利用OASIS HLB固相萃取柱富集和净化样品,采用高效液相色谱分离,紫外及荧光检测,建立了不同饮料基质中的壬基酚(NP)、辛基酚(OP)和双酚A(BPA)等类雌激素的测定方法,并用该方法对市售部分产品进行了测定。不同加标水平的BPA,NP和OP的回收率为91.08%~103.19%,其相对标准偏差为0.5%~5.49%。该方法具有操作简单、灵敏度高和重现性好等优点。  相似文献

A procedure has been developed for the determination of bisphenol A (BPA) in foods by gas chromatography/mass spectrometry as 2,2-bis-(4-(isopropoxycarbonyloxy)phenyl)propane formed in the reaction with isopropyl chloroformate. Optimal conditions have been found for BPA derivatization, providing its quantitative conversion into diether derivative in aqueous media. The concentration of BPA has been determined in some samples of canned foods and beverages (from 2.15 to 42.91 ng/g). The detection limit is 0.05–0.1 ng/g.  相似文献

An improved analytical method enabling rapid and accurate determination and identification of bisphenol F diglycidyl ether (novolac glycidyl ether 2-ring), novolac glycidyl ether 3-ring, novolac glycidyl ether 4-ring, novolac glycidyl ether 5-ring, novolac glycidyl ether 6-ring, bisphenol A diglycidyl ether, bisphenol A (2,3-dihydroxypropyl) glycidyl ether, bisphenol A (3-chloro-2-hydroxypropyl) glycidyl ether, bisphenol A bis(3-chloro-2-hydroxypropyl) ether, and bisphenol A (3-chloro-2-hydroxypropyl) (2,3-dihydroxypropyl) ether in canned food and their contact packaging materials has been developed by using, for the first time, ultra-performance liquid chromatography coupled with tandem mass spectrometry. After comparison of electrospray ionization and atmospheric pressure chemical ionization in positive and negative-ion modes, tandem mass spectrometry with positive electrospray ionization was chosen to carry out selective multiple reaction monitoring analysis of novolac glycidyl ethers, bisphenol A diglycidyl ether, and its derivatives. The analysis time is only 5.5 min per run. Limits of detection varied from 0.01 to 0.20 ng g(-1) for the different target compounds on the basis of a signal-to-noise ratio (S/N) = 3; limits of quantitation were from 0.03 to 0.66 ng g(-1). The relative standard deviation for repeatability was <8.01%. Analytical recovery ranged from 87.60 to 108.93%. This method was successfully applied to twenty samples of canned food and their contact packaging materials for determination of migration of NOGE, BADGE, and their derivatives from can coatings into food.  相似文献

Computationally and spectroscopically assisted analytical comparative investigation into the extraction of bisphenol A using three cyclodextrins, that is, α, β, and γ respectively, were performed. A simple, self-tailored μ-solid-phase extraction podium was used to extract bisphenol A from water samples, and high-performance liquid chromatography-ultraviolet was used for the qualitative and quantitative analysis of bisphenol A. Density functional theory first principle calculations, attenuated total reflectance Fourier-transform infrared spectroscopy and Fourier-transform Raman spectroscopy data supports the analytical selection of β-cyclodextrin as the adsorbent for bisphenol A extraction. Analytical optimization of various parameters including sample volume, sample pH, eluting solvent and its volume was performed to discover the most proper conditions for maximum extraction. Under the optimized conditions, a limit of detection value of 0.70 ng/ml and a limit of quantification value of 2.31 ng/ml was achieved with β-cyclodextrin, with recovery (%) values over 98.40–102.50 in real source water samples. Overall, well assisted by comprehensive computational and spectroscopic studies, a novel, simple, sensitive and economic analytical method was developed for the extraction of bisphenol A from source water using cyclodextrin.  相似文献

Due to regulation of the use of bisphenol A, several analogs serving as bisphenol A replacements have drawn substantial attention for their adverse health effects. To investigate their occurrence in humans and identify possible pollution sources, it is necessary to develop a sensitive method for total bisphenols detection. Thus, a method based on enzymolysis and liquid‐liquid extraction followed by molecularly imprinted polymer solid‐phase extraction and pre‐column derivatization with high‐performance liquid chromatography and tandem mass spectrometry was proposed. The developed method exhibited superior selectivity and sensitivity. The matrix effect can be eliminated to a great extent. The method detection limits for eight bisphenols were 0.05~0.19 ng/mL. Satisfactory recoveries (71~119%) were obtained by spiking bovine serum at three levels (0.8, 8 and, 20 ng/mL). The method was successfully applied to determine total bisphenols in the serum samples of children. Bisphenol A, bisphenol F, bisphenol S, bisphenol B and bisphenol F were detected with concentrations from below the method detection limit to 1.65, 0.45, 0.79, 2.04 and 0.17 ng/mL, respectively. These results indicate that bisphenol A remains the major pollutant among the studied bisphenols in children, whereas threats from bisphenol A analogs should also be monitored.  相似文献

建立了纺织品和食品包装材料中壬基酚、辛基酚和双酚A的液相色谱-串联质谱分析方法。不同类型的纺织品和食品包装材料样品采用加速溶剂萃取法,以无水乙醇为提取剂,在10.3 MPa和120℃下静态循环提取2次,提取液经Supelclean Envi-Carb石墨化碳黑固相萃取柱净化,收集甲醇-二氯甲烷(1∶4,V/V)洗脱液,采用Waters XBridge C18色谱柱,以甲醇-0.1%氨水溶液为流动相,梯度洗脱分离后,在LC/MS/MS多反应监测模式下进行定性与定量分析。壬基酚、辛基酚和双酚A的方法检出限为0.5μg/kg,在0.5~10μg/kg的3个添加水平范围内,纺织品样品的平均回收率为86.9%~92.5%,相对标准偏差均小于9.1%;食品包装材料样品的平均回收率为87.8%~93.0%,相对标准偏差均小于8.8%。本方法准确、快速、灵敏度高,可用于纺织品和食品包装材料的实际检验。  相似文献

A sensitive method for the analysis of bisphenol A and 4-nonylphenol is developed by means of the optimization of solid-phase microextraction using Uniform Experimental Design methodology followed by high-performance liquid chromatographic analysis with fluorescence detection. The optimal extraction conditions are determined based on the relationship between parameters and the peak area. The curve calibration plots are linear (r2>or=0.9980) over the concentration range of 1.25-125 ng/mL for bisphenol A and 2.59-202.96 ng/mL for 4-nonylphenol, respectively. The detection limits, based on a signal-to-noise ratio of 3, are 0.097 ng/mL for bisphenol A and 0.27 ng/mL for 4-nonylphenol, respectively. The validity of the proposed method is demonstrated by the analysis of the investigated analytes in real water samples and sensitivity of the optimized method is verified by comparing results with those obtained by previous methods using the same commercial solid-phase microextraction fiber.  相似文献

A fast screening method consisting of off-line solid-phase microextraction coupled to HPLC and fluorescence detection, suitable for the analysis of several bisphenol derivatives and their degradation products in aqueous solution, has been developed. Detection limits of 0.7 ng ml(-1) for 2,2-bis[4-(glycidyloxy)phenyl]propane, 0.9 ng ml(-1) for bisphenol A bis(3-chloro-2-hydroxypropyl)ether, 1.1 ng ml(-1) for 2,2-bis(4-hydroxyphenyl)propane and 2.4 ng ml(-1) for bisphenol F diglycidyl ether have been achieved working in the linear range 10-500 ng ml(-1). The good analytical features achieved make the proposed method an interesting option for the direct determination of these compounds in aqueous canned food such as peas, tuna, olives, maize, artichokes or palm hearts. Both the optimization process and the results, including the analysis of real samples, are given and discussed.  相似文献

Bisphenol A and other hydroxylated diphenylalkanes (generally known as bisphenols) have been identified as potential estrogenic substances. In this paper, a specific polyclonal antibody was produced against these compounds by immunization of rabbits with a conjugate of 4,4-bis (4-hydroxyphenyl) valeric acid and bovine serum albumin (BHPVA-BSA). The polyclonal antibody showed specific recognition of the bisphenol structure, while the cross reactions of other common phenolic compounds such as phenol, hydroquinol and p-hydroxybenzoic acid were all lower than 1%. The linear range of bisphenol A calibration curve was 1–10 000 ng ml−1. Real water samples and mouse serum samples were spiked with known amount of bisphenol A and measured by the competitive ELISA. Recoveries were between 92 and 105%. The detection limits were found to be 0.1 ng ml−1 for real water samples and 2 ng ml−1 for serum samples. The method is very useful for monitoring bisphenol compounds in environmental and biological samples.  相似文献

The purpose of this study was to develop an analytical method for determination of bisphenol A (BPA) from fruits and vegetables. The present method developed for extraction of BPA from samples was based on solid-phase extraction (SPE) method and solvent extraction. Recovery results in the samples spiked with a 10 ng/ml BPA [no detection (<1 ng/g) to 77%] were lower than those in the samples with a 50 ng/ml BPA (26-96%). The fact that the low recovery results were caused by BPA degradation by enzymes is found. These problems were proved by the pH (pH ≤3) and the heating treatment (at ≥80 °C for 5 min). However, because the heating treatment at temperatures of ≥80 °C for 5 min is more difficult and time-consuming method than the pH control, we suggest that the pH control is useful to prevent BPA degradation. Good recovery results (82-101%) were obtained from all fruit and vegetable samples after pH treatment (pH ≤3). Effective elimination of impurities and a good detection limit (1 ng/g) were obtained with a method involving two SPE cartridges (OASIS HLB and Sep-Pak Florisil cartridge).  相似文献

建立了测定食品接触材料中6种双酚-二环氧甘油醚(双酚A二缩水甘油醚(BADGE)及其衍生物双酚A(2,3-二羟丙基)甘油醚(BADGE•H2O)、双酚A(3-氯-2-羟丙基)甘油醚(BADGE•HCl)、双酚A(3-氯-2-羟丙基)(2,3-二羟丙基)醚(BADGE•H2O•HCl)和双酚F二缩水甘油醚(BFDGE)及其衍生物双酚F双(3-氯-2-羟丙基)甘油醚(BFDGE•2HCl))迁移到食品中的迁移量的高效液相色谱-串联质谱法(HPLC-MS/MS)。样品以叔丁基甲醚(MTBE)为提取溶剂,超声提取,提取液经多壁碳纳米管(MWCNTs)固相萃取(SPE)柱富集、净化。以COSMOSIL C18为分析柱,流动相为0.1%甲酸的5 mmol/L醋酸铵溶液和甲醇。6种双酚-二环氧甘油醚在1.0~100 μg/L范围内线性关系良好(r2>0.9991)。在3个添加水平下,6种目标化合物的回收率范围为78.6%~89.9%,相对标准偏差小于10%。方法检出限范围为0.5~1.5 μg/L。该方法操作简单,灵敏度高,可应用于食品接触材料中双酚-二环氧甘油醚迁移量的快速检测。  相似文献

Decanoic acid reverse micelle-based coacervates were proposed for the extraction of bisphenol A (BPA) from canned vegetables and fruits prior to its determination by liquid chromatography and fluorescence detection at lambda(exc) = 276 nm and lambda(em) = 306 nm. The procedure involved the extraction of minute quantities (300-700 mg) of homogenized food sample with an aqueous solution containing 10% of THF and 0.5% of decanoic acid, conditions under which the coacervate (around 340 microL) formed in situ and instantaneously. The overall sample treatment, which included extraction and centrifugation, took about 25-30 min, and several samples could be simultaneously treated using conventional lab equipment. No clean-up or solvent evaporation were required. Extraction efficiencies mainly depended on the decanoic acid and THF concentration in the aqueous solution and were not affected by the pH or the temperature in the ranges studied (1-4 and 20-60 degrees C, respectively). Recoveries in samples ranged between about 81 and 96%. The precision of the method, expressed as relative standard deviation, was about 3% and the quantitation limit was around 9 ng g(-1), which was far below the current specific migration limit (SML) set for BPA by the EU Commission (600 ng g(-1)). The method was successfully applied to the determination of BPA in the solid content of canned fruit salad, peaches in syrup, mango slices, red peppers, sweetcorn, green beans and peas. BPA was present at concentrations in the range from 7.8 to 24.4 ng g(-1) in canned fruits and from 55 to 103 ng g(-1) in canned vegetables.  相似文献

The aim of this work was to develop and validate a method to determine aflatoxin M1 (AFM1) in cheese, yogurt, and dairy beverages. The method consisted of aqueous methanol extraction, immunoaffinity column purification and isolation, RPLC separation, and fluorescence detection. The four types of cheese samples were classified according to moisture and fat content. The mean recoveries were 71% for cheese at spiked levels from 100 to 517 ng/kg, and 76% for yogurt and dairy beverages spiked at levels from 66 to 260 ng/kg. The mean RSDs were 5.9% for cheese, and 10% for yogurt and dairy beverages. The LOD was 3 ng/kg and the LOQ was 10 ng/kg for all test commodities. To test the applicability of the developed method, a small survey of the presence of AFM1 in cheese, yogurt, and dairy beverages purchased in Ribeir?o Preto-SP, Brazil, was conducted. AFM1 was detected (> 3 ng/kg) in all samples. Twenty cheese samples (83%) were contaminated with AFM1 in the range of 13-304 ng/kg. In yogurt and dairy beverages, the contamination was lower (13-22 ng/kg) in five samples (42%). The results indicated that the method is adequate for the determination of AFM1 in these four types of cheese, as well as in yogurt and dairy beverages.  相似文献

The present study illustrates the possibilities of capillary isotachophoresis (CITP) online coupled with capillary zone electrophoresis (CZE) and hyphenated with fiber-based spectrophotometric diode array detection (DAD) for the direct, highly reliable, and ultrasensitive determination of quinine (QUI) in real multicomponent ionic matrices (beverages). Here, the CITP provided an effective online sample pretreatment (preseparation and preconcentration) prior to the CZE separation. Due to the CITP sample preconcentration, a simple UV-visible absorbance spectrophotometric detection was sufficient for obtaining very low concentration limits of detection (~2.3 ng/mL). Enhanced separation selectivity due to the combination of different separation mechanisms (CITP vs. CZE) enabled to obtain a pure analyte zone, suitable for its detection and quantitation in the directly injected real samples. The spectrophotometric DAD, unlike single wavelength UV detection, enabled to characterize the purity (i.e. spectral homogeneity) of the analyte zone and preliminary data indicate structurally related compounds via characteristic spectra recorded in the interval of 200-600 nm. The proposed CITP-CZE-DAD method was characterized by favorable performance parameters (sensitivity, linearity, precision, recovery, accuracy, robustness, and selectivity) and successfully applied to the control of QUI and potential QUI impurities in commercial beverages. This method is proposed as a routine automatized method for the highly reliable quality food control.  相似文献

Ochratoxin A (OTA) is a mycotoxin produced by fungi species belonging to the genera Aspergillus and Penicillium being isolated in alcoholic beverages. The aim of this work is developed and applied a procedure for the analysis of OTA in wines. An analytical method based on immunoaffinity column (IAC) for clean-up, liquid chromatography with fluorescence detection (LC-FD), and LC-FD after of OTA methylation was used to determine the occurrence of OTA in wines. Recoveries of this mycotoxin spiked to red wines at 0.5 ng/ml level were >90% with an average of relative standards deviations of 4%. Furthermore, 116 wine samples from designation of origin (DO) and three samples from food stores of Valencian Community (Spain) were examined for the occurrence of OTA being the levels of this mycotoxin ranged from <0.01 to 0.76 ng/ml. Finally, the estimated daily intake of OTA in this study was 0.15 ng/kg bw per day.  相似文献

A method for the determination of endocrine disruptors, bisphenol A and 17alpha-ethinylestradiol, in environmental water samples was developed using in-tube solid-phase microextraction followed by liquid chromatography and fluorescence detection. A poly(acrylamide-vinylpyridine) monolithic capillary column was applied as the extraction media in view of its greater phase ratio than open-tubular capillaries and thus higher extraction efficiency. After optimizing the extraction conditions, bisphenol A and 17alpha-ethinylestradiol were extracted directly from water samples in a wide dynamic linear range of 0.5-1000 ng mL(-1), with the detection limits obtained as 0.064 and 0.12 ng mL(-1), respectively. The precision of the method was satisfactory with the intraday and interday RSD values smaller than 7.2%. Environmental water samples of different sources were successfully analyzed with the presented method and the monolithic capillary was proved to be robust and reusable in analyzing real water samples.  相似文献

基于在线富集-高效液相色谱-荧光法检测法,以C18柱分别作为富集柱和分析柱,经优化荧光波长、色谱分离和富集条件,建立了同时快速检测塑料儿童水杯浸出液中9种双酚类物质的方法。方法学考察结果显示,所建方法符合含量测定要求。9种双酚类物质在各自范围内线性关系良好,相关系数(r2)均大于0.998,检出限为0.13~66.7 ng/L。9种双酚类物质的回收率为90.7%~112.4%(RSD<11.3%,n=6)。结果显示,除双酚芴外,其他双酚类物质均有检出,浸出量随盛放时间增加而增大,高温多次浸泡后部分双酚类浸出量降低。该方法灵敏度高,操作简便,环境友好,可实现多种新型双酚类物质的同时快速检测。  相似文献

Ricin is a toxic ribosome inactivating protein (RIP-II) present in beans of the castor plant, Ricinus communis. Its potential as a biodefense threat has made the rapid, sensitive detection of ricin in food important to the U.S. Food and Drug Administration. Samples of juice, dairy products, soda, vegetables, bakery products, chocolate, and condiments were spiked with varying concentrations of ricin and analyzed using a 96-well format, electrochemiluminescence (ECL) immunoassay. Assay configurations included the use of a monoclonal capture antibody coupled with either a polyclonal or monoclonal detector antibody. The samples and detector antibodies were either added sequentially or in combination during the capture step. Using the polyclonal antibody, 0.04 ng/mL ricin was detected in analytical samples prepared from several beverages. By simultaneously incubating the sample with detector antibody, it was possible to decrease the assay time to a single 20 min incubation step with a limit of detection <10 ng/mL. Assays run according to this single incubation step exhibited a hook effect (decrease in signal at high concentrations of ricin), but because of the large signal-to-noise ratio associated with the ECL assay, the response remained above background and detectable. Thus, the ECL assay was uniquely suited for the screening of samples for ricin.  相似文献

We have developed a simple HPLC method for the microanalysis of bisphenol A (BPA), which is often contained in environmental water and is known as an endocrine disrupter. HPLC coupled with electrochemical detection requires a simpler procedure of pretreatment compared to GC-MS. In this study, we analyzed BPA using molecularly imprinted polymer as an on-line pretreatment device. This polymer has molecular recognition sites and provides specific selectivity in extraction process. Due to this effect, the detection limit obtained with this HPLC was 0.36 ng/l. This method applied to environmental water and purified water samples containing 2-70 ng/l of BPA successfully. Furthermore, UV detection was performed in some actual analyses.  相似文献
