Copper complexes of chiral iminopyridines prepared from camphane-derived ketones and picolylamine catalyzed the enantioselective Henry (nitroaldol) reaction between nitromethane and a number of aromatic and aliphatic aldehydes with high yields and good enantioselectivities. Iminopyridines derived from (1R)-(+)-camphor and (1S)-(+)-ketopinic acid gave the best results to afford the opposite enantiomers in each case, despite the fact they have the same stereochemical pattern at the camphane skeleton. The reactions were carried out without air or moisture exclusion.  相似文献

An enantioselective Henry reaction was efficiently carried out under mild reaction conditions in the presence of catalytic 9-epi and natural Cinchona alkaloids and copper (II) acetate. The best catalytic performance was observed for native quinine (12 mol %) and Cu(OAc)2 (10 mol %). Aromatic and aliphatic aldehydes with nitromethane and its α-substituted derivatives provided the corresponding β-nitroalcohols in good to reasonable yields, high syn-diastereoselectivity, and (S)-enantioselectivity of up to 94% ee.  相似文献

A series of chiral tridentate Schiff-base ligands and their polymer-supported ligands were conveniently prepared and introduced as copper(II) chiral complexes for the asymmetric Henry reaction. The structures of these ligands have been characterized by IR, 1H NMR, 13C NMR and MS. The experimental results showed that the corresponding β-nitro alcohols were obtained in moderate to high yields (up to 98%) with up to 98% ee under mild conditions. The complex catalyst forming from copper(II) polymer-supported ligands could be recycled by a simple filtration and reused 6 times at least with similar good catalytic effect (about 94% yield and 90% ee).  相似文献

Enantiopure macrocyclic ligands were synthesized from (1R,2R)‐(+)‐ and (1S,2S)‐(?)‐diphenylethylenediamine with 3,3'‐methylenebis(5‐(tert‐butyl)‐2‐hydroxybenzaldehyde) and characterized. The chirality transfer and chiral inversion from ligand to copper(II) metal centre were studied using circular dichroism spectroscopy. The enantiopure binuclear copper(II) complexes (ΔΔ and ΛΛ) were used as catalysts for asymmetric nitroaldol reaction to generate β‐nitroalcohol with 88% yield and 67% enantioselectivity. A family of enantiopure β‐aminoalcohols based on aziridine backbones were synthesized, and examined as chiral ligands for the copper(II)‐catalyzed asymmetric Henry reaction of aromatic aldehydes with nitromethane, giving β‐nitroalcohols in excellent yields (up to 93%) and moderate to good enantioselectivities (up to 82%). Ferrocenyl-substituted aziridinylmethanol (Fam-1) was used as a catalyst with zinc for the asymmetric nitroaldol (Henry) reaction. This catalyst worked with a variety of aldehydes (aromatic, aliphatic, alpha,beta-unsaturated, and heteroaromatic) and alpha-ketoesters to give the nitroaldol product in up to 97% yield and 91% ee. The chiral ligand can be recovered and recycled without losing its activity.  相似文献

A new chiral N,N'-dioxide-Cu(I) catalyst has been developed for the asymmetric Henry (nitroaldol) reaction. The approach benefited from the easy modification of the chiral space. As the highly effective N-oxide ligand, 1d has been adopted for the Henry reaction with both aromatic and heteroaromatic aldehydes. The corresponding nitro-alcohol products were obtained in good yields with high enantiomeric excesses up to 98%. Moreover, alpha-ketoesters were also catalyzed by this catalyst to give attractive optically active alpha-hydroxy beta-nitro esters containing chiral quaternary carbon centers (up to 99% ee). On the basis of a combination of several techniques including the 1H NMR, ESI-HRMS, and MM2 calculations, the proposed mechanism was presented to explain the origin of reactivity and asymmetric inductivity.  相似文献

Tributyltin hydride-mediated cyclizations of 1-nitro-2-acetoxy-5-hexenes 7a-g having multiple substitutions on carbons 1 and 6 result in 2,3-substituted-1-acetoxycyclopentanes 1a-g. The substrates were prepared by nitroaldol reactions of silyloxyaldehydes followed by acetylation, desilylation, and oxidation to the acetoxynitroaldehydes 6a-e. Wittig olefination of aldehydes 6a-e then afforded substrates for the radical cyclizations. The overall scheme gave a diverse array of cyclopentanes, including gem-disubstituted cyclopentanes having substitution on three contiguous carbons.  相似文献

Phosphonium ionic liquids exchanged with bicarbonate and methylcarbonate anions (CILs) exhibit catalytic performances comparable to those of sterically hindered (non nucleophilic) organosuperbases such as DBU. At 25-50 °C, under solventless conditions, CILs efficiently catalyze the Henry addition of different aldehydes and ketones to nitroalkanes: not only they allow the selective formation of nitroaldols but they unlock a novel high-yielding access to dinitromethyl derivatives of ketones.  相似文献

The development of highly enantioselective and general catalytic nitroaldol (Henry) reactions with ketones is a challenging yet desirable task in organic synthesis. In this communication, we report an asymmetric nitroaldol reaction with alpha-ketoesters catalyzed by a new C6'-OH cinchona alkaloid catalyst. This is the first highly efficient organocatalytic asymmetric Henry reaction with ketones. This reaction is operationally simple and affords high enantioselectivity as well as good to excellent yield for a broad range of alpha-ketoesters.  相似文献

Ten optically pure substituted 2-(pyridin-2-yl)imidazolidin-4-ones, 1a-d, 2a-4a, and 2b-4b, were prepared and characterized. The absolute configurations of individual ligands were determined by X-ray analysis or NOESY experiments. The Cu(II) complexes of the respective ligands were studied as enantioselective catalysts of the nitroaldol (Henry) reaction of aldehydes with nitromethane, giving the corresponding substituted 2-nitroalkanols. In the case of an anti arrangement of the imidazolidin-4-one ring, the obtained result was 91-96% ee, whereas in the case of syn arrangement, a significant drop to 25-27% ee was observed.  相似文献

A small library of C(1)-symmetric chiral diamines (L1-L9) was constructed via condensing exo-(-)-bornylamine or (+)-(1S,2S,5R)-menthylamine with various Cbz-protected amino acids. Among them, ligand L1/CuCl(2)·2H(2)O complex (2.5 mol %) shows outstanding catalytic efficiency for Henry reaction between a variety of aldehydes and nitroalkanes to afford the expected products in high yields (up to 98%) with excellent enantioselectivities (up to 99%) and moderate to good diastereoselectivities (up to 90:10). This process is air- and moisture tolerant and has been applied to the synthesis of (S)-2-amino-1-(3,4-dimethoxyphenyl)ethanol (9), a key intermediate for (S)-epinephrine and (S)-norepinephrine. On the basis of HRMS and X-ray diffraction analysis of the L1/CuCl(2) complex, a transition-state model was proposed to explain the origin of asymmetric induction. The low catalyst loading, excellent yields and enantioselectivities, inexpensive copper salt, and mild reaction conditions make our catalytic system to be practically useful.  相似文献

We report the mechanism of asymmetric nitroaldol (Henry) reaction catalyzed by a dinuclear Zn complex using density functional theory. The experimentally proposed catalytic cycle is validated, in which the first step is the deprotonation of nitromethane by the ethyl anion of the catalyst, subsequently a C? C bond formation step, and then the protonation of the resulting alkoxide. Three mechanistic scenarios (differing in binding modes) have been considered for the C? C bond formation step. The origin of the enantioselectivity is discussed.
