Summary A colorimetric method for estimating hydroxyproline in the range 10–150 ng is described. Chloramine-T is used for the oxidation stage and good colour production with adequate chromogen stability is obtained.

Zusammenfassung Eine Methode zur kolorimetrischen Bestimmung von 10–150 ng Hydroxyprolin wurde beschrieben. Chloramin-T wird als Oxydationsmittel verwendet. Dabei entsteht eine gut meßbare und beständige Färbung.

     

Bestimmung einiger Aminosäuren unter Verwendung der Ringofenmethode

Zusammenfassung Die mit Hilfe der Ringofensegmenttechnik im Nanogrammbereich durchgeführten Bestimmungen der Aminosäuren Tryptophan, Prolin, Hydroxyprolin, Arginin, Histidin, Tyrosin, Cystein und Cystin wurden beschrieben. Die Fehler liegen innerhalb der bei der Ringofenmethode üblichen Grenzen.Die bereits früher entwickelte Segmenttechnik dient zur Bestimmung von Substanzen in all jenen Fällen, bei denen das Reaktionsprodukt einer zeitlichen Veränderung unterworfen ist. Dabei ist keine haltbare Standardskala notwendig.

Determination of some amino acids by means of the king oven

Summary Determinations of the amino acids tryptophane, proline, hydroxyproline, arginine, histidine, tyrosine, cysteine and cystine in the nanogram range with the aid of the ring oven segment technique are described. The range of error of these determinations is about the same as usual in ring colorimetry.The segment technique already earlier described serves for the determination of substances in cases, where the product of reaction is unstable; this is possible because a stable standard scale is not necessary.

Acylation of silylenol ethers1 and9 yield with 2-alkoxy-2-alkyl(or aryl)-1,3-dioxolanes5 in a simple way by zinc dichloride-diethyl ether-catalysis regioselectively the - and -protected dicarbonyl derivatives6,7, and10. The enhanced reactivity of the cyclic orthoesters5 in this reaction is discussed in comparison with acyclic reagents. The yield is influenced by steric effects at the reaction center.

Zusammenfassung Aus Isatosäureanhydriden erhält man durch Umsetzen mit 1,2-Dimethylhydrazin die entsprechenden 1,2-Dimethylanthranilsäurehydrazide, die sich mit Phosgen zu 1,3,4-Benzotriazepin-2,5-dionen cyclisieren lassen.

Synthesis of 1.3.4-benzotriazepine-2.5-diones

By reaction of isatoic anhydrides with 1.2-dimethylhydrazine the corresponding 1.2-dimethylanthranilic acid hydrazides are formed. The reaction of the latter with phosgene yields 1.3.4-benzotriazepine-2.5-diones.

Hydroxyproline quantification for the estimation of collagen in tissue using multiple reaction monitoring mass spectrometry

Collagens are highly abundant mammalian proteins that contain a high content of hydroxylated amino acids such as hydroxyproline. We have exploited the high hydroxyproline content of collagen and developed a method for hydroxyproline quantification as a measure of collagen content in muscle samples. The novel method utilizes a highly selective and sensitive method of multiple reaction monitoring (MRM) by mass spectrometry. The analytical method is simple, rapid (5min), convenient (no derivatization), precise (<17% RSD), accurate (90-108%), sensitive (4.88nmol/L) and linear (R(2)>0.999) over three orders of magnitude (5-5000nmol/L).

Syntheses of the title compound and of some of its derivatives are described. Homocamphenilanic acid could not be obtained by aDiels-Alder reaction of cyclohexadiene with an appropriate dienophile but is easily accessible via homocamphene by hydroboration of this homoterpene and subsequent oxidation of the reaction product.

Cathodic reduction of SO2 in the presence of organic dihalides

The reaction of the electrolytically generated SO ₂ ^– anion radical with 1,-dihalides is used for the synthesis of sulfur containing heterocycles like oxathiolane-, oxathiane-, thiane- and thiepane-oxides. 1,2-Dihalides are reductively deblocked to give olefines.

Zusammenfassung Die Reaktion des elektrolytisch erzeugten SO ₂ ^– -Anionradikals mit 1,-Dihalogeniden führt zu schwefelhaltigen Heterocyclen wie Oxathiolan-, Oxathian-, Thian- und Thiepanoxiden. 1,2-Dihalogenide geben durch reduktive Eliminierung Olefine.

     

Die Farbreaktion von Arylhydrazinen nach Postowsky

Zusammenfassung Es wurde bewiesen, daß bei der Farbreaktion von Arylhydrazinen mit Selendioxid und 1-Naphthylamin die entsprechenden Diazoniurasalze gebildet werden. Diese wurden nach der Oxydation mit SeO₂ papierchromatographisch nachgewiesen und parallel wurden die gleichen Diazoniumsalze, die durch Diazotierung der betreffenden Amine dargestellt wurden, chromatographiert. Auch die nach der Reaktion mit 1-Naphthylamin gebildeten Azofarbstoffe wurden dünnschichtchromatographisch verglichen. Bei 18 Arylhydrazinen war die Farbreaktion positiv, eine negative Reaktion gaben N-Acetyl- und N-Formyl-N-phenylhydrazin, N,N-Diphenyl-hydrazin, p-Nitrophenylhydrazin und das 2,4-Dinitrophenylhydrazin.

The colour reaction of arylbydrazines according to Postowsky

Summary Paper chromatography was used to proof that diazonium salts are formed during the oxidation of arylhydrazines with SeO₂. They were identified by comparison with those obtained by diazotization of the corresponding amines. The azo dyes formed after the reaction with 1-naph-thylamine were compared by thin-layer chromatography. Eighteen tested arylhydrazines gave a positive colour reaction. A negative result was obtained with N-acetyl- and N-formyl-N-phenylhydrazines, N,N-diphenyl-hydrazine, p-nitrophenylhydrazine and 2,4-dinitrophenylhydrazine.

Application of fluorescence reactions in the ring oven segment technique

Summary The ring oven segment technique already described earlier has been applied to the determination of ng- tog-amounts of organic and inorganic substances using fluorescence reactions. The methods proposed here are based either on self-fluorescence or on appearance or disappearance of fluorescence following a chemical reaction.

Zusammenfassung Die bereits früher beschriebene Ringofen-Segmenttechnik wurde auf die Bestimmung organischer und anorganischer Substanzen im ng- bisg-Bereich mit Hilfe von Fluoreszenzreaktionen angewendet. Fluorimetrische Reaktionsdetektoren zur HPLC-Analyse von Steroiden

Zusammenfassung Acht bekannte Umsetzungsmöglichkeiten von Steroiden zu fluoreszierenden Derivaten wurden auf automatische, luftsegmentierte Reaktionssysteme übertragen. Die Reaktionen mit Isonicotinoylhydrazid sowie mit Periodat/Acetylaceton lassen sich auch in Verbindung mit HPLC-Trennungen in einem Reaktionsdetektor unter Verwendung von Teflonschläuchen einsetzen. Die Nachweisgrenzen für das Corticosteron liegen bei 7 bzw. 100 ng im Reaktionssystem. Die fluorimetrische Bestimmung mehrerer Steroide in Urinextrakten nach Trennung an einer chemisch-gebundenen Nitrophase mit dem Isonicotinoylhydrazid-Reaktionsdetektor wird gezeigt.

Fluorimetric reaction detectors for the HPLC analysis of steroids

Summary Eight known reaction possibilities for steroids to fluorescent derivatives were transferred to automatic, air-segmented reaction systems. The reactions with isonicotinoylhydrazide and with periodate/acetylacetone under application of teflon tubes are suitable for application in a reaction detector in connection with HPLC separations. The determination limits for corticosterone are 7 resp. 100 ng in the reaction system. The fluorimetric determination of some steroids in urine extracts after separation on a chemical-bonded nitrophase by the isonicotinoylhydrazide reaction detector is demonstrated.

     

Combustion of mixtures containing NaClO4, S and carbon

The thermoanalytical techniques e.g. DSC, TG/DTG and Hot Stage Microscopy have been used to examine the system of NaClO₄, S and charcoal mixtures. The results show that there is no reaction between NaClO₄ and S alone, but the presence of charcoal activates their reaction. Charcoal is oxidised by NaClO₄ prior to the melting of NaClO₄, showing a solid phase reaction. The presence of S activates the whole of the reaction to such a level that the mixture burns around 300C.

Zusammenfassung Mittels DSC, TG/DTG und HSM wurde das System von Gemischen aus NaClO₄, S und Holzkohle untersucht. Die Ergebnisse zeigen, da\ sich zwischen NaClO₄ und S allein keine Reaktion abspielt, da\ diese Reaktion aber durch Gegenwart von Holzkohle aktiviert wird. Noch vor dem Schmelzen von NaClO₄ wird Holzkohle in einer Feststoffreaktion von NaClO₄ oxydiert. Die Gegenwart von S aktiviert die gesamte Reaktion in dem Ma\e, da\ das Gemisch bei 300C verbrennt.

1H and13C NMR studies on lanthanide complexes with proline and hydroxyproline

The formation and the mode of coordination of rare earth (Y, La, Pr, Nd, Sm, Eu, Gd, Tb, Dy, Ho, Er, Tm, Lu) complexes with proline and hydroxyproline have been investigated by¹H and¹³C NMR spectral techniques. It has been established that the nitrogen and the carboxyl group of the ligands are involved in complexation, and that the OH^– group of hydroxyproline does not participate in coordination.

¹H und¹³C NMR Untersuchungen an Lanthanid-Komplexen mit Prolin und Hydroxyprolin

Zusammenfassung Die Bildung und die Koordination von seltenen Erden (Y, La, Pr, Nd, Sm, Eu, Gd, Tb, Dy, Ho, Er, Tm, Lu) in Komplexen mit Prolin und Hydroxyprolin wurden mit Hilfe von¹H- und¹³C-NMR-Spektroskopie untersucht. Es wurde festgestellt, daß in den Komplexen der Stickstoff und der Carboxylsauerstoff der Liganden koordinieren. Die OH^–-Gruppe von Hydroxyprolin nimmt keinen Anteil an der Koordinierung.

     

Über die umsetzung von N-fluorsulfonylisocyanat mit verschiedenen phosphorverbindungen

Zusammenfassung N-Fluorsulfonylisocyanat bildet mit Triphenylphosphin, Phosphorigsäuretrisdimethylamid und Phosphorigsäuretriäthylester 11-Addukte unterschiedlicher Stabilität. Die Bildung von N-Fluorsulfonylcarbodiimiden aus N-Fluorsulfonylisocyanat und dem Phospholenoxid 7 gelingt nicht, da die Reaktion auf der Stufe des N-Fluorsulfonylphospholenimins stehenbleibt.

Reaction of N-fluorosulfonyl isocyanate with phosphorus compounds

11-adducts of different stability are the products of the interaction of N-fluorosulfonylisocyanate with triphenylphosphine, phosphorous acid tris(dimethylamide), and triethyl phosphite. No N-fluorosulfonylcarbodiimides are formed from N-fluorosulfonylisocyanate and phospholene oxide (7), since the reaction does not proceed beyond the intermediate N-fluorosulfonylphospholene imine.

     

Spectrophotometric determination of some steroid sex-hormones

Summary A detailed spectrophotometric study of the reaction of estrogens and androgens with 98% sulphuric acid at 20° C and of the effect of various dilutions with water on the nature of the spectra is presented. Dilution of the androgens-acid and estrogens-acid solutions with water to 40% and 80% v/v acid, respectively, increases the absorption intensity 3 to 11 times. The optimum conditions for determination of down to 2g/ml of estrone, estradiol, ethinylestradiol, testosterone, and rostanalone and progesterone are reported. The results show a relative standard deviation of +-1.5 %.

Spektralphotometrische Bestimmung einiger Steroid-Sexualhormone

Zusammenfassung Die Reaktion der Östrogene und Androgene mit 98%iger Schwefelsäure bei 20° C und der Einfluß von Verdünnungen. mit Wasser auf die Art der Spektren wurden eingehend untersucht. Verdünnungen auf einen Säuregehalt von 40% und 80% bewirken eine 3–11fache Zunahme der Absorption. Die optimalen Bedingungen zur Bestimmung von >2g/ml Östron, Östradiol, Äthinylöstradiol, Testosteron, Androstanalon und Progesteron wurden ausgearbeitet. Die relative Standardabweichung beträgt+-1,5%.
