The one-step synthesis of 8-t-butoxycarbonyl-6,7-dimethyl-2-oxo-1,2,3,4-tetrahydropyrrolo[1,2-a]pyrimidine from acetyl methyl carbinol, 3-aminopropionic acid, and t-butyl cyanoacetate is reported. Utilizing a similar technology under basic conditions, 7-substituted 5,6-dimethyl-2-oxo-2,3-dihydro-(1H)-pyrrolo[1,2-a]imidazole is synthesized from acetyl methyl carbinol, ethyl glycinate, and the appropriate acetonitrile.  相似文献

Substituted pyrrolo[1,2-a]imidazoles and imidazo[1,2-a]imidazoles are prepared in a simple one pot reaction sequence from esters of heterocyclic or aromatic α-amino acids. A new method is proposed for the synthesis of pyrrolo[1,2-a]pyrazines and pyrrolo[1,2-a]-quinoxalines. By the alkylation of sodium derivatives of 2-acylpyrroles with -bromo carbonyl compounds or their acetals and subsequent treatment of the reaction products with ammonium acetate in acetic acid, a number of derivatives of pyrrolo[1,2-a]pyrazine, including the first member of the class, pyrrolo[1,2-a]pyrazine itself, have been obtained. Similarly, from 2-benzoylpyrrole and the dimethyl ketal of -bromocyclohexanone was obtained 4-phenyltetrahydropyrrolo[1,2-a]quinoxaline, which readily dehydrogenates in the presence of Raney nickel to form 4-phenylpyrrolo[1,2-a]quinoxaline.Translated from Khimiya Geterotsiklicheskikh Soedinenii, Vol. 6, No. 8, pp.1048–1050, August, 1970.  相似文献

The acylation of pyrrolo[1,2-a]pyrazines with acetic anhydride and the acid chlorides of various carboxylic acids has been studied. It has been shown that pyrrole[1,2-a]pyrazines are selectively acylated at the α-position of the pyrrole ring when it is free. The closeness of the electronic structures of the ions formed in the first act of disintegration of the ions is responsible for the monotypic character of the subsequent fragmentation of pyrrolo[1,2-a]benzimidazole and imidazo[1,2-a]benzimidazole derivatives. Reactions of 2-(3,3-diamino-2-cyanoprop-2-enylidene)indolin-3-ones with amide acetals afforded derivatives of 3-aminomethyleneamino-2-cyano-9-oxo-9H-pyrrolo[1,2-a]indole and 3-amino-2-cyano-9-oxo-9H-pyrrolo[1,2-a]indole. The rate and pathway of the reaction depend on the reagents and solvents. The formation schemes for the compounds obtained were proposed. Their structures were proven by IR and 1H NMR spectroscopy and mass spectrometry and confirmed by independent syntheses. Published in Russian in Izvestiya Akademii Nauk. The protonation of a number of pyrrolo[1,2-a]imidazole derivatives in trifluoroacetic acid was studied by PMR spectroscopy. The 5,7-unsubstituted compounds form a mixture of two forms of cations, the structures of which correspond to the addition of a proton to the C5 and C7 atoms of the two-ring system with predominance (60–90%) of the 5-C cation. The introduction of a CH3 group into the 5 position changes the direction of protonation to favor predominant (95%) formation of the 7-C cation. The 7-methyl derivatives of pyrroloimidazole are protonated exclusively at the C5 atom. It is demonstrated that the basicity of the pyrroloimidazoles considerably surpasses the basicity of indolicine derivatives. The protonation of a number of pyrrolo[1,2-a]benzimidazole derivatives in trifluoroacetic acid was studied by PMR spectroscopy. 1,3-Unsubstituted compounds are protonated exclusively at the C1 atom. Under similar conditions, pyrrolobenzimidazoles that have a methyl group in the 1 position form a mixture of two protonated forms, which correspond to the addition of a proton to C1 and C3, respectively. The relative percentage of the C3-protonated form decreases successively (from 81 to 18%) on passing from the 3-unsubstituted compound to the corresponding 3-phenyl and 3-methyl derivatives. The basicity constants of the pyrrolobenzimidazoles decrease symbatically with an increase in the relative percentage of this form. A transition metal-free process for the regioselective synthesis of pyrrolo[1,2-a]quinoxalines under mild conditions in one-pot is described. The reaction afforded a variety of products in good to excellent yields. Indolo[1,2-a]quinoxalines were also synthesized from indole-2-carboxamides under the same conditions.  相似文献

The fragmentation mechanisms of 11H-dibenzo[c,f][1,2]diazepine (I), its 3,8-dichloro derivative (II), 3,8-dichlorodibenzo[c,f] [1,2]diazepin-11-one (III) and 3,8-dichloro-11H-dibenzo-[c,f][1,2]diazepin-N-oxide (IV) are discussed. The initial loss of molecular nitrogen is characteristic of I, II and III. Compound IV has a strong molecular ion, that competitively eliminates cither NO or Cl- and N2O. The common radical ion, m/166 e present in the mass spectra of I, fluorene, 2-methyl-9,10-anthraquinone and 2-methylbenzo[c]cinnoline, appears to be formed in different states.  相似文献

Several 1 1-amino-5H-pyrrolo[2,1-c][1,4]benzodiazepines have been used as starting material to prepare a number of derivatives of 9H-imidazo[1,2-a]pyrrolo[2,1-c][1,4]benzodiazepines and 10H-pyrimido[1,2-a]pyrrolo[2,1-c][1,4]benzodiazepines. The imidazole nucleus was built by reaction of amidines with ethyl bromopyruvate or aminoacetaldehyde dimethylacetal. Several derivatives of imidazo[1,2-a]pyrrolo[2,1-c][1,4]benzodiazepine have been prepared by formylation of the pyrrole ring followed by formation of thioamides. Condensation of 11-amino-5H-pyrrolo[2,1-c][1,4]benzodiazepines with diethyl ethoxymethylenemalonate afforded intermediate diesters which were transformed into the corresponding 10H-pyrimido[1,2-a]pyrrolo[2,1-c]-benzodiazepines.  相似文献

Background: Neurotic disturbances, anxiety, neurosis-like disorders, and stress situations are widespread. Benzodiazepine tranquillizers have been found to be among the most effective antianxiety drugs. The pharmacological action of benzodiazepines is due to their interaction with the supra-molecular membrane GABA-a-benzodiazepine receptor complex, linked to the Cl-ionophore. Benzodiazepines enhance GABA-ergic transmission and this has led to a study of the role of GABA in anxiety. The search for anxiolytics and anticonvulsive agents has involved glutamate-ergic, 5HT-ergic substances and neuropeptides. However, each of these well-known anxiolytics, anticonvulsants and cognition enhancers (nootropics) has repeatedly been reported to have many adverse side effects, therefore there is an urgent need to search for new drugs able to restore damaged cognitive functions without causing significant adverse reactions. Objective: Considering the relevance of epilepsy diffusion in the world, we have addressed our attention to the discovery of new drugs in this field Thus our aim is the synthesis and study of new compounds with antiepileptic (anticonvulsant) and not only, activity. Methods: For the synthesis of compounds classical organic methods were used and developed. For the evaluation of biological activity some anticonvulsant and psychotropic methods were used. Results: As a result of multistep reactions 26 new, five-membered heterocyclic systems were obtained. PASS prediction of anticonvulsant activity was performed for the whole set of the designed molecules and probability to be active Pa values were ranging from 0.275 to 0.43. The studied compounds exhibit protection against pentylenetetrazole (PTZ) seizures, anti-thiosemicarbazides effect as well as some psychotropic effect. The biological assays evidenced that some of the studied compounds showed a high anticonvulsant activity by antagonism with pentylenetetrazole. The toxicity of compounds is low and they do not induce muscle relaxation in the studied doses. According to the study of psychotropic activity it was found that the selected compounds have an activating behavior and anxiolytic effects on the models of “open field” and “elevated plus maze” (EPM). The data obtained indicate the anxiolytic (anti-anxiety) activity of the derivatives of pyrimidines, especially pronounced in compounds 6n, 6b, and 7c. The studied compounds increase the latent time of first immobilization on the model of “forced swimming” (FST) and exhibit some antidepressant effect similarly to diazepam. Docking studies revealed that compound 6k bound tightly in the active site of GABAA receptor with a value of the scoring function that estimates free energy of binding (ΔG) at −7.95 kcal/mol, while compound 6n showed the best docking score and seems to be dual inhibitor of SERT transporter as well as 5-HT1A receptor. Conclusions: Тhe selected compounds have an anticonvulsant, activating behavior and anxiolytic effects, at the same time exhibit some antidepressant effect.  相似文献
