Abstract The separation of three closely related tetrahydrocannabinol isomers differing only in the position of the double bond in ring C was achieved by HPLC using a μBondapak C18 column and a ternary mobile phase of acetonitrile/tetrahydrofuran/water. Abstract Soil organic matter was extracted from field soils using mild pretreatment and a Chelex 100 resin bed. The extracts were separated by molecular weight using a Sephadex G-25 gel permeation chromatography column. Reverse-phase high performance liquid chromatography and o-phthalialdehyde precolumn derivatization were used to rapidly determine the primary amino compounds present in the 6 N HCl-soluble fractions. Twenty amino-N compounds were quantitatively identified in the hydrolysates. 相似文献

Monolithic molecularly imprinted columns are a new class of column that emerged in the early 1990s. These monolithic materials are typically prepared directly inside stainless steel columns or capillary columns without the tedious procedures of grinding, sieving, and column packing. Furthermore, the preparation of this type of MIP is more cost-efficient, because the amount of template molecules required is much lower. In recent years monolithic supports as stationary phases in high-performance liquid chromatography (HPLC) and capillary electrochromatography (CEC) have attracted significant interest because of their ease of preparation, high reproducibility, versatile surface chemistry, and rapid mass transport. 相似文献

Abstract A method is described for the analysis of 16 major nucleotides including NAD, IDP, GDP, cAMP, cGMP and succinyl AMP by reverse phase HPLC. The use of 65mM KH2PO4, at pH 3.2, low concentrations of the ion pairing reagent tetrabutylammonium phosphate and acetonitrile allowed the simultaneous separation of these nucleotides by isocratic or gradient elution in 18 and 28 minutes respectively. Stainless steel and radially compressed columns were compared and a similar separation profile was obtained. The latter columns increased retention and improved the efficiency of separation. 相似文献

Abstract Of the columns investigated, the graphitised carbon column provided the best chromatographic characteristics for the highly water-soluble compound ethylenethiourea (ETU). The stability of the carbon column in strongly acidic media permitted the incorporation of the phosphoric acid electrolyte into the 5% acetonitrile-in-water mobile phase. ETU eluted from the column in 200 s as a sharp symmetrical peak at a mobile phase flow rate of 1 mL/min and a column temperature of 35°C. The k' value was 1.72. ETU peak retention times and responses showed excellent repeatability with coefficients of variation of 0.28 and 1.40%, respectively, for 6 replicates with the high performance liquid chromatographic-electrochemical system using the graphitised carbon column. Although ETU eluted as a sharp symmetrical peak with the cyclodextrin chiral columns, their instability at low pH required post-column addition of the phosphoric acid electrolyte solution. ETU chromatographed poorly or degraded on the polymer columns. The chromatographic separation of ETU on the C-8 reverse-phase bonded silica column appeared to be due mainly to residual silanol groups. With the NH2 bonded silica column ETU eluted immediately after the injection solvent. 相似文献

The influence of aminoacid and peptide type buffers as the separation electrolyte to the resolution of underivatized neutral carbohydrates in capillary electrophoresis was investigated. With the use of plain glycylglycine as the background electrolyte, a noticeable improvement in the resolution of carbohydrates was observed. Without any additive, 50 mmol L−1 glycylglycine electrolyte at pH 12 provides both the fast separation and indirect detection of sugars. The electrophoretic mobilities of 16 sugars were calculated at this buffer and the combinations of 10 sugars were simultaneously detected. The method was applied to the determination of sucrose, glucose, and fructose in commercial juice samples. 相似文献

Abstract New bifunctional reagents, 2-amino-6-carboxyethylpyridine and 2-amino-6-cyanoethylpyridine, were designed and synthesized in order to provide a novel procedure for preparation of neoglycoconjugates from fluorescence-labeled and purified sugar chains. Labeling of model sugar chains with these reagents was effected by reductive amination using BH3.Me2NH to give corresponding 6-carboxyethylpyridylaminated (CEPA-) and 6-cyanoethylpyridylaminated (CNEPA-) derivatives, which were readily purified by reversed phase HPLC. The reagent parts of the labeled products possess the functional groups which then serve as linkers for coupling with matrices such as proteins and polymers. A CEPA-derivative of glucose was directly coupled with the ε-amino group of a Lys derivative to give a neoglycoprotein model. CNEPA-sugars were hydrogenated to give 6-aminopropylpyridylaminated (APPA-) derivatives, which can then be readily used for the preparation of various types of neoglycoconjugates by use of appropriate spacers as exemplified by the coupling of APPA-maltotriose with bovine serum albumin (BSA), biotin, and acrylic acid. The reaction of APPA-maltotriose with succinimidyl 3-(3-nitro-2-pyridyldithio)propionate gave the corresponding amide to be used for a disulfide formation with BSA. Condensation with biotin was effected by use of N-hydroxysuccinimidobiotin. The conjugation of APPA-maltotriose with acrylic acid was carried out by use of 4-acryloyloxyphenyldimethylsulfonium methylsulfate to give the corresponding acrylamide, which can be used for the preparation of sugar-acrylamide copolymers. 相似文献

分别用乙二胺、二乙胺、三乙胺将自制的以甲基丙烯酸缩水甘油酯(GMA)为单体、乙二醇二甲基丙烯酸酯(EDMA)为交联剂的整体柱修饰为弱、强阴离子交换整体柱。考察了该整体柱的性能,选择出分离蛋白质(牛血清白蛋白、溶菌酶和谷胱甘肽)的最佳实验条件,并在最佳分离条件下考察了这些蛋白质在整体柱上的色谱行为和该整体柱对纤维素降解酶的分离纯化情况。实验结果表明,该整体柱性能良好,可以实现对纤维素降解酶的快速分离与纯化。同时,实验也证明采用梯度洗脱可以实现对某些蛋白质的分离纯化。 相似文献

The steroids were derivatised with aroyl chloride to yield the corresponding benzoates. Among them, their 4-methoxybenzoates were obtained in greatest yield and were analyzed by HPLC with the smallest detection limit (10 ng) by means of a UV detector (254 nm). This method of derivatization can be applied for analysis of steroids and triterpenoids from crude products with the HPLC technique. 相似文献
