Some oxyhalides can be found in drinking waters as inorganic disinfection byproducts. An on-line coupled capillary isotachophoresis—capillary zone electrophoresis (CITP-CZE) method was developed for the analysis of chlorate, chlorite and bromate in water. The optimized CITP-CZE electrolyte system consisted of the following: 10 mM—HCl+20 mM—β-Alanine (leading electrolyte), 5 mM—succinic acid (terminating electrolyte), and 10 mM—succinic acid +5 mM—β-Alanine +0.1% HPMC (carrier electrolyte). A clear separation of oxyhalides from other components of drinking water was achieved within 25 min. Method characteristics, i.e., linearity (0–200 ng/mL), accuracy (88–110%), intra-assay (3–5%), quantification limit (5–15 ng/mL), and detection limit (2–5 ng/mL), were determined. Minimum labor requirements, sufficient sensitivity and low running cost are important attributes of this method. It was found that the developed method is useful for the routine analysis of oxyhalides in water.  相似文献

The polymerization of o-phenylenediamine (OPD) on l-tyrosine (Tyr) functionalized glassy carbon electrode (GCE) and its electro-catalytic oxidation towards ascorbic acid (AA) had been studied in this report. l-Tyrosine was first covalently grafted on GCE surface via electrochemical oxidation, which was followed by the electrochemical polymerization of OPD on the l-tyrosine functionalized GCE. Then, the poly(o-phenylenediamine)/l-tyrosine composite film modified GCE (POPD-Tyr/GCE) was obtained. X-ray photo-electron spectroscopy (XPS), field emission scanning electron microscope (SEM), and electrochemical techniques have been used to characterize the grafting of l-tyrosine and the polymerization and morphology of OPD film on GCE surface. Due to the doping of the carboxylic functionalities in l-tyrosine molecules, the POPD film showed good redox activity in neutral medium, and thus, the POPD-Tyr/GCE exhibited excellent electrocatalytic response to AA in 0.1 mol l−1 phosphate buffer solution (PBS, pH 6.8). The anode peak potential of AA shifted from 0.58 V at GCE to 0.35 V at POPD-Tyr/GCE with a greatly enhanced current response. A linear calibration graph was obtained over the AA concentration range of 2.5 × 10−4–1.5 × 10–3 mol l−1 with a correlation coefficient of 0.9998. The detection limit (3δ) for AA was 9.2 × 10−5 mol l−1. The modified electrode showed good stability and reproducibility and had been used for the determination of AA content in vitamin C tablet with satisfactory results.  相似文献

The combination of capillary isotachophoresis (ITP) and capillary zone electrophoresis (CZE) in the column-coupling configuration has been optimized in a mode in which the background electrolyte employed in the CZE step was different from the leading and terminating electrolytes of the ITP step. The optimum composition of the electrolyte system was 0.01 M HCl, 0.02 M IMI, 0.2% HEC, pH 7.2 (leading electrolyte), 0.01 M HEPES, pH 8.2 (terminating electrolyte), and 25 mM MES, 50 mM TRIS, 30 mM boric acid, 0.2% HEC, pH 8.3 (background electrolyte). All solutions contained 20% methanol. The timing of the transfer of isotachophoretically stacked analyte zones into the CZE column was also optimized. An ITP–CZE method with UV detection at 270 nm was developed for separation of nine phenolic acids (protocatechuic, syringic, vanillic, cinnamic, ferulic, caffeic, ρ-coumaric, chlorogenic, and gentisic acids) in a model mixture and used for assay of some of these acids in a methanolic extract of herba epilobi. Application of ITP–CZE resulted in 100-fold better sensitivity than conventional CZE; limits of detection ranged between 10 and 60 ng mL−1. When MES–TRIS–borate-based buffer, pH 8.3, was used in the CZE separation step the linearity of the ITP–CZE response was satisfactory (correlation coefficients were from 0.9937 to 0.9777). Summary A capillary zone electrophoresis (CZE) method was developed for the simultaneous assay of three bioactive components—baicalin, chlorogenic acid and caffeic acid—in seven traditional Chinese medicinal preparations. The analytes were separated successfully within 3.5 min using 10 mM borate buffer (pH8.6). Regression equations revealed linear relationships (correlation coefficients 0.9942–0.9996) between the peak area and concentration of the three analytes. The relative standard deviations of the migration times and the peak areas of the three constituents were 1.12–2.68% and 1.62–5.73%, respectively. Recovery of the three constituents ranged from 89 to 107%. The extraction efficiencies of different extraction solutions are also discussed. The contents of the three components in seven different Chinese medicinal preparations containing Honeysuckle flower and/orScutellariae radix were determined by the CZE method with satisfactory results.  相似文献

A supramolecular catalytic kinetic spectrofluorimetric method was developed for the determination of platinum(IV) and the possible mechanism of catalytic reaction was discussed. The method was based on the fluorescence-enhancing reaction of salicylaldehyde furfuralhydrazone (SAFH) with potassium bromate, which was catalysed by platinum(IV) in a water–ethanol medium. β–Cyclodextrin (β-CD) obviously sensitized the determination at pH 5.20 and 25°C. Under optimum conditions, the β-CD–platinum–KBrO3–SAFH supramolecular kinetic catalytic reaction system had excitation and emission maxima at 372 and 461 nm, respectively. The linear range of this method was 0.60–180 ng ml−1 with a relative standard deviation of 1.2%, and the detection limit was 0.18 ng ml−1. Investigation of the mechanism and the effects of interferences is presented. The proposed method was applied successfully to determine trace platinum(IV) in the chemotherapeutic drug cisplatin and serum from patients with satisfactory results.   相似文献

A reverse phase HPLC method was developed and validated for the simultaneous determination of 2-hydroxy-4-methoxybenzaldehyde and 2-hydroxy-4-methoxybenzoic acid in the root extracts of Hemidesmus indicus. A comprehensive validation of the method including sensitivity, linearity, reproducibility, accuracy, limit of detection (LOD) and limit of quantification (LOQ) was conducted using the optimized chromatographic conditions. The method was found to be linear (r > 0.998) in the range of 5–350 μg mL−1 for 2-hydroxy-4-methoxybenzaldehyde and for 2-hydroxy-4-methoxybenzoic acid (r > 0.999) in the range 10–300 μg mL−1. The method was found to be precise with inter-day precision values (% RSD) in the ranges of 0.61–1.76% for 2-hydroxy-4-methoxybenzaldehyde and 1.3–2.8% for 2-hydroxy-4-ethoxybenzoic acid while intra-day precisions (% RSD) of two analytes were in the range of 0.41–1.07 and 0.95–2.5%. The limits of detection (LODs) for 2-hydroxy-4-methoxybenzaldehyde and 2-hydroxy-4-methoxybenzoic acid were 0.84 and 2.34 μg mL−1. The described method was fast, sensitive and reproducible, and thus well suited for routine analysis of these two compounds from root extracts of H. indicus and other plants.  相似文献

A new method is described for the determination of endogenous jasmonic acid (JA) in Lemna minor plant extracts using liquid chromatography (LC) with fluorescence detection. Plant tissues were extracted and derivatised using 9-anthryldiazomethane (ADAM reagent) prepared in situ. Accuracy and precision were improved by using the internal standard dihydrojasmonic acid (dh-JA) for the correction of JA losses during sample preparation steps. Liquid chromatography–mass spectrometry (LC/MS) analysis of ADAM derivatives of JA and dh-JA confirmed that a single molecule of JA and dh-JA was coupled with one molecule of reagent. Derivatives of JA and dh-JA were separated with gradient elution on a C18 reversed-phase column using acetonitrile/water as a mobile phase and detected by a fluorescence detector at excitation and emission wavelengths of 254 and 412 nm, respectively. The detection limits of JA and dh-JA were 2.9 ng mL−1 and 3.7 ng mL−1 per 50-μL injection. The method is reproducible and selective and yields single peaks for each compound regardless of isomer. The specificity and accuracy of the proposed LC/FD method was confirmed by liquid chromatography–TurboIon Spray tandem mass spectrometric (LC/MS/MS) analysis of free JA in Lemna minor samples under multiple reaction monitoring conditions.  相似文献

Simultaneous determination of arbutin (ART) and l-ascorbic acid (AA) by HPLC with chemiluminescence detection is proposed for the first time. This method is based on the CL reaction of acidic potassium permanganate with ART and AA in the presence of formaldehyde as enhancer. The separation was performed on a C18 column with a 90:10 (v/v) mixture of 0.02 M phosphate buffer and methanol as mobile phase. The effects of several conditions on HPLC resolution and CL emission were studied systematically. The linear ranges were 0.5–50 and 1–200 μg mL−1 for ART and AA, respectively. The detection limits were 0.2 and 0.3 μg mL−1, respectively. The method was successfully applied to the determination of ART and AA in whitening cosmetics.  相似文献

Two in-line preconcentration capillary zone electrophoresis (CZE) methods (field amplified sample injection (FASI) and stacking with sample matrix removal (LVSS)) have been evaluated for the analysis of acrylamide (AA) in foodstuffs. To allow the determination of AA by CZE, it was derivatized using 2-mercaptobenzoic acid. For FASI, the optimum conditions were water at pH > or = 10 adjusted with NH3 as sample solvent, 35 s hydrodynamic injection (0.5 psi) of a water plug, 35 s of electrokinetic injection (-10 kV) of the sample, and 6s hydrodynamic injection (0.5 psi) of another water plug to prevent AA removal by EOF. In stacking with sample matrix removal, the reversal time was found to be around 3.3 min. A 40 mM phosphate buffer (pH 8.5) was used as carrier electrolyte for CZE separation in both cases. For both FASI and LVSS methods, linear calibration curves over the range studied (10-1000 microg L(-1) and 25-1000 microg L(-1), respectively), limit of detection (LOD) on standards (1 microg L(-1) for FASI and 7 microg L(-1) for LVSS), limit of detection on samples (3 ng g(-1) for FASI and 20 ng g(-1) for LVSS) and both run-to-run (up to 14% for concentration and 0.8% for time values) and day-to-day precisions (up to 16% and 5% for concentration and time values, respectively) were established. Due to the lower detection limits obtained with the FASI-CZE this method was applied to the analysis of AA in different foodstuffs such as biscuits, cereals, crisp bread, snacks and coffee, and the results were compared with those obtained by LC-MS/MS.  相似文献

A new and efficient Hg(II) back-elution method for the desorption of Cd, Cu, and Pb from Chelex-100 chelating resin was developed. A smaller eluent volume and shorter elution time can be achieved using an Hg(II) containing eluent rather than pure nitric acid. Owing to the remaining Hg(II) ion in the effluent, a mercury thin-film electrode is formed in-situ during the anodic stripping voltammetric determination without any further addition of Hg(II). The results indicate that all the analytes in seawater matrix can be completely adsorbed on Chelex-100 resin from the sample at pH 6.5, and subsequently eluted from the resin with an acid solution of 5 × 10–4 mol/L Hg2+ + 1 mol/L HClO4. The detection limits obtained from the differential-pulse anodic (μg L–1 to ng L–1) stripping voltammetry are at sub-ppb to ppt (μg L–1 to ng L–1) levels permitting to determine Cd, Cu and Pb traces in seawater. The analytical reliability was confirmed by the analysis of the certified reference material CASS-II (open ocean seawater). Metabolism of four tobacco-specific N-nitrosamines (TSNAs), N′-nitrosonornicotine (NNN), N′-nitrosoanatabine (NAT), N′-nitrosoanabasine (NAB), and 4-(methylnitrosamino)-1-(3-pyridyl)-1-butanone (NNK) has been studied by solid-phase extraction (SPE) and liquid chromatography–tandem mass spectrometry (LC–MS–MS). 4-(Methylnitrosamino)-4-(3-pyridyl)-1-butanol (iso-NNAL) was used as internal standard. SPE and LC–MS–MS was found to be a rapid, simple, sensitive, and selective method for analysis of TSNAs in rabbit serum. The relative standard deviation (R.S.D., n = 6) for analysis of 5 ng mL−1 and 0.5 ng mL−1 standards and of serum sample spiked with 5 ng mL−1 standards of five TSNAs was 2.1–11% and recovery of 5 ng mL−1 standards from serum was 100.2–112.9%. A good linear relationship was obtained between peak area ratio and concentration in the range of 0.2–100 ng mL−1 for NNAL and 0.5–100 ng mL−1 for other four TSNAs, with correlation coefficients (R 2) >0.99 (both linear and log–log regression). Detection limits for standards in solvent were between 0.04 and 0.10 ng mL−1. Doses of TSNAs administered to rabbits via the auricular vein were 4.67 μg kg−1 and 11.67 μg kg−1, in accordance with the different levels in cigarettes. Metabolic curves were obtained for the four TSNAs and for 4-(methylnitrosamino)-1-(3-pyridyl)-1-butanol (NNAL), a metabolite of NNK; on the basis of these curves we modeled metabolic kinetic equations for these TSNAs by nonlinear curve fitting.  相似文献

A liquid chromatography–mass spectrometry (LC-MS) method was developed and validated for the simultaneous determination of alisol A and alisol A 24-acetate from Alisma orientale (Sam.) Juz. in rat plasma using diazepam as an internal standard. A 200-μl plasma sample was extracted by methyl tert-butyl ether and the separation was performed on Kromasil C18 column (150 × 4.6 mm, 5 μm) with the mobile phase of acetonitrile (containing 0.1% of formic acid)–water (73:27, v/v) at a flow rate of 0.8 ml/min in a run time of 10 min. The two analytes were monitored with positive electrospray ionization by selected ion monitoring mode. The lower limit of quantitation for both alisol A and alisol A 24-acetate were 10 ng/ml. The calibration curves were linear in the measured range 10–1,000 ng/ml for alisol A and 10–500 ng/ml for alisol A 24-acetate. The mean extraction recoveries were above 74.7% for alisol A and above 72.4% for alisol A 24-acetate from biological matrixes. The intra- and inter-day precision for all concentrations of quality controls was lower than 14.1% (RSD %) for each analyte. The accuracy ranged from −12.3% to 9.8% (RE %) for alisol A, and −8.6% to 14.2% (RE %) for alisol A 24-acetate. The method was successfully applied to the study on the pharmacokinetics of alisol A and alisol A 24-acetate in rat plasma.  相似文献

A new flow injection chemiluminescent immunoassay was developed for the detection of 17β-estradiol (E2). The method uses p–iodophenol (PIP) as enhancer and is based on a solid-phase immunoassay format in which an E2–OVA immobilized immunoaffinity column inserted in the flow system is used to trap unbound horseradish peroxidase (HRP)-labeled anti-E2 antibody after an off-line incubation of E2 with HRP-labeled anti-E2 antibody. The trapped enzyme conjugate was detected by injecting substrates to produce an enhanced chemiluminescence (CL) response. The linear range for E2 was 10.0–1,000.0 ng mL−1 with a correlation coefficient of 0.996 and a detection limit of 3.0 ng mL−1. The sampling and chemiluminescence detection time for one sample was 400 s after a pre-incubation procedure of 30 min. Serum samples detected by this method were in good agreement with the results obtained by EIA with E2–biotin.   相似文献

A single optosensing device based on lanthanide-sensitized luminescence was developed for determination of p-aminobenzoic acid (PABA). The method is based on the formation of a complex between PABA and Tb(III) immobilized on the solid phase (QAE A-25 resin) placed inside the flow cell. NaCl (1 M) was used as carrier solution and HCl (0.05 M) as eluent. The sample solutions of PABA (100 μL) containing Tb(III) and buffered at pH = 6.0 were injected into the carrier stream and the luminescence was measured at λ ex = 290 nm and λ em = 546 nm. The method shows a linear range from 0.2 to 6.0 μg mL−1 with an RSD of 1.2% (n = 10) and a sampling frequency of 22 h−1. A remarkable characteristic of the method is its high selectivity which allows it to be satisfactorily applied to the analysis of PABA in pharmaceutical samples without prior treatment. In this study, the development and validation of an analytical method for triptolide in whole blood using high-performance liquid chromatography coupled with atmospheric-pressure chemical ionization ion trap tandem mass spectrometry (LC–APCI-IT-MS-MS) is reported. This is the first report of the systematic development and validation of an LC–MS-MS method for the quantitation of triptolide in human whole blood using prednisolone as an internal standard (IS). Prior to LC–MS-MS analysis, liquid–liquid extraction with ethyl acetate was used to isolate them from the biological matrix. Validation parameters such as specificity/selectivity, limit of quantitation (LOQ), linearity, precision, accuracy and stability are evaluated for this method. The calibration curve was linear (r 2 = 0.9973) in the concentration range of 0.5–100.0 ng mL−1 in human whole blood with a lower limit of quantitation of 0.5 ng mL−1. Intra- and inter-day relative standard deviations (RSDs) were less than 8.6 and 11.7%, respectively. Extraction recoveries of triptolide ranged from 81.5 to 88.1%. In this study, the determination of 4-Bromoaniline (4-BA) in green algae Chlamydomonas reinhardtii (C. reinhardtii) was investigated by applying continuous-flow microextraction (CFME) combined with high-performance liquid chromatography (HPLC). Continuous-flow microextraction was conducted in a homemade glass chamber, i.e. the sample solution flowed through a constant volume drop of solvent in the chamber at a constant flow rate. The effects of different factors on extraction efficiencies were also investigated and these factors included the kind of extraction solvent, solvent drop volume, sample flow rate, extraction time and addition amount of salt. Under the optimum extraction conditions (extraction solvent, carbon tetrachloride; solvent drop volume, 3.5 μL; sample flow rate, 1.0 mL min−1; extraction time, 10 min; no addition of salt), the calibration plot was set up by plotting peak area against a series of 4-Bromoaniline concentrations (0.01–10 μg mL−1) in aqueous solution. The correlation coefficient (r) was 0.9990. The limit of detection (LOD) was 0.6 ng mL−1. The precision of this method was obtained by successive five time analyses of 100-ng mL−1 standard solution of 4-Bromoaniline, and the relative standard deviation (RSD) was 3.5%. The concentration factor was calculated by the ratio of peak area of the analyte obtained after and before extraction and found to be 10.6. 4-Bromoaniline residues in Chlamydomonas. reinhardtii cells and tap water samples were satisfactorily analyzed according to the method described above.  相似文献

The component composition of essential oils produced by steam distillation from flower heads, leaves, and stems of Salvia anatolica (Lamiaceae), a recently described new species endemic from Turkey, was studied by GC/FID and GC/MS. A total of 127 volatile components representing 96% of the oil was identified in essential oil from flower heads and leaves. It was found that the principal oil components of flower heads and leaves were α-pinene (10.9%), β-pinene (6.7%), α-copaene (6.3%), heptacosane (6.2%), and hexadecanoic acid (5.0%). A total of 109 volatile compounds representing 87.9% of the oil was characterized in essential oil isolated from stems. Radix Scrophulariae (Xuanshen) is one of the famous Chinese herbal medicines widely used to treat rheumatism, tussis, pharyngalgia, arthritis, constipation, and conjunctival congestion. Harpagoside and cinnamic acid are the main bioactive components of Xuanshen. The purpose of this study was to develop an HPLC–UV method for simultaneous determination of harpagoside and cinnamic acid in rat plasma and investigate pharmacokinetic parameters of harpagoside and cinnamic acid after oral administration of Xuanshen extract (760 mg kg−1). After addition of syringin as internal standard, the analytes were isolated from plasma by liquid–liquid extraction. Separation was achieved on a Kromasil C18 column, and detection was by UV absorption at 272 nm. The described assay was validated in terms of linearity, accuracy, precision, recovery, and limit of quantification according to the FDA validation guidelines. Calibration curves for both analytes were linear with the coefficient of variation (r) for both was greater than 0.999. Accuracy for harpagoside and cinnamic acid ranged from 100.7–103.5% and 96.9–102.9%, respectively, and precision for both analytes were less than 8.5%. The main pharmacokinetic parameters found for harpagoside and cinnamic acid after oral infusion of Xuanshen extract were as follows: C max 1488.7 ± 205.9 and 556.8 ± 94.2 ng mL−1, T max 2.09 ± 0.31 and (1.48 ± 0.14 h, AUC0–24 10336.4 ± 1426.8 and 3653.1 ± 456.4 ng h mL−1, 11276.8 ± 1321.4 and 3704.5 ± 398.8 ng h mL−1, and t 1/2 4.9 ± 1.3 and 2.5 ± 0.9 h, respectively. A very sensitive and selective catalytic adsorptive cathodic stripping procedure for trace measurements of cobalt is presented. The method is based on adsorptive accumulation of cobalt-CCA (calcon carboxylic acid) complex onto a hanging mercury drop electrode followed by reduction of the adsorbed species by voltammetric scan using differential pulse modulation. The reduction current is enhanced catalytically by nitrite. The effect of various parameters such as pH, concentration of CCA, concentration of nitrite, accumulation potential and accumulation time on the selectivity and sensitivity were studied. The optimum condition for the analysis of cobalt, include pH 5.2 (Acetate buffer), 2.1 μM clacon carboxylic acid, 0.032 M sodium nitrite and an accumulation potential of 0.05 V (versus Ag/AgCl). Under these optimum conditions and for an accumulation time of 60 s, the measured peak current at −0.480 V is proportional to the concentration of cobalt over the entire concentration range tested 0.003–2.0 ng ml−1 with a detection limit of 1 pg ml−1 for an accumulation time of 60 s and 2.0–10.0 ng ml−1 for an accumulation time of 40 s. The relative standard deviations for ten replicate measurement of 0.5 ng ml−1 of cobalt were 3.1%. The main advantage of this new system is the microtrace Co(II) determination by ASV. The method was applied to determination of cobalt in a water sample and some analytical grade salts with satisfactory results. A simple, rapid and specific chemiluminescence (CL) method for the determination of glutathione has been developed. The method is based on the enhanced CL of the reaction between Ru(phen)3 2+ (phen = 1,10-phenanthroline) and KMnO4 by glutathione in HCl medium. Under the optimum conditions, the response is linearly proportional to the concentration of glutathione between 1.5 × 10−7 and 1.0 × 10−5 mol L−1. The dection limit for glutathione (5.8 × 10−8 mol L−1) is about 10 and 200 times better than those of the spectrophotometric method using Ellman regent and the Lucigenin – CL method, respectively. The final procedure allows the determination of glutathione in human serum with recoveries of 92%–108%. A satisfactory agreement was obtained with a mean relative difference of 2.5% compared to the HPLC method.  相似文献
