A new an innovative separation method has been developed using N-n-octylaniline (liquid anion exchanger) coated on silica gel. Ruthenium(III) has been extracted quantitatively from 0.05 M hydrochloric acid, striped with aqueous solution of various acids and then determined by spectrophotometric method. Mutual separation scheme for ruthenium(III), iridium(III) and osmium(VIII) has been developed. Ruthenium(III) has been separated from commonly associated elements. Different parameter are studied viz effect of acid concentration, reagent concentration, striping agent, flow rate, foreign ion and synthetic mixture corresponding to alloys. Validity of method has been verified by separation of ruthenium(III) from fissium alloy.  相似文献

In recent years, materials science has propelled to the research forefront. Ionic liquids with unique and fascinating properties have also left their footprints to the developments of materials science during the last years. In this review we highlight some of their recent advances and provide an overview at the current status of ionic liquid-modified materials applied in solid-phase extraction, liquid and gas chromatography and capillary electrochromatography with reference to recent applications. In addition, the potential of ionic liquids in the modification of capillary inner wall in capillary electrophoresis is demonstrated. The main target material modified with ionic liquids is silica, but polymers and monoliths have recently joined the studies. Although imidazolium is still clearly the most commonly used ionic liquid for the covalently modification of materials, the exploitation of pyridinium and phosphonium will most probably increase in the future.  相似文献

In countercurrent chromatography (CCC) both stationary and mobile liquids undergo intense mixing in the variable force field of a coil planet centrifuge and the separation process, like the separation in conventional solvent extraction column, is influenced by longitudinal mixing in the phases and mass transfer between them. This paper describes how the residence time distribution (or the elution profile) of a solute in CCC devices and the interpretation of experimental peaks, can be described by a recently developed cell model of longitudinal mixing. The model considers a CCC column as a cascade of perfectly mixed equal-size cells, the number of which is determined by the rates of longitudinal mixing in the stationary and mobile phases. Experiments were carried out to demonstrate the validation of the model and the possibility of predicting the partitioning behaviour of the solutes. The methods for estimating model parameters are discussed. Longitudinal mixing rates in stationary and mobile phases have been experimentally determined and experimental elution profiles are compared with simulated peaks. It is shown that using the cell model the peak shape for a solute with a given distribution constant can be predicted from experimental data on other solutes.  相似文献

The surface derivatization of poly(p-phenylene terephthalamide) fiber was studied. The obtained surface-derivatized filaments were packed into a fused-silica capillary to evaluate its surface characteristics by using GC. As the stationary phase for GC the surface-derivatized fibers showed higher retention for alkanes and alkylbenzenes than that with the untreated Kevlar fiber. The improvements on the retention power and the peak shape were observed on the benzyl-modified fibrous stationary phase. The derivatized fibrous materials were also evaluated as the extraction medium in fiber-in-tube SPE, and the effect of the surface modification on the extraction power was compared to the parent fiber. The results indicated that the modified fiber possessed a higher extraction power than the untreated fiber. Based on the facts, the successful modification of the fiber surface was estimated.  相似文献

Cloud point extraction (CPE) was used to extract and separate lanthanum(III) and gadolinium(III) nitrate from an aqueous solution. The methodology used is based on the formation of lanthanide(III)-8-hydroxyquinoline (8-HQ) complexes soluble in a micellar phase of non-ionic surfactant. The lanthanide(III) complexes are then extracted into the surfactant-rich phase at a temperature above the cloud point temperature (CPT). The structure of the non-ionic surfactant, and the chelating agent-metal molar ratio are identified as factors determining the extraction efficiency and selectivity. In an aqueous solution containing equimolar concentrations of La(III) and Gd(III), extraction efficiency for Gd(III) can reach 96% with a Gd(III)/La(III) selectivity higher than 30 using Triton X-114. Under those conditions, a Gd(III) decontamination factor of 50 is obtained.  相似文献

Cyclitols and sugars were obtained as a mixture from Medicago sativa L., in a comparative study by using maceration, and pressurized liquid extraction, as a modern and green extraction techniques. The influence of extraction parameters including: extraction temperature, time and number of cycles on the content of sugars and cyclitols was investigated based on response surface methodology. The highest total amount of sugars and cyclitols (62.27 ± 2.30 and 50.35 ± 0.77 mg/g of dry material, respectively) was obtained when extraction was performed at 88°C, for 22 min, in two cycles. The methodology used involved extraction, purification, selective separation (using yeast and anion exchange resin) and derivatization, followed by gas chromatography ‐mass spectrometry analysis. The use of yeast treatment realized an effective fractionation of cyclitols and sugars, which allowed the removal of most sugars. The involvement of anion exchange resin after yeast allowed the removal of sugar alcohols and lactose, together with other sugar traces remained and to obtain a solution containing six cyclitols. The recrystallization of dry residue after solvent evaporation, from ethanol, allowed us to obtain 14.65 mg of white pure crystals identified with NMR spectroscopy, liquid chromatography with mass spectrometry, gas chromatography with mass spectrometry, optical rotation and melting point as analysis D‐pinitol.  相似文献

Implementation of an uncomplicated SPE process for the rapid extraction and preconcentration of the alkaloids, colchicine, strychnine, aconitine, and nicotine, from water, apple juice, and nonfat milk samples is presented. When coupled to analysis via micellar EKC (MEKC), the total analysis time per sample was less than 15 min for the water and juice samples and less than 20 min for the milk. The SPE process allowed for anywhere from a three to a fourteen-fold improvement in the LOD for each alkaloid when compared to detecting the alkaloids in a nontreated water sample matrix. Following SPE, the LODs for colchicine, strychnine, and nicotine were sufficient to meet levels from 150 to 5000 times more dilute than the LD(50) for a 50 kg individual drinking 12 oz of a contaminated beverage. Aconitine, on the other hand, was detected at approximately the LD(50) level. The percent recoveries for the SPE ranged from 37% to as high as 99%. Nicotine attained the highest recovery efficiencies, followed by colchicine, and finally, aconitine and strychnine, which were nearly identical. The greatest recovery efficiencies were achieved from apple juice and water, whereas nonfat milk yielded the lowest.  相似文献

Countercurrent chromatography (CCC) is a separation technique in which two immiscible liquid phases are used for the preparative purification of synthetic and natural products. In CCC the number of repetitive mixing and de-mixing processes, the retention of the stationary phase and the mass transfer between the liquid phases are significant parameters that influence the resolution and separation efficiency. Limited mass transfer is the main reason for peak broadening and a low number of theoretical plates along with impaired peak resolution in CCC. Hence, technical improvements with regard to column design and tubing modifications is an important aspect to enhance mixing and mass transfer.  相似文献

Summary A semi-automatic reversed-phase HPLC system is described for the direct determination of conjugated and unconjugated bilirubin in human plasma. An Advanced Automated Sample Processor (AASP) is used for sample preparation, and for controlling sample injection and elution. The new sequential centrifugal layer chromatography extends the possibilities for preparative separation of furocoumarin isomers. In the present work, isobergapten, pimpinellin, bergapten, isopimpinellin, and sphondin were isolated from a prepurified extract of Heracleum sphondylium roots using two different techniques, namely, centrifugal layer chromatography (CLC) and sequential centrifugal layer chromatography (SCLC). The former technique was carried out with four plates, whereas the latter method required only one plate, using the same mobile phase in each case. A further advantage of the SCLC method was that all completely separated compounds could be quickly and efficiently eluted in small volumes of mehtanol.  相似文献

A surface plasmon resonance (SPR) biosensor screening assay was developed and validated to detect 11 benzimidazole carbamate (BZT) veterinary drug residues in milk. The polyclonal antibody used was raised in sheep against a methyl 5(6)-[(carboxypentyl)-thio]-2-benzimidazole carbamate protein conjugate. A sample preparation procedure was developed using a modified QuEChERS method. BZT residues were extracted from milk using liquid extraction/partition with a dispersive solid phase extraction clean-up step. The assay was validated in accordance with the performance criteria described in 2002/657/EC. The limit of detection of the assay was calculated from the analysis of 20 known negative milk samples to be 2.7 μg kg−1. The detection capability (CCβ) of the assay was determined to be 5 μg kg−1 for 11 benzimidazole residues and the mean recovery of analytes was in the range 81-116%. A comparison was made between the SPR-biosensor and UPLC-MS/MS analyses of milk samples (n = 26) taken from cows treated different benzimidazole products, demonstrating the SPR-biosensor assay to be fit for purpose.  相似文献

A simple method was developed to separate Pu and Am using single column extraction chromatography employing N,N,N′,N′-tetra-n-octyldiglycolamide (DGA) resin. Isotope dilution measurements of Am and Pu were performed using accelerator mass spectrometry (AMS) and alpha spectrometry. For maximum adsorption Pu was stabilized in the tetra valent oxidation state in 8 M HNO3 with 0.05 M NaNO2 before loading the sample onto the resin. Am(III) was adsorbed also onto the resin from concentrated HNO3, and desorbed with 0.1 M HCl while keeping the Pu adsorbed. The on-column reduction of Pu(IV) to Pu(III) with 0.02 M TiCl3 facilitated the complete desorption of Pu. Interferences (e.g. Ca2+, Fe3+) were washed off from the resin bed with excess HNO3. Using NdF3, micro-precipitates of the separated isotopes were prepared for analysis by both AMS and alpha spectrometry. The recovery was 97.7 ± 5.3% and 95.5 ± 4.6% for 241Am and 242Pu respectively in reagents without a matrix. The recoveries of the same isotopes were 99.1 ± 6.0 and 96.8 ± 5.3% respectively in garden soil. The robustness of the method was validated using certified reference materials (IAEA 384 and IAEA 385). The measurements agree with the certified values over a range of about 1–100 Bq kg−1. The single column separation of Pu and Am saves reagents, separation time, and cost.  相似文献

建立了酚法提取-二维液相色谱分离-高分辨质谱分析水稻叶片蛋白质组的方法。水稻叶片蛋白质经过酚法提取,酶解肽段脱盐后用离线反相-反相二维液相色谱分离,然后用线性离子阱/静电场轨道阱组合式高分辨质谱分析,共鉴定到2712种蛋白质。比较了液相色谱分离系统(一维液相色谱与二维液相色谱)和水稻叶片蛋白质提取方法(酚法、十二烷基硫酸钠法(SDS法)和三氯乙酸/丙酮法(TCA/丙酮法))对鉴定蛋白质数量的影响,结果表明:在二维液相色谱条件下,酚法、SDS法和TCA/丙酮法鉴定到的蛋白质数目为2712、2415和1914,分别是一维液相色谱条件下鉴定到的蛋白质数目的2.7、2.5和1.9倍。二维液相色谱条件下,酚法鉴定到的蛋白质数目比SDS法和TCA/丙酮法分别多297和798。与SDS法和TCA/丙酮法相比,酚法不但鉴定到的蛋白质数量多,而且能够鉴定到一些极端蛋白质,如酸性、碱性及高等电点的蛋白质。此外,对二维液相色谱条件下3种蛋白质提取方法提取到的蛋白质进行生物学功能分类,发现3种方法鉴定到的蛋白质的功能存在互补性,但酚法鉴定到的蛋白质功能种类最多。该法为水稻蛋白质组学研究提供了技术支撑,同时也为其他作物的蛋白质组学研究技术提供重要的借鉴。  相似文献

Summary The aim of this study has been the evaluation of an automated system for on-line sample preparation using solid phase extraction and HPLC purification for the measurement of prostanoids in urine. We have established the optimum precolumn and column conditions for this analysis. The manual extraction —HPLC procedure furnishes lower recoveries and higher coefficients of variation than those obtained by the automated on-line procedure. The automated system has been applied to prostanoid analysis of human urine samples from subjects exposed to lead.  相似文献

Capillary RPLC/ESI-MS (cRPLC/ESI-MS) is one of the most powerful analytical tools for current proteomic research. The development of cRPLC techniques coupled online to a mass spectrometer has focused on increasing the separation efficiency, detection sensitivity, and throughput. Recently, the use of high-pressure (over 10,000 psi) LC systems that utilize long, small inner diameter capillary columns has gained much attention for proteomic analyses. In this study, we developed an ultrahigh-pressure dual online SPE/capillary RPLC (DO-SPE/cRPLC) system. This LC system employs two online SPE columns and two capillary columns (75 microm inner diameter x 1 m length) in a single separation system, and has a maximum operating pressure of 10,000 psi. This DO-SPE/cRPLC system is capable of providing high-resolution separation in addition to several other advantageous features, such as high reproducibility in terms of the LC retention time, rapid sample injection, online desalting, online sample enrichment of dilute samples, and increased throughput as a result of essentially removing the column equilibration time between successive experiments. We coupled the DO-SPE/cRPLC system online to a tandem mass spectrometer to allow high-throughput proteomic analyses. In this paper, we demonstrate the efficiency of this DO-SPE/cRPLC/MS/MS system by its use in the analyses of proteomic samples exhibiting different levels of complexity.  相似文献

Summary L-N-(3,5-dimethoxyoxybenzoyl)isoleucine, ionically bonded to γ-aminopropyl silica, has been tested as a chiral stationary phase for the separation of racemates by HPLC. The phase shows good selectivity towards different types of racemates and in particular for those having an electron-poor aromatic group in their molecule. Summary Several polystyrene gels of different pore sizes were packed into a 500 mm×2.1 mm I.D. column. Semi-micro size-exclusion chromatography (SEC) using these columns was carried out with a system consisting of a triple piston pump, a micro loop injector and a flow cell with 1.0-μl cell volume constructed for semi-micro HPLC, because the dead volume of the injector and the cell volume of flow cell for conventional HPLC caused a significant loss in column efficiency. The effects of sample amount, injection volume and mobile phase flow rate on column efficiency and retention volume were examined and the optimized operational variables of the sample amount (below 500 μg), the injection volume (less than 15 μl) and the flow rate range (30–70 μl/min) determiend for semi-micro SEC. Oligostyrene, epoxy resin, phenol-formaldehyde resin and phthalates were analyzed by the optimized semi-micro SEC system under the given conditions. In addition, molecular weight distribution of four different poly(ethylene terephthalate) films was successfully measured by using a mixture of chloroform and hexafluoroisopropanol as the eluent.  相似文献

A CPE-HPLC (UV) method was developed for the determination of Sudan (I-IV) dyes, non-ionic surfactant Triton X-100 was used to extract and preconcentrate Sudan dyes from chilli powder prior to their determination by HPLC-UV. The separation and determination of Sudan dyes was then carried out in the HPLC-UV system with isocratic elution, and the detector was set at 500 nm. The parameters and variables that affect the extraction were investigated. Under optimum conditions: 3% of Triton X-100 (W/V), 10% of Na2CO3 (W/V), heat-assisted at 70 °C for 30 min. Recoveries of the Sudan dyes ranged from 80.70% to 85.45% in chilli powder by CPE method, with all the relative standard deviations of less than 3%. Limit of detection (LOD) and limit of quantification (LOQ) were in the range of 2.0-4.0 and 7.0-12.0 μg kg−1 in chilli powder, respectively.  相似文献

Gas-stirred propylene carbonate (PC) extraction can be performed by bubbling nitrogen without loss of volume. This extraction technique has proved to be highly useful for the separation and subsequent direct polarographic estimation of Tl(III) in the recovered PC phase. Tl(III) can be determined by differential-pulse polarography over the concentration range of 0.4 10 g ml–1 without any interferences due to most metal ions such as Pb(II), Bi(III), Cu(II), Sb(III) and As(III). This proposed technique would be possible to use as a simple extraction step prior to the final estimation by instrumental method of analysis such as atomic absorption spectrometry or spectrophtometry.  相似文献
