A highly useful methodology that allows the determination of the absolute configuration of aliphatic and aromatic chiral amines based on the rationalization of stereoselective three-point interactions during chromatography on a rationally designed chiral stationary phase and conformational analysis of the elutes was developed. This approach is based on the broadly accepted chiral recognition mechanism of the Whelk-O 1 CSP and requires a facile derivatization of the amine and the determination of the lowest energy conformation of the corresponding t-Boc- or Z-derived carbamates. The absolute configuration determined by employing chiral structure activity relationships in HPLC analysis was verified by single-crystal X-ray analysis or studies with carbamoyl derivatives of amines of known configuration. The general validity and applicability of this methodology was demonstrated by analysis of seven carbamates derived from aliphatic and aromatic amines. Due to its simplicity and time-efficiency, i.e., ease of derivatization of amines and fast HPLC method development, this approach can be considered a useful supplement to established techniques such as NMR spectroscopy.  相似文献

A wide range of nucleophiles, such as amines and alcohols, reacted intermolecularly with various allenes in the presence of gold catalysts to give the corresponding hydrofunctionalization products in high yields. The intermolecular hydroamination of chiral allenes with aromatic and aliphatic amines proceeded with high to good enantioface selectivities to afford the corresponding chiral allylic amines. On the other hand, in the case of the intermolecular hydroalkoxylation of chiral allenes, no chirality transfer was observed. This marked contrast on the chirality transfer indicates that the mechanisms of gold-catalysis between hydroamination and hydroalkoxylation are different.  相似文献

A new chiral stationary phase for gas chromatography was prepared by covalently attaching a diproline chiral selector that has proven to be effective in liquid chromatography to a methylhydrosiloxane-dimethylsiloxane copolymer. With this new chiral stationary phase for GC, racemic aromatic alcohols could be resolved without derivatization. Racemic aromatic and aliphatic amines could also be resolved after derivatization of the amino groups with trifluoroacetic anhydride or isopropyl isocyanate. On this stationary phase, the isopropyl isocyanate derivatives of amines showed higher enantioselectivity than the trifluoroacetic anhydride derivatives. In both the enantiomeric separations of alcohols and derivatized amines, the aromatic racemic analytes showed higher enantioselectivities than their aliphatic analogs. Some of the alpha-amino and alpha-hydroxy aromatic acids could also be separated after derivatization to N-trifluoroacetyl methyl esters for amino acids or O-trifluoroacetyl methyl esters for hydroxyl acids.  相似文献

A 3-component cascade synthesis of bis(2-arylallyl) tertiary amines from aryl iodide, allene and primary aliphatic amines is described; chiral amines give analogous products with no detectable racemisation; mixtures of two different aryl iodides can be utilised to give the mixed tertiary amines as the sole, or major, product; the reaction is sensitive to stereoelectronic effects which lead to mono(2-arylallyl) secondary amines.  相似文献

A Ni-catalyzed asymmetric ring-opening reaction of 2-substituted cyclopropane-1,1-dicarboxylates with aliphatic amines has been accomplished using the chiral indane-trisoxazoline (In-TOX) ligand. This highly enantioselective reaction provides an efficient approach to a variety of chiral γ-substituted γ-amino acid derivatives, which are readily transformed into multifunctionalized piperidines and γ-lactams. The single-crystal X-ray structure of the TOX-Ni complex is provided, and the role of the side arm in the chiral ligand is discussed.  相似文献

A highly active organocatalyst has been shown to affect the asymmetric reductive amination of ketones producing both aromatic and aliphatic amines. At 1 mol% catalyst loading, a series of structurally diverse chiral amines were quickly and economically prepared with good enantioselectivity and generally useful yield. The efficient synthesis of the calcimimetic (+)-NPS R-568 (67%, 89% ee) demonstrated the synthetic applicability of this methodology.  相似文献

In the course of our studies on Cerpegin analogues synthesis, a serendipitous reactivity of enaminolactone nitrile has been observed. Instead of expecting iminocerpegins, we have gained new class of substituted 2-aminopyridines. The methodology has been applied on a wide range of primary amines, as aliphatic, aromatic, heteroaromatic and also, diamines, hydrazines and chiral amines.  相似文献

Palladium-catalyzed hydroaminocarbonylation reactions of olefins using aliphatic amines were performed under carbon monoxide atmosphere. Despite the strong basicity of the applied nucleophiles, the targeted amides were successfully synthesized in the absence of acidic additives. Styrene, oct-1-ene and isoprene were transformed to the corresponding amide isomers in moderate to good isolated yields under optimized reaction conditions. Various aliphatic amines were used as N-nucleophiles. The effect of chiral diphosphines on product distribution, that is, on chemo-, regio- and enantioselectivities was also studied. Plausible explanation was given for the additive-free hydroaminocarbonylation reaction.  相似文献

An efficient strategy for preventing the β-hydride elimination of alkylpalladium species by ligation of the palladium with adjacent amino-group was developed, which enabled a novel palladium-catalyzed ring-closing aminoalkylative amination of unactivated aminoenynes. The reaction is amenable to aminals, as well as aliphatic aldehydes with secondary amines, which provides straightforward access to structurally diverse exocyclic allenic amines bearing 5 to 12-membered N-heterocycles. With chiral phosphoramidite-ligated palladium complex as the catalyst, an enantioselective variant was achieved with up to 93 % ee. Simultaneously, synthetic transformations of the chiral products were also conducted to afford structurally unique spirodiamines including one pharmaceutically active molecule via axial-to-central chirality transfer.  相似文献

The ring opening of N-tosylaziridines with aliphatic amines can be efficiently catalyzed by lithium perchlorate to provide derivatives of the trans-1,2-diamine in high yields. The reaction was used in desymmetrization of several cyclic N-tosylaziridines using chiral amines. Using this strategy, an efficient synthesis of chiral vicinal C2 symmetric bis(sulfonamide) and unsymmetrical bis(sulfonamide) ligands based on trans-1,2-cyclohexanediamine was developed.  相似文献

The catalytic asymmetric aziridination of imines and diazo compounds (AZ reaction) mediated by boroxinate catalysts derived from the VANOL and VAPOL ligands was investigated with chiral imines derived from five different chiral, disubstituted, methyl amines. The strongest matched and mismatched reactions with the two enantiomers of the catalyst were noted with disubstituted methyl amines that had one aromatic and one aliphatic substituent. The synthetic scope for the AZ reaction was examined in detail for α-methylbenzyl amine for cis-aziridines from α-diazo esters and for trans-aziridines from α-diazo acetamides. Optically pure aziridines could be routinely obtained in good yields and with high diastereoselectivity and the minor diastereomer (if any) could be easily separated. The matched case for cis-aziridines involved the (R)-amine with the (S)-ligand, but curiously, for trans-aziridines the matched case involved the (R)-amine with the (R)-ligand for imines derived from benzaldehyde and n-butanal, and the (R)-amine with the (S)-ligand for imines derived from the bulkier aliphatic aldehydes pivaldehyde and cyclohexane carboxaldehyde.  相似文献

The kinetic resolution of 2‐aryl‐N‐tosylaziridines and the asymmetric desymmetrization of meso ‐N‐tosylaziridines by ring openings with various primary and secondary anilines, and aliphatic amines as nucleophile have been realized by using a single silver(I)/chiral diphosphine complex as catalyst for the first time. The simple starting materials, broad scope, and easy scalability render this protocol a practical way to chiral vicinal diamine derivatives.  相似文献

New 1-alkoxy-3-alkyl-4,5-dimethylimidazolium bromides were synthesized by alkylation of the corresponding 1-alkylimidazole 3-oxides, which were conveniently prepared via condensation of α-(hydroxyimino)ketones, primary aliphatic amines, and formaldehyde. By using enantiomerically pure chiral amines, optically active imidazolium salts were obtained. Treatment with sodium tetrafluoroborate in acetone yielded the corresponding imidazolium tetrafluoroborates. All these compounds, with only one exception, were obtained as oils, which are considered as potential ionic liquids and ‘chiral ionic liquids’. The reduction of the chiral or non-chiral 1-alkylimidazole 3-oxides with Raney-Ni, followed by alkylation with alkyl bromides and subsequent ion exchange to tetrafluoroborates, gave the corresponding 1,3-dialkylimidazolium salts, most of them showing properties of ionic liquids. The alkylation of 1-butyl-4,5-dimethylimidazole 3-oxide and the corresponding imidazole, respectively, with 1,3-dibromopropane led to the first bis-imidazolium dibromides and bis-tetrafluoroborates.  相似文献

A new chiral stationary phase (CSP) was developed by bonding isopropyl-carbamate functionalized cyclofructan6 (IP-CF6) to the silica gel. It was evaluated by injecting 119 racemic primary amine-containing compounds. This CSP showed pronounced enantioselectivity toward all types of primary amines, separating 93% of all tested compounds. Baseline separation was achieved even for some simple aliphatic racemic amines that contained no other functionality. The polar organic mode was shown to be the effective mobile phase owing to higher efficiency. This new chiral stationary phase showed great potential for preparative-scale separations. It is also interesting that the chiral selector, R-naphthylethyl-carbamate functionalized CF6 (RN-CF6), was found to provide complementary selectivity for the relatively few amine analytes that did not separate on IP-CF6. Thus between the two CSPs, 98% of attempted amine compounds were separated.  相似文献

An efficient and reproducible synthesis of various α-ethoxycarbamates is described via a microwave heating mode. Compared to the thermal process, the microwave dielectric heating induces a dramatic reduction of the reaction time and the improvement of the yields. The reaction is general since applicable to aromatic and aliphatic aldehydes with various primary amines. Several examples involving chiral aldehydes have also been considered.  相似文献

The racemization of nonactivated aliphatic amines has been mediated with alkanethiols and with methyl thioglycolate in the presence of AIBN. The process involves reversible H-abstraction at the chiral center, in a position alpha relative to nitrogen, by thiyl radical. The knowledge of the reaction enthalpy is critical to select the appropriate thiol. In the absence of experimental values, theoretical calculations of the alpha-C-H BDEs and the S-H BDE provide a useful guide.  相似文献

A metal-free, mild and chemodivergent transformation involving nitroalkanes has been developed. Under optimized reaction conditions, in the presence of trichlorosilane and a tertiary amine, aliphatic nitroalkanes were selectively converted into amines or nitriles. Furthermore, when chiral β-substituted nitro compounds were reacted, the stereochemical integrity of the stereocenter was maintained and α-functionalized nitriles were obtained with no loss of enantiomeric excess. The methodology was successfully applied to the synthesis of chiral β-cyano esters, α-aryl alkylnitriles, and TBS-protected cyanohydrins, including direct precursors of four active pharmaceutical ingredients (ibuprofen, tembamide, aegeline and denopamine).  相似文献

The development of a catalytic asymmetric method for the direct aminoallylation of aldehydes is described that gives high asymmetric inductions for a broad range of substrates including both aromatic and aliphatic aldehydes. This method allows for direct isolation of unprotected analytically pure homoallylic amines without chromatography. The unique catalyst system developed for this process involves the synergistic interaction between a chiral and a nonchiral Br?nsted acid.  相似文献

The chiral phosphorus derivatizing agent (CDA) 1 was prepared from optically pure (S)‐1,1‐bis‐2‐naphthol. It was first used in the determination of the enantiomeric excess of chiral alcohols and amines by means of 31P NMR spectroscopy. It showed that, for the chiral aromatic alcohols, no apparent kinetic resolution was noted and good base‐line separation was observed. Furthermore, the chemical shift difference (Δδ) of 31P NMR spectroscopy was much larger than those determined by the use of other chiral phosphorus derivatizing agents reported previously. However, for aliphatic alcohols, it showed not only obvious kinetic resolutions but incomplete base‐line separation. Moreover, we also found that the use of CDA 1 was suitable for the determination of enantiomeric excess of chiral primary amines. © 2002 John Wiley & Sons, Inc.
