The reaction between p-(dimethylamino)benzaldehyde and 2-acylmethyl-1H-benzimidazoles and 6-amino-1,3-dimethylpyrimidino-2,4(1H,3H)-dione has given previously unknown 5-unsubstituted 6-(1H-benzimidazol-2-yl)pyrido[2,3-d]pyrimidino-2,4(1H,3H)-diones. A three component cyclocondensation of p-(dimethylamino)benzaldehyde with 3,5-dimethoxyaniline and 2-phenacyl-1H-benzimidazoles gives previously unknown 2-aryl-3-(1H-benzimidazol-2-yl)-5,7-dimethoxyquinolines. The Hantzsch three-component cyclization of 4-(dimethylamino)benzaldehyde, cyclohexane-1,3-diones, and some 1,3-[N,C]-dinucleophiles (5-amino-3-methyl-1-phenylpyrazole, 3,5-dimethoxyaniline, and 4-amino-1,3-dimethylpyrimidine-2,6-dione) proceeds in boiling acetic acid with the splitting off N,N-dimethylaniline and the formation of polycondensed heterocyclic sytems with a γ-unsubstituted pyridine ring (pyrazolo[3,4-b]quinoline, acridine, and pyrimido[4,5-b]quinoline). 3-Amino-4,6-dimethylpyrazolo[3,4-b]pyridine was used for the preparation of some novel heterocycles of pharmaceutical interest. The starting material reacted with 2-cyano-3,3-bis(methylthio)acrylate, chloroacetyl chloride, phenyl isothiocyanate, carbon disul-fide, and aromatic aldehydes to give the novel heterocycles. A series of 1-adamantylimidazo[1,2-a]pyridines has been synthesized. The negative influence of electron-acceptor substituents in the pyridine ring on the alkylation of substituted 2-aminopyridines with bromomethyl (adamantan-1-yl) ketone has been demonstrated. Bromination of adamantylimidazo-[1,2-a]pyridines on boiling in liquid bromine or in the presence of a Lewis acid resulted in insertion of a bromine atom in the imidazole ring without affecting the adamantane nucleus. 7-[1,3-Dithiolan-2-ylidene]-3-phenyl-5,6,7,8-tetrahydro[1,2,4]triazolo[3,4-b][1,3,4]thiadiazepine-6,8-dione and 7-[5-oxo-1,3-dithiolan-2-ylidene]-3-phenyl-5,6,7,8-tetrahydro[1,2,4]triazolo[3,4-b][1,3,4]-thiadiazepine-6,8-diones were obtained by treating 3-phenyl-5,6,7,8-tetrahydro[1,2,4]triazolo-[3,4-b][1,3,4]thiadiazepine-6,8-diones with CS2 and chloroacetyl chloride, respectively. Treatment of the above compounds with mercaptoacetic acid gave 1,2-dibromoethane or the corresponding spiro polyfused heterocycles. Three tetrahydrobenzo[e]pyrano[4,3-b]pyridines formed in a diethyl 2,4,6-trioxoheptanedicarboxylic ether-substituted salicylic aldehyde-ammonium acetate system were studied by single crystal X-ray diffraction. After the introduction of a methoxy group in the tricyclic system, the tetrahydropyridine and pyrane rings adopted the half-chair conformation, while in the unsubstituted compound, the tetrahydropyridine ring has a distorted boat conformation, and the pyrane ring has a C-envelope conformation. In the compounds, the N-H?O and O-H?O intermolecular hydrogen bonds give rise to the development of chain structures. In two of the three compounds examined, the hydrogen atoms at the chiral centers are in the trans-position, as in the structure of natural tetrahydrocannabinols.  相似文献
