The synthesis, characterization and photophysical properties of two perfluoroalkyl (5,10,15,20-tetrakis-[trifluoromethyl]- and [heptafluoropropyl]-porphyrin) and two perfluoroaryl (5,10,15,20-tetrakis-[2,6-difluorophenyl]- and [pentafluorophenyl]-porphyrin) are described, with reference to their potential in both photodynamic therapy (PDT) and in vivo imaging by fluorescence and 19F nuclear magnetic resonance spectroscopy. Absorption and fluorescence spectra, fluorescence lifetimes and triplet-singlet difference spectra are reported. Triplet yields have been obtained by flash photolysis and pulse radiolysis, whereas yields of sensitized singlet oxygen formation have been determined by time-resolved phosphorimetry. All four compounds show high yields of triplet formation and singlet oxygen sensitization. The spectral properties, stability and attractive solubility characteristics of the perfluoroalkyl derivatives make them particularly suitable candidates for future study for applications in PDT.  相似文献

PHOTODYNAMIC INACTIVATION OF LYSOZYME BY EOSIN   总被引:2,自引:0,他引:2  
Abstract— It has been demonstrated that singlet oxygen is the major oxidizing entity in the photo-dynamic inactivation of hen egg white lysozyme by eosin, using D2O to enhance the solvent-induced decay lifetime, and azide ion as a specific scavenger. Two regimes of inactivation can be distinguished depending on whether the sensitizer is free or complexed to the enzyme. The kinetic analysis for free dye sensitization, based on photostationary measurements and inactivation quantum yields, indicates that at least 1 in 15 singlet oxygen interactions with lysozyme leads to loss of lytic activity. The direct attack of triplet eosin makes a lesser overall contribution in air-saturated solutions, where 1 in 4 reactions induces inactivation. Lysozyme binds 1 eosin molecule from pH 4 to 12, leading to almost total quenching of the tryptophyl residue fluorescence without inhibition of the enzymic activity. The inactivation quantum yields indicate that singlet oxygen generated from the bound dye is the inactivating agent, but the dominant attack takes place with the complexed fraction of lysozyme molecules. The tryptophyl residue loss is the same or smaller in changing from H2O to D2O despite the 5–10 times increase in quantum yield, indicating that singlet oxygen inactivates also by reacting with residues other than tryptophan. The photochemical and fluorescence results are consistent with the the identification of tryptophyl site 108 with the eosin binding site and a reaction target for singlet oxygen. In a re-examination of earlier work on eosin-sensitized photo-oxidation of I", it has been found that singlet oxygen is the oxidizing agent in aerobic solutions.  相似文献

Abstract— The o-, m-, and p-isomers of 5, 10, 15, 20- tetra(hydroxyphenyl)-porphyrin have been of recent interest as potential second-generation sensitisers in tumour phototherapy. Fluorescence spectroscopy, nanosecond laser flash photolysis and pulse radiolysis have been used to characterise the singlet and triplet excited states of tetraphenylporphyrin and the o, m-, and p-isomers of tetra(hydroxyphenyl)porphyrin. This has included evaluation of fluorescence yields and lifetimes, triplet spectra, lifetimes, oxygen quenching rate constants, extinction coefficients, and yields and singlet oxygen yields. Whilst the fluorescence quantum yields were low, the triplet yields were all 0.7 ± 10% and the singlet oxygen yields 0.6 ± 10%: all these parameters are in the ranges shown by other efficient porphyrin photosensitisers. The similar photophysical properties found for these compounds suggest that their differing tumour sensitising efficiencies are likely to be due to other factors.  相似文献

The synthesis and characterization of new peripherally tetra-4-benzyloxybenzoxy substituted metal-free, zinc and lead phthalocyanines are described for the first time in this study. The influence of various organic solvents and the nature of the central metal ion on the spectroscopic, photophysical and photochemical properties has been investigated. General trends are described for photodegradation, singlet oxygen and fluorescence quantum yields, and fluorescence lifetimes of these compounds in different solvents. Photophysical and photochemical properties of phthalocyanine compounds are very useful for photodynamic therapy applications. Especially high singlet oxygen quantum yields are very important for Type II mechanism. The studied phthalocyanine compounds showed good singlet oxygen generation and these compounds show potential as Type II photosensitizers. The fluorescences of the studied compounds are effectively quenched by 1,4-benzoquinone in different solvents.  相似文献

A complete set of platinum(II) solketal substituted phthalocyanines has been synthesized and characterized. To evaluate their potential as Type II photosensitizers for photodynamic therapy, comparative studies of their photophysical and photochemical properties with analogous zinc(II) series have been achieved: electronic absorption, fluorescence quantum yields, lifetimes, and fluorescence quenching by benzoquinone, as well as singlet oxygen generation and photodegradation. It appears that platinum(II) phthalocyanines are worth being used as Type II photosensitizers, as they exhibit good singlet oxygen generation and appropriate photodegradation.  相似文献

Peptide-coated quantum dot-photosensitizer conjugates were developed using novel covalent conjugation strategies on peptides which overcoat quantum dots (QDs). Rose bengal and chlorin e6, photosensitizers (PSs) that generate singlet oxygen in high yield, were covalently attached to phytochelatin-related peptides. The photosensitizer-peptide conjugates were subsequently used to overcoat green- and red-emitting CdSe/CdS/ZnS nanocrystals. Generation of singlet oxygen could be achieved via indirect excitation through F?rster (fluorescence) resonance energy transfer (FRET) from the nanocrystals to PSs, or by direct excitation of the PSs. In the latter case, by using two color excitations, the conjugate could be simultaneously used for fluorescence imaging and singlet oxygen generation. Singlet oxygen quantum yields as high as 0.31 were achieved using 532-nm excitation wavelengths.  相似文献

Terpolymers of ethylene, propylene and ethylidene norbornene (EN) rapidly react with singlet oxygen generated by dye sensitization. The singlet oxygen attacks the ethylidene double bond forming at least two different allylhydroperoxides. The instability of these hydroperoxides causes crosslinking of the photo-oxidized terpolymers at relatively low temperatures. Reduction of the hydroperoxide groups with triphenylphosphine yields a thermally stable hydroxylated polymer. Terpolymers of ethylene, propylene and dicyclopentadiene (DCPD) are inert towards singlet oxygen.  相似文献

Dendrimers with a C60 core have been obtained by cyclization of dendritic bis-malonate derivatives at the carbon sphere. The resulting bis-methanofullerene derivatives have been characterised by electrospray (ES) and/or MALDI-TOF mass spectrometries. UV-VIS absorption spectra, fluorescence spectra, and fullerene singlet excited state lifetimes have been determined in solvents of different polarity (toluene, dichloromethane, acetonitrile). These data suggest a tighter core/periphery contact upon increase of solvent polarity and dendrimer size. In all the investigated solvents, the fullerene triplet lifetimes are steadily increased with the dendrimer volume, reflecting lower diffusion rates of O2 inside the dendrimers along the series. Measurements of quantum yields of singlet oxygen sensitization indicate that longer lived triplet states generate lower amounts of singlet oxygen (1O2) in dichloromethane but not in apolar toluene suggesting a tighter contact between the dendritic branches and the fullerene core in CH2Cl2. In acetonitrile, the trend in singlet oxygen production is peculiar. Effectively, enhanced singlet oxygen production is monitored for the largest dendrimer. This reflects specific interactions of excited 1O2 molecules with the dendritic wedges, as probed by singlet oxygen lifetime measurements, possibly as a consequence of trapping effects.  相似文献

We report the synthesis, crystallographic, optical, and triplet and singlet oxygen generation properties of a series of BF2‐rigidified partially closed chain bromotetrapyrroles as near infrared emitters and photosensitizers. These novel dyes were efficiently synthesized from a nucleophilic substitution reaction between pyrroles and the 3,5‐bromo‐substituents on the tetra‐ and hexabromoBODIPYs and absorb in the near‐infrared region (681–754 nm) with high molar extinction coefficients. Their fluorescent emission (708–818 nm) and singlet oxygen generation properties are significantly affected by alkyl substitutions on the two uncoordinated pyrrole units of these dyes and the polarity of solvents. Among them, dyes 4 ca and 4 da show good singlet oxygen generation efficiency and good NIR fluorescence emission (fluorescence quantum yields of 0.14–0.43 in different solvents studied).  相似文献

The new compounds have been characterized by elemental analysis, IR, 1H and 13C NMR spectroscopy, electronic spectroscopy and mass spectra. General trends are described for photodegradation, singlet oxygen, triplet state and fluorescence quantum yields, and triplet and fluorescence lifetimes of these compounds in dimethylsulfoxide (DMSO). Photophysical and photochemical properties of phthalocyanine complexes are very useful for PDT applications. The effects of the substituents on the photophysical and photochemical parameters of the zinc(II) phthalocyanines (3a, 5a and 6a) are also reported. The singlet oxygen quantum yields (Phi(Delta)), which give an indication of the potential of the complexes as photosensitizers in applications where singlet oxygen is required (Type II mechanism) ranged from 0.60 to 0.72. Thus, these complexes show potential as Type II photosensitizers. The fluorescence of the complexes was quenched by benzoquinone (BQ).  相似文献

2,4-Bis(4-dimethylaminophenyl)-6-alkylthiopyrylium and selenopyrylium dyes are essentially nonfluorescent (phi F < 0.001) and are poor generators of singlet oxygen in aqueous solution. However, upon complexation to calf thymus DNA, quantum yields for both fluorescence and generation of singlet oxygen increased dramatically. Irradiation of the dye-DNA complexes produced strand breaks in the DNA. The photodamage is not observed in the absence of oxygen and is suppressed by the addition of the singlet oxygen quencher imidazole. The inactivation of the pseudo-rabies virus upon treatment of oxygenated leukodepleted 20% hematocrit red blood cell suspensions with the chalcogenopyrylium dyes and light followed the same trend observed with quantum yields for the generation of singlet oxygen in the dye-DNA complexes.  相似文献

The synthesis, photophysical and photochemical properties of the tetra-substituted aryloxy gallium(III) and indium(III) phthalocyanines are reported for the first time. General trends are described for photodegradation, singlet oxygen, fluorescence, and triplet quantum yields and triplet lifetimes of these compounds. The introduction of phenoxy and tert-butylphenoxy substituents on the ring resulted in lowering of fluorescence quantum yields and lifetimes, and triplet quantum yields, and an increase of kIC, kISC, and kF. Photoreduction of the complexes was observed during laser flash photolysis. The singlet oxygen quantum yields (ΦΔ), which give an indication of the potential of the complexes as photosensitizers in applications where singlet oxygen is required (Type II mechanism) ranged from 0.41 to 0.91. Eight differently substituted title dye compounds have been investigated regarding intersystem crossing, triplet state, fluorescence and singlet excited state pKa properties. In general, non-halogenated oxazines and thiazines as well as a mono bromooxazine show very low triplet quantum yields, phi tau (less than 0.03) and relatively long triplet lifetimes (approximately 40 microseconds) in acidic methanol. The phi tau data correlate well with known singlet oxygen yields. In basic methanol no triplet transient is observed but a significant yield of a ground state transient protonated (base dye) form is produced with a short lifetime, approximately 400 ns. Fluorescence can be seen simultaneously from both the excited base and the protonated base dye forms in basic methanol. For iodinated oxazine or thiazines, the triplet yield increases and can be as high as 0.5 (diiodo case) in acidic methanol. The triplet lifetimes are further shortened to approximately 10 microseconds compared to the non-iodinated derivatives above. The triplet yields of the iodo compounds are higher or equal to known singlet oxygen yields. In basic methanol triplet yields up to 0.2 can be seen, the triplet lifetime are shortened still further to 1 microsecond but no observable protonated form is produced (in distinction to the non-iodinated cases). Consideration is given to the correlation of triplet and singlet oxygen yields, ground and excited pKa properties, spin-orbit coupling and internal conversion properties, solvent effects, and phototherapeutic activity of these dyes.  相似文献

The synthesis and photophysical properties of a new series of zinc(II) phthalocyanines (ZnPcs) bearing multiple hydroxy and tert-butyl groups are reported. The X-ray structures of two phthalonitriles and one ZnPc are presented. All hydroxy-substituted ZnPcs show low fluorescence quantum yields in DMSO and complete fluorescence quenching in aqueous solutions, but high singlet oxygen quantum yields in DMSO (0.2-0.7). Our results suggest that the tetra- and octa-hydroxy ZnPcs might find application as photosensitizers in the PDT treatment of cancer.  相似文献

The spectral–luminescent, photophysical, and photochemical properties of dichloro-, dibromo-, and diiodo-derivatives of boron dipyrromethenate (BODIPY) have been studied, as well as the feasibility of generating singlet oxygen (1O2) via its photosensitization by the dihalogenated derivatives of BF2 dipyrromethene in solutions. Quantum yields of singlet oxygen have been determined using 1,3-diphenylisobenzofuran as the 1O2 trap. The lowest fluorescence quantum yields have been shown to correspond to the maximum yields of singlet oxygen. It has been found that the best 1O2 photosensitizer among the three test dihalotetraphenylaza- BODIPY is dibromotetraphenylaza-BODIPY, which in addition possesses the highest photostability. Diiodotetramethyl-BODIPY results in the singlet oxygen yield close to unity, but it has significantly lower photostability. The yield of singlet oxygen is affected by the solvent. Dibromtetraphenylaza-BODIPY and diiodotetramethyl-BODIPY may find use as a medium in photodynamic therapy and photocatalysis of oxidation reactions.  相似文献

We report the singlet oxygen sensitization properties of a series of bis-cyclometalated Ir(III) complexes (i.e., (bt)2Ir(acac), (bsn)2Ir(acac), and (pq)2Ir(acac); bt = 2-phenylbenzothiazole, bsn = 2-(1-naphthyl)benzothiazole, pq = 2-phenylquinoline, and acac = acetylacetonate). Complexes with acetylacetonate ancillary ligands give singlet oxygen quantum yields near unity (PhiDelta = (0.7-1.0) +/- 0.1), whether exciting the ligand-based state or the lowest energy excited state (MLCT + 3LC). The singlet oxygen quenching rates for these beta-diketonate complexes were found to be small [(5 +/- 2) x 105 to (6 +/- 0.2) x 106 M-1 s-1], roughly 3 orders of magnitude slower than the corresponding phosphorescence quenching rate. Similar complexes were prepared with glycine or pyridine tethered to the Ir(III) center (i.e., (bsn)2Ir(gly) and (bt)2Ir(py)Cl; gly = glycine and py = pyridine). The glycine and pyridine derivatives give high singlet oxygen yields (PhiDelta = (0.7-1.0) +/- 0.1).  相似文献

Synthesis, photophysical and metal ion recognition properties of a series of amino acid‐linked free‐base and Zn‐porphyrin derivatives (5–9) are reported. These porphyrin derivatives showed favorable photophysical properties including high molar extinction coefficients (>1 × 105 m ?1 cm?1 for the Soret band), quantum yields of triplet excited states (63–94%) and singlet oxygen generation efficiencies (59–91%). Particularly, the Zn‐porphyrin derivatives, 6 and 9 showed higher molar extinction coefficients, decreased fluorescence quantum yields, and higher triplet and singlet oxygen quantum yields compared to the corresponding free‐base porphyrin derivatives. Further, the study of their interactions with various metal ions indicated that the proline‐conjugated Zn‐porphyrins (6 and 9) showed high selectivity toward Cu2+ ions and signaled the recognition through changes in fluorescence intensity. Our results provide insights on the role of nature of amino acid and metallation in the design of the porphyrin systems for application as probes and sensitizers.  相似文献

A new series of aza‐BODIPY derivatives ( 4 a – 4 c , 5 a , c , and 6 b , c ) were synthesized and their excited‐state properties, such as their triplet excited state and the yield of singlet‐oxygen generation, were tuned by substituting with heavy atoms, such as bromine and iodine. The effect of substitution has been studied in detail by varying the position of halogenation. The core‐substituted dyes showed high yields of the triplet excited state and high efficiencies of singlet‐oxygen generation when compared to the peripheral‐substituted systems. The dye 6 c , which was substituted with six iodine atoms on the core and peripheral phenyl ring, showed the highest quantum yields of the triplet excited state (ΦT=0.86) and of the efficiency of singlet‐oxygen generation (ΦΔ=0.80). Interestingly, these dyes were highly efficient as photooxygenation catalysts under artificial light, as well as under normal sunlight conditions. The uniqueness of these aza‐BODIPY systems is that they are stable under irradiation conditions, possess strong red‐light absorption (620–680 nm), exhibit high yields of singlet‐oxygen generation, and act as efficient and sustainable catalysts for photooxygenation reactions.  相似文献

This work gives an overview of what is currently known about the mechanisms of the photosensitized production of singlet oxygen. Quenching of pi pi* excited triplet states by O2 proceeds via internal conversion of excited encounter complexes and exciplexes of sensitizer and O2. Both deactivation channels lead with different efficiencies to singlet oxygen generation. The balance between the deactivation channels depends on the triplet-state energy and oxidation potential of the sensitizer, and on the solvent polarity. A model has been developed that reproduces rate constants and efficiencies of the competing processes quantitatively. Sensitization by excited singlet states is much more complex and hence only qualitative rules could be elaborated, despite serious efforts of many groups. However, the most important deactivation paths of fluorescence quenching by O2 are again directed by excess energies and charge-transfer interactions similar to triplet-state quenching by O2. Finally, two recent developments in photosensitization of singlet oxygen are reviewed: Two-photon sensitizers with particular application potential for photodynamic therapy and fluorescence imaging of biological samples and singlet oxygen sensitization by nanocrystalline porous silicon, a material with very different photophysics compared to molecular sensitizers.  相似文献
