Preparation and characterization of ruthenium complexes with the new 4,4′,4″-tri-tert-butylterpyridine ligand and with 4,4′-di-tert-butylbipyridine

An efficient procedure for the preparation of 4,4′,4″-tri-tert-butylterpyridine (trpy*) which is formed together with 4,4′-di-tert-butylbipyridine (bipy*) is described, and the preparation of the complexes (trpy*)RuCl₃, [(trpy*)L₂RuCl]PF₆ (L₂ = bipy, bipy*), and (bipy*)₂RuCl₂ and their characterization by cyclic-voltametry, UV—vis and ¹H NMR spectroscopy are reported. It is shown that introduction of tert-butyl substituents increases the solubility of the resultant complexes and enhances the electron donating influence of the trpy ligand.     

Influence of the smectic A-nematic transitional order on the formation of the smectic phases of 4-n-hexyloxybenzylidene-4′-n-butyIaniline and 4-n-butyloxybenzylidene-4′-n-octyIaniline from an electrically deformed nematic phase

Abstract

The crucial role of the smectic A-nematic transitional order for the formation of the smectic A, B and G phases from an electrically deformed nematic phase of the liquid crystal 4-n-hexyloxy-benzylidene-4′-n-butylaniline (6O.4) with a typical smectic A-nematic first order transition and the formation of the smectic A and B phases from an electrically deformed nematic phase of the liquid crystal (4-n-butyloxy-benzylidene-4′-n-octylaniline (40.8) with a smectic A-nematic second order transition has been demonstrated. The nematic phase was deformed by an AC voltage of 2U,_th 5U _th and 10U _th, where U _th is the threshold voltage which causes the appearance of the Fréedericksz transition in the homeotropic nematic layer. The smectic textures have been observed on free cooling of the nematic phase or after the use of an oven. The smectic A phase of the liquid crystal 60.4 was observed with the formation of a clear smectic A-nematic phase boundary while the smectic A phase of the liquid crystal 40.8 has been formed from intermediate pretransitional stripes, observed by Cladis and Torza [1]. The homeotropic anchoring of the direction was crucial for the formation of the smectic phases of the liquid crystal 40.8 but not significant for the liquid crystal 60.4.     

On the influence of the chiral side linking bridge type upon the synclinic vs. anticlinic balance in the case of 2′,3′-difluoroterphenyl derivatives

A homologous series of new ferro- and antiferroelectric liquid crystals, containing 2′,3′-difluorosubstituted terphenyl core as a mesogenic core, has been recently synthesised. Synthetic routes and mesomorphic properties were described. Liquid crystal phases were preliminarily determined by thermomicroscopic and microcalorimetric measurements and verified by dielectric measurements and miscibility methods. Temperatures and enthalpies of phase transitions are given.     

Synthesis and antiviral evaluation of 2′,2′,3′,3′-tetrafluoro nucleoside analogs

Herein, we report the synthesis of novel 2′,2′,3′,3′-tetrafluorinated nucleoside analogs along with their phosphoramidate prodrugs. A tetrafluoro ribose moiety was coupled with different Boc/benzoyl-protected nucleobases under Mitsunobu conditions. After deprotection, tetrafluorinated nucleosides 13b, 14b, 20b-22b were reacted with phenyl-(isopropoxy-l-alaninyl)-phosphorochloridate to afford corresponding monophosphate prodrugs 24b–28b. All synthesized compounds were evaluated against several DNA and RNA viruses including HIV, HBV, HCV, Ebola and Zika viruses.     

Resolution and determination of the absolute configuration of 3,3′,4,4′-tetramethyl-1,1′-diphosphaferrocene-2-carboxaldehyde

Racemic 3,3′,4,4′-tetramethyl-1,1′-diphosphaferrocene-2-carboxaldehyde 1 was resolved via the formation of diastereomeric dioxolanes with (S)-(+)-1-phenyl-1,2-ethanediol. Four stereoisomers were separated by column chromatography. The absolute configuration of one of them (2′′R, 4′′S,1R) was established by X-ray diffraction. Acid hydrolysis of the dioxolanes afforded quantitatively (R)- and (S)-enantiomers of 1. Optical rotatory dispersion (ORD) spectra of both enantiomers are also reported.     

Synthesis and anti-inflammatory-analgesic activity of 2′,4′-difluoro-3-（carbamoyl）biphenyl-4-yl benzoates

Eighteen 2′,4′-difluoro-3-（carbamoyl）biphenyl-4-yl benzoates were synthesized from diflunisal in three steps with total yields from 72% to 86%. All compounds were identified by IR, 1^H NMR, MS and elemental analysis. The anti-inflammatory activity and analgesic activity for 18 compounds were evaluated. The preliminary assay results showed that compounds 4a and 4p exhibited potent anti-inflammatory-analgesic activity.     

X-ray investigation of the nematic and reentrant nematic phases of N-[(4-n-octyloxybenzoyloxy)-salicilidene]-4′-cyanoaniline

We have performed an X-ray scattering study of the nematic-smectic A and reentrant nematic-smectic A phase transitions in N-[(4-n-octyloxybenzoyloxy)-salicilidene]-4′-cyanoaniline (OOBOSCA). A diffractometer with a linear position sensitive detector was used. The results show that the smectic phase in OOBOSCA is of the A_d type with an interlayer spacing incommensurate with the molecular length L; d ∽ 1·2L. In the reentrant nematic phase two types of fluctuation modes were found. One of them corresponds to the monolayer wavevector q ₁ ∽ 2π/L, and the other is due to the partial bilayer fluctuations with the wavevector q ₂ ∽ 0·8q ₁. The temperature dependences of the interlayer spacing, X-ray scattering intensity and longitudinal correlation length for both types of layering in the reentrant nematic phase are presented. The change of the fluctuation regime from S_Ad, to S_Cd type with decreasing temperature in the reentrant nematic phase of OOBOSCA was found. The results are discussed on the basis of models with competing order parameters. The influence of alkyl chain flexibility on the stability of a partial bilayer smectic phase is also considered.     

Synthesis of 2-fluoro-substituted and 2,6-modified purine 2′,3′-dideoxy-2′,3′-difluoro-d-arabinofuranosyl nucleosides from d-xylose

A series of novel purine-modified 2′,3′-dideoxy-2′,3′-difluro-D-arabinonucleosides, including fluorinated analogs of fludarabine and nelarabine, have been prepared via anion glycosylation reactions of salts of 2-fluoropurine derivatives with the glycosyl bromide. A short and efficient synthetic route to the carbohydrate precursor 5-O-benzoyl-2,3-difluoro-α-d-arabinofuranosyl bromide was developed in five steps from d-xylose. Improved synthesis of methyl 5-O-benzoyl-2,3-difluoro-α-d-arabinofuranoside based upon the study of diethylaminosulfur trifluoride (DAST)-reactions with 5-O-protected methyl D-xylosides was explored using mild reaction conditions on the key step. New peculiarities for selective fluorinations of 5-O-benzoylated α- and β-D-pentofuranosides with DAST leading to the formation of mono and difluoro-furanoside derivatives are reported.     

The effect of solvents and organic acids on the p-doping behaviors of poly(3′,4′-Ethylenedioxy-2,2′:5′,2″-terthiophene)

Poly(3??,4??-Ethylenedioxy-2,2??:5??,2??-terthiophene) was synthesized by chemical oxidative polymerization of 3??,4??-Ethylenedioxy-2,2??:5??,2??-terthiophene using FeCl₃ as an oxidant. The resulting polymer was characterized by FTIR and ¹H NMR. p-doping behaviors of substituted polyterthiophene with different organic sulfonic acids (methanesulfonic acid, p-toluene sulfonic acid, and ??-naphthalene sulfonic acid) in different solvents (CH₃CN, CHCl₃, THF and DMF) were studied by UV-VIS spectroscopy. The results from structural characterizations showed that the ??,????-bonding predominates in polyterthiophene. The spectroscopic changes in UV-VIS showed that ??-naphthalene sulfonic acid was an efficient doping acid for polymer while varying the solvents. Meanwhile, CH₃CN showed a high efficiency for p-doping of polyterthiophene while varying the doping acids.

Zusammenfassung Die Synthese von geschützten 2,3-Dideoxy-2-hydroxymethyl-nucleosiden wird beschrieben. Die durch ein Mehrstufenverfahren aus Isopropylidenglycerol erhaltenen Nucleoside können als Bausteine zur Darstellung von Oligonucleotiden verwendet werden, deren 2- und 5-Positionen über eine Etherbrücke verbunden sind.

Synthesis of 2,3-dideoxy-2-hydroxymethyl nucleosides

Summary The synthesis of protected 2,3-dideoxy-2-hydroxymethyl nucleosides is presented. The nucleosides, obtained in a multi-step procedure starting from isopropylideneglycerol, may be used as building blocks for the synthesis of 2,5-ether linked oligonucleotides.

     

Mechanism of Formation of Ribonucleoside Cyclic 2′,3′-Phosphates in the Reaction of Appropriate 3′,5′-Phos-Phorothioates with Epoxides

Abstract

It has been recently demonstrated in our laboratory CD that ribonucleoside cyclic 3′,5′-phosphorothioates react with epoxides to give as a major product corresponding cyclic 5′,3′-phosphates, We now report the results OF our ste-reochemical studies OF this reaction. WE have Found that the diastereomerically pure (Sp) 3′.5′-CUFLPS when treated with styrene [¹⁰O]-oxide in ethanol gives the same isotopomer OF 2′,3′-[¹⁰O]cUMP as that obtained From the reaction of endo-2′.3′-cUMPS with styrene [¹⁰O]-oxide. This result strongly supports our previous assumption that reaction of 3′,5′-cNMPS with epoxides proceeds via 3′-oxathiaphosphorylated intermediate (L) which is Formed with inversion and decomposes to 2′,3′-cNMP with retention of configuration at phosphorus atom.

Dimorphism in a 2,2′,2″-terpyridyl compound

Chloro-(2,2,2-terpyridine)platinum(II) chloride dihydrate, [Pt(terpy)Cl]Cl · 2H₂O, crystallizes from aqueous EtOH (1:1) as a yellow dimorph, but from dilute (1 M) HCl as its less stable red dimorph. X-ray powder photography shows that, above 100 °C, the red form converts into the yellow form.     

Synthesis of 2′,3′-didehydro-3′-deoxythymidine and its activity against HIV

2,3-Didehydro-3-deoxythymidine has been obtained with an overall yield of 10% by the condensation of 1,2-di-O-acetyl-3,5-di-O-benzoyl-D-xylofuranose with bis-(dimethylsilyl)thymine and a series of subsequent transformations. The anti-HIV activity of the compound obtained was studied on the model of MT-4 lymphoid cells primarily infected with HIV-1.
