Studies on pyrrolidinones. A convenient synthesis of 2-methyl-5-(5-oxo-1-benzyl-2-pyrrolidinyl)-1,3,4-oxadiazole

2-Methyl-5-(5-oxo-1-benzyl-2-pyrrolidinyl)-1,3,4-oxadiazole is easily obtained by dehydration of N-acetyl-1-benzylpyroglutamic acid hydrazide with a methanesulfonic acid/phosphoric anhydride mixture.

Reactions of 3-methyl-1-(2-pyridyl)-4-chloro-5-formyl-6,7-dihydroindazole

The reactions of 3-methyl-1-(2-pyridyl)-, 3-methyl-1-phenyl-, and 3-methyl-1,6-diphenyl-4-chloro-5-formyl-6,7-dihydroindazoles with guanidine and benzo- and 3- and 4-pyridinecarbamidines gave the corresponding 8-substituted 1-methyl-3-(2-pyridyl)-and 1-methyl-3-phenyl-4,5-dihydropyrazolo[5,4-h]quinazolines. With acetic anhydride the same indazole derivatives gave the 4-acetoxy-5-formyl derivatives, and with hydroxylamine they gave4-chloro-5-hydroxyiminomethyl-6,7-dihydroindazoles. Thereactionof4-acetoxyl-1-(2-pyridyl)-5-formyl-6,7-dihydroindazole with hydroxylamine gave 8-methyl-6-(2-pyridyl)-4,5-dihydroisoxazolo[5,4-e]indazole, while dehydration of 5-hydroxyiminomethyl-3-methyl-4-chloro-6,7-dihydroindazole gave the 4-chloro-5-cyano derivative. Synthesis of new modified aza heterocycles on the basis of 5-(2-chloro-1-nitroalkyl)-3-phenyl-and 5-(2-chloro-1-nitroalkyl)-3-methyl-1,2,4-oxadiazoles

3-Phenyl-and 3-methyl-5-(2-chloro-1-nitroalkyl)-1,2,4-oxadiazoles reacted with piperidine, pyrrolidine, and morpholine to give the corresponding 5-(2-amino-1-nitroalkyl) derivatives, while their reactions with sodium p-toluenesulfinate led to the formation of 2-[3-methyl(or phenyl)-1,2,4-oxadiazol-5-yl]-2-nitroethyl p-tolyl sulfones.     

Synthesis and reactions of 1-(2-pyridyl)-3-methyl-4-chloro-5-formyl-6,7-dihydroindazoles

The Vilsmeier formylation of 1-(2-pyridyl)-3-methyl-4-oxo-4,5,6,7-tetrahydroindazole and its 6-phenyl derivative gives 1-(2-pyridyl)-3-methyl-4-chloro-5-formyl-6,7-dihydroindazoles. Reactions of these derivatives with different N- and C-nucleophilic agents, including bisnucleophiles, were studied as a means of obtaining new 4- and 5-functional derivatives of indazole and its condensed systems.Riga Technical University, Riga LV-1658, Latvia. 4-取代苯次甲亚胺-5-(1-苯基-3-甲基-5-氯吡唑)-2H-1,2,4-三唑-3-硫酮的合成

以乙醇为溶剂,冰醋酸为催化剂,4-氨基-5-(1-苯基-3-甲基-5-氯吡唑)-1,2,4-三唑-3-硫酮(1)与芳醛经缩合反应合成了7个新型的4-取代苯次甲亚胺-5-(1-苯基-3-甲基-5-氯吡唑)-4H-1,2,4-三唑-3-硫酮(3a~3g),收率66%~74%,其结构经1H NMR,IR及元素分析表征。合成4-(苯次甲亚胺)-5-(1-苯基-3-甲基-5-氯吡唑)-2H-1,2,4-三唑-3-硫酮(3a)的最佳反应条件为:以乙醇为溶剂,乙酸为催化剂,1 10 mmol,n(苯甲醛)∶n(1)=1.2,于75℃反应3 h,产率74%。     

Synthesis of 2-and 6-chloro-3-(5-methyl-1,3,4-oxadiazol-2-yl)pyridines

A convenient synthesis of 2- and 6-chloro-3-(5-methyl-1,3,4-oxadiazol-2-yl)pyridines, 3 and 10 , utilizing the readily available amide 7 is described. Incorporation of the 3-t-butylamino-2-hydroxypropoxy side chain onto 3 provided 2 , a potential bioisostere of 1 . The antihypertensive activities of 2 and 12 were evaluated in the spontaneously hypertensive rat model.
