/L范围内,DDN荧光强度(F)与Cr3+离子浓度[Cr3+]呈良好的线性关系,线性方程为F=9430.3527+14600.2973[Cr3+],相关系数r=0.9976。检测限为4.3×10-7mol/L。  相似文献

Based on a change in structure between spirocyclic (non-fluorescent) and ring-open (fluorescent) forms of rhodamine-based dyes, a new fluorescent and colorimetric Cu(2+) probe was designed and synthesized. Upon treatment with Cu(2+), the weakly fluorescent probe exhibited a strong fluorescence response with high selectivity.
