杏鲍菇多糖的提取及含量测定   总被引:4,自引:0,他引:4  
采用苯酚-硫酸分光光度法对杏鲍菇中多糖含量进行了测定。测定波长485 nm,多糖换算因子f=1.57,在5.00~70.00μg/mL范围内吸光度与被测物含量呈良好的线性关系,相关系数r=0.999 8,方法的回收率在98.6%~102.3%之间,相对标准偏差RSD在2.01%~2.94%之间。测定结果表明,杏鲍菇中含有丰富的多糖。该法简便、准确、重现性好,可作为杏鲍菇多糖含量的测定方法。  相似文献

Pleurotus eryngii (PE) is an edible mushroom with high nutritional value. Pleurotus eryngii polysaccharides (PEPs) are one of the main active ingredients and manifest a great variety of biological activities. This study mainly focused on the chemical characterization and biological activities of PEPs, which were separated into two fractions (named WPS and P-1). WPS is mainly dominated by β-glycosidic bonds and contains α-glycosidic bonds, and P-1 only contains α-glycosidic bonds. The molecular weights of WPS and P-1 were 4.5 × 105 Da and 2.2 × 104 Da. The result of GC indicated that two the fractions were composed of rhamnose, arabinose, xylose, mannose, glucose, and galactose, with a ratio of 0.35:0.24:0.45:0.24:28.78:1.10 for WPS and 0.95:0.64:0.66:1.84:60.69:0.67 for P-1. The advanced structure studies indicated that the two fractions had no triple-helical structure, where WPS had a dense structure and P-1 had a loose structure. In addition, the antioxidant activity of WPS surpassed P-1, and the two fractions also exhibited a high hypoglycemic activity via inhibiting α-glycosidase activities and promoting the expression of PI3K-AKT signaling pathway based on in vitro assay and cell experiments.  相似文献

采用苯酚-硫酸法测定了景宁产杏鲍菇中多糖的含量,并采用等离子体发射光谱法(ICP-AES法)测定了其微量元素的含量.结果表明,多糖测定波长为490nm,在2.04~10.20μg/mL范围内吸光度与被测物含量呈良好的线性关系,r=O.9992,方法的平均回收率为100.8%,RSD=1.3%.杏鲍菇含有丰富的多糖及铜、铁、锰、锌、硒等微量元素,药食两用,具有很高的经济价值和药用价值.多糖测定方法简便、准确、重现性好,可作为杏鲍菇多糖含量的测定方法.  相似文献

A crude polysaccharide possessing antitumour, radiation-resistant and anti-ageing attributes was extracted from Tricholoma matsutake by water extraction and alcohol precipitation. From this crude polysaccharide, a homogeneous polysaccharide, TMP-5II, was successfully purified by Sephacryl S-300 column chromatography. The average molecular weight (Mw) of TMP-5II was 15.76 kDa. Monosaccharide analysis indicated that the homogeneous polysaccharide contained four different residues: d-glucose, d-galactose, d-mannose and d-fucose. Attenuated total reflectance infrared spectroscopy revealed characteristics typical of carbohydrate polymers and a peak typical of a β-type glycosidic bond. TMP-5II was selected for structural characterisation by nuclear magnetic resonance (NMR) analysis. According to 1H NMR, 13C NMR and two-dimensional-NMR analysis, TMP-5II contains two kinds of linkages, β and α, at a ratio of 4:1. Preliminary results indicated that the polysaccharide had (1-4)-beta-pyran glucose as the main chain, and a branched chain in the O-6 location with fucose (1-2) mannose (1-3)-alpha-pyran galactose.  相似文献

采用热水浸提法从杏鲍菇中提取粗多糖, 经纯化得到一种新的多糖WPP2.通过紫外光谱、色谱、质谱、核磁共振、红外光谱、环境扫描电子显微镜(ESEM)和原子力显微镜(AFM)等技术相结合的方法, 对WPP2结构进行了表征, 并对其体外抗氧化和抗肿瘤活性进行了初步探讨.结果表明, WPP2的平均分子量为4.499×105, 主要由Glc组成, 每个重复单元存在2条支链,→1,2,3)-β-D-Manp和→1,2,6)-β-D-Manp位于支链位点, 支链由→1,6)-β-D-Glcp和→1,6)-β-D-Galp构成, 非还原末端基为→1)-β-D-Glcp, 主链主要由→1,3)-β-D-Glcp,→1,6)-β-D-Galp和→1,3)-β-D-Manp糖残基组成.刚果红实验结果表明其具有三螺旋结构; 用ESEM和AFM首次在纳米尺度观察到WPP2具有多分枝结构; 活性实验结果表明WPP2具有一定的抗氧化和抗肿瘤活性.  相似文献

Abstract

Juniperus tibetica Kom., a common forest species in Tibet, has been traditionally used in Tibetan medicine to treat rheumatic arthritis. In order to investigate the effective substances of J. tibetuca, anticomplent assay-guided fractionation using gradient anion-exchange chromatography and gel permeation chromatography was applied to the crude polysaccharides of the twigs and leaves of J. tibetica and led to the isolation of a homogenous polysaccharide, YB-PS. It exhibited a robust anticomplement activity (CH50: 461?±?10?μg/mL) through the classical pathway of complement system activation by targeting complement components C1q, C3, C4 and C5. The structure of YB-PS was studied by monosaccharide composition and methylation analyses, 1?D, 2?D NMR data and scanning electron microscope. The results indicated that YB-PS was composed of Ara, Gal, and Glc with a molar ratio of 5.8:3.0:1.2. Its backbone was comprised of 1,3-linked α-Araf, 1,3,5-linked α-Araf, 1,2,4-linked α-Galp, and 1,3-linked α-Glcp with terminal α-Araf, β-Galp and 1,4-linked β-Galp as branches. YB-PS was found to have a sheet structure with smooth surface. This study provided scientific foundation for YB-PS as a potential complement inhibitor to treat diseases involved with excessive activation of the complement system.  相似文献

A novel heteroglycan TLH-G was successfully isolated from Tricholoma lobayense cultivated in south China. TLH-G was composed of five monosaccharides, namely, mannose, rhamnose, glucuronic acid, glucose and galactose, and consisted of →1)-β-D-Glcp-(6→ and →1)-α-D-Galp-(3→ motifs. TLH-G had excellent scavenging activities towards 2,2-diphenyl-1-picrylhydrazyl (DPPH) and 2,2′-azino-bis(3-ethylbenzothiazoline-6-sulphonic acid) (ABTS) radicals. Structure-activity relationship analysis demonstrated that glycosidic bond types, monosaccharide composition, and branching degree were more important for the antioxidant activity of polysaccharides from Tricholoma lobayense than other structural factors. All the above results showed that the growth environment played a crucial role in the structure and antioxidant activity of polysaccharides from Tricholoma lobayense.  相似文献

In recent years, the structure of selenium-enriched polysaccharides and their application in immunomodulation have attracted much attention. In previous studies, we extracted and purified a novel selenium-enriched Pleurotus ostreatus polysaccharide called Se-POP-21, but its structure and immunomodulatory activity were still unclear. In this study, the main structural unit formula of Se-POP-21 was characterized by methylation analysis and an NMR experiment. The results showed that the backbone of Se-POP-21 was →[2,6)-α-D-Galp-(1→6)-α-D-Galp-(1]4→2,4)-β-L-Arap-(1→[2,6)-α-D-Galp-(1→6)-α-D-Galp-(1]4→, branched chain of β-D-Manp-(1→ and β-D-Manp-(1→4)-β-L-Arap-(1→ connected with →2,6)-α-D-Galp-(1→ and →2,4)-β-L-Arap-(1→,respectively, through the O-2 bond. In vitro cell experiments indicated that Se-POP-21 could significantly enhance the proliferation and phagocytosis of RAW264.7 cells, upregulate the expression of costimulatory molecules CD80/CD86, and promote RAW264.7 cells to secrete NO, ROS, TNF-α, IL-1β, and IL-6 by activating the NF-κB protein. The results of this study indicate that Se-POP-21 can effectively activate RAW264.7 cells. Thus, it has the potential to be used in immunomodulatory drugs or functional foods.  相似文献

A water-soluble polysaccharide (CPS-0) was obtained from the root of Cudrania tricuspidata (Carr.) Bur., by hot water extraction (70°C), deproteination using enzymolysis and Sevag method, precipitation with ethanol, and fractionation through DEAE-Sephadex A-50 chromatography. The purity of CPS-0 was determined by HPLC and the structure was elucidated by monosaccharide composition analysis, methylation analysis, GC, GC-MS, NMR spectral (1H-NMR, 13C-NMR, HMQC), UV, IR, and elemental analysis. The CPS-0 was found to contain glucose residues only. In this work, the optimization of extraction of polysaccharides from Citrullus colocynthis curd polysaccharides (CCPs) is investigated by using the analysis of variance. For this aim, a chemical extraction method has been developed in order to be able to isolate CCPs at high level of purity. The extraction was performed by using boiling water at different extraction times (30, 60 and 120?min), extraction temperatures (50, 75 and 100°C), and a material ratio (30, 60 and 100?g/ml), for maximum polysaccharide extraction. The 3-D response surface plot and the contour plot derived from the mathematical models are used to determine the optimal conditions. The optimum extraction conditions are as follows: extraction temperature 100°C, extraction time 120?min and the ratio of water 30?ml/g. Under these conditions, the experimental final mass is 210.6?mg which is very close to the predicted value (209.46?mg) with a regression coefficient of 94.01%. In addition, the existence of the polysaccharide has been confirmed by the characterization of the galactomannan obtained from C. colocynthisis obtained by Fourier transform infrared-attenuated total reflection, IR-Raman spectroscopy and X-ray diffraction.  相似文献

One main fraction of Selenium-enriched Pleurotus ostreatus (P. ostreatus) polysaccharide (Se-POP-1) was extracted and purified by DEAE-52 and sephadex G-100. Se-POP-1 was an approximate homogenous polysaccharide with an average molecular weight of 1.62 × 104 Da, and mainly composed of glucose, mannose and galactose, with molar ratio of 5.30:1.55:2.14. The absorption peaks at 941 cm?1 and 1048 cm?1 in FT-IR analysis were ascribed as C-O-Se and Se=O bonds. Pr-treatment of Se-POP-1 (400 μg/mL) increased the cell survival of H2O2-stimulated PC12 cells and inhibited intrinsic apoptosis and oxidative stress in H2O2-stimulated PC12 cells via limiting DNA degradation and decreasing the reactive oxygen species (ROS) generation. In addition, up-regulation of anti-apoptotic protein Bcl-2, down- regulation of pro-apoptotic protein Bax, cleaved caspase 3, and cytochrome c were also observed. Se-POP-1 presented an obvious effect to alleviate oxidative damage and apoptosis in PC12 cells induced by H2O2. Therefore, Se-POP-1 possessed potent antioxidant and biological activities with the ability to prevent oxidation via scavenging ROS and free radicals in cells. It could be developed as organic selenium dietary supplement and functional food.  相似文献

The structural properties and Angiotensin-I converting enzyme (ACE) inhibition activities of a polysaccharide (PGE) extracted from Gastrodia elata Blume were investigated. PGE was extracted using hot water and purified by Sephadex G-200 followed by ultra-filtration. The structural characterisation of PGE was analysed by FT–IR, NMR spectroscopy, specific rotation determination, periodate oxidation-smith degradation, methylation analysis, GC–MS and Congo red test. The results revealed that PGE was composed by glucose, with an average molecular weight of 1.54 × 103 kDa. The structure of PGE was 1→3 and 1→4,6-branched-glucopyranose that had a linear backbone of (1 → 4)-linked-d-glucopyranose (Glcp). ACE-inhibitory activity results showed that PGE was efficient to inhibit ACE and the IC50 value was 0.66 mg/mL.  相似文献

A polysaccharide named PNP was extracted and purified from Pholiota nameko. The total sugar content of PNP was 95.29% and the molecular weight was 1.89 × 103 kDa. The structural features of PNP were investigated by the combination of chemical and instrumental analysis such as UV spectrophotometer, specific rotation determination, FT-IR, methylisation analysis and Congo red. The results showed that the optical rotation of PNP was +120° and that it had a triple-helical structure. Besides, PNP was mainly composed of glucose and mannose at the molar ratio of 4.24:1.00. The backbone of PNP was composed of (1→3)-linked-Glc and (1→3)-linked-Man whereas the branches of (1→3,6)-linked- Glc, (1→3,6)-linked-Man and T- Glc. Consistenting with the results of UV–Vis spectra, FT-IR spectroscopy and 1H NMR, indicated that PNP was a complex of polysaccharides and polyphenols. In vitro antioxidant results suggested that PNP was processed with certain scavenging capacity.  相似文献

An organic-assisted alkaline extraction method was developed for the determination of hexavalent chromium (Cr(VI)) in plastics. The solubilization of polymer as a pre-step of the alkaline extraction provided good extraction efficiency of Cr(VI) from the sample. The optimization of the experimental conditions affecting the extraction and UV–vis spectrophotometric analysis was accomplished by evaluating the recovery rate of Cr(VI) through the analysis of Cr(VI) in in-house polymer reference materials (RMs). With the proposed method, most of the Cr(VI) in polymers was released within a short extraction time of 30 min and the Cr(III)-DPCO complex can be kept stable for 24 h. The heating for the extraction of the Cr(VI) was not necessary. The optimal pH of the final solution was fixed at 2.0. The proposed extraction method was applied successfully for the determination of Cr(III) and Cr(VI) in spiked samples. The practical applicability of this new method was evaluated through the analysis of Cr(VI) in in-house polymer RMs. The good linearity was demonstrated at desired concentrations of the range 0–3.3 mg L−1. The detection limits were quite low, varying from 0.0061 to 0.0285 mg L−1. The recovery of Cr(VI) was between 97 and 106%, and the relative standard deviation (R.S.D.) was below 6%.  相似文献

Abstract

The enhanced osteoblast differentiation is beneficial to the prevention of osteoporosis. In this study, a homogeneous polysaccharide (LRP-S2A) with the potential of promoting osteoblast differentiation was obtained from the fruits of Lycium ruthenicum, a traditional herb for treatment of postmenopausal metabolic disorders. Structural identification indicated that LRP-S2A, with a relative molecular weight of 2.65 × 106 Da and an uronic acid content of 41.8%, contained Rha, Ara, Gal, Glc and GlcA in a molar ratio of 1.00 : 2.07 : 0.57 : 2.59 : 4.33 and was composed of a backbone consisting of 6-O-Me-α-(1→4)-D-GlcpA, 2-O-acetyl-α-(1→4)-D-Glcp, α-(1→2,4)-L-Rhap, β-(1→3)-D-Galp andα-(1→3,5)-L-Araf, and some branches consisting of 6-O-Me-α-(1→4)-D-GlcpA and terminal α-L-Araf. These results suggested that LRP-S2A with the potential of promoting osteoblast differentiation was a new acidic polysaccharide.  相似文献

从猴头发酵菌丝体水提后残渣中用碱提出了水溶性粗多糖,对其分级纯化得到纯样HPP,SepharoseCL-6B柱层析测得其相对分子质量约为6 53×104,G C 分析表明,其单糖组成为Glc;经部分酸水解、高碘酸氧化、Smith降解和甲基化分析确定此多糖为多分枝结构,其中(1→6)Glc构成主链的核心结构;分支点为O-3;支链部分由(1→3)Glc构成,并连接一端基Glc.  相似文献
