The ring-opening abilities of amines toward 1,3,4-oxadiazolines, 2-phenyl-1,3,4-oxadiazolin-5-one ( 1a ) and 2-phenyl-1,3,4-oxadiazoline-5-thione ( 1b ), were investigated with relation to their basicities or pKb values. Oxadiazolines 1a and 1b were easily reacted with amines such as benzylamine and aniline, but not with p-nitroaniline, to form the corresponding ring-opening adducts. The reactions of both 1a and 1b with o-phenylenediamine produced benzodiazoles with the liberation of benzoylhydrazide, whereas the reactions with o-aminobenzamide furnished quinazolines with the liberation of ammonia. o-Aminophenol and o-aminothiophenol were also reacted with 1a and 1b both of them giving 1,5-dibenzoylcarbohydrazide from 1a and 1,2-dibenzoylhydrazine from 1b. From the conditions affording the corresponding ring-opening adducts or reaction products, the ring-opening abilities of the amines toward 1a and 1b are in good correlation with the strength of their basicities or pKb values. The ring-opening of oxadiazolines were proved to occur with anilines. Therefore, the other reactions are also supposed to proceed via the ring-opening steps.  相似文献

Starting from readily available methyl 5-methyloxazole-4-carboxylate ( 1 ) and 4-methyl-5-oxazolylcar-boxylic acid hydrazide ( 11 ) the title compounds were prepared. The reaction of compound 1 with hydrazine hydrate afforded the corresponding hydrazide 2 . The reaction of compound 2 with formic acid yielded 1-formyl-2-(5-methyloxazole-4-carboxyl)hydrazine ( 3 ). Refluxing of the latter with phosphorus pentasulfide in xylene gave compound 5 in 62% yield. The reaction of compound 3 with phosphorus pentoxide afforded compound 4 . Starting from hydrazide 11 , compounds 13 and 14 were prepared similarly. Reaction of compound 2 with substituted isothiocyanate yielded compound 9 which was cyclized in basic medium to 4-alkyl-5-(5-methyl-4-oxazolyl)-2,4-dihydro-3H-1,2,4-triazole-3-thione ( 10 ). The isomer 19 was prepared similarly. Methylation and subsequent oxidation of compound 19 gave compound 21 . Reaction of the acid 7 with thiosemicarbazide in the presence of phosphorus oxychloride gave 2-amino-5-(5-methyl-4-oxazolyl)1,3,4-thiadiazole ( 8 ). 2-Amino-5-(4-methyl-5-oxazolyl)-1,3,4-thiadiazole ( 17 ) was prepared from acyl chloride 15 by the usual method.  相似文献

Acyclic diphosphanes, R1(H)PCH2ZCH2P(H)R2 (with Z = O, S; R1 = R2 = H, Me, Ph; R1 = H, R2 = Me), and 1,3,4-Oxadiphospholanes as well as 1,3,4-Thiadiphospholanes are obtained from the esters by reduction.  相似文献

A series of thermally stable and semiconducting polyazomethines containing 1,3,4-oxadiazole or 1,3,4-thiadiazole ring in the polymer backbone were synthesized by the simple solution poly-condensation of dialdehydes with the preformed nuclei with aromatic diamines under mild conditions. To elucidate the structure and also structure-property relationships of the polymers, model compounds were prepared under the same reaction conditions. These polyazomethines having a wholly conjugated system were yellow powders and had reduced viscosities up to 0.38 dL/g in concentrated sulfuric acid and electric conductivity as high as 10?11 S cm?1 at room temperature. Thermogravimetry showed that all the polymers were heat resistant up to around 400°C, in both air and nitrogen atmospheres. Their completely black colored charge-transfer complexes were prepared by iodine-doping of the polymers. The room temperature conductivity of the polymers was found to be markedly increased up to the orders of 10?6–10?7 S cm?1 upon doping. The highest value attained was 4.8 x 10?6 S cm?1. Comparison of electronic spectra of the polymers with those of the model compounds indicated the π-electrons in the polymers are extensively delocalized along the polymer main chain.  相似文献

Since the introduction of the first antibiotic (penicillin, 1942) into medical practice, to date, there has been an ongoing “race” between scientists creating new drugs and pathogenic bacteria. Antibiotic-bacteria are becoming progressively common, and to make matters worse, more and more bacteria are becoming resistant to all known antibiotics. The traditional method for this problem is to introduce new antibiotics that kill the resistant mutants. This specific “arms race” resulted into thousands of potentially active chemicals are synthesized in laboratories around the world every day.1,3,4-Oxadiazole; 1,3,4-thiadiazole; 1,2,4-triazole and some of their derivatives are involved in modifications at the following axes: First, attaching a thio-group into heterocyclic rings. Second, introducing different substitutions at position 5 which often are the residuals of the synthetic starting materials such as simple aliphatic, substituted aliphatic chains, aromatic carbocyclic and heterocyclic residues.  相似文献

对氟苯甲酰腙与乙酸酐或丙酸酐反应后再脱水环化制得2-苯基-3-乙酰基-5-对氟苯基-1,3,4-噁唑啉(3)或2-苯基-3-丙酰基-5-对氟苯基-1,3,4-噁唑啉(4).N-对氟苯甲酰基-N′-苯甲酰肼在POCl3存在下脱水环化制得2-苯基-5-对氟苯基-1,3,4-噁二唑(6).新化合物3,4和6的结构经1H NMR,IR,MS和元素分析表征.用MTT方法评价了其对HepG-2,A549-1和231-2癌细胞株的体外生长抑制活性.结果表明,3,4和6均具有潜在的体外抑制癌细胞生长活性,其中3的活性最强.  相似文献

The structures of 2-amino-5-methyl-1,3,4-thia­diazo­le, C3H5N3S, and 2-amino-5-ethyl-1,3,4-thia­diazo­le, C4H7N3S, have been determined for comparison with unsubstituted 2-amino-1,3,4-thia­diazo­le. Despite their different space groups (P21/n and Pbca, respectively), the packing modes of the methyl and ethyl derivatives are similar, with comparable three-dimensional hydrogen-bonding associations. This is in contrast to the hydrogen-bonding network in 2-amino-1,3,4-thia­diazo­le, which is one-dimensional and has denser packing. It is shown that both packing forms are different polymorphs of a specific subunit of each array.  相似文献

The reactions of trichloromethylarenes with excess hydrazine hydrate in ethanol gives symmetrical 2,5-diaryl-1,3,4-oxadiazoles in 68–96% yields. The reaction of 1,4-bis(trichloromethyl)benzene with acylhydrazines in an ethanol-pyridine mixture gives the corresponding substituted or unsubstituted 1,4-phenylenebis-1,3,4-oxadiazoles in 35–51% yields. The mass spectra of 2,5-diaryl-1,3,4-oxadiazoles and 1,4-phenylenebis-1,3,4-oxadiazoles were studied. Translated fromIzvestiya Akademii Nauk. New methods for the synthesis of 1,3,4-oxadiazole and 1,3,4-thiadiazole have been described. No cyclizations took place in the absence of ZnCl2. 1,3,4-Thiadiazoles are formed in the presence of ZnCl2 alone, whereas oxadiazoles are produced when a base such as Et3N or KOH was used along with ZnCl2. % Yields are optimized.  相似文献
