羟丙基三甲基氯化铵壳聚糖制备的可控性研究   总被引:5,自引:0,他引:5  
用壳聚糖与缩水甘油三甲基氯化铵反应制备羟丙基三甲基氯化铵壳聚糖,所得产物的结构受壳聚糖分子量和脱乙酰度、反应温度、反应时间、壳聚糖与缩水甘油三甲基氯化铵投料比的影响。实验结果表明:随着反应温度升高,羟丙基三甲基氯化铵壳聚糖的取代度增加,在70~80℃达最大;反应时间增加,取代度增加,产物分子量降低。缩水甘油三甲基氯化铵与壳聚糖比例达3:1前取代度随比例升高而增加。脱乙酰度和分子量越大的壳聚糖其季铵盐取代度越高。控制反应温度在30~90℃,反应时间3~10h,投抖比为1:1~4:1,可以得到取代度15%~909/6,分子量1万到100万的羟丙基三甲基氯化铵壳聚糖。  相似文献

The emulsification of sunflower oil by chitosan solutions with deacetylation degrees (DD) between 73 and 95% was studied using different techniques. The droplet size distributions, conductivity, ageing behavior and viscosity of emulsions were studied as functions of the chitosan DD. All DD gave stable polydisperse water-in-oil-in-water emulsions with different viscosities. Two optimum DD values were found, 81 and 88%, giving complete emulsification without residual oil or sedimentation. Chitosans with intermediate DD were less effective emulsifiers. 建立了一种基于光纤折射率传感技术的壳聚糖脱乙酰度测定方法. 利用光纤折射率传感器监测酸碱滴定过程中溶液折射率的变化, 根据折射率变化转折点之间碱的用量来计算壳聚糖的脱乙酰度. 该方法测得的3种不同含量实际样品的脱乙酰度与氢核磁共振波谱(1H NMR)方法测定结果相符, 验证了方法的可靠性. 该方法具有用量少、 结构简单、 准确、 重复性好和转折点明显等优点, 可应用于工业生产中壳聚糖脱乙酰度的测定.  相似文献

研究了酸碱滴定中壳聚糖溶液电导的变化,利用电导变化两个转折点之间碱的用量来计算壳聚糖的脱乙酰度,消除了壳聚糖吸附的残留酸或碱的影响。该方法具有准确可靠、重复性好、转折点明显等优点。用于实际样品的测定,结果较好,相对标准偏差小于0.6%。  相似文献

Chitosans having three different degrees of acetylation (DA) were studied in acid solution using the uranyl staining technique and electron microscopy. Strings of approximately spherical aggregates were seen. The aggregates were interpreted as micelle-like agglomerates formed by almost fully acetylated polysaccharide, interconnected by blocks of almost fully deacetylated polysaccharide stretched by electrostatic repulsion. These agglomerates include NH3+ groups which produce electrostatic swelling of the agglomerates, giving a radius proportional to the degree of deacetylation. The length of the strings are also proportional to the deacetylation degree. These strings are extended because of the electrostatic repulsion between charged ammonium groups.  相似文献

A purified acetyl esterase (AE), isolated from a commercial enzyme preparation, released acetic acid from water-soluble and water-insoluble cellulose acetates (CAs), native and chemically acetylated xylan as well as acetylated starch. The AE specifically cleaved off the acetyl substituents from the C2- and C3-positions from CAs of DS <1.8 and left the acetyl substituents at the C6-positions intact without degrading the polysaccharide. The activity of endoglucanase was enhanced by the presence of acetyl esterase, while the acetyl esterase derived no advantage from the presence of the endoglucanase; it was able to function independently.  相似文献

Inhibition of radial growth and spore germination of Aspergillus niger in media with added chitosan were detected. The highest radial growth inhibition (73%) was determined at 24 h with 3 g · L?1 of chitosan, and the percent inhibition of spore germination was 40% after 13 h of inoculation. Further, the CC50, that is, the concentration at which spore germination is inhibited by 50%, was estimated by probit analysis (3.5 g · L?1). The activation energies, EA were estimated by an Arrhenius model in control and amended chitosan media, obtaining 35.6 and 36.6 kcal · mol?1, respectively. These values were in the same order of magnitude because chitosan as inhibitor was more effective at low temperature (≤ 18 °C). Hence synergism of temperature and chitosan were only observed at 12 and 18 °C. Therefore, the maximal percentage of germinated spores, Smax was also affected by low temperatures in chitosan‐amended media with estimated values lower than 70% at temperatures < 37 °C whereas in control media Smax reached values close to 100%. Scanning electron micrographs showed that chitosan produced spore aggregation and morphological anomalies affecting swelling, germ tube emergence, and polarization.

用NaOH作为催化剂, 在异丙醇悬浮体系中环氧丙烷(PO)与壳聚糖(CS)在60 ℃下反应8 h, 制备取代度超过0.8的羟丙基壳聚糖(HPCS). HPCS在水溶液中与氯乙酸反应, 制备了一种结构新颖的两性聚合物N,O-羧甲基化羟丙基壳聚糖(HPCMS), 羧甲基取代度可控制在0.42~1.38之间. 采用NMR和FTIR对产物结构进行表征. 结果表明, 在壳聚糖的羟丙基化改性过程中, C6位羟基首先与环氧丙烷反应, 生成HPCS; 在与氯乙酸反应过程中, HPCS上的羟基和氨基同时与氯乙酸发生取代反应.  相似文献

研究了吸附-絮凝耦合的方法固定脂肪酶的工艺条件.结果表明:在33℃下,用0.03mol/L的磷酸二氢钾?氢氧化钠缓冲液控制体系pH为7.0,酶与树脂(质量比1∶8)作用吸附1h后,用0.2mL絮凝剂聚丙烯酰胺(w(PAM)=0.5%)处理,得到活力较高的固定化脂肪酶.固定化酶最适pH9.0,最适温度为45℃,活力为405U/g,酶活回收率可以达到40%.固定化脂肪酶制备简便,可重复使用,稳定性较高.  相似文献

The abundant biopolymer chitin, found mainly in crustaceous exoskeleton, such as crab, shrimp and lobster, can be deacetylated to yield chitosan. This slightly different biopolymer is more reactive than chitin, being more effective for many applications in fields as environmental remediation, biomedical sciences, catalysis and so on. The main process for chitin deacetylation used sodium hydroxide solutions at high temperatures for long times to obtain chitosan with high deacetylation degree (DD). The present study has evaluated the effect from room temperature (RT), 363 and 393 K, hydroxide concentration (2.0 or 10.0 mol dm3) and time (3 and 24 h) on shrimp chitin deacetylation. Similar amounts of chitin and sodium hydroxide solutions were stirred jointly and the resultant solids were filtered and washed until pH 7, than dried at environmental conditions. The obtained samples were characterized by several techniques, such as elemental analysis, X-rays diffraction (XRD), laser scattering and absorption spectroscopy at infrared region with Fourier transform (FTIR), which was used for DD calculation. The results showed that all chitin-chitosan samples did not reach DD > 90%, as observed for some good commercial chitosans. The highest DD was obtained by the sample prepared at more drastic conditions, as expected, however the higher sodium hydroxide concentration leads to decrease of molecular mass when associated with high temperatures. The crystallinity was influenced mostly by reaction time, which change the positions and intensities as indicated by XRD main peaks, located at 9.3 and 19.4° 2Θ. Particle sizes were strongly diminished by treatment at 393 K, what imply also some increase at the pressure, favoring chain dissociation reactions. This work mapped several properties for chitin-chitosan samples achieved by the described conditions.  相似文献

研究了以刚果红分光光度法测定壳聚糖,探讨了最佳实验条件及干扰因素的影响。在pH9.7时,刚果红和壳聚糖形成有色物质,检测波长为543.5nm,线性方程为:A=0.0200ρ+0.0156,R^2=0.9978。在0~20.00μg/mL范围内呈现良好的线性关系,平均回收率为99.20%。采用该方法测定了两种复方样品中的壳聚糖,方法的选择性和灵敏度良好。该方法可用于检测复方样品中少量的壳聚糖。  相似文献

The contents of the exoskeleton of Parapenaeus longirostris from Moroccan local sources were analyzed and the percentages of inorganic salt, protein, lipid, and chitin were determined. Chitin in the α form was extracted from Parapenaeus longirostris shells by 0.25 M HCl and 1 M NaOH treatment for demineralization and deproteinization, respectively. The obtained chitin was converted into the more useful soluble chitosan. The chemical structure and physico-chemical properties of chitin and chitosan were characterized using Fourier transform-infrared (FT-IR) spectroscopy, nuclear magnetic resonance (NMR) spectroscopy, X-ray diffractometry (XRD), and scanning electron microscopy (SEM). The molecular weight (MW) of chitosan was determined by viscometric methods. The degree of acetylation (DA) of chitin and chitosan was determined by the 1H NMR technique. To the best of our knowledge this is the first report on the extraction and characterization of chitin and chitosan from Parapenaeus longirostris.  相似文献

对黑曲霉木聚糖酶同工酶的酶学性质及圆二色谱(CD)进行了研究.结果表明,黑曲霉木聚糖酶粗酶液经离心、超滤及SephadexG-75凝胶层析后,分离得到了两个黑曲霉木聚糖酶同工酶组分XⅠ,XⅡ,其最适反应温度均为45℃~55℃,最佳反应pH值均为4 8~5 5,SDS-PAGE法测得XⅠ,XⅡ的相对分子质量分别为2.2×104和4.0×104.XⅠ与XⅡ的CD光谱表明,XⅠ,XⅡ分别在218和215nm处有最大负峰,说明β-折叠结构是组分XⅠ,XⅡ具有木聚糖酶活性的主要结构,也是它们发挥催化功能的结构基础.  相似文献

Abstract

Graphite furnace atomic absorption spectroscopic (GFAAS) analysis of oil samples from the onshore and offshore fields in the Niger Delta was carried out in order to determine their trace element contents. The concentrations of the elements (Cr, Co, Cu, Fe, Mn, Ni, Pb, and V) determined ranged from 0.151–941.199 ppb with an average of 30.190 ppb (% RSD<5) for the oil samples analyzed. The pattern of occurrence of each element was in agreement with studies conducted employing neutron activation analysis and flame atomic absorption spectroscopy. From the cluster analysis of the data, two groups of oils were evident. The groups are correlated with each other, indicating common or similar genetic origin. Trace metal parameters indicate that the oils were generated from organic matter of strong terrestrial input. GFAAS proved to be a good method for the analysis of elemental composition and classification of Niger Delta oils.  相似文献

Abstract

Twenty four extracts from Bignoniaceae plants of northwest Argentina were tested for antifungal activity against Aspergillus species responsible of the grape black rot. Stems and leaves of Amphilophium cynanchoides, Macfadyena cynanchoides, Tecoma stans and Jacaranda mimosifolia were separately extracted with solvents of increasing polarity to obtain the dichloromethane (fCH2Cl2), ethyl acetate (fEtOAc) and methanol extracts (fMeOH). The fCH2Cl2 from stem of M. cynanchoides had the lowest IC50 (1.0–1.2?mg/mL) and MID values (0.6–1.2?mg) and the highest ID values (5.0–6.8?mm) on A. niger and A. carbonarius. The main contributors of the antifungal activity of fCH2Cl2 were identified as lapachol (MIC?=?0.25–1.00?mg/ml) and 1-hydroxy-4-methylanthraquinone (MIC?=?0.0625–0.125?mg/mL). These compounds synergized the antifungal activity of sodium metabisulfite and showed an additive effect in mixtures with propiconazol. They might be used as additives of commercial antifungals to protect grapes against A. niger and A. carbonarius.  相似文献
